$\gamma$-PGA는 우리 전통 콩 발효식품인 청국장의 끈적끈적한 점액성 성분의 하나로, 매우 다양한 기능을 가지고 있는 천연물질이다. 이러한 $\gamma$-PGA의 아토피발진 억제 효과를 알아보고자 NC/Nga 생쥐를 사용하여 in vivo 실험을 하였다. BMAC로 유도된 NC/Nga 아토피 피부발진 생쥐에 분자량 300 kDa인 $\gamma$-PGA(PGA-HM)와 이를 저분자화 시킨 저분자 $\gamma$-PGA(PGA-LM)을 경구 투여한 결과 PGA-LM 투여군에서 clinical skin severity score가 유의성 있게 감소하였다. 혈청 IgE 수준은 PGA-LM이 대조군에 비하여 유의적으로 감소하였고, 혈청 IgG1 수준은 대조군에 비하여 감소하였으나 두 군 모두 유의성이 없었다. $CD4^+CD25^+foxp3^+$ Treg 세포가 유도되는 것을 확인하기 위하여, PGA-HM와 PGA-LM를 투여한 NC/Nga 아토피 피부발진 생쥐의 등 부위에서 mRNA를 분리히여 real-time PCR로 foxp3 mRNA 유전자 발현량을 측정한 결과는 대조군에 비하여 PGA-LM 투여군이 약 2배 이상 증가를 나타내었다. 또한 등피부 조직의 조직검사에서도 epidermis의 두께, 비만세포 침윤, 그리고 $CCR3^+$ 세포수 등이 대조군에 비하여 현저히 억제됨을 관찰할 수 있었다. 이상의 결과로 PGA-HM보다 PGA-LM이 BMAC로 아토피 피부염이 유발된 NC/Nga 생쥐에 $CD4^+CD25^+foxp3^+$ Treg 세포를 활성화하여 IgE 및 염증 사이토카인의 생산 및 $CCR3^+$ 호산구의 활성화가 억제되어 아토피조절 효과를 나타내는 것으로 사료되었다.
Objectives : HRHDT has been known as a useful prescription with antibiotic, anti-inflammatory, antioxidative and immunosuppressive activity. To evaluate anti-atopic dermatitis effect of HRHDT, we treated HRHDT in NC/Nga mice model skin induced contact hypersensitivity. Methods : Contact hypersensitivity, a local inflammatory response of skin, was induced by spreading the back skin of NC/Nga mice with 0.4${\sim}$1% DNCB. HRHDT was prepared by dissolving 3% HRHDT in solution and treated 3 weeks on the back skin. Results : HRHDT significantly reduced TEWL and erythema by 0.4${\sim}$1% of DNCB treatment compared to control group. HRHDT also reduced IgE on serum obtained from blood of DNCB-treated mice. Conclusions : These results showed that HRHDT could be used as a pharmaceutical material with anti-atopic dermatitis effect by reducing IgE in contact, hypersensitivity atopic dermatitis NC/Nga mice model by DNCB.
Purpose : To investigate the effects of the lavender, lemon and eucalyptus oil mixture on the atopy dermatitis skin lesions induced on NC/Nga Mice by dinitrochlorobenzene (DNCB). Material and Method : For this purpose, we fabricated the oil mixture blending three essential oils (lavender, lemon, eucalyptus : ELL) with one carrier oil (jojoba) and apply it on the atopic dermatitis skin lesions of NC/Nga Mice. Atopic dermatitis in NC/Nga Mice was induced by DNCB treatment on the dorsal skin of mice for 8 weeks. The mixture of ratio of each essential oil drop was 1 (eucalyptus) : 2 (lemon) : 2 (lavender) and this mixture was blended with jojoba oil 50ml (0.025%). The ELL-ointment was supplied for 8 weeks. We evaluated the effects of ELL on cell viability of mouse lung fibroblast, clinical skin features and severity, the level of serum Immunoglobulin (Ig) E & Ig G1, Interleukin (IL)-4, IL-13 and Interferon (IFN)-$\gamma$. Results : ELL showed safety on the cell viability of mouse lung fibroblast compared with control group. The cell viability was measured by SRB method. The effects of ELL on clinical skin features and severity in DNCB-induced dermatitis model of NC/Nga mice was significant compared with control group. EEL also showed significant effects on clinical symptom score compared with control group. Serum IgE & IgG1 level and development of atopy dermatitis skin lesions were evaluated. Serum IgE & IgG1 production was significantly down-regulated in EEL group compared with control group. ELL also down-regulated the levels of IL-4 and IL-13, and up-regulated the level of IFN-$\gamma$ compared with control group significantly. Conclusion : ELL was effective on atopy dermatitis by modulating Th2 related factors.
