Occurrence of Nitrosamines in Nakdong River Basin (낙동강 수계에서의 Nitrosamines 검출 현황)

  • The survey of nitrosamine occurrence at Nakdong river is conducted in this study. According to the study results, six nitrosamine compounds (NDEA as N-nitrosodiethylamine, NDPA as N-Nitrosodi-n-propylamine, NDMA as N-nitrosodimethylamine, NMEA as N-nitrosomethylethylamine, NDBA as N-nitrosodi-n-butylamine, and NDPHA as N-Nitrosodiphenylamine) were detected at the Nakdong river. Among these, NDEA and NDPA are the most important compounds in terms of the nitrosamine contamination of Nakdong river. The detected concentration of NDEA exceeded the CDHCS (California Department of Health Care Services) response level of 100 ng/L at several sites. The detected concentration of NDPA approached the response level (500 ng/L) at few sites. When all nitrosamine concentrations were summed up, the maximum concentration of 735.7 ng/L was detected at the Nakdong river.

The Formation of N-nitrosamine in yellow Corvenia During its Processing (굴비 가공중 N-nitrosamine의 생성)

  • Gulbi, salted and dried yellow corvenia, is a favorite diet food in Korea from the ancient times. A Few studies have dealt with sanitary concerns related to its products, while a number of investigators studied its taste compounds. This study attemps to establish the basic data for ensuring the safety of Gulbi. The contents of nitrate and nitrite were detected 1.2 and <0.1 mg/kg in raw sample, but their contents were detected 3.6~3.9 and 1.2~2.0 mg/kg during its processing and storage of Gulbi, respectively. TMAO contents decreased while betaine, TMA, DMA and formaldehyde increased in yellow corvenia during its processing and storage. Recovery from raw, salted, salt-dried and stored sample spiked with 10$\mu\textrm{g}$/kg for NDBA was 83.2~102.7% average 92.7%). N-nitrosamine such as NDMA, NDEA and NDPA was not detected in raw sample and drastically increased during Gulbi processing and storage. The levels of NDMA, NDEA and NDPA in Gulbi stored for 30 days were about 2.7,4.5 and 5.3 times higher than those in salted sample, respectively. Regardless of cooking methods, NDMA, NDEA and NDPA during cooking when sample were cooked using direct heating methods such as a gas and a briquet fire than when sample were cooked using indirect methods such as an microwave oven and a fried pan. Indirect cooking methods was effective to minimize the N-nitrosamine formation such as NDMA, NDEA and NDPA during cooking of Gulbi.

  • In 7 kinds of Gulbi purchansed from Yosu, Sunchon and Chinju, N-nitrosamine (NA) such as N-nitrosodimethylamine (NDMA), N-nitrosodiethylamine (NDEA) and N-nitrosodipropylamine(NDPA), were detected and their content was 2.8∼78.3$\mu\textrm{g}$/kg, trace and 0∼1.4$\mu\textrm{g}$/kg, respectively. This experiment was conducted to study the effect of processing conditions on NA formation in the salted and dried yellow corvenia(Guibi), prepared by using the different salting method like dry and brine salting by pure and crude salt. NDMA, NDEA and NDPA were not detected in raw sample, but NDMA content increased in yellow corvenia during its salting (27.6∼37.4$\mu\textrm{g}$/kg), and then drastically decreased in Gulbi stored for 40 days, ranging from 2.8 to 4.3$\mu\textrm{g}$/kg. Content of above NA was inhibited more effectively in the samples prepare with brine salting method than with the dry salting method during its processing and storage. Especially, NA in the sample prepared with brine salted and dried yellow corvenia using the pure salt was detected in the lower concentrations, and that of NDMA, NDEA and NDPA was 2.8∼27.6, trance and ND-2.7$\mu\textrm{g}$/kg, respectively.

Investigating of Nitrosamines in Small tributary rivers, Sewage Tretment Plants and Drinking Water Treatment Plants (하천수 및 상하수도처리공정에서의 니트로사민류 조사)

