전립선 암조직에서의 사람파필로마바이러스의 감염과 p53단백질의 발현에 대한 연구 (Human Papillomavirus Infections and p53 Expression in Prostatic Carcinoma)

  • Prostatic carcinoma is the leading second cause of cancer in men. Previous epidermiological studies implicated human papillomavirus as an infectious agent. Since there are only limited studies on the association of HPV to prosate cancer, we examined the prevalence of HPV infections in korean prostate cancer patients. We observed that out of 26 cases, 4 cases and 5 cases were infected by HPV 16(27%) and HPV 18 (31%), respectively and 3 cases by both (46%) and at least 18 were positive for HPV (69%). For these samples, immunohistochemical detection of the p53 and proliferative cell nuclear antigen (PCNA) were also studied, using monoclonal antibodies. Sixteen of 26 (61%) showed immunostaining for p53 protein. While 8 samples with no HPV infection (100%) showed all positive for p53 protein staining, less than half of the 18 patients with any HPV infection (44%) showed p53 protein staining. These findings indicate that altered expression of p53 protein occurs in the more than half of prostate cancers, however, p53 expression is less frequent in HPV infected tissues. This implies that there might be an inverse correlation in general between HPV infection and p53 amplification. However, while 50% (4 of 8) of HPV negative prostate cancer was positive for PCNA staining, 13 out of 18 HPV infected patients (72%) were positive. Therefore HPV infection is more strongly associated with increase proliferation. In addition HPV infected cancer patients are generally in more advanced status implying that HPV infection plays a role in the development of highly malignant prostatic carcinomas, eventhough the statistical significance of this interpretation might be waited for the analysis of more cases.

낙동강 본류 측정소들의 생물학적 산소요구량 수치에 대한 비모수적 회귀분석과 특이점분석 (Lowess and outlier analysis of biological oxygen demand on Nakdong main stream river)

  • 본 연구는 국립환경과학원의 물환경정보시스템에서 제공한 자료를 사용하였다. 자료는 낙동강 본류 (낙본, nb)의 수질측정소 A지역에서 측정소 N지역까지 2003년 1월부터 2013년 8월까지 측정한 월별 수질데이터를 이용하였다. 생물학적 산소요구량 BOD (biological oxygen demand)의 통계학적 수질분석은 월별, 연도별, 지역별로 R 프로그래밍을 이용하여 구체화 하였다. 낙본지역 측정소들의 BOD에 대하여 탐색적 자료분석 (exploratory data analysis) 방법과 비모수 회귀분석방법 중 하나인 Lowess (locally weighted scatter plot smoother) 경향분석법을 이용하여 장기수질경향과 지역별 수질분포의 현황을 분석하였다. 그리고 특이점 (outlier)이 가장 많이 발생하는 시기와 낙본 측정지역들을 분석하였다. 그 결과 낙본하류지역인 부산광역시 강서구 명지동 측정소 nbM의 BOD 수질환경 보다 낙본중류지역인 대구광역시 달성군 구지면의 측정소 nbG와 경상남도 창원시의 측정소 nbI 지역의 수질오염이 보다 심각한 문제점들이 있는 것으로 나타난다.

Reversibility and safety of KISS1 metastasis suppressor gene vaccine in immunocastration of ram lambs

  • Objective: The aim of this study was to investigate the reversibility and safety of KISS1 metastasis suppressor (KISS1) gene vaccine in immunocastration. Methods: Six eight-week old ram lambs were randomly divided into vaccinated and control groups. The vaccine (1 mg/ram lamb) was injected at weeks 0, 3, and 6 of the study. Blood samples were collected from the jugular vein before primary immunization and at weeks 2, 4, 6, 10, 14, 22, and 30 after primary immunization. All ram lambs were slaughtered at 38 weeks of age, and samples were collected. Results: The specific anti-KISS1 antibody titers in vaccinated animals were significantly higher and the serum testosterone level was significantly lower than those in the control groups from week 4 to 14 after primary immunization (p<0.05). No significant difference was observed at weeks 22 and 30 after the primary immunization. Similar results were also found for scrotal circumference, testicular weight, length, breadth, and spermatogenesis in seminiferous tubules in week 30 after primary immunization. KS (KISS1-hepatitis B surface antigen S) fusion fragment of KISS1 gene vaccine was not detected in host cell genomic DNA of 9 tissues of the vaccinated ram lambs by polymerase chain reaction. Conclusion: The effects of KISS1 gene vaccine in immunocastration were reversible and no integration events were recorded.