아토피 피부염에서 유산균의 유익성에 대해서는 이미 많이 알려져 있고 우리나라 전통식품인 김치유산균의 유익성에 대한 연구도 많이 진행되었으나 아토피 피부염에서 김치 유산균의 효과에 관한 연구는 아직 미비한 실정이다. 본 연구에서는 아토피 동물 모델인 NC/Nga mice에 김치유산균에서 추출한 유산균(L.plantarum K8)의 파쇄물과 유산균 및 ${\gamma}$-리놀렌산 복합체(L.rhamnosus GG 생균, B. lactis Bb-12Lb 생균, L. plantarum K8생균, L. plantarum K8 파쇄물, ${\gamma}$-리놀렌산)를 경구 투여한 후 혈청 면역지표(IgE, IL-4, IL-5, IFN-${\gamma}$)와 비장 배양액에서 IgE 농도를 측정하였다. 실험동물의 체중 및 사료섭취량은 사육기간이 지남에 따라 증가하는 경향을 보였으며 각 군 간에 유의적인 차이는 관찰되지 않았다. 실험 시작 시 혈청 IgE 농도는 각 군 간에 차이가 나타나지 않았으나 실험 전 기간 4주 동안 유산균 파쇄물(DPF), 유산균 및 ${\gamma}$-리놀렌산 복합체(DPOC), 항히스타민(DAH)을 섭취한 군에서 증류수(DDW)만을 섭취군에 비하여 유의적으로 낮게 나타났다. 혈청 IFN-${\gamma}$ 농도는 각 군에서 유의적인 차이가 나타나지 않았으나 혈청 IL-4 농도는 유산균 및 ${\gamma}$-리놀렌산 복합체를 섭취한 군(DPOC)에서 대조군에 비하여 유의적으로 낮게 나타났다. 혈청 IL-5 농도는 유산균 파쇄물(DPF), 유산균 및 ${\gamma}$-리놀렌산 복합체(DPOC), 항히스타민(DAH)을 섭취한 군에서 모두 대조군에 비하여 유의적으로 낮았다. 비장 임파구 배양 상층액의 IgE 농도는 각 군에서 군 간 유의적인 차이가 나타나지 않았다. 결론적으로, 아토피 동물모델 NC/Nga mice에 유산균 파쇄물(DPF)과 유산균 및 감마리놀렌산 복합체(DPOC)의 경구투여는 혈청 IgE, IL-4, IL-5를 감소시키는 것으로 나타났으며, 김치로부터 추출한 프로바이오틱스를 아토피 환자에게 섭취시켰을 때 IgE, IL-4, IL-5 감소와 같은 긍정적인 효과를 볼 수 있을 것으로 기대한다. 여러 종류의 프로바이오틱스는 아토피의 증상완화 또는

Objective : This Experimental study was done to investigate the Effect of Taklisodok-um and Hwangryunhaedok-tang(TH) on Atopic Dermatitis. Methods : We assessed effects of TH on the IgE, IL-4, IL-5, IL-6, IgM, IgG1, IFN-${\gamma}$ in vivo, on the IL-4, IL-5, CCR3 in the skin tissues of ear and dorsum with NC/Nga mice. And we assessed effects of TH on the COX-2, IL-$1{\beta}$, TNF-${\alpha}$, IL-6 with RAW 264.7 cell. Results and Conclusion : 1. IgE, IL-4, IL-5, IL-6, IgM, IgGl levels in the serum of TH treated NC/Nga mouse group were decreased compared to the untreated control mice. IFN-r showed a increase in the experimental group compared to the untreated control group. The spleen weight of TH treated NC/Nga mice was decreased compared to the untreated control group. 2. mRNA expression levels of IL-4, IL-5 and CCR3 in the skin tissues of TH treated NC/Nga mice were decreased, and expression levels of IL-6 in the skin tissues of TH treated NC/Nga mice were decreased compared to the untreated control group. IFN-${\gamma}$ mRNA expression levels were increased compared to the untreated control group. 