  • This study was investigated nine nitrosamines in small tributary rivers, sewage treatment plants (STPs) and drinking water treatment plants. They are N-nitrosodimethylamine (NDMA), N-nitrosomethylethylamine (NMEA), N-nitrosodiethylamine (NDEA), N-nitrosopyrrolidine (NPYR), N-nitrosodi-n-propylamine (NDPA), N-nitrosomorpholine (NMOR), N-nitrosopiperidine (NPIP), N-nitrosodi-n-butylamine (NDBA) and N-nitrosodiphenylamine (NDPHA). The nine nitrosamines were analyzed by gas chromatography mass spectrometry (GC/MS) using solid phase extraction (SPE) with a coconut charcoal cartridge. Among the nine nitrosamines, NDMA, NMEA, NDEA, NDPA NDBA and NDPHA were detected in small tributary rivers and sewage tretment plants. In small tributary rivers, NDMA, NMEA, NDEA, NDPA, NDBA and NDPHA were obtained as ND~16.4 ng/L, ND~17.7 ng/L, ND~102.4 ng/L, ND~455.4 ng/L, ND~330.1 ng/L and ND~161.0 ng/L, respectively. Also NDMA, NMEA, NDEA, NDPA and NDBA were investigated ND~821.4 ng/L, 22.5~55.4 ng/L, 53.2~588.5 ng/L, ND~56.6 ng/L and ND~527.9 ng/L in STPs, respectively. In drinking water treatment plants, NMEA and NDEA concentration were increased to as high as 38.8 ng/L after ozonation process. However nitrosamines were decreased subsequent biological activated carbon (BAC) treatment process. It was supposed that nitrosamines were formed by $O_3$ oxidation and were removed by biodegradation of BAC.

Analysis Method of N-Nitrosamines in Human Urine by LC-MS/MS System (LC-MS/MS 시스템을 이용한 소변 중 N-니트로사민류 분석법 확립)

  • N-nitrosamines are the nitroso compounds which are produced by nitrosation reactions of the secondary amine and nitrite under acidic conditions. Approximately 300 species of N-nitrosamine have been tested for carcinogenicity in laboratory experiments, with 90% of them demonstrated carcinogenic effects different animal species, including higher primates. In 1978, IARC classified NDMA and NDEA as Group 2A, and NDPA, NDBA, NPIP, NPYR and NMOR as Group 2B. In this study, we established pretreatment and analytical method for N-nitrosamines (NDMA, NDEA, NMEA, NDPA, NDBA, NPIP, NPYR and NMOR) in human urine for biological monitoring of N-nitrosamines. The analytes were extracted using solid phase extraction (SPE), then quantitative analysis was performed by LC-(APCI)-MS/MS. The accuracies of the established method were between 85.8~108.7% and precisions were lower than 20%. The limit of detection (LOD) were between 0.0002 (NDBA) and 0.0793 (NDMA) ng/ml. The linearity obtained was satisfying for the 8 N-nitrosamines, with a coefficient of determination ($r^2$) higher than 0.999. The mean concentrations of N-nitrosamines in the urine were 2.645 mg/g creatinine for NDMA, 0.067 mg/g creatinine for NDEA, 0.009 mg/g creatinine for NMEA, 0.011 mg/g creatinine for NDBA, 0.271 mg/g creatinine for NPIP and 0.413 mg/g creatinine for NPYR. NDPA and NMOR were not detected. It can be used as a instrumental methodology for evaluation and risk assessment of human exposure to N-nitrosamines for the further research.

Accelerated Aging Characteristics of a Double Base Propellant (복기추진제의 가속 노화 특성 연구)

  • Accelerated aging test for the double base propellant was carried out at three different temperatures (60, 70, and $75^{\circ}C$) for over a year. To evaluate the aging characteristics of the double base propellant, the stabilizer contents and thermal decomposition kinetics were analyzed by using high performance liquid chromatography (HPLC) and AKTS-Thermokinetics software. As a result, stabilizer contents in the double base propellant gradually decreased according to the aging temperature and aging duration. The consumption rate of 2-NDPA in the accelerated aged propellants showed that it was two times faster at $75^{\circ}C$ in compared with ther rate at $70^{\circ}C$. These experimental values were simulated by the SB kinetic model, and it was shown that the two-step model with constant reaction orders n1=1 and n2=0 best describes the process of the stabilizer depletion for the double base propellant.

  • ABSTRACT : The aging characteristics of NEPE propellant is studied by HPLC analysis, cube crack test, swelling, and mechanical property test. The depletion of mixed stabilizer NMAI2-NDPA is analysed by Berthelot's law. The stabilizer NMA was completely comsumed after 16 weeks at $70{\circ}$ and after 6 weeks at $80{\circ}$ and the cracks developed in the sample. The aging trend of the propellant was shown hardening phenomena in the range of 20-$60{\circ}$ due to evaporation of energetic plasticizers.