자연산과 양식산 동자개 Pseudobagrus fulvidraco 정소의 생식소발달 비교 연구 (A Comparative Study of Male Gonadal Development between Wild and Cultured Yellow Catfish, Pseudobagrus fulvidraco)

  • 양식업에서 인기 있는 어종 중 하나인 동자개를 자연산과 양식산을 구분하여 생식소 발달을 관찰하였다. 자연산과 양식산 동자개는 2018년 4월에서 10월에 채집하여 수컷의 생식소 지수, 비만도지수 및 생식소 발달을 연구하였다. 광학현미경으로 관찰된 월별 생식소 발달과 GSI를 기반으로 생식소 발달 단계는 자연산과 실외 사육장은 4월 성숙기, 5월 산란기, 6월에서 7월 퇴화기, 8월에서 10월 휴지기로 관찰되었다. 실내 사육장은 4월에서 6월까지 성숙기, 7월에서 8월 산란기, 9월 퇴화기, 10월 휴지기로 구분되었다. GSI 값은 자연산과 실외 사육장은 5월에 가장 높은 값을 보였고, 실내 사육장은 9월에 가장 높은 값을 나타내 차이를 보였다. CF는 자연산과 양식산 사이에 유의한 차이가 없었으나 자연산은 6월에 가장 높은 값을 보였으며, 실외 사육장은 11월에 가장 높은 값을 보였다. 본 실험 결과, 실외 사육장 동자개 수컷을 인공 수정에 사용하는 것이 적합하며, 자연산 생식주기와 같은 5월에 인공 수정을 하는 것이 효율적일 것으로 판단된다.

삼백초 추출물의 자궁경부암세포 억제 효능 (The Efficacy of Saururus chinensis on Cervical Cancer Cells : The Inhibitory Effect on the Function of E6 and E7 Oncogenes of HPV Type 16)

  • Cervical cancer is one of the leading causes of female death from cancer worldwide with about 500,000 deaths per year. A strong association between certain human papilloma viruses (HPV type 16 and 18) and cervical cancer has been well known. An extract of Saururus chinensis, named as PE-46, has been used to investigate whether this agent has the ability of inhibiting the oncogenes E6 and E7 of HPV type 16. PE-46 inhibited the proliferation of human cervical cancer cell lines in a dose response manner. PE-46 also inhibited the in vitro binding of E6 and E6AP which are essential for the binding and degradation of the tumor suppressor p53. In addition, PE-46 inhibited the in vitro binding of E7 and Rb which is essential tumor suppressor for the control of cell cycle. The levels of mRNA for E6 and E7 were also decreased by PE-46. SiHa cells treated with PE-46 induced G0/G1 arrest, resulting in inhibition of growth. Our study showed that the PE-46 can inhibit the cervical carcinomas via both inhibition of bindings between oncogenes and tumor suppressors, and inhibition of G1longrightarrowS transition. PE-46 inhibited the oncogenecity of E6 and E7 of HPV 16 type, thus could be used as a putative modulating agent for the treatment of cervical carcinomas caused by HPV.

空間活動保險法律問題及中國狀況:基於空間商業化最新發展的分析 (Legal Aspects of Insurance Regarding Space Activities and the Situation in China: an Analysis Based on the New Development of Space Commercialization)

