MtMKK5 inhibits nitrogen-fixing nodule development by enhancing defense signaling

  • The mitogen-activated protein kinase (MAPK) signaling cascade is essential for a wide range of cellular responses in plants, including defense responses, responses to abiotic stress, hormone signaling, and developmental processes. Recent investigations have shown that the stress, ethylene, and MAPK signaling pathways negatively affect the formation of nitrogen-fixing nodules by directly modulating the symbiotic signaling components. However, the molecular mechanisms underlying the defense responses mediated by MAPK signaling in the organogenesis of nitrogen-fixing nodules remain unclear. In the present study, I demonstrate that the Medicago truncatula mitogen-activated protein kinase kinase 5 (MtMKK5)-Medicago truncatula mitogen-activated protein kinase 3/6 (MtMPK3/6) signaling module, expressed specifically in the symbiotic nodules, promotes defense signaling, but not ethylene signaling pathways, thereby inhibiting nodule development in M. truncatula. U0126 treatment resulted in increased cell division in the nodule meristem zone due to the inhibition of MAPK signaling. The phosphorylated TEY motif in the activation domain of MtMPK3/6 was the target domain associated with specific interactions with MtMKK5. I have confirmed the physical interactions between M. truncatula nodule inception (MtNIN) and MtMPK3/6. In the presence of high expression levels of the defense-related genes FRK1 and WRKY29, MtMKK5a overexpression significantly enhanced the defense responses of Arabidopsis against Pseudomonas syringae pv. tomato DC3000 (Pst DC3000). Overall, my data show that the negative regulation of symbiotic nitrogen-fixing nodule organogenesis by defense signaling pathways is mediated by the MtMKK5-MtMPK3/6 module.

Association of Chicken Growth Hormones and Insulin-like Growth Factor Gene Polymorphisms with Growth Performance and Carcass Traits in Thai Broilers

  • Molecular marker selection has been an acceptable tool in the acceleration of the genetic response of desired traits to improve production performance in chickens. The crossbreds from commercial parent stock (PS) broilers with four Thai synthetic breeds; Kaen Thong (KT), Khai Mook Esarn (KM), Soi Nin (SN), and Soi Pet (SP) were used to study the association among chicken growth hormones (cGH) and the insulin-like growth factor (IGF-I) genes for growth and carcass traits; for the purpose of developing a suitable terminal breeding program for Thai broilers. A total of 408 chickens of four Thai broiler lines were genotyped, using polymerase chain reaction-restriction fragment length polymorphism methods. The cGH gene was significantly associated with body weight at hatching; at 4, 6, 8, 10 weeks of age and with average daily gain (ADG); during 2 to 4, 4 to 6, 0 to 6, 0 to 8, and 0 to 10 weeks of age in $PS{\times}KM$ chickens. For $PS{\times}KT$ populations, cGH gene showed significant association with body weight at hatching, and ADG; during 8 to 10 weeks of age. The single nucleotide polymorphism variant confirmed that allele G has positive effects for body weight and ADG. Within carcass traits, cGH revealed a tentative association within the dressing percentage. For the IGF-I gene polymorphism, there were significant associations with body weight at hatching; at 2, 4, and 6 weeks of age and ADG; during 0 to 2, 4 to 6, and 0 to 6 weeks of age; in all of four Thai broiler populations. There were tentative associations of the IGF-I gene within the percentages of breast muscles and wings. Thus, cGH gene may be used as a candidate gene, to improve growth traits of Thai broilers.

Vitellogenin as a Biomarker of Endocrine Disrupter in the Aquatic Environment

  • A number of chemical released into the environment eliciting their effects by disrupting normal hormonal pathways. Endocrine disrupting compounds are present in the aquatic environment and pose potential health consequences to wildlife and humans. This review are designing fur xenobiotic estrogens based on induction of the egg-yolk precursor protein vitelloge-nin. In fish of aquatic environment, it may result in decrease fertility and egg production in females or lead to reduced gonad size or feminization of genetic male fish. It has been known that male fish exposed to estrogenic compounds show induced production of vitellogenin. Vitello-genin production is normally restricted to adult females, which have elevated estrogen levels during egg production. However, vitellogenin can be induced in males by Pollution of environmental endocrine disruptors. Consequently, the presence of vitellogenin in male fish can serve as an indicator of exposure of environmental endocrine disrupting compounds. In immature fish pol-luted at low levels of environmental endocrine disrupter, vitellogenin can serve as a reliable biomarker for exposure to endocrine disrupter. This review demonstrates the utility of vitellogenin as a biomarker fur exposure to estrogenin agents in auqatic environment.

