Effect of Protease on the Extraction and Properties of the Protein from Silkworm pupa (Protease 처리가 누에번데기 단백질 추출 및 기능성에 미치는 영향)

  • To extract insoluble proteins from silkworm pupa meal, the meal was treated with pretense produced by Bacillus sp. JH-209. The extraction of insoluble silkworm pupa protein was enhanced at alkaline pHs ranged from 7 to 11 by treatment with the protease. The optimum extraction temperature was $40^{\circ}C$ for in soluble protein treated with pretense. The optimum protease treatment time for extraction of protein was 11 hrs and optimum amount of enzyme treated for extraction of protein was 60 Unit, respectively. The treatment of enzyme extracted more protein than ordinary extraction method without pretense. The foaming capacity, foaming stability, emulsion capacity, and emulsion stability of silkworm pupa meal protein extracted by the treatment of the enzymes increased at all pH ranges. Further more oil absorption as well as water absorption capacities of the protein extracted by the treatment of the enzymes were also increased.

Effect of the Ethanol Extract of Vitis labrusca Root on Apoptosis in Hep G2 Cells (포도근 에탄올 추출물이 Hep G2 세포의 자연사에 미치는 효과)

  • The root of Vitis labrusca, is used as a source of health promoting drug in Korean traditional medicine. It has been reported that root of Vitis labrusca has antioxidant, anti lipid peroxidation and anti-reactive nitrogen species (RNS) activities. The aim of this study was to elucidate the molecular changes of apoptotic signaling pathways in phorbol 12-myristate 13 acetate (PMA)-induced human hepatocellular carcinoma cell line (Hep G2). The root of Vitis labrusca, ethanol extract (RVLEE) was tested for cell viability on Hep G2 cell using the MTT assay. RVLEE exhibited weak cytotoxic activity. However, treatment of Hep G2 cells with RVLEE suppressed PMA-induced cell proliferation. Also, dramatic changes of cell death signals in cellular molecules such as Chk2/Cds1, CIDE-B, CLIMP-63, Bax, Bcl-xL, C-myc, Bcl-2, Bric-5, NIP-3, TRAF2 and BAR but not CIDE-B and DR4. Futhermore, our results showed that the treatment of Hep G2 cells with 25 and $50\; {\mu}g/ml$ of RVLEE suppressed PMA-induced COX-2 gene activity. These data suggest that RVLEE have inhibitory effect of cell proliferation, induction of apoptosis and, thus, may offer therapeutic potential in Hep G2.

The Effects of Ink Emulsion on Printed Mottle (잉크 유화가 인쇄 모틀에 미치는 영향)

  • Lithography like off-set printing is processed using the repellent properties between water and oil, so all inks for off-printing must work with dampening solution(water). The dampening water may cause the emulsification of ink by the printing pressure in the printing nip. This study aimed to investigate the effect of emulsified inks on print mottle. The cyan ink was emulsified artificially with the different IPA(isopropyl alcohol) content in this study. We evaluated the print mottle by densitometer and image analysis method. The print mottle phenomenon was directly affected by IPA content. The emulsification of inks also had an influence on flow properties of inks and it increased ink transfer rate. It, however, caused low ink density. Moreover the emulsified inks were spreaded to around dots and cause the thinning density on the non-printing area like print mottle. The trial printing showed that the emulsified inks also cause scumming on the printing result with little mistake of adjusting dampening solution and mostly decreasing dot reproduction. We could, therefore, find out the obvious effect of emulsified inks on print mottle.

