Identification of a Variant Form of Cellular Inhibitor of Apoptosis Protein (c-IAP2) That Contains a Disrupted Ring Domain

  • Among the members of the inhibitor of apoptosis (IAP) protein family, only Livin and survivin have been reported to have variant forms. We have found a variant form of c-IAP2 through the interaction with the X protein of HBV using the yeast two-hybrid system. In contrast to the wild-type c-IAP2, the variant form has two stretches of sequence in the RING domain that are repeated in the C-terminus that would disrupt the RING domain. We demonstrate that the variant form has an inhibitory effect on TNF-mediated $NF-{\kappa}B$ activation unlike the wild-type c-IAP2, which increases TNFmediated $NF-{\kappa}B$ activation. These results suggest that this variant form has different activities from the wild-type and the RING domain may be involved in the regulation of TNF-induced $NF-{\kappa}B$ activation.

Anti-Inflammatory Effect of Fermented Artemisia princeps Pamp in Mice

  • Essential oil-excluded Artemisia princeps Pamp var Ssajuarissuk (AP) was fermented with Lactobacillus brevis K-1, which was isolated from cabbage Kimchi, and the anti-inflammatory effects of AP and fermented AP (FAP) on lipopolysaccharide (LPS)-induced inflammatory response in peritoneal macrophages were investigated. AP and FAP inhibited LPS-induced TNF-$\alpha$, IL-$1{\beta}$, COX-2, iNOS and COX-2 expression, as well as NF-${\kappa}B$ activation. AP and FAP also reduced ear thickness, inflammatory cytokine (TNF-$\alpha$, IL-$1{\beta}$ and IL-6) expression and NF-${\kappa}B$ activation with 12-O-tetradecanoylphorbol-13-acetate (TPA) induced dermatitis in mice. Furthermore, AP and FAP also reduced exudate volume, cell number, protein amount, inflammatory cytokines (TNF-$\alpha$, IL-$1{\beta}$ and IL-6) expression and NF-${\kappa}B$ activation in carrageenan-induced air pouch inflammation in mice. The inhibitory effects of FAP were more potent than those of non-fermented AP. Based on these findings, we propose that FAP can improve inflammatory diseases, such as dermatitis, by inhibiting the NF-${\kappa}B$ pathway.

Oak Wood Vinegar Suppresses the Expression of Cyclooxygenase-2 Induced by TLR4 Agonist

  • Toll-like receptors (TLRs) recognize molecular structures derived from microbes including bacteria, viruses, yeast, and fungi. TLRs have emerged as a major signaling component of the mammalian host defense. TLR4 is a member of the Toll family that senses lipopolysaccharide (LPS), a cell wall component of gram negative bacteria. LPS recognition by TLR4 requires an additional accessory molecule, MD-2. LPS induces the activation of NF-${\kappa}B$ and IRF3 through MyD88 or TRIF-dependent pathways. The activation of NF-${\kappa}B$ leads to the induction of inflammatory gene products including cytokines and cyclooxygenase-2 (COX-2). This study was carried out to investigate the anti-inflammatory effects of oak wood vinegar. Oak wood vinegar inhibits the NF-${\kappa}B$ activation and COX-2 expression induced by LPS. These results provide new ideas to understand the mechanism of oak wood vinegar for its anti-bacterial and anti-inflammatory activities.

Ginsenoside compound K inhibits nuclear factor-kappa B by targeting Annexin A2

  • Background: Ginsenoside compound K(C-K), a major metabolite of ginsenoside, exhibits anticancer activity in various cancer cells and animal models. A cell signaling study has shown that C-K inhibited nuclear factor-kappa B ($NF-{\kappa}B$) pathway in human astroglial cells and liver cancer cells. However, the molecular targets of C-K and the initiating events were not elucidated. Methods: Interaction between C-K and Annexin A2 was determined by molecular docking and thermal shift assay. HepG2 cells were treated with C-K, followed by a luciferase reporter assay for $NF-{\kappa}B$, immunofluorescence imaging for the subcellular localization of Annexin A2 and $NF-{\kappa}B$ p50 subunit, coimmunoprecipitation of Annexin A2 and $NF-{\kappa}B$ p50 subunit, and both cell viability assay and plate clone formation assay to determine the cell viability. Results: Both molecular docking and thermal shift assay positively confirmed the interaction between Annexin A2 and C-K. This interaction prevented the interaction between Annexin A2 and $NF-{\kappa}B$ p50 subunit and their nuclear colocalization, which attenuated the activation of $NF-{\kappa}B$ and the expression of its downstream genes, followed by the activation of caspase 9 and 3. In addition, the overexpression of Annexin A2-K320A, a C-K binding-deficient mutant of Annexin A2, rendered cells to resist C-K treatment, indicating that C-K exerts its cytotoxic activity mainly by targeting Annexin A2. Conclusion: This study for the first time revealed a cellular target of C-K and the molecular mechanism for its anticancer activity.