3. Judged from that IL-$1{\beta}$, TNF-${\alpha}$, IL-6 express of gene, effect of inflammatory Cytokines revelation were decreased compared to the untreated control group. 4. Depend on the strength of TH, inflammatory RAW 264.7 in the serum of TH were inhibited compared to the untreated control serum that leaded a COX-2 activity model. 5. Histological observation of the ear and skin tissues showed that the extents of inflammation and infiltrated immune cells in the epidermis and dermis of TH treated NC/Nga mice were highly reduced compared to the untreated control group.
본 실험에서는 CGT의 항염증과 관련한 다양한 실험을 실시하였다. In-vitro 실험에서 MTT, NO, ROS와 같은 염증 매개 물질 실험은 RAW 264.7 세포를 이용하여 실시하였다. In-vivo 실험은 아토피피부염 동물 모델인 NC/Nga 생쥐에서 항염증과 관련한 인자 및 조직학적 변화 등을 관찰하였다. CGT는 RAW 264.7 세포에서 100% 이상의 생존율을 나타냈으며, LPS를 이용한 NO와 ROS의 검사에서도 효과를 나타내었다. CGT 처치 그룹은 혈청 내 IL-$1{\beta}$와 IL-6, TNF-흰 에서 53%, 43%, 57%의 유의성 있는 감소를 나타내었고, IgE 역시 56%의 감소를 보였다. 또한, 피부에 지방세포의 침윤을 억제하고, 표피 및 진피의 두께 역시 감소하였다. 그 결과 CGT는 NC/Nga 생쥐에서 항염증에 효과를 나타내었다. 따라서 CGT는 아토피피부염과 염증 치료에 좋은 효과가 있다고 제안하는 바이다.
Objective : This study was carried out to compare Saengmaeksan-gamibang(SG) to Saengmaeksan(SM) on the efficacy of moisturization, skin whitening, anti-inflammation, and anti-allergy in atopic dematitis using NC/Nga mice model. Methods : We assessed the effects of SG and SM on tyrosinase, filaggrin, serine-palmitoyl transferase(SPT), COX-2, AP-1 in vitro, on IgE, IL-4, IL-5, IL-6, IgM, IgG1, IFN-${\gamma}$ in vivo with NC/Nga mice. Results : 1. SG increased the tyrosinase inhibition activity and the levels of filaggrin and SPT, decreasing the level of COX-2. 2. On the other hand, SM decreased the tyrosinase inhibition activity and the levels of filaggrin and SPT, increasing the level of COX-2. 3. Serum IgE, IL-4, 5, 6, IgM, and IgG1 were decreased in both SG group and SM group compared to the control group. The decrease degree in these factors was higher in SG than in SM. 4. Serum IFN-${\gamma}$ was increased in both SG group and SM group compared to the control group. The increase degree in IFN-${\gamma}$ was higher in SG than in SM. Conclusion : As the result of the above experiments, it was proved that SG has the moisturization, skin whitening, anti-inflammation, and anti-allergy effects, which are greater than those of SM, and provides the significant efficacy on atopic dermatitis treatment.
