Antioxidative Activity of Phenolic Compounds in Roasted Safflower (Carthamus tinctorius L.) Seeds

  • Antioxidative compounds contained in roasted safflower seeds were investigated. Six phenolic compounds, N-feruloylserotonin, N-(p-coumaroyl) serotonin, matairesinol, 8'-hydroxyarctigenin, acacetin 7-Ο-β-D-glucoside(tilianine) and acacetin were isolated and identified from the extract of seeds. The inhibitory effects of six phenolic compounds on 1,1-dipheny1-2-picrylhydrazyl(DPPH) radical and lipid peroxidation induced by H₂O₂/FeSO₄in rat liver microsomes were determined. Two serotonins showed more potent DPPH radical scavenging activity, and a stronger inhibitory effect on the lipid peroxidation than that of α-tocopherol. In addition, acacetin and matairesinol also considerably inhibited lipid peroxidation, while 2-hydroxy-arctigenin and tilianine were inactive. These results suggest that phenolic compounds, including serotonins, lignans and flavonoids in the roasted safflower seeds can be used as potential dietary natural antioxidants.

홍화(Carthamus tinctorius L.)씨와 발아홍화씨의 화학성분 비교 (Chemical Comparison of Germinated- and Ungerminated-Safflower(Carthamus tinctorius) Seeds)

  • 홍화씨를 이용한 골다공증 및 고지혈증 예방용 건강기능식품을 개발하기 위한 연구의 일환으로 홍화씨의 소화성, 기능성 및 기호성을 증대시킬 수 있는 방안으로 발아홍화씨를 제조하여 일반성분 및 기능성성분(지방산, tocopherols, 수용성아미노산 및 페놀화합물)의 함량을 홍화씨와 비교 분석하였다. 홍화씨는 발아하면서 조단백질과 조지방은 감소한 반면, 가용성 무질소물, 조섬유소 및 회분은 다소 증가하는 경향을 나타내었으며, linoleic 및 oleic aicds가 약 80% 이상 거의 대부분을 차지하고 있었으며, 그 외 palmitic, stearic 및 arachidic acids가 주요 지방산으로 나타났고, 발아에 따른 지방산의 조성 비율은 거의 변화가 없었다. 홍화씨와 발아홍화씨의 $\alpha$-tocopherol 함량은 각각 744.7 및 809.0 mg%로서 발아 후 64.3 mg% 증가하였으며, 홍화씨의 주된 아미노산으로 asparagine, arginine, proline, glutamic acid가 차지하고 있었으며, 발아함에 따라 그들의 함량이 크게 증가하였고, 특히 threonine, valine, leucine, isoleucine, phenylalanine, lysine 및 histidine 등의 필수아미노산 함량이 크게 증가하였다. 홍화씨에는 3종의 리그난(8'-hydroxyarctigenin 4'-O-$\beta$-D-glucoside, 8'-hydroxyarctigenin 및 matairesinol)과 4종의 세로토닌유도체[N-feruloylserotonin 5-O-$\beta$-D-glucoside, N-feruloylserotonin, N-(pcoumaroyl)-serotonin 5-O-$\beta$-D-glucoside, N-(p-coumaroyl)serotonin] 그리고 2종의 플라보노이드(acacetin 7-O-$\beta$-D-glucuronide, acacetin) 화합물이 존재하였으며, 발아함에 따라 세로토닌유도체를 함유한 모든 페놀화합물의 함량은 다소 감소하는 경향을 나타내었다. 이와 같이 홍화씨를 발아함에 따라 섬유소 및 유용 기능성성분의 증가와 더불어 설사를 유발하는 고미성분의 감소 그리고 소화성 및 기호성을 떨어뜨리는 홍화씨 껍질의 분리 및 효율성 확대를 이룰 수 있기에 향후 발아홍화씨는 항골다공증 및 항고지혈증 건강기능식품 소재로서 개발 가능성이 높다고 사료된다.

국외 수집 홍화 유전자원의 항산화 활성 및 세로토닌 유도체 함량 분석 (Analysis of Antioxidant Activity and Serotonin Derivatives in Safflower (Carthamus tinctorius L.) Germplasm Collected from Five Countries)

  • 본 연구는 이란 등 5개국에서 수집한 홍화자원을 대상으로하여 천연항산화제로써 홍화종자의 항산화 활성을 평가하고, 세로토닌 유도체인N-(p-Coumaroyl) serotonin과N-feruloylserotonin의 함량을 측정하여 항산화 활성과의 상관관계를 조사하였다. 총폴리페놀함량은 $28.25{\sim}90.53{\mu}g\;GAE/mg\;DE$의 범위를 나타내었고, 색소화합물인 DPPH 라디칼과$ABTS^{+}$을 이용한 유리라디칼소거능을 측정하는 DPPH assay와 ABTS assay의 값은 18.76 ~ 93.98 및 $48.91{\sim}163.73{\mu}g\;ASCE/mg\;DE$의 범위를 나타내었다. 시료의 환원능을 측정하는 방법인 FRAP assay과 RP assay는 3.80 ~ 132.29 및 $26.32{\sim}80.08{\mu}g\;ASCE/mg\;DE$의 범위로 나타내었다. UHPLC를 이용하여 CS와 FS를 분석한 결과, CS 함량은 2.56 ~ 64.99 mg/g DE이고, FS 함량값은 1.92 ~ 65.36 mg/g DE이었다. 5개국 중 이란 원산종은 다른 나라에 비해 세로토닌 유도체 함량과 항산화 활성의 평균값이 높게 측정되었다. 항산화 활성과 세로토닌 유도체 함량간의 상관관계 분석 결과, CS는 TPC와 ABTS, DPPH와 높은 상관성을 보였고(r = 0.673, 0.727, 0.820), FS는 DPPH와 높은 상관성을 보였다(r = 0.740). RACI를 통하여 선발된 IT321214과 IT321215는 항산화 활성이 높은 자원으로 천연항산화제 소재 탐색 및 개발에 기초자료로 활용할 수 있을 것으로 생각된다.

Preparation of High Quality Safflower (Carthamus tinctorius L.) Seed Extract by High-Pressure Extraction Process

  • Safflower seed extract was prepared by a high-pressure extraction technology and its quality characteristics were compared to that of other conventional extraction techniques, such ultrasonic and reflux extractions. Safflower seeds were extracted with 80% aqueous ethanol by three above extraction methods, and further fractionated with Diaion HP-20 column chromatography to obtain a partially purified safflower seed extract (PPSSE). Among the three extraction techniques examined, the reflux extraction showed the higher yields of EtOH extract and PPE than the ultrasonic and high-pressure extractions. Levels of most phenolic compounds in the EtOH extract of safflower seed are higher in reflux and ultrasonic extractions than the high pressure extraction, but levels of two serotonin aglycones, N-(p-coumaroyl)serotonin (CS) and N-feruloylserotonin (FS), in PPSSE were higher in the high pressure extraction than the reflux and ultrasonic extractions. In addition, color values (L and a) of the PPSSE were higher