Naringenin이 NF-$\kappa$B, AP-1 억제를 통한 MMP-9 활성 및 발현 억제 효과 (Inhibitory Effect of Naringenin on MMP-9 Activity and Expression in HT-1080 Cells)

  • Naringenin은 flavonoid구조의 감귤류 과피에 다량 함유되어 있으며 항암 및 항산화 등의 다양한 생리활성을 가지는 것으로 보고되었다. 이에 본 연구에서는 HT-1080 섬유육종세포의 전이에 대한 영향을 조사하였다. 먼저 Naringenin이 암세포의 이동에 미치는 영향을 알아보기 위해 migrationassay를 한 결과, Naringenin이 암세포의 이동을 농도 의존적으로 억제시켰다. 암의 전이에 있어서 매우 중요한 역할을 하는 단백질분해효소인 matrix metalloproteinase-9 (MMP-9)의 활성도를 측정하기 위해 gelatin zymography를 한 결과, Naringenin이 PMA에 의해 증가된 MMP-9의 효소 활성도를 농도 의존적으로 감소시켰다. 또한 MMP-9와 TIMP-1의 유전자 발현에 대한 Naringenin의 영향을 RT-PCR방법으로 조사한 결과, Naringenin이 PMA에 의해 증가된 MMP-9의 mRNA발현을 농도 의존적으로 감소시켰으나 TIMP-1의 mRNA발현에는 변화가 없었다. MMP-9발현 감소에 관여하는 전사조절인자를 확인하기 위해 promoterassay를 한 결과, Naringenin이 PMA에 의해 증가된 MMP-9 및 NF-$\kappa$B, AP-1의 Promoter활성을 농도 의존적으로 감소시 켰다. 또한 MMP-9의 전자조절인자인 NF-$\kappa$B와 AP-1의 결합 활성도를 electrophoretic mobility shift assay로 확인한 결과 Naringenin이 PMA에 의해 증가된 NF-$\kappa$B와 AP-1의 결합 활성도를 농도 의존적으로 감소시켰다. 결론적으로 Naringenin이 전사조절인자인 NF-$\kappa$B와 AP-1의 활성을 억제함으로써 MMP-9의 유전자 발현 및 효소 활성을 억제하고 그 결과 암세포의 이동과 침윤을 억제하는 것을 알 수 있다.

LPS로 활성화된 복강 대식세포에서 신이 추출물의 염증성 사이토카인 및 NO 억제 효과 (Water Extract of Flowers of Magnolia Denudata Inhibits LPS-induced Nitric Oxide and Pro-inflammatory Cytokines Production in Murine Peritoneal Macrophage by Inhibiting $NF-{\kappa}B$ Activation)

  • Flowers of Magnolia denudata has been reported to possess a variety of pharmacological activities. In this study, we investigated the anti-inflammatory effects and mechanism of the water extract of Flowers of Magnolia denudata(MD) in lipopolysacchride (LPS)-mediated inflammatory mediators in murine peritoneal macrophages. MD itself does not have any toxic effects in murine peritoneal macrophages. MD inhibits LPS-induced nitric oxide (NO), tumor necrosis factor $(TNF)-{\alpha}$, IL-6 and IL-12 production in murine peritoneal macrophages. Furthermore, we have found that MD inhibited LPS-induced $NF-{\kappa}B$ but not c-Jun N-terminal kinase (JNK), p38 and extracellular signal-ragulated kinase (ERK) activation. These results suggested that MD inhibit LPS-induced production of $TNF-{\alpha}$, IL-6 and IL-12 via suppression of the $NF-{\kappa}B$ activation.

Propenone 유도체의 $NF-{\kappa}B$ 활성 억제 및 IL-8 유도에 의한 단핵구의 장 상피세포 부착 억제 효과 (Inhibitory Effects of Propenone Derivatives on $NF-{\kappa}B$ activity and IL-8-Induced Monocyte Adhesion to Colon Epithelial Cells)

  • In this study, we examined the inhibitory effects of propenone derivatives, 1,3-diphenyl-propenone (DPhP), 3-phenyl-1-thiophen-2-yl-propenone (PhT2P), 3-phenyl-1-thiophen-3-yl-propenone (PhT3P) and 1-furan-2-yl-3-phenyl-propenone (FPhP), on $TNF-{\alpha}$-induced nuclear factor (NF)-${\kappa}B$ activity and interleukin (IL)-8-induced monocyte adhesion to colon epithelial cells. 1-Furan-2-yl-3-pyridin-2-yl-propenone (FPP-3) that is previously reported as a $NF-{\kappa}B$ inhibitor suppressed $TNF-{\alpha}$-induced monocyte-epithelial cell adhesion in a concentration-dependent manner. The propenone derivatives, DPhP, PhT2P, PhT3P, FPhP, also inhibited $TNF-{\alpha}$-induced $NF-{\kappa}B$ activation in a similar degree to FPP-3. In a DPPH radical scavenging assay, none of the compounds showed DPPH radical scavenging activity, indicating that the inhibitory actions of the propenone derivatives on redox-sensitive $NF-{\kappa}B$ activity is not due to a simple free radical scavenging activity. In addition, the propenone derivatives also suppressed the IL-8-induced monocyte adhesion to colon epithelial cells. Furthermore, the effective concentrations of the propenone derivatives on both $NF-{\kappa}B$ activation as well as IL-8 induced monocyte-epithelial cell adhesion were 1000 times lower than 5-aminosalicylic acid (5-ASA), a clinically used drug for inflammatory bowel disease. These results suggest that the propenone derivatives may be a potential lead having a strong inhibitory activity against inflammatory cytokine-induced epithelial inflammation.