  • The media transport system is used in a printer, a ATM(Automated Tellor Machine), and so on. The media transport system has many problems through miniaturization and rapid transportation of these machines. In the paper feeding mechanism, it is important to feed the sheet without jamming under any conditions. To avoid sheet jamming, first we need to predict the behavior of the sheet exactly. In this paper, the analysis of media behavior is based on J. Stolte's studies. In all of OA machines, a flexible beam or plate is pushed from the channel. The motion may be constrained by guides. This leads to a transient and geometrically nonlinear problem. The behavior of paper is simulated by dynamic elastica theory. The shape of guide is represented by parametric cubic curve. But J. Stolte's studies did not considered contact condition between sheet and guide. So Klarbring's Model. will be applied. And the analysis of flexible media has to include aerodynamic effect for more exact behavior analysis, because the flexible media can be deformed drastically by a little force. Therefore aerodynamic force must be applied to the governing equation. Lastly, the simulation of this model is performed, and the experiment is performed for verification of this model. The experimental results of low exit velocity are consistent with the simulation results, however experimental results of high exit velocity do not agree well with analytical results. The reason is that there may be other effects like nip Phenomena

Effect of Cacao Nip Extracts (CEs) on Quality Characteristics of Pork Patties during Cold Storage Period

  • Cacao has been shown to have antioxidant effects and health benefits. However, the applicability of cacao as a meat preservative has not been thoroughly evaluated. Here, we examined the effects of cacao nib extracts (CEs) on suppression of fat oxidation and enhancement of quality characteristics of pork patties. Cacao nib powder was extracted in distilled water or 50%, 70%, or 99% ethanol. CEs prepared using 70% ethanol had the highest total phenolic and total flavonoid contents, and the highest 1,1-diphenyl-2-picrylhytdrazyl radical and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activities. Decompression-concentrated CEs prepared using 70% ethanol and 0.1% ascorbic acid were added to pork patties, and the physiochemical properties of the patties were measured. The pH of all pork patties increased during storage, but tended to decrease according to the CEs content. CEs enhanced the preservation of redness and texture of the pork patties during storage. Analysis of thiobarbituric acid reactive substances (TBARS) in patties revealed that fat oxidation was highly suppressed in all treatment groups containing CEs during storage, and TBARS values decreased according to CE content. Treatment with 0.1% CE reduced fat oxidation to a level similar to that of treatment with 0.1% ascorbic acid. Consumer flavor preference increased according to CE content, and overall preference was the highest for patties prepared with 0.05% and 0.075% CEs. Overall, 70% ethanol was found to be the optimal concentration for extraction of cacao nibs, and adding 0.05% or 0.075% CEs to pork patties yielded the highest quality.

  • This paper proposes a method of recognizing that a person doesn't put on a safety cap using image processing method in DSP(Digital Signal Processor). It processes inputted images by image input devices that equipped in a industrial settings. If the method recognizes a person that doesn't put on a safety cap, a system transfers relevant recognition result to a supervisor and takes proper measures. If an accident happens and someone doesn't put on a safety cap, additional casualities could be. Proposed method can nip additional casualties in the bud. To recognize that a person don't put on a safety cap, images are processed by object abstraction, removal of noise, decision of a thing or a person, abstraction of a head part in a image, recognizing whether a man puts on a safety cap using HSV color space or not, and so on. Image input and image process are processed by DSP. And C language-based codes are optimized by an eignefunction(Intrinsics) for speed improvement of algorithms.

Development of a precision machining process for the outer cylinder of vacuum roll for film transfer (필름 이송을 위한 진공 롤 외통의 정밀가공 공정개발)

  • Unlike the roll-to-roll process that uses a steel roll and a nip roll, a vacuum roll can hold and transfer a thin film using a single roll. To precisely manufacture a vacuum roll, a thin outer cylinder must be machined, which is assembled on the outside of the roll and contacts the film via vacuum pressure. In this study, the effects of jaw width and chucking force on the deformation of the outer cylinder during the turning process were investigated using analysis, and a precision machining and burr removal process was developed. The deformation of the outer cylinder decreased almost linearly with increasing jaw width and increased with higher chucking force and larger cylinder diameter. Additionally, the deflection due to the weight of the outer cylinder was approximately three times greater than that caused by film tension. For the machined outer cylinder, a burr removal experiment was conducted, and concentricity and cylindricity were measured. Using a device that removes burrs by rotating a wheel connected to the main shaft at high speed, it was found that burrs generated on the inner diameter could be removed very efficiently. On the vacuum side, the concentricity errors of the inner and outer diameters were 0.015 mm and 0.014 mm, respectively, and on the opposite side, they were 0.006 mm and 0.010 mm, respectively. Additionally, the measurement of Total Indicator Runout (TIR) according to the angle showed that the maximum cylindricity of the outer and inner diameters was 0.02 mm and 0.025 mm, respectively. Finally, through burr-height measurement at the hole boundary, it was found that the heights were within 0.05 mm.