This study was conducted to determine if YJ-1, a novel herbal complex from a mixture of six oriental herbs (Hydnocarpi Semen, Sesami Semen, Dictamni Radicis Cortex, Momordicae Semen, Xanthii Fructus, and Sophorae Radix), has therapeutic properties for the treatment of atopic dermatitis (AD). Using AD like symptom-induced NC/Nga mice by 1-chloro 2,4-dinitrobenzene (DNCB), the effectiveness of YJ-1 on AD was evaluated. Elidel cream$^{(R)}$ (1% pimecrolimus) was used as a control. Dermal application of YJ-1 reduced major clinical signs of AD such as erythema, pruritus, lichenification, edema/escoriations, and dryness. Interestingly, YJ-1 more improved AD-related symptoms including decrease of spleen weight, IL-4, and IgE level in the serum as well as reduction of scratching counts and clinical skin severity in the NC/Nga AD mouse model. Especially, treatment of YJ-1 at 20% in NC/Nga mice more effected than Elidel cream. These results suggest that the ointment of YJ-1 may enhance the process of AD healing by alleviating allergic reaction and has potential for therapeutic reagent for the treatment of AD.
Objective : Yukmijihwang-tang is one of the important medicines for blood- deficiency and yin-deficiency. Atopic dermatitis usually shows dampness-heat pattern in its acute stage and blood-deficiency or yin-deficiency pattern in its chronic stage. Therefore, I hypothesized that Yukmijihwang-tang is effective on atopic dermatitis and investigated the effects of Yukmijihwang-tang on NC/Nga mice's atopic dermatitis induced by DNCB. Methods : The NC/Nga mice with atopic dermatitis were divided into four groups: three experimental groups and one control group. The experimental groups were respectively put on 2.5g(YM-2.5), 5g(YM-5) and 10g(YM-10) of Yukmijihwang-tang extract per their weight once a day for 10 days while the control group was fed normal saline. After 10 days, I measured TEWL(transepidermal water loss), observed scratching behaviors, conducted a skin biopsy and checked levels of Total IgE, IL-4 and $IFN-\gamma$ on NC/Nga mice. Results : 1. Yukmijihwang-tang significantly suppressed skin dryness. In particular, YM-5 and YM-10 had better skin hydration than YM-2.5. 2. Yukmijihwang-tang significantly suppressed pruritus while there was no significant difference among the experimental groups. 3. Yukmijihwang-tang retained skin structure(epidermis, dermis and subcutaneous fat). 4. Yukmijihwang-tang reduced Total IgE level while there was no significant difference between the control group and the experimental groups. 5. Yukmijihwang-tang did not reduce IL-4(Th2 cytokine) level. 6. Yukmijihwang-tang significantly reduced $IFN-\gamma$(Th1 cytokine) level. In particular, YM-2.5 and YM-5 had lower $IFN-\gamma$ level than YM-10. Conclusion : The results suggest that Yukmijihwang-tang suppresses skin dryness and pruritus, retains skin structure and is effective on chronic atopic dermatitis which is associated with Th1 cytokines in the immune response.
Atopic dermatitis (AD) is a chronic inflammatory skin disease characterized by inflammatory cell infiltration in the skin. This study was performed to assess the therapeutic effects of BHOSB on the DNCB-induced dermatitis in NC/Nga mice, characterized by the onset of AD along with an increase the number of inflammatory cells and dysregulation of inflammatory mediators including cytokines and chemokines. BHOSB administration significantly reduced clinical dermatitis severity including pruritus, edema, eczematous and erythema. Histological findings indicated that the thickening of epidermis/dermis and dermal infiltration of inflammatory cells including mast cells were dramatically reduced. The suppression of dermatitis by BHOSB was accompanied by a decrease in the number of CD11b$^+$/Gr-1$^+$ immune cells in skin but not CD3$^+$/CCR3$^+$ cells. However, the number of CD3$^+$ cells was increased in BHOSB administrated NC/Nga mice. Oral administration of BHOSB significantly reduced the level of IL-6, TNF-a and eotaxin 2 mRNA in skin. These data suggest that BHOSB may be effective therapeutic agents for the treatment of AD.