Sulforaphane Inhibits Ultraviolet B-induced Matrix Metalloproteinase Expression in Human Dermal Fibroblasts

  • Sulforaphane [1-isothiocyanato-4-(methylsulfinyl)-butane] is one of the most abundant isothiocyanates in some cruciferous vegetables, especially broccoli. Sulforaphaene has been shown to exhibit many pharmacological activities, including anti-oxidant, anti-inflammatory and anti-microbial activities. However, the anti-skin photoaging effects of sulforaphane have not yet been reported. In the present study, we investigated the inhibitory effects of sulforaphane on MMP-1 and -3 expressions of the human dermal fibroblasts via various in vitro experiments and elucidated the pathways of inhibition. Western blot analysis and real-time PCR revealed sulfiraphane inhibited UVB-induced MMP-1 and -3 expressions in a dose-dependent manner. UVB strongly activated nuclear factor-${\kappa}B$ (NF-${\kappa}B$) activity, which was determined by NF-${\kappa}B$ DNA binding activity. UVB-induced NF-${\kappa}B$ activation and MMP expression were completely blocked by sulforphane. These findings suggest that sulforaphane could prevent UVB-induced MMPs expressions through inhibition of NF-${\kappa}B$ activation.

  • 선천성 면역 반응을 위해 중요한 역할을 하는 TLRs가 외부 병원성 물질에 자극을 받게 되면 NF-${\kappa}$B를 활성화시키며, 그 결과로 염증을 유도하는 COX와 같은 유전자를 발현한다. 이번 연구에서, 옛날부터 지금까지 전통적인 약재로써 질병 치료에 다양하게 쓰이고 있는 감초의 뿌리에서 추출한 성분 중의 하나인 ILG가 NF-${\kappa}$B활성과 COX 발현을 어떻게 조절하여 항염증 효과를 가지고 있는지 알아보았다. ILG는 TLR agonists인 MALP-2, Poly[I:C], 그리고 LPS에 의해 유도된 NF-${\kappa}$B 활성화와 COX-2 발현을 억제시켰다. 또한 ILG는 리간드(ligand)에 독립적인 TLRs signaling downstream molecules인 MyD88, IKK ${\beta}$, 그리고 p65에 의해서 유도된 NF-${\kappa}$B 활성을 억제시켰다. 이러한 결과는 한약재로서 많이 이용되는 감초가 단지 한약의 쓴맛을 줄이기 위함이 아니라 TLRs 신호전달 체계를 조절하여 항염증 효과를 가지고 있다는 것을 보여주는 것이라 할 수 있겠다.

Nitric Oxide Prevents the Bovine Cerebral Endothelial Cell Death Induced by Serum-Deprivation

  • Endothelial cells play a central role in the inflammatory processes, and activation of nuclear factor kappa B ($NF-_{\kappa}B$) is a key component in that inflammatory processes. Previously, we reported that tumor necrosis factor alpha($TNF{\alpha}$) had protective effect of cell death induced by serum deprivation and this protection was related to $NF-_{\kappa}B$ activation. Inducible nitric oxide synthase (iNOS) is a member of the molecules which transcription is regulated mainly by $NF-_{\kappa}B$. And the role of nitric oxide (NO) generated by iNOS on cell viability is still controversial. To elucidate the mechanism of $TNF{\alpha}$ and $NF-_{\kappa}B$ activation on cell death protection, we investigate the effect of NO on the cell death induced by serum- deprivation in bovine cerebral endothelial cells in this study. Addition of $TNF{\alpha}$, which are inducer of iNOS, prevented serum-deprivation induced cell death. Increased expression of iNOS was confirmed indirectly by nitrite measurement. When selective iNOS inhibitors were treated, the protective effect of $TNF{\alpha}$ on cell death was partially blocked, suggesting that iNOS expression was involved in controlling cell death. Exogenously added NO substrate (L-arginine) and NO donors (sodium nitroprusside and S-nitroso-N-acetylpenicillamine) also inhibited the cell death induced by serum deprivation. These results suggest that NO has protective effect on bovine cerebral endothelial cell death induced by serum-deprivation and that iNOS is one of the possible target molecules by which $NF-_{\kappa}B$ exerts its cytoprotective effect.
