Characterization of B- , T- , and NK-like Cells in Nile Tilapia (Oreochromis nilotica)

  • It has been very difficult to develop and evaluate efficient fish vaccines because fish immune cells have not been properly characterized. In this study, we investigated the cell-mediated immunological properties of B- and T-like cells in Nile tilapia (Oreochromis nilotica). Surface immunoglobulin negative ($slg^{-}$) cell population proliferated in response to mammalian T-cell mitogens PHA and Con A, while surface immunoglobulin positive ($slg^{+}$) cells responded to the B-cell mitogen LPS. The slg$^{[-10]}$ cells from hemocyanin (HC)-immunized Tilapia, compared to the non-immunized control, reacted more to PHA than to Con A. Unexpectedly, antigen (Ag)-specific response was observed in both $slg^{-}$ and $slg^{-}$cells. Regardless of HC immunization, whole leukocytes from 8 head kidney of fish showed natural killer (NK)cell activity. Especially, NK cell activity was much higher in slg$^{[-10]}$ cells than in slg$^{+}$cells, indicating the possibility that fish NK cells were not at least associated with slg$^{+}$ cell population and not activated by Ag. Further understanding of functional fish immune cells will help to evaluate and develop effective vaccines for fishes and to monitor the course of therapy In infected fishes.hes.

Aureobasidium pullulans으로 부터 분리한 β-1,3/1,6-glucan의 면역활성의 연구 (An in vitro study of immune activity by β-1,3/1,6-glucan isolated from Aureobasidium pullulans)

  • ${\beta}$-Glucan은 효모나 버섯의 세포벽이나 곡물의 섬유에 들어있는 천연 화합물이며, 면역 시스템을 활성화 한다고 알려져 있다. Aureobasidium pullulans 수용성의 ${\beta}$-1,3/1,6-glucan을 생성한다. 본 연구에서는 A. pullulans 로부터 생성된 ${\beta}$-1,3/1,6-glucan의 면역활성에 관한 효과를 조사하기 위하여, 면역활성에 관여하는 NK 세로의 활성과 대식세포에 미치는 영향을 관찰하였다. NK 세포에 의한 Yac-1 세포의 사멸율은 ${\beta}$-1,3/1,6-glucan을 처리한 군은 처리하지 않은 대조군에 비하여 63-39% 증가하는 것으로 나타났고, 대식세포의 탐식작용은 zymosan을 처리한 군보다 15-21% 증가함을 보였다. LP-BM5 바이러스 복제 억제능을 확인하기 위하여 SC-1/LP-BM5 세포주에 ${\beta}$-1,3/1,6-glucan을 처리한 결과 $200{\mu}g/mL$ 처리에서 LP-BM5 바이러스 복제능이 최대 74% 유의적으로 감소하였다. 이 결과는 ${\beta}$-1,3/1,6-glucan이 면역활성에 관여하는 NK 세포와 대식세포를 활성화하여 면역체계가 민감하게 반응할 수 있도록 만들고, LP-BM5 바이러스 복제를 억제하여 면역력을 증진할 수 있다는 것을 보여준다.

Effects of Kimchi Extracts on Interleukin-2 Production and Natural Killer Cell Activity in Mice

