Non-Destructive Sorting Techniques for Viable Pepper (Capsicum annuum L.) Seeds Using Fourier Transform Near-Infrared and Raman Spectroscopy

  • Purpose: This study examined the performance of two spectroscopy methods and multivariate classification methods to discriminate viable pepper seeds from their non-viable counterparts. Methods: A classification model for viable seeds was developed using partial least square discrimination analysis (PLS-DA) with Fourier transform near-infrared (FT-NIR) and Raman spectroscopic data in the range of $9080-4150cm^{-1}$ (1400-2400 nm) and $1800-970cm^{-1}$, respectively. The datasets were divided into 70% to calibration and 30% to validation. To reduce noise from the spectra and compare the classification results, preprocessing methods, such as mean, maximum, and range normalization, multivariate scattering correction, standard normal variate, and $1^{st}$ and $2^{nd}$ derivatives with the Savitzky-Golay algorithm were used. Results: The classification accuracies for calibration using FT-NIR and Raman spectroscopy were both 99% with first derivative, whereas the validation accuracies were 90.5% with both multivariate scattering correction and standard normal variate, and 96.4% with the raw data (non-preprocessed data). Conclusions: These results indicate that FT-NIR and Raman spectroscopy are valuable tools for a feasible classification and evaluation of viable pepper seeds by providing useful information based on PLS-DA and the threshold value.

흡착조건이 키토산의 지방질 흡착 특성에 미치는 영향 (Effects of Adsorption Condition on Fat-binding Characteristics of Chitosan)

  • To study the lipid adsorption characteristic of chitosans with different molecular weights and the degrees of deacetylation, in vitro test and near-infrared (NIR) spectroscopic analysis have been performed for the measurement of lipid adsorption characteristics of chitosan. The degrees of deacetylation in chitosans were $70{\%},\;85{\%}\;and\;92{\%}$ at different deacetylation times (1 hr, 2 hrs, 3 hrs), respectively. The molecular weight of each chitosan was controlled by enzymatic hydrolysis, and then the molecular weight of the chitosan was 4 kDa. The bulk density, water holding capacity and fat binding capacity of each chitosan powder were $96.2-504.0{\%},\;374.4-1217.9{\%},\;and\;307.0-659.3{\%}$, respectively. The higher molecular weight of chitosan was exhibited the lower bulk density and the higher water and fat binding capacities. Bindinf capacities of chitosan powders to bile salts, cholesterol and linoleic acid were $41.2-63.3{\%},\;40.8-67.4{\%},\;42.6-72.6{\%}$, respectively. In NIR spectrum of lipid adsorbed chitosan the occurrence static eletronical binding between chitosan and lipid was identified by NIR spectrum peak induced from combination of carboxylic group in lipid and amino group in chitosan. In conclusion, the higher degree of deacetylation and molecular weight of chitosan showed the higher lipid binding capacity and the lipid adsorption of chitosan were occurred by combination of carboxylic group in lipids and amino group in chitosan.

Non-destructive and Rapid Prediction of Moisture Content in Red Pepper (Capsicum annuum L.) Powder Using Near-infrared Spectroscopy and a Partial Least Squares Regression Model

  • Purpose: The aim of this study was to develop a technique for the non-destructive and rapid prediction of the moisture content in red pepper powder using near-infrared (NIR) spectroscopy and a partial least squares regression (PLSR) model. Methods: Three red pepper powder products were separated into three groups based on their particle sizes using a standard sieve. Each product was prepared, and the expected moisture content range was divided into six or seven levels from 3 to 21% wb with 3% wb intervals. The NIR reflectance spectra acquired in the wavelength range from 1,100 to 2,300 nm were used for the development of prediction models of the moisture content in red pepper powder. Results: The values of $R{_V}{^2}$, SEP, and RPD for the best PLSR model to predict the moisture content in red pepper powders of varying particle sizes below 1.4 mm were 0.990, ${\pm}0.487%$ wb, and 10.00, respectively. Conclusions: These results demonstrated that NIR spectroscopy and a PLSR model could be useful techniques for measuring rapidly and non-destructively the moisture content in red pepper powder.

Nondestructive Evaluation for the Viability of Watermelon (Citrullus lanatus) Seeds Using Fourier Transform Near Infrared Spectroscopy

  • Purpose: Conventional methods used to evaluate seeds viability are destructive, time consuming, and require the use of chemicals, which are not feasible to implement to process plant in seed industry. In this study, the effectiveness of Fourier transform near infrared (FT-NIR) spectroscopy to differentiate between viable and nonviable watermelon seeds was investigated. Methods: FT-NIR reflectance spectra of both viable and non-viable (aging) seeds were collected in the range of 4,000 - 10,000 $cm^{-1}$ (1,000 - 2,500 nm). To differentiate between viable and non-viable seeds, a multivariate classification model was developed with partial least square discrimination analysis (PLS-DA). Results: The calibration and validation set derived from the PLS-DA model classified viable and non-viable seeds with 100% accuracy. The beta coefficient of PLS-DA, which represented spectral difference between viable and non-viable seeds, showed that change in the chemical component of the seed membrane (such as lipids and proteins) might be responsible for the germination ability of the seeds. Conclusions: The results demonstrate the possibility of using FT-NIR spectroscopy to separate seeds based on viability, which could be used in the development of an online sorting technique.