A Nationwide Survey on the Child Day Care and Common Infectious Diseases (영유아의 보육시설 이용과 감염성 질환 실태 연구)

  • Purpose : As the number of children who attend child care centers has increased, concerns has increased about the effect of child day care on childhood illness. This study was conducted to examine the relationship between experience in child care and common infectious diseases in children under 5 years of age. Methods : Data were collected by surveying 1,000 respondents with children under age 5 through online interviews using a structured questionnaire. The contents of the survey were composed of demographic characteristics, child care facilities usage, experience in infectious diseases, and immunization status Results : Among the 1,000 children <5 years of age, 78.5% attended a child care facility. Rates of common communicable illnesses were higher in children in child care than for children reared exclusively at home. The predominant communicable diseases which the respondents' children experienced, in order of decreasing frequency, were gastroenteritis (47.1%), otitis media (41.8%) and pneumonia (19.1%). The immunization rate of vaccines that are not included the national immunization program (NIP) (Haemophilus influenzae type b vaccine - 76.6%, hepatitis A vaccine - 63.3%, pneumococcal vaccine - 59.4%, rotavirus vaccine - 43.1%) was lower than that of the NIP vaccines (90.4%) Conclusion : Children in child care experience more bouts of common infectious disease, so nationwide policies to prevent or to control the spread of infectious agents in a child-care should be available and appropriate immunization should be emphasized as the most effective method for the control of infectious disease for children.

Identification of Matrix Mineralization-Related Genes in Human Periodontal Ligament Cells Using cDNA Microarray (cDNA microarray에 의한 치주인대세포의 광물화 결절형성에 관여하는 유전자들의 분석)

  • Periodontal ligament (PDL) cells have been known as multipotential cells, and as playing an important rolesin periodontal regeneration. The PDL cells are composed of heterogeneous cell populations which have the capacity to differentiate into either cementoblasts or osteoblasts, depending on needs and conditions. Therefore, PDL cells have the capacity to produce mineralized nodules in vitro in mineralization medium which include ascorbic acid, ${\beta}$-glycerophosphate and dexamethasone. In spite of these well-known osteoblast like properties of PDL cells, very little is known about the molecules involved in the formation of the mineralized nodules in the PDL cells. In the present study, we analysed gene-expression profiles during the mineralization process of cultured PDL cells by means of a cDNA microarray consisting of 3063 genes. Nodules of mineralized matrix were strongly stained with alizarin red S on the PDL cells cultured in the media with mineralization supplements. Among 3,063 genes analyzed, 35 were up-regulated more than two-fold at one or more time points in cells that developed matrix mineralization nodules, and 38 were down-regulated to less than half their normal level of expression. In accord with the morphological change we observed, several genes related to calcium-related or mineral metabolism were induced in PDL cells during osteogenesis, such as IGF-II and IGFBP-2. Proteogycan 1, fibulin-5, keratin 5, ,${\beta}$-actin, ${\alpha}$-smooth muscle actin and capping protein, and cytoskeleton and extracellular matrix proteins were up-regulated during mineralization. Several genes encoding proteins related to apoptosis weredifferentially expressed in PDL cells cultured in the medium containing mineralization supplements. Dkk-I and Nip3, which are apoptosis-inducing agents, were up-regulated, and Btf and TAXlBP1, which have an anti-apoptosis activity, were down-regulated during mineralization. Also periostin and S100 calciumbinding protein A4 were down-regulated during mineralization.