Fermented Unpolished Black Rice (Oryza sativa L.) Inhibits Melanogenesis via ERK, p38, and AKT Phosphorylation in B16F10 Melanoma Cells

  • Melanin is a major factor that darkens skin color as one of the defense systems to prevent the harmful effects of UV light. However, darkened skin from the localized or systemic accumulation of melanin is viewed in many cultures as an esthetic problem. Consequentially, searching for anti-melanogenic agents from natural sources is very popular worldwide. Previous screening of fermented rice products, obtained from various rice cultivars fermented with different sources of loog-pang (Thai traditional fermentation starter), revealed that the highest ability to reduce the melanin content in B16F10 melanoma cells was from unpolished black rice fermented with a defined starter mixture of microbes isolated from loog-pang E11. The aim of this study was to investigate the mechanism of the fermented unpolished black rice (FUBR) on the inhibition of melanogenesis in B16F10 melanoma cells. The strongest reduction of cellular melanin content was found in the FUBR sap (FUBRS). The melanin reduction activity was consistent with the significant decrease in the intracellular tyrosinase activity. The FUBRS showed no cytotoxic effect to B16F10 melanoma or Hs68 human fibroblast cell lines. It also significantly reduced the transcript and protein expression levels of tyrosinase, tyrosinase-related protein 1 (TYRP-1), TYRP-2, and microphthalmia-associated transcription factor. Furthermore, it induced a significantly increased level of phosphorylated ERK, p38 and Akt signaling pathways, which likely contributed to the negative regulation of melanogenesis. From these results, a model for the mechanism of FUBRS on melanogenesis inhibition was proposed. Moreover, these results strongly suggested that FUBRS possesses anti-melanogenesis activity with high potential for cosmeceutical application as a skin depigmenting agent.

Degradation of Chlorinated Phenolic Compounds by Soil Actinomycetes Isolated from the Contami-nated Soil Nearby the Kyung-An River (경안천 유역 오염토양에서 분리한 방선균의 염화 페놀계 화합물 분해)

  • Lignin-peroxidase (LiP) has been considered as one of the most important industrial enzymes for biodegradation of various recalcitrant toxic compounds such as chlorinated aromatic hydrocarbons and azo-dyes. Recently, several soil actinomycetes have been reported to secrete a functionally-similar lignin-peroxidase called actinomycetes lig-nin-peroxidase (ALiP). In this manuscript, we isolated over 100 morphologically distinct actinomycetes from the contaminated soils around 10 different gas stations located nearby the Kyung-An river. Among these actinomycetes screened based on the congo-red dye-decolorization activities, one newly-isolated actinomycetes named SMA-2 showed the most significant dye-decoloring activity on the congo-red plate as well as a significant ALiP activity in a yeast-extract-malt-extract liquid media supplemented with starch. The optimum SMA-2 culture condition fur ALiP production was determined and the kinetic parameters fur the SMA-2 AkIP activity were characterized. The optimally-cultured SMA-2 also exhibited the oxidation activities toward various recalcitrant aromatic compounds including phenol, 2- chlorophenol, 4- chlorophenol, 2,4- dichlorophenol ,2,6- dichlorophenol, and 2,4, f-trichlorophe - not, suggesting a potential application of SMA-2 for contaminated soil bioremediation.

Color Image Analysis of Histological tissue Sections (해부병리조직에 대한 칼라 영상분석)

  • In this paper, we suggest a new direct method for mage segmentation using texture and color information combined through a multivariate linear discriminant algorithm. The color texture is computed in nin 3${\times}$3 masks obtained from each 3${\times}$3${\times}$3 spatio-spectral neighborhood in the image using the classical haralick and Pressman texture features. Among these 9${\times}$28 texture features the best set was extracted from a training set. The resulting set of 10 features were used to segment an image into four different regions. The resulting segmentation was Compared to classical color and texture segmentation methods using both box classifiers and maximum likelihood classification. It compared favourably on the test image from a Fastred-Lightgreen stained prostatic histological tissue section based on visual inspection. The classification accuracy of 97.5% for the new method obtained on the training data was also among the best of the tested methods. If these results hold for a larger set of images, this method should be a useful tool for segmenting images where both color and texture are relevant for the segmentation process.

Suppression of CYP1A1 Expression by Naringenin in Murine Hepa-1c1c7 Cells

  • Naringenin, dietary flavonoid, is antioxidant constituents of many citrus fruits. In the present study, we investigated the effect of naringenin on 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)-inducible CYP1 A 1 gene expression in mouse hepatoma Hepa-1c1c7 cells. Naringenin alone did not affect CYP1A1-specific 7-ethoxyresorufin O-deethylase (EROD) activity. In contrast, the TCDD-inducible EROD activities were markedly reduced upon concomitant treatment with TCDD and naringenin in a dose dependent manner. TCDD-induced CYP1A1 mRNA level was also markedly suppressed by naringenin. A transient transfection assay using dioxin-response element (DRE)-linked luciferase and electrophoretic mobility shift assay revealed that naringe-nin reduced transformation of the aryl hydrocarbons receptor(AhR) to a form capable of specif-ically binding to the DRE sequence in the promoter of the CYP1A1 gene. These results suggest the down regulation of the CYP1A1 gene expression by either naringenin in Hepa-1c1c7 cells might be antagonism of the DRE binding potential of nuclear AhR.