  • To determine the immune effect of kimchi extracts in mice, 0.5mg/day of the extracts from kimchis, which were prepared with conventionally (general kimchi)and organically(organic kimchi) cultivated ingredients, were treated orally to male BALB/c mice. Following 1, 3 and 5 weeks of treatment , the Interleukin-2(IL-2) production in the presence (con-A-stimulated )or the absence(spontaneous)of con A 95 $\mu\textrm{g}$/ml) and the natural killer cell (NK) activity of the splenocytes were measured. The IL-2 production in most of treatments with methanol extract from general kimchi were significantly higher than those of control(p<0.05).And at the 3 weeks of treatment, the spontaneous or con A-stimulated IL-2 productions from splenocytes of mice treated with it increased more than those of control group, by 2.8 and 2.2 times, respectively. However, the longer the treatment with methanol extracts from organic kimchi showed the higher the enhancing effect on the IL-2 production. The spontaneous or con A-stimulatdIL-2 productions form splenocytes of mice treated with dicholoromethyane fraction from general kimchi also increased at 5 weeks of treatment compared to those of control group, by 2.7 and 2.5 times, respectively. The natural killer cell activity of splenocytes from mice treated with methano lextracts from general kimchi for 1 ~5 weeks significantly higher than that of control goup (p<0.01). The effect of methano extracts from general kimchi was the highest at 3 weeks of treatment, as same as in the IL-2 production. The enhancing effect of methano extracts from organic kimchi on the NK cell activity was the highest at 5 weeks of treatment . The NK cell activity of splenocytes from mice treated with dichloromethane fraction from general kimchi for 5 weeks was significantly higher than those in control and 3 weeks of treatment. These results showed that the effects of kimchi extracts on the IL-2 production and the NK cell activity in mice were profound in long term of treatment (3 and 5 weeks than 1 week) . We suggest that kimchi extracts might have an immune effect in part due to its enhancing action on the IL-2 production and the NK cell activity.

  • Natural killer (NK) cells, key antitumor effectors of the innate immune system, are endowed with the unique ability to spontaneously eliminate cells undergoing a neoplastic transformation. Given their broad reactivity against diverse types of cancer and close association with cancer prognosis, NK cells have gained considerable attention as a promising therapeutic target for cancer immunotherapy. NK cell-based therapies have demonstrated favorable clinical efficacies in several hematological malignancies but limited success in solid tumors, thus highlighting the need to develop new therapeutic strategies to restore and optimize anti-tumor activity while preventing tumor immune escape. The current therapeutic modalities yielding encouraging results in clinical trials include the blockade of immune checkpoint receptors to overcome the immune-evasion mechanism used by tumors and the incorporation of tumor-directed chimeric antigen receptors to enhance NK cell anti-tumor specificity and activity. These observations, together with recent advances in the understanding of NK cell activation within the tumor microenvironment, will facilitate the optimal design of NK cell-based therapy against a broad range of cancers and, more desirably, refractory cancers.

Interleukin-18 Synergism with Interleukin-2 in Cytotoxicity and NKG2D Expression of Human Natural Killer Cells

  • Natural killer (NK) cells play an important role in anti-tumor immunity. Interleukin (IL)-18 is an immunoregulatory cytokine that induces potent NK cell-dependent anti-tumor responses when administrated with other cytokines. In this study, we explored the effects of combining IL-18 and IL-2 on NK cytotoxicity as well as expression levels of the NK cell receptor NKG2D in vitro. Freshly isolated PBMCs were incubated for 48 h with IL-18 and IL-2, then CD107a expression on $CD3^-CD56^+$ NK cells was determined by three-colour flow cytometry to evaluate the cytotoxicity of NK cells against human erythroleukemia K562 cells and human colon carcinoma HT29 cells. Flow cytometric analysis was also employed to determine NKG2D expression on NK cells. The combined use of IL-18 and IL-2 significantly increased CD107a expression on NK cells compared with using IL-18 or IL-2 alone, suggesting that the combination of these two cytokines exerted synergistic enhancement of NK cytotoxicity. IL-18 also enhanced NKG2D expression on NK cells when administered with IL-2. In addition, blockade of NKG2D signaling with NKG2D-blocking antibody attenuated the up-regulatory effect of combining IL-18 and IL-2 on NK cytolysis. Our data revealed that IL-18 synergized with IL-2 to dramatically enhance the cytolytic activity of human NK cells in a NKG2D-dependent manner. The results appear encouraging for the use of combined IL-18 and IL-2 in tumor immunotherapy.