  • Insurances of space activities are divided into satellite insurance, astronaut insurance and third party liability insurance. Against the background of the rapid development of space commercialization, especially the increasing participation of private entities in space affairs, the present international and domestic mechanisms of space insurance are challenged. As a space-faring state which is in the process of developing space businesses, the regulations of space insurance in China are deserved to be discussed. Satellites insurance is at present well-developed, the "pre-launch", "launch" and "in-orbit" phases of satellites are all possible to be insured by related companies. China created the CAIA in 1997 to provide insurance for Chinese satellites. However, with more private entities start to involve in space as well as satellite industry, the regime established under the framework of CAIA is necessary to be modified, and the mechanism relating to space insurance brokers should be promoted. The astronauts are recognized as the envoy of humankind, and relevant international regulations are made to provide assistance to them in emergency circumstances. From the domestic perspective, astronauts will be fully insured. China creates a particular type of insurance for astronauts. However, once space tourism becomes a business, the insurance of the tourist will be demanded to be created. In order to promote China's space tourism, it is recommended to take the "Astronaut Group Insurance" as an optional model to space tourists, if the tourists are customers of a governmental-owned space company. Once private involvement of providing orbital/suborbital tourism service becomes a reality, new rules are required. Getting a third party liability insurance is deemed as an indispensable precondition for an applicant to get a launch permission. Domestic space laws will include provisions for the third party liability insurance. China's "Interim Measures" of 2002 realizes the importance of third party liability insurance and requires the permit holder to get it before entering the launching site. This regulation is different from the practices of other states. Concerning that China is the sponsor of APSCO, for the purpose of promoting commercial space cooperation, a harmonized approach to domestic law is recommended to be found.

Comparison of In Vitro Cell Transformation Assay Using Murine Fibroblasts and Human Keratinocytes

  • The in vitro cell transformation assays (CTA) were performed using BALB/3T3 murine fibroblasts and HaCaT human keratinocytes in order to evaluate concordance between both in vitro CTAs and carcinogenicity with compounds differing in their genotoxic and carcinogenic potential. Six test articles were evaluated, two each from three classes of compounds: genotoxic carcinogens (2-amino-5-nitrophenol and 4-nitroquinoline-N-oxide), genotoxic noncarcinogens (8-hydroxyquinoline and benzyl alcohol), and nongenotoxic carcinogens (methyl carbamate and N-nitrosodiphenylamine). Any foci of size $\geq$2 mm regardless of invasiveness and piling was scored as positive in CTA with BALB/3T3. As expected, four carcinogens regardless of their genotoxicity had positive outcomes in two-stage CTA using BALB/3T3 cells. However, of the two genotoxic noncarcinogens, benzyl alcohol was positive CTA finding. We concluded that, of the 6 chemicals tested, the sensitivity for BALB/3T3 system was reasonably high, being 100%. The respective specificity for BALB/3T3 assay was 50%. We also investigated the correlation between results of BALB/3T3 assay and results from HaCaT assay in order to develop a reliable human cell transformation assay. However, evaluation of staining at later time points beyond the confluency stage did not yield further assessable data because most of HaCaT cells were detached after $2{\sim}3$ days of confluency. Thus, after test article treatment, HaCaT cells were split before massive cell death began. In this modified protocol for this HaCaT system, growing attached colonies were counted instead of transformed foci 3 weeks since last subculture. Compared to BALB/3T3 assay, HaCaT assay showed moderate low sensitivity and high specificity. Despite these differences in specificity and sensitivity, both cell systems did exhibit same good concordance between in vitro CTA and rodent carcinogenicity findings (overall 83% concordant results). At present the major weakness of these in vitro CTA is lack of validation for regulatory acceptance and use. Thus, more controlled studies will be needed in order to be better able to assess and quantitatively estimate in vitro CTA data.

Evaluation of Potential Biomarkers for Thioacetamide-induced Hepatotoxicity using siRNA