근적외선 분광분석법을 이용한 벌꿀의 품질평가 (Determination of Honey Quality by Near Infrared Spectroscopy)

  • 다양한 품질특성을 가지는 230점의 벌꿀시료를 선발하여 품질평가 요소에 대한 각각의 검량식을 작성하였으며, 검량식 평가를 위한 벌꿀시료는 50점을 사용하였다. 근적외선 스펙트럼에 대한 수학적 처리를 하여 검량식을 얻었으며 회귀분석방법은 PLS법이 가장 적합하였다. 이화학적 분석과 NIR을 이용해 얻은 분석치를 비교한 결과 수분의 RSQ는 0.997, SEP는 0.10%로 매우 신뢰성 있는 결과를 얻을 수 있었다. fructose와 glucose는 RSQ가 0.926, 0.951로 나타났으며 SEP는 각각 0.54%, 0.52%이었으며, sucrose와 maltose는 SEP가 각각 0.25%, 0.22%로 나타났다. HMF의 SEP는 2.96 mg/kg이었으며 산도는 SEP가 0.73 meq/kg이었다. 탄소동위원소비율은 SEP가 $1.08{\textperthousand}$로 오차가 컸으나 RSQ값이 0.950으로 비교적 안정된 결과를 얻을 수 있어 근적외선 분광분석법으로도 꽃꿀의 순도를 판별할 수 있는 가능성이 제시되었다.

Transferring Calibrations Between on Farm Whole Grain NIR Analysers

  • On farm analysis of protein, moisture and oil in cereals and oil seeds is quickly being adopted by Australian farmers. The benefits of being able to measure protein and oil in grains and oil seeds are several : $\square$ Optimize crop payments $\square$ Monitor effects of fertilization $\square$ Blend on farm to meet market requirements $\square$ Off farm marketing - sell crop with load by load analysis However farmers are not NIR spectroscopists and the process of calibrating instruments has to the duty of the supplier. With the potential number of On Farm analyser being in the thousands, then the task of calibrating each instrument would be impossible, let alone the problems encountered with updating calibrations from season to season. As such, NIR technology Australia has developed a mechanism for \ulcorner\ulcorner\ulcorner their range of Cropscan 2000G NIR analysers so that a single calibration can be transferred from the master instrument to every slave instrument. Whole grain analysis has been developed over the last 10 years using Near Infrared Transmission through a sample of grain with a pathlength varying from 5-30mm. A continuous spectrum from 800-1100nm is the optimal wavelength coverage fro these applications and a grating based spectrophotometer has proven to provide the best means of producing this spectrum. The most important aspect of standardizing NIB instruments is to duplicate the spectral information. The task is to align spectrum from the slave instruments to the master instrument in terms of wavelength positioning and then to adjust the spectral response at each wavelength in order that the slave instruments mimic the master instrument. The Cropscan 2000G and 2000B Whole Grain Analyser use flat field spectrographs to produce a spectrum from 720-1100nm and a silicon photodiode array detector to collect the spectrum at approximately 10nm intervals. The concave holographic gratings used in the flat field spectrographs are produced by a process of photo lithography. As such each grating is an exact replica of the original. To align wavelengths in these instruments, NIR wheat sample scanned on the master and the slave instruments provides three check points in the spectrum to make a more exact alignment. Once the wavelengths are matched then many samples of wheat, approximately 10, exhibiting absorbances from 2 to 4.5 Abu, are scanned on the master and then on each slave. Using a simple linear regression technique, a slope and bias adjustment is made for each pixel of the detector. This process corrects the spectral response at each wavelength so that the slave instruments produce the same spectra as the master instrument. It is important to use as broad a range of absorbances in the samples so that a good slope and bias estimate can be calculated. These Slope and Bias (S'||'&'||'B) factors are then downloaded into the slave instruments. Calibrations developed on the master instrument can then be downloaded onto the slave instruments and perform similarly to the master instrument. The data shown in this paper illustrates the process of calculating these S'||'&'||'B factors and the transfer of calibrations for wheat, barley and sorghum between several instruments.