시호 약침제제가 생쥐의 면역활성에 미치는 영향 (Effect of Bupleuri Radix Aqua-Acupuncture Solution on Immune Response in Mice)

  • In order to investigate the effects of Bupleuri Radix aqua-acupuncture solution (BRAS) on immuno suppression induced by glucocorticoid, ICR mice were administrated with glucocorticoid (80 mg/kg) for 7 days, and immunized with hapten, methamphetamine-horseradish peroxidase $(10\;{\mu}g/mouse)$. And then, BRAS (0.2ml/mouse) injected into $CV_4\;and\;BL_{23}$, which are the classical acupuncture points in traditional medicine, for 7 days. And then B and T cells proliferation and cytolytic activity of natural killer (NK) cells were measured. Intraperitoneal injection of glucocorticoid decreased lysozyme activity in macrophage and cytolytic activity of NK cell. B and T cell proliferation were significantly increased in aqua-acupuncture group compared to normal group. On the other hand, BRAS significantly increased the lysozyme activity in macrophage, and the cytolytic activity of purified NK cell on K562. These results suggest that BRAS at $CV_4\;and\;BL_{23}$ may proliferate B and T cells that are suppressed by glucocorticoid and activate NK cell activity.

Effects of Cordyceps Militaris Extract on Tumor Immunity

  • Background and Aims : Even though various strategies for cancer treatment have advanced with the remarkable development of genomic information and technology, it is far from giving relief to cancer patients. Recently there is accumulating evidence that the immune system is closely connected to anti-tumor defense mechanisms in a multistage process. This includes tumorigenesis, invasion, growth and metastasis. Cordyceps Militaris, a well-known oriental herbal medicine, is a parasitic fungus that has been used as an immune enhancing agent for a long period of time. However, little is known about the cancer-related immunomodulatory effects and anti-tumor activities. In the present study, we aimed to investigate the effects of Cordyceps Militaris extract (CME) on immune modulating and anti-tumor activity. Materials and Methods : To elucidate the effects of CME on macrophage and natural killer (NK) cell activity, we analyzed nitric oxide (NO) production, NK cytotoxicity and gene expression of cytokines related with macrophages and NK cell activity. Results and Conclusions : CME activated and promoted macrophage production of NO. It also enhanced gene expression of IL-1 and iNOS in RAW 264.7 cells. CME promoted cytotoxicity of NK cells against YAC-1 cells and enhanced NK cell related gene expression such as IL-1, IL-2, IL-12, iNOS, IFN-${\gamma}$ and TNF-${\alpha}$ in mice splenocytes. It also Promoted protein expression of IL-10, IL-12, IFN-${\gamma}$ and TNF-${\alpha}$ in mice splenocytes and inhibited lung tumor metastasis induced by CT-26 cell line compared with the control group. From these results, it could be concluded that CME is an effective herbal drug for modulating the immune system and anti-cancer treatment by promoting macrophage and NK cell activity.

말초혈액 자연살해세포 분획 및 세포용해 활성도 분석을 통한 습관성 유산 위험군의 진단적 유용성에 관한 연구 (Increased Peripheral NK Cell Fraction and Their Cytolytic activity in Patients with History of Recurrent Spontaneous Abortion)