  • In our previous publication we compared the gene expression profiles on hepatotoxicants exposure to assess the comparability between in vivo and in vitro test systems. We investigated global gene expression from both mouse liver and mouse hepatic cell line treated with thioacetamide (TAA) and identified several common genes. In this study, we selected genes to validate them as potential biomarkers for hepatotoxicity on the relevance of in vitro and in vivo system. Three up-regulated, aquaporin 8 (Aqp8), glutathione peroxidase 1 (Gpx1), succinate-CoA ligase, GDP-forming, alpha subunit (Suclg1) and two down-regulated, DnaJ (Hsp40) homolog subfamily C member 5 (Dnajc5) and tumor protein D52 (Tpd52) genes were tested for their effects in vitro. For characterization of gene function, short interfering RNA (siRNA) for each gene was synthesized and transfected in mouse hepatic cell line, BNL CL.2. Cell viability, mRNA expression level and morphological alterations were investigated. We confirmed siRNA transfection against selected five genes induced down-regulation of respective mRNA expression. siRNA transfection in general decreased cell viability in different degrees and induced morphological changes such as membrane thickening and alterations of intracellular structures. This suggests that these genes could be associated with TAA-induced toxicity. Furthermore, these genes may be used in the investigation of hepatotoxicity for better understanding of its mechanism.

Effect of Age on Energy Requirement for Maintenance and Growth of Dorper and Hu Crossbred F1 Ewes Weighing 20 to 50 kg

  • This research aimed to define the energy requirement of Dorper and Hu Hybrid $F_1$ ewes 20 to 50 kg of body weight, furthermore to study energy requirement changes with age and evaluate the effect of age on energy requirement parameters. In comparative slaughter trial, thirty animals were divided into three dry matter intake treatments (ad libitum, n = 18; low restricted, n = 6; high restricted, n = 6), and were all slaughtered as baseline, intermediate, and final slaughter groups, to calculate body chemical components and energy retained. In digestibility trial, twelve ewes were housed in individual metabolic cages and randomly assigned to three feeding treatments in accordance with the design of a comparative slaughter trial, to evaluate dietary energetic values at different feed intake levels. The combined data indicated that, with increasing age, the net energy requirement for maintenance ($NE_m$) decreased from $260.62{\pm}13.21$ to $250.61{\pm}11.79kJ/kg^{0.75}$ of shrunk body weight (SBW)/d, and metabolizable energy requirement for maintenance (MEm) decreased from $401.99{\pm}20.31$ to $371.23{\pm}17.47kJ/kg^{0.75}$ of SBW/d. Partial efficiency of ME utilization for maintenance ($k_m$, 0.65 vs 0.68) and growth ($k_g$, 0.42 vs 0.41) did not differ (p>0.05) due to age; At the similar condition of average daily gain, net energy requirements for growth ($NE_g$) and metabolizable energy requirements for growth ($ME_g$) for ewes during late fattening period were 23% and 25% greater than corresponding values of ewes during early fattening period. In conclusion, the effect of age upon energy requirement parameters in the present study were similar in tendency with previous recommendations, values of energy requirement for growth ($NE_g$ and $ME_g$) for Dorper and Hu crossbred female lambs ranged between the NRC (2007) recommendation for early and later maturating growing sheep.

Real-Time PCR Analysis of Metabolic Pathway of PHB in Acidiphilium cryptum DX1-1

  • The time, yield, and related genes expression of PHB accumulation of Acidiphilium cryptum DX1-1 were investigated under four different initial C/N ratios, 1.2, 2.4, 7.5, and 24. The results of time and yield of poly-$\beta$-hydroxybutyrate (PHB) accumulation show that the initial C/N ratio of 2.4 was optimum for strain DX1-1 to accumulate PHB, but both higher and lower initial C/N ratios did not favor that process. Based on the genome of Acidiphilium cryptum JF-5, 13 PHB accumulation related genes in strain JF-5 were chosen and successfully cloned from strain DX1-1. The differential expressions of the 13 functional genes, in different C/N ratios as cited above, were then studied by real-time PCR. The results show that all the 13 genes were most upregulated when the initial C/N ratio was 2.4, and among which the gene Acry_3030 encoding poly-$\beta$-hydroxybutyrate polymerase and Aery_0626 encoding acetyl-CoA synthetase were much more upregulated than the other genes, which proved that they play the most important role for PHB accumulation, and acetate is the main initial substance for PHB accumulation for strain DX1-1. Potential regulatory motifs analysis showed that the genes related to PHB accumulation are regulated by different promoters and that the motif had weak similarity to the model promoters, suggesting that PHB metabolism in Acidiphilium cryptum may be mediated by a different mechanism.
