Development of Novel Microsatellite Markers for Strain-Specific Identification of Chlorella vulgaris

  • A strain-specific identification method is required to secure Chlorella strains with useful genetic traits, such as a fast growth rate or high lipid productivity, for application in biofuels, functional foods, and pharmaceuticals. Microsatellite markers based on simple sequence repeats can be a useful tool for this purpose. Therefore, this study developed five novel microsatellite markers (mChl-001, mChl-002, mChl-005, mChl-011, and mChl-012) using specific loci along the chloroplast genome of Chlorella vulgaris. The microsatellite markers were characterized based on their allelic diversities among nine strains of C. vulgaris with the same 18S rRNA sequence similarity. Each microsatellite marker exhibited 2~5 polymorphic allele types, and their combinations allowed discrimination between seven of the C. vulgaris strains. The two remaining strains were distinguished using one specific interspace region between the mChl-001 and mChl-005 loci, which was composed of about 27 single nucleotide polymorphisms, 13~15 specific sequence sites, and (T)n repeat sites. Thus, the polymorphic combination of the five microsatellite markers and one specific locus facilitated a clear distinction of C. vulgaris at the strain level, suggesting that the proposed microsatellite marker system can be useful for the accurate identification and classification of C. vulgaris.

Teaching Location Using Newpaper Articles: Venture Firms as an Example (신문기사를 활용한 입지론 수업 : 벤처기업의 입지 사례)

  • Location is the key theme in geography and various location theories are well established for diverse economic activities. As normative theory, the logical presentation of the location theories tends to make students appraise the class difficult and lose interests. This study presents a teaching materials and inductive method for the location of venture firms using newspaper articles as an example of the knowledge and information industry. Presented are various newspaper articles about venture firms in which the locational notions are selected to find out general location factors. And then their locational preference of large cities, diseconomies of agglomeration, the possibilities of deconcentration from large cities and new establishments in peripheral regions are presented in order. The inductive way of teaching location from specific examples to general tendencies using NIE could bring students' interests and participation which is in line with the student-oriented inquiry-based teaching emphasized recently in educational methods. It could be an effective teaching method for the location theories in economic geography and other difficult subjects.

Effective Stiffness of Composite Beams Considering Shear Slip Effects (전단슬립 효과를 고려한 합성보의 유효강성)

  • This study investigated the effects of a shear slip on the deflection of steel-concretecomposite beams with partial shear interaction. Under the guidance of various current design codes, this deflection was related to the strength of shear connectors in the composite beams. In this paper, a shear connector stiffness based on exact solutions, regardless of loading conditions, was developed. The equivalent rigidity of composite beams that considered three different loading types was first derived, based on equilibrium and curvature compatibility, from which a general formula accounting for slips was developed. To validate this approach, the predicted maximum deflection under the proposed method was compared against currently used equations to calculate beam effective stiffness (AISC)Nie's equations, which have recently been proposed. For typical beams that were used in practice, shear slips might result in stiffness reduction of up to 18% for short-span beams. For full composite sections, the effective section modulus with the AISC specifications was larger than that of the present study, which meant that the specifications were not conservative. For partial composite sections, the AISC predictions were more conservative than those in the present study.

  • This study is considered how to use newspapers to apply education by the way of analyses of environment coverage in newspapers. Data for the study were gathered by content analyses of KINDS(Korean Integrated News Database System) established by Korean Press Institute. The environment coverage is mainly placed in social and regional magazines of newspapers, and the news story are mainly assigned to straight/feature magazines in type. The news of environment coverage is mostly gathered from data by government-informer, and the news is positive/agreement or negative/disagreement in tenor. The news gathering methods of planning/magazine newspaper serial are scientific and objective, and they are of the firsthand data by news reporter, contributions by experts and interviews. The spaces of the news are specially edited. The environment news is often negative/disagreeable in tenor because the news is mostly of straight ones written by non-experts. Applying newspapers in education is a useful learning method which students could develop thinking power and induce concerning and interest by themselves. From the results of the study, the useful suggestions to apply newspapers to learning are as follows. At first, spaces and types of news must be read in detail. Secondly, it is hopeful that indirect news by not writer himself might be possibly avoided in learning. Thirdly, the themes of news would be picked up in relation with learning contents. Lastly, it suggests that the tenor of news is neutral or, in cases, positive and negative together possibly.