  • 목 적: 임신 전 $CD3^-/CD56^+/CD16^+$ 말초혈액 자연살해세포 (pbNK cell)의 분획과 세포용해 활성도를 정상군과 습관성 유산의 기왕력을 가진 환자군으로 나누어 비교, 분석하고 습관성 유산의 위험도를 제시할 수 있는 각각의 cut-off value를 설정하고자 하였다. 연구방법: 전향적 연구로서 습관성 유산의 기왕력이 있는 여성을 환자군 (n=35)으로 하였으며, 대조군으로 불임이나 습관성 유산의 기왕력이 없으며 정상아의 출산 경험이 있는 여성을 대조군 (n=15)으로 설정하였다. 유세포분석기를 이용하여 pbNK cell 분획 및 세포용해 활성도를 측정 후 그 결과를 비교 분석하였다. 결 과: pbNK cells의 분획은 습관성 유산 환자군에서 대조군에 비해 통계적으로 유의하게 높은 결과를 보였다($14.2{\pm}5.2$ vs. $9.4{\pm}3.7%$, p=0.002, 95% confidence interval [CI] 1.8~7.8). Receiver operating characteristic curve (ROC) 곡선을 이용하여 pbNK cell의 분획에 대한 cut-off values을 12.1%로 정하였을 때 습관성 유산의 위험도는 8.4배 증가하였다. pbNK cell의 K562 세포용해 활성도를 3가지 다른 Effector to Target (E:T) 비율 (50:1, 25:1, 12.5:1)을 사용하여 측정한 결과 각각의 경우에 있어 습관성 유산 환자군에서 대조군에 비해 통계적으로 유의하게 증가된 결과를 보였다 ($48.3{\pm}19.0$ vs. $31.3{\pm}11.9%$ in 50:1 ratio, p=0.002; $37.0{\pm}18.1$ vs. $20.2{\pm}9.2%$ in 25:1 ratio, p<0.001; $23.5{\pm}12.7$ vs. $12.4{\pm}7.3%$ in 12.5:1 ratio, p=0.001). ROC 곡선을 이용하여 각각 E:T 비율에서 세포용해 활성도의 cut-off values (43.1% in 50:1, 26.9% in 25:1, and 17.4% in 12.5:1)을 설정하여 분석한 결과 습관성 유산의 위험도는 각각 10.0배, 11.4배, 그리고 15.0배 증가된 결과를 보였다. 결 론: 원인이 분명하지 않은 습관성 유산 환자에서 pbNK cell의 분획과 세포용해 활성도를 측정하는 것은 면역학적 원인, 특히 동종면역 요인에 의한 습관성 유산의 유용한 진단 지표로 이용될 수 있을 것으로 사료된다. 향후 동종 면역반응에 의한 습관성 유산 환자에서 면역학적 원인의 치료 전, 후 pbNK cell의 분획과 세포용해 활성도를 측정, 비교하여 그 효과를 증명하는 연구가 필요할 것으로 생각된다.

  • The effects of Buthus martensi Karsch (BMK) on natural killer (NK) cell activity in mouse spleen were studied. Water extracted solution of BMK was orally administrated to Balb/c mice for 2 weeks. Among splenic cells, T cell fractions were separated by Nylon wool column. Furthermore, NK cell purification was performed 4.5% percoll gradients methods. The cytotoxcity of NK cell to K562 cell was determined by lactic acid dehydrogenase and $[^3H]-thymidine $ incorporation methods. And the cytotoxicity of effector cell was most effectively induced in a ration of 50:1 (effector/target cell). As a result, cytotoxicity of NK cells was significantly increased compared with control group both in vivo and in vitro systems. The similar cytotoxic effect was shown in $[^3H]-thymidine $ incorporation methods. This suggests that when BMK is administrated to mice with malignant tumors, an increase in NK cell activity may occur and affect K562 tumor cells.

Effect of pregabalin on nociceptive thresholds and immune responses in a mouse model of incisional pain

  • Background: It is known that some analgesics as well as pain can affect the immune system. The aim of this study was to investigate the analgesic effect and immunomodulation of pregabalin (PGB) in a mouse incisional pain model. Methods: A postoperative pain model was induced by hind paw plantar incision in male BALB/c mice. Mice were randomly divided into four groups (n = 8): a saline-treated incision (incision), PGB-treated incision (PGB-incision), sham controls without incision or drug treatment (control), and a PGB-treated control (PGB-control). In the PGB treated groups, PGB was administered intraperitoneally (IP) 30 minutes before and 1 hour after the plantar incision. Changes of the mechanical nociceptive thresholds following incision were investigated. Mice were euthanized for spleen harvesting 12 hours after the plantar incision, and natural killer (NK) cytotoxicity to YAC 1 cells and lymphocyte proliferation responses to phytohemagglutinin were compared among these four groups. Results: Mechanical nociceptive thresholds were decreased after plantar incision and IP PGB administration recovered these decreased mechanical nociceptive thresholds (P < 0.001). NK activity was increased by foot incision, but NK activity in the PGB-incision group was significantly lower than that in the Incision group (P < 0.001). Incisional pain increased splenic lymphocyte proliferation, but PGB did not alter this response. Conclusions: Incisional pain alters cell immunity of the spleen in BALB/c mice. PGB showed antinocieptive effect on mouse incisional pain and attenuates the activation of NK cells in this painful condition. These results suggest that PGB treatment prevents increases in pain induced NK cell activity.