근적외 분광법을 이용한 양념 야채의 건조 불균일성 관찰 (Observation for drying non-uniformity of allium vegetables using NIR spectroscopy)

  • 본 연구에서는 근적외 분광법을 이용하여 양념용 채소로 많이 사용되는 파, 마늘, 양파의 건조 불균일성을 관찰하였다. 대류식 온풍건조기에서 건조 위치에 따른 양념채소류의 건조 상태 차이를 조사한 결과, 건조 트레이의 높이나 위치에 따라 바람의 온도와 습도에 차이가 발생하여 건조가 균일하게 되지 않았다. 건조 정도가 다른 양념채소류의 2차 미분 근적외 스펙트럼을 비교한 결과 1,390 nm에서 1,400 nm 부근의 수소결합이 약한 물이 먼저 증발되었고 1,420 nm 부근의 수소결합이 강한 물은 건조 후에도 잔존함을 관찰할 수 있었다. 초분광 NIR 이미징 기법으로 측정한 스펙트럼을 주성분분석함으로써 건조채소의 건조도를 가시적으로 판별할 수 있었다.

ICE ABSORPTION FEATURES IN NIR SPECTRA OF GALACTIC OBJECTS

  • We present results of AKARI/IRC near-infrared (NIR) slit-spectroscopy ($2.5-5.0{\mu}m$, R ~ 100) of Galactic sources, focusing on ice absorption features. We investigate the abundance of $H_2O$ and $CO_2$ ices and other ice species (CO and XCN ices) along lines of sight towards Galactic H $\small{II}$ regions, massive YSOs, and infrared diffuse sources. Even among those different kinds of astronomical objects, the abundance ratio of $CO_2$ to $H_2O$ ices does not vary significantly, suggesting that the pathway to $CO_2$ ice formation driven by UV irradiation is not effective at least among the present targets.

  • The availability of in vivo and in sacco degradability values are limited because those methods require work with fistulated animals and are rather complicated, labour intensive and expensive. That is to say, the dynamics and logistics of the methodology result in considerable work, due to limitations on the amount of samples, number of bags that can be placed in an animal and different time intervals to perform kinetic studies. Therefore, a simpler method is necessary to estimate the degradation characteristics of the feed. In this way, near infrared reflectance spectroscopy has been used to predict degradation characteristics of forages. In other hand, the possibility of achieving successful transfer of spectra and equations between instruments is closely related. The objective of this study was to confirm the potential of NIR to optimize work conditions to avoid duplicated efforts in collaborative trials on animal feeds evaluation between research institutions. For this purpose, one set with forty hays and dehydrated forages samples from SERIDA and ten samples with the same characteristics from SIA, were be used to create a spectral database. A calibration was developed using samples from degradation essays made in SERIDA to predict dry matter and crude protein degradability. With the addition of five samples from SIA in original calibration set, the effect of different origin and location was compensated.

근적외선분광법을 이용한 이탈리안 라이그라스, 페레니얼 라이그라스,톨 페스큐 종자의 초종 판별 (Discrimination of Pasture Spices for Italian Ryegrass, Perennial Ryegrass and Tall Fescue Using Near Infrared Spectroscopy)

  • 본 연구는 근적외선분광법을 이용하여 우리나라에서 재배되고 있는 목초류 중 외형적 특성이 유사한 이탈리안 라이그라스, 페레니얼 라이그라스와 톨 페스큐 종자의 초종판별 가능성을 검토하고자 수행되었다. 근적외선분광기를 이용하여 목초류 종자를 가시파장 대역대(680~1,099 nm), NIRS 파장 대역대(1,100-2,500 nm) 및 NIRS 전체 파장 대역대(680-2,500 nm)로 구분하여 스펙트라를 얻은 후 1차 미분과 8 nm gap으로 수 처리를 수행하였으며 부분최소자승(PLS) 회귀분석법을 통해 초종판별 검량식을 개발하고 판별 정확성을 검증하였다. 목초류의 초종판별 정확성은 가시파장대역에서 SECV 1.732, $R^2cv$ 0.96으로 가장 판별 정확성이 낮았으며 NIRS 전체 파장대역에서 SECV 1.182, $R^2cv$ 0.98로 가장 높은 판별 정확성을 나타내었다. 파장대역별 예측 정확성은 NIR 파장대역(1,100-2,500 nm)에서 교차검증오차(SECV) 1.319에서 예측 오차(SEP) 1.288로 낮아졌으며 가시영역대(680~1,099)는 SECV 1.732에서 SEP 1.749로 약간 높아졌다. Discrimination equation 분석법에 의한 NIRS 전체 파장대역별 목초류 초종의 판별 결과는 초종간에 판별 정확성의 차이가 크게 나타났으며 이탈리안 라이그라스의 'Hits'는 68%로 가장 낮았으며 페레니얼 라이그라스가 78%의 정확성으로 가장 높게 나타났다. 따라서 NIRS를 이용한 목초류 초종의 판별분석이 가능할 것으로 판단되었다.