Carbon Budget in Campus of the National Institute of Ecology (국립생태원 캠퍼스 내 주요 식생의 탄소수지)

  • This study was conducted to quantify a carbon budget of major vegetation types established in the campus of the National Institute of Ecology (NIE). Carbon budget was measured for Pinus thunbergii and Castanea crenata stands as the existing vegetation. Net Primary Productivity (NPP) was determined by applying allometric method and soil respiration was measured by EGM-4. Heterotrophic respiration was calculated as 55% of total respiration based on the existing results. Net Ecosystem Production (NEP) was determined by the difference between NPP and heterotrophic respiration (HR). NPPs of P. thunbergii and C. crenata stands were shown in $4.9ton\;C\;ha^{-1}yr^{-1}$ and $5.3ton\;C\;ha^{-1}yr^{-1}$, respectively. Heterotrophic respirations of P. thunbergii and C. crenata stands were shown in $2.4ton\;C\;ha^{-1}yr^{-1}$ and $3.5ton\;C\;ha^{-1}yr^{-1}$, respectively. NEPs of P. thunbergii and C. crenata stands were shown in $2.5ton\;C\;ha^{-1}yr^{-1}$ and $1.8ton\;C\;ha^{-1}yr^{-1}$, respectively. Carbon absorption capacity for the whole set of vegetation types established in the NIE was estimated by applying NEP indices obtained from current study and extrapolating NEP indices from existing studies. The value was shown in $147.6ton\;C\;ha^{-1}yr^{-1}$ and it was calculated as $541.2ton\;CO_2ha^{-1}yr^{-1}$ converted into $CO_2$. This function corresponds to 62% of carbon emission from energy that NIE uses for operation of various facilities including the glass domes known in Ecorium. This carbon offset capacity corresponds to about five times of them of the whole national territory of Korea and the representative rural area, Seocheongun. Considered the fact that ongoing climate change was originated from imbalance of carbon budget at the global level, it is expected that evaluation on carbon budget in the spatial dimension reflected land use pattern could provide us baseline information being required to solve fundamentally climate change problem.

Structure of Export Competition between Asian NIEs and Japan in the U.S. Import Market and Exchange Rate Effects (한국(韓國)의 아시아신흥공업국(新興工業國) 및 일본(日本)과의 대미수출경쟁(對美輸出競爭) : 환율효과(換率效果)를 중심(中心)으로)

