Effects of Scutellaria scordifolia Fisch. ex Schrank Extracts on Biofilm Formation and the Activities of Klebsiella pneumoniae (Klebsiella pneumoniae균의 바이오 필름 형성과 활성에 대한 병두황진 추출물의 효과)

  • The emergence of biofilms have generated urgent alarm in clinical and medicine manufacturing fields engaged in the search for novel antimicrobials from ethno-medicinal plants. The National Institutes of Health (NIH) has estimated that 70% of all microbial infections in the world are associated with biofilms. In addition, the emergence of strains resistant to conventional antibiotics has become a serious threat to global public health. Therefore, finding alternative medicines is a major issue in the field of integrative medicine. In this study, four different herb extracts were screened for biofilm formation and the activities of Klebsiella pneumoniae. Of them, Scutellaria scordifolia Fisch. ex Schrank extracts had inhibitory effects on bacterial growth and biofilm formation. The Scutellaia scordifolia Fisch. ex Schrank extracts did not cause any cytotoxicity to L929 cells. The growth of K. pneumoniae was inhibited compared to other comparators in the experimental group containing Scutellaia scordifolia Fisch. ex Schrank. In a group of experiments with plant extracts, a maximum of 60 times the level of living bacteria was confirmed compared to the controls without the addition of the Scutellaia scordifolia Fisch. ex Schrank extracts. In a group of experiments with a significantly lower level of fluorescence extraction, differential interference contrast imaging showed that the number of K. pneumonae was reduced. These results suggest that extracts of this plant be applied as antimicrobial agents against K. pneumoniae, particularly in biofilm forms.

  • In order to develop convenient and reproducible methods for identification of ginseng drugs at a DNA level, RAPD (randomly amplified polymorphic DNA) and PCR-RFLP (PCR-Restriction fragment length polymorphism) analysis were applied within Panax species. To authenticate Panax ginseng betvyeen Chinese and Korean ginseng population, RAPD analysis were carried out using 20 mer-random primer. The similarity coefficients among the DNA of ginseng plants analyzed were low, ranging from 0.197 to 0.491. In addition, using PCR-RFLP analysis, very different fingerprints were obtained within Korean ginseng plants. These results suggest that these methods are able to authenticate the concerned Panax species. Broader application of this approach to authenticate other morphologically similar medicinal materials is rationalized.

  • 최근의 생물학 연구를 위한 기기의 자동화 및 고속화는 생물학 관련 정보량의 급증을 가져오고 있다. 예를 들어, DNA chip에서 얻어지는 마이크로어레이(microarray)는 수천 종류의 유전자의 발현량을 동시에 측정한다. 이러한 기술들은 생물의 세포나 조직에서 일어나는 일련의 다양한 현상을 전체적으로 조망하는 관점에서 관찰할 수 있는 기회를 제공하고 있으며, 이를 통한 생명공학의 전반적인 발전이 기대되고 있다. 따라서 대량의 생물학 관련 정보의 분석이나 데이터 마이닝이 행해지고 있으며 이를 위한 대표적인 기법들로는 각종 클러스터링(clustering) 및 신경망 계열의 모델 등이 있다. 본 논문에서는 확률그래프모델의 하나인 베이지안망(Bayesian network)을 생물정보분석에 이용한다. 구체적으로 유전자 발현패턴과 약물의 활성패턴 및 암 종류 사이의 확률적 관계를 모델링한다. 이러한 모델은 NCI60 dataset(http://discover.nci.nih.gov)에서 베이지안망을 학습함으로써 구성된다. 분석의 대상이 되는 데이터가 sparse하기 때문에 발생하는 어려움들을 해결하기 위한 기법들이 제시되며 학습된 모델에 대한 검증은 이미 생물학적으로 확인되어 있는 사실과의 비교를 통해 이루어진다. 학습된 베이지안망 모델은 각각의 유전자 간, 혹은 유전자와 처리된 약물 간의 실제 생물학적 관계를 다수 표현하며, 이는 제시되는 방법이 생물학적으로 유의미한 가설을 데이터 분석을 통해 효율적으로 생성하는데 유용하게 활용될 수 있음을 보인다.