Reaction Sintering and Thermal Conductivity of AIN Ceramics with $\textrm{Al}_2\textrm{O}_3$ Additions ($\textrm{Al}_2\textrm{O}_3$를 함유하는 AIN세라믹스의 반응소결 및 열전도도)

  • 5-64.3mol% AI$_{2}$O$_{3}$를 함유하는 AIN(1wt% $Y_{2}$O$_{3}$)의 1650-190$0^{\circ}C$ 상압소결에 따른 치밀화 거동, 미세구조, 열전도도가 검토 되었다. XRD 분석결과, AION(5NIN \ulcorner9 AI$_{2}$O$_{3}$ ), 27R AIN다형, AIN이 소결체의 주상으로서 동정되었다. AI$_{2}$O$_{3}$ 의 함량이 증가할수록 소결체의 부피밀도는 증가 하였다. AION을 기지상으로 하는 물질($\geq$ 30mol% AI$_{2}$O$_{3}$ )인 경우는 175$0^{\circ}C$ 소결에서 최대의 부피밀도를 나타내었으며, AIN을 기지상으로 하는 경우(5mol% AI$_{2}$O$_{3}$ ) 는 소결온도가 증가할수록 밀도가 감소하였다. $Y_{2}$O$_{3}$의 존재하에서 주로 185$0^{\circ}C$이상에서 AI$_{2}$O$_{3}$ 와 AIN의 반응에 으해서 액상이 생성되었다. AION을 기지로 하는 물질의 치밀화는 주로 액상의생성 및 AION의 입성장에 의해서 지배되었으나, AIN을 기지로 하는 물질에 있어서는 1$650^{\circ}C$에서 액상이 생성되었고, 소결온도가 190$0^{\circ}C$까지 상승할 동안 AIN의 입성장은 크게 일어나지 않았다. AI$_{2}$O$_{3}$ 함량이 증가할수록 낮은 열도도를 갖는 다량의 AION 및 액상의 생성으로 인하여 소결체의열전도도는 감소 하였다. 5mol% AI$_{2}$O$_{3}$ 를 함유한 190$0^{\circ}C$ 소결체가 최대의 열전도도(77.9W/(m\ulcornerk))를 나타내었다.

A Homeotic Gene, Hoxc8, Regulates the Expression of Proliferating Cell Nuclear Antigen in NIH3T3 Cell

  • Hoxc8 is one of the homeotic developmental control genes regulating the expression of many downstream target genes, through which animal body pattern is established during embryonic development. In previous proteomics analysis, proliferating cell nuclear antigen (PCNA) which is also known as cyclin, has been implied to be regulated by Hoxc8 in F9 murine embryonic teratocarcinoma cell. When the 5' upstream region of PCNA was analyzed, it turned out to contain 20 Hox core binding sites (ATTA) in about 1.17 kbp (kilo base pairs) region ($-520{\sim}-1690$). In order to test whether this region is responsible for Hoxc8 regulation, the upstream 2.3 kbp fragment of PCNA was amplified through PCR and then cloned into the pGL3 basic vector containing a luciferase gene as a reporter. When the luciferase activity was measured in the presence of effector plasmid (pcDNA : c8) expressing murine Hoxc8, the PCNA promoter driven reporter activity was reduced. To confirm whether this reduction is due to the Hoxc8 protein, the siRNA against Hoxc8 (5'-GUA UCA GAC CUU GGA ACU A-3' and 5'-UAG UUC CAA GGU CUG AUA C-3') was prepared. Interestingly enough, siRNA treatment up regulated the luciferase activity which was down regulated by Hoxc8, indicating that Hoxc8 indeed regulates the expression of PCNA, in particular, down regulation in NIN3T3 cells. These results altogether indicate that Hoxc8 might orchestrate the pattern formation by regulating PCNA which is one of the important proteins involved in several processes such as DNA replication and methylation, chromatin remodeling, cell cycle regulation, differentiation, as well as programmed cell death.

Inhalational Deep Sedation Using Sevoflurane in Pediatric Dental Patients (소아치과 환자에서의 세보플루란을 이용한 흡입 심진정법)

  • Background: Sevoflurane, a rotatively new inhalational anesthetic. has non-pungent odor and is less reluctant to pediatric patients. The purpose of this study is to examine the feasibility of sevoflurane in inhalational sedation instead of the nitrous oxide for short and simple dental treatments in pediatric patients. Patients and Methods: Fifteen healthy children, whose dental treatment was abandoned due to their little or no cooperation, were selected with their caregivers' written permission. Deep sedation was induced and maintained with oxygen and 1-5% sevoflufane via specially designed nasal mask. Blood pressure, heart rate, oxygen saturation, and electrocardiogram were monitored at 3-nin interval. A dental anesthesiologist, who was independent of dental treatments, was wholly responsible for the sedation procedure. Post-sedation complications and operator's and caregiver's acceptability of this type of inhalational sedation were also investigated. Results: The systolic and diastolic blood pressure, heart rate, and oxygen saturation was significantly depressed during the deep sedation using sevoflurane (P < 0.05). No severe post-sedation complications were found, however, bradycardia was reported in 3 patients. Almost all the operators and caregivers answered that they would adapt this sedation procedure again if possible. Conclusion: In this study, inhalational deep sedation using sevoflurane for dental treatments was found to be very useful. Furthermore, the application of sevoflurane to conscious sedation for pediatric and adult dental patients should be added.