  • This paper analyzes U.S. demand for imports from Asian NIEs and Japan, utilizing the Almost Ideal Demand System (AIDS) developed by Deaton and Muellbauer, with an emphasis on the effect of changes in the exchange rate. The empirical model assumes a two-stage budgeting process in which the first stage represents the allocation of total U.S. demand among three groups: the Asian NIEs and Japan, six Western developed countries, and the U.S. domestic non-tradables and import competing sector. The second stage represents the allocation of total U.S. imports from the Asian NIEs and Japan among them, by country. According to the AIDS model, the share equation for the Asia NIEs and Japan in U.S. nominal GNP is estimated as a single equation for the first stage. The share equations for those five countries in total U.S. imports are estimated as a system with the general demand restrictions of homogeneity, symmetry and adding-up, together with polynomially distributed lag restrictions. The negativity condition is also satisfied for all cases. The overall results of these complicated estimations, using quarterly data from the first quarter of 1972 to the fourth quarter of 1989, are quite promising in terms of the significance of individual estimators and other statistics. The conclusions drawn from the estimation results and the derived demand elasticities can be summarized as follows: First, the exports of each Asian NIE to the U.S. are competitive with (substitutes for) Japan's exports, while complementary to the exports of fellow NIEs, with the exception of the competitive relation between Hong Kong and Singapore. Second, the exports of each Asian NIE and of Japan to the U.S. are competitive with those of Western developed countries' to the U.S, while they are complementary to the U.S.' non-tradables and import-competing sector. Third, as far as both the first and second stages of budgeting are coneidered, the imports from each Asian NIE and Japan are luxuries in total U.S. consumption. However, when only the second budgeting stage is considered, the imports from Japan and Singapore are luxuries in U.S. imports from the NIEs and Japan, while those of Korea, Taiwan and Hong Kong are necessities. Fourth, the above results may be evidenced more concretely in their implied exchange rate effects. It appears that, in general, a change in the yen-dollar exchange rate will have at least as great an impact, on an NIE's share and volume of exports to the U.S. though in the opposite direction, as a change in the exchange rate of the NIE's own currency $vis-{\grave{a}}-vis$ the dollar. Asian NIEs, therefore, should counteract yen-dollar movements in order to stabilize their exports to the U.S.. More specifically, Korea should depreciate the value of the won relative to the dollar by approximately the same proportion as the depreciation rate of the yen $vis-{\grave{a}}-vis$ the dollar, in order to maintain the volume of Korean exports to the U.S.. In the worst case scenario, Korea should devalue the won by three times the maguitude of the yen's depreciation rate, in order to keep market share in the aforementioned five countries' total exports to the U.S.. Finally, this study provides additional information which may support empirical findings on the competitive relations among the Asian NIEs and Japan. The correlation matrices among the strutures of those five countries' exports to the U.S.. during the 1970s and 1980s were estimated, with the export structure constructed as the shares of each of the 29 industrial sectors' exports as defined by the 3 digit KSIC in total exports to the U.S. from each individual country. In general, the correlation between each of the four Asian NIEs and Japan, and that between Hong Kong and Singapore, are all far below .5, while the ones among the Asian NIEs themselves (except for the one between Hong Kong and Singapore) all greatly exceed .5. If there exists a tendency on the part of the U.S. to import goods in each specific sector from different countries in a relatively constant proportion, the export structures of those countries will probably exhibit a high correlation. To take this hypothesis to the extreme, if the U.S. maintained an absolutely fixed ratio between its imports from any two countries for each of the 29 sectors, the correlation between the export structures of these two countries would be perfect. Therefore, since any two goods purchased in a fixed proportion could be classified as close complements, a high correlation between export structures will imply a complementary relationship between them. Conversely, low correlation would imply a competitive relationship. According to this interpretation, the pattern formed by the correlation coefficients among the five countries' export structures to the U.S. are consistent with the empirical findings of the regression analysis.

Study of Cell-mediated Response in Mice by HPV16 L1 Virus-like Particles Expressed in Saccharomyces cerevisiae

  • The first vaccine against human papillomaviruses (HPV) formulated with HPV16 L1 virus-like particles (VLPs) produced in yeast was approved by the FDA in June 2006. Nevertheless, there have been few studies of the immunogenicity in mice of VLPs. In this study, we evaluated the cell-mediated immune response to VLPs produced in Saccharomyces cerevisiae. After immunization of mice with HPV16 L1 VLPs, we measured splenocytes proliferation and the levels of IFN$_{\gamma}$, IL2, IL4, and IL5. Splenocytes proliferation was significantly increased and a mixed Th1/Th2 response was indicated. IgG subtype immunoresponses were strongly induced and IgG1 titers were higher than those of IgG2a.

  • Cancer is a menace fast gaining momentum in Nigeria and other developing countries. It is an expensive disease requiring a major financial and human resources for prevention, diagnosis and treatment. With no national policy on cancer control in the conntry, incidence (111.7/100,000 population) and mortality (86.6/100,000) rates in Nigeria are spiraling beyond control. This literature search study was primarily aimed at providing recommendations on cost-effective strategies for development interventions to promote self-management for cancer survivors in Nigeria with a goal to improve quality of life and overall survival.

Packet Scheduling with QoS and Fairness for Downlink Traffic in WiMAX Networks

  • The IEEE 802.16 standard is supposed to provide a wide-range broadband wireless service, but it leaves the implementation of the wireless resource scheduler as an open issue. We have studied the scheduling problem and propose a two level scheduling (TLS) scheme with support for quality of service and fairness guarantees for downlink traffic in a WiMAX network. A central controller Base Station has a number of users, and each mobile subscriber station has different channel conditions. The same mobile subscriber station may have different service requirements at different times in the WiMAX network. Based on OPNET simulation, the results show our scheduling algorithm can increase the network throughput, maintain relative fairness, and lower delay over the round robin and weighted round robin algorithms.