Induction of the Nuclear Proto-Oncogene c-fos by the Phorbol Ester TPA and c-H-Ras

  • TPA is known to cooperate with an activated Ras oncogene in the transformation of rodent fibroblasts, but the biochemical mechanisms responsible for this effect have not been established. In the present study we used c-fos promoter-luciferase constructs as reporters, in transient transfection assays, in NIH3T3 cells to assess the mechanism of this cooperation. We found a marked synergistic interaction between TPA and a transfected v-Ha-ras oncogene in the activation of c-fos promoter and SRE. SRE has binding sites for TCF and SRF. A dominant-negative Ras (ras-N17) inhibited the TPA-Ras synergy by blocking the PKC-MAPK-TCF pathway. Dominant-negative RhoA and Rac1 (but not Cdc42Hs) inhibited the TPA-Ras synergy by blocking the Ras-Rho-SRF signaling pathway. Constitutively active $PKC{\alpha}$ and $PKC{\varepsilon}$ showed synergy with v-Ras. These results suggest that the activation of two distinct pathways such as Ras-Raf-ERK-TCF pathway and Rho-SRF pathway are responsible for the induction of c-fos by TPA and Ras in mitogenic signaling pathways.

Assessment of the Measurement Method of the Bone Mineral Density on Cu-Equivalent Image (구리당량 영상작성에 의한 골밀도계측방법의 평가)

  • Purpose : The effects of step numbers of copper wedge and exposure on the coefficient of determination (r²) of the conversion equation to Cu-equivalent image and on the Cu-equivalent value (mmCu) and it's coefficient of variation measured at each copper step and the mandibular premolar area were evaluated. Method: Digital image analyzing system consisted of scanner, personal computer, and a stepwedge with 10 steps of 0.03 mm copper in thickness as reference material was prepared for quantitative assessment of the bone mineral density. NIH image program was used for analyzing images. Results : The film having moderately high film density showed the discrepancy between the real thickness and the measured Cu-equivalent value of each copper step. The Cu-equivalent image was dependent on the determinational coefficient of the conversion equation than the coefficient of variance of the measured value. Conclusion : Obtaining conversion equation with high coefficient of determination and proper film exposure are supposed to be neccessary for quantitative assessment of bone density. Multiple steps in the range of the corresponding copper thickness to the bone density of the area to be measured should be prepared.

The determination of reference material for bone density by using bone phantom (골판톰을 이용한 골밀도측정 참조체의 결정)

  • Purpose: To determine the proper reference step wedge for digital Cu-Equivalent Image analyzing systems for measurement of bone density. Meterials and Methods : Radiograms of lumbar vertebrae phantom (1g/㎠) with 3 test copper step wedges of 0.03, 0.05 and, 0.1 mm thickness unit were taken and analyzed using NIH image software on a Macintosh personal computer. Measured densities of the lumbar areas in the Cu-Equivalent images made by utilizing 3 different copper stepwedges were compared with a known bone density. Results: The values of r2 for all copper equivalent images were over 0.99. The mean Cu-Eq value of lumbar in copper equivalent image made by a 0.1 mm copper stepwedge was 0.22 ± 0.06 mm and converted to hydroxyapatite density of 1.03 g/㎠. The stepwedges of 0.03 and 0.05 mm produced results having higher values than the actual known bone density. They did not show the blue and green color level that appeared in lumbar on color enhanced image. Conclusion : A copper stepwedge of adequate thickness and range of steps which can express the range of density of bone being measured should be used.