Studies on N-Nitrosamine of Korean Ordinary Soysauce (한국 재래식 간장의 니트로소화합물에 관한 연구)

  • In recent years, the presence of N-nitrosamine, which was produced by the interaction of nitrite and secondary amine, in the fermented foods has been the subject of considerable interest and controversy. In this experiment, the levels of N-nitrosamine such as N-nitrosodimethylamine(NDMA), N-nitrosodiethylamine(NDEA) and N-nitrosodipropylamine(NDPA) in the Korean ordinary soysauce, which were added with ascorbic acid, sorbic acid, and sodium benzoate in the making of it were analyzed by low resolution mass spectrometry, and then the changes of dimethylamine(DMA), nitrate and nitrite nitrogen during the fermentation of it were observed. The contents of DMA nitrogen increased during the fermentation of Korean ordinary soysauce, continuously, but those of DMA nitrogen in the soysauce which had been added with ascorbic acid were inhibited, considerably, until the fermentation of 70days. The levels of nitrate nitrogen during the fermentation of Korean ordinary soysauee decreased, while those of nitrite nitrogen increased. The soysauce which had been incoporated with ascorbic acid in the making of it showed low amounts of nitrite. The concentration of NDMA in the control sample were 2.7 and $8.5{\mu}g/kg$ after the fermentation of 30 and 60 days, respectively, those of NDMA increased during the fermentation of Korean ordinary soysauce, but NDEA and NDPA in all of the soysauce were not detected. The samples were treated with ascorbic acid, sorbic acid, and sodium benzoate in the making of Korean ordinary soysauce were turn out to be effective in preventing the formation of NDMA. Inhibitive actions from food additives as above were, respectively, $82.2{\sim}87.0%$(ascorbic acid), $25.9{\sim}65.4%$(sorbic acid) an $13.2{\sim}63.5%$ (sodium benzoate) in comparison with control sample during the fermentation of Korean ordinary soysauce. NDMA contents were detected below $1.5{\mu}g/kg$ in the soysauce, which food additives were mixed to the pure NaCI in the brewing of it. Free amino acids such as glutamic acid, proline, and histidine were proved to be inhibiting the formation of NDMA during the fermentation of Korean ordinary soysauce. This might be due to the reaction above amino acids and nitrite by Van Slyke reaction.

Adsorption Characteristics for Nitrosamines in Granular Activated Carbon Process (입상활성탄 공정에서의 nitrosamine류 흡착 특성)

  • This study accessed the adsorption characteristics of the nine nitrosamine species on coal-based granular activated carbon (GAC). The breakthrough appeared first for NDMA and sequentially for NMOR, NPYR, NMEA, NDPA, NDEA, and NPIP. On the other hand, NDBA and NDPHA were not detected in the treated effluent for the operation period. The maximum adsorption capacity (X/M) for the seven nitrosamine species with apparent breakthrough points ranged from $27.5{\mu}g/g$ (for NDMA) to $671.0{\mu}g/g$ (for NPIP). Carbon usage rate (CUR) for NDMA was 1.07 g/day, 13.4 times higher than that for NPIP (0.08 g/day). The X/M values for the seven nitrosamine species were fitted well with a linear regression ($r^2$ = 0.94) by their octanol-water partitioning coefficient ($K_{ow}$).

Determination of N-nitrosamines in Water by Gas Chromatography Coupled with Electron Impact Ionization Tandem Mass Spectrometry (EI-GC/MS/MS를 이용한 니트로사민류의 수질분석)

  • This study assessed analysis of N-nitrosamines by separation, identification, and quantification using a gas chromatography (GC) mass spectrometer (MS) with electron impact (EI) mode. Samples were pretreated by a automated solid phase extraction (SPE) and a nitrogen concentration technique to detect low concentration ranges. The analysis results by EI-GC/MS (SIM) and EI-GC/MS/MS (MRM) on standard samples with no pretreatment exhibited similar results. On the other hand, the analysis of pretreated samples at low concentrations (i.e. ng/L levels) were not reliable with a EI-GC/MS due to the interferences from impurity peaks. The method detection limits of eight (8) N-nitrosamines by EI-GC/MS/MS analysis ranged from 0.76 to 2.09 ng/L, and the limits of quantification ranged from 2.41 to 6.65 ng/L. The precision and accuracy of the method were evaluated using spiked samples at concentrations of 10, 20 and 100 ng/L. The precision were 1.2~13.6%, and the accuracy were 80.4~121.8%. The $R^2$ of the calibration curves were greater than 0.999. The recovery rates for various environmental samples were evaluated with a surrogate material (NDPA-$d_{14}$) and ranged 86.2~122.3%. Thus, this method can be used to determine low (ng/L) levels of N-nitrosamines in water samples.