  • Bacterial lipopolysaccharide(LPS) is can stimulate the most LPS-responsive cells in the mammalian host. The macrophage response to LPS can protect the host from infection but high levels, contribute to systemic inflammatory response syndrome and destruction of host itself, The previously study, secretory leukocyte pretense inhibitor (SLPI) was known LPS-induced product of macrophage and had the function that antagonizes their LPS-induced activation of pro-inflammation signaling factors. Purpose of this study was to identify the expression of SLPI involving the infection in various cell lines including odontoblast cell line. Therefore, we conducted in vitro researches, which treated the LPS to the MDPC-23, and compared to NIH3T3, RAW264.7. To investigate the expressionof SLPI in mRNA level, the methods was used RT-PCR and western blotting for protein expression of SLPI. Moreover, we performed the scanning electron microscopic (SEM) observation for the morphological change. This work was supported by Korea Science and Engineering Foundation.

Effects of Abeliophyllum distichum Nakai Flower Extracts on Antioxidative Activities and Inhibition of DNA Damage

  • The present study attempts to evaluate antioxidant activities of extracts from Abeliophyllum distichum. Nakai flower. The samples were collected in Jangyyeon-myeon, Goesan-gun, Korea and extracted with either hot-water or ethyl acetate (EtOAC). In DPPH, hydroxyl radical scavenging activity and $Fe^{2+}$ chelating activity of EtOAC extracts were 93.41%, 98.43%, and 7.38%, while those of hot-water extracts were 86.93%, 41.33% and 47.68% at 200 ${\mu}g/ml$, respectively. In ${\varphi}X$-174 RF I plasmid DNA cleavage assay, the protective effects of EtOAC and hot-water extracts against oxidative DNA damage were 82% and 17% at 200 ${\mu}g/ml$, respectively. Both extracts showed the protective effect of DNA migration by oxidative stress in intracellular DNA migration assay. Both extracts had no cytotoxity in NIH3T3 cells. Several polyphenolic compounds were identified such as 2-methoxy-benzoic acid, vanillic acid, phytol and pulegone by GC/MS. These results indicated that extracts of Abeliophyllum distichum Nakai flower showed antioxidant activities and protective activities against oxidative DNA damage and showed the possibility to be used as an effective natural antioxidants.

  • Radiation-induced life shortening, carcinogenesis and other pathological changes were investigated in NIH(GP) mice after $^{60}Co-{\gamma}$ ray irradiation(100~700rads). The results were summarized as follows: 1. There were little difference in body weights, hematological examination and other clinical findings between normal and irradiated groups. 2. Mean survival time of the mice after irradiation were decreased dose-dependently. 3. Main gross findings of the mice irradiated were appeared as enlargement of spleen, thymus and lymph nodes, tumorous nodules of lung and cyst of ovary. Especially, enlargement of thymus was promineort in high dose group. 4. Microscopically, there were various findings including myeloid leukemia, thymic lymphoma, lung adenoma, adenosquamous cell carcinoma of pancreas, pneumonia and other pathological changes. Especially thymic lymphoma was highly frequent in the 700 rads group.

Nonviral Gene Delivery by a Novel Protein Transduction Domain

  • Gene therapy using nonviral gene delivery carriers has focused on the development and modification of synthetic carriers such as liposomes and polymers. Most polymers that are commercially used are taking advantage of their polycationic character which allows not only strong ligand-DNA affinity but also competent cell penetration. Despite the relatively high transfection efficiencies, high cytotoxicity is continuously pointed out as one of the major shortcomings of polycationic polymers such as PEI. Studies on the utilization of peptides have therefore been carried out recently to overcome these problems. For these reasons, the human transcription factor Hph-1, which is currently known as a protein transduction domain (PTD), was investigated in this study to evaluate its potential as a gene delivery carrier. Although its transfection efficiency was about 10-fold lower than PEI, it displayed almost no cytotoxicity even at concentrations as high as $100{\mu}M$. Hph-1 was oxidatively polymerized to yield poly-Hph-1. The cell viability of poly-Hph-1 transfected U87MG and NIH-3T3 cells was almost as high as the control (untreated) groups, and the transfection efficiency was about 10-fold higher than PEI. This study serves as a preliminary evaluation of Hph-1 and encourages further investigation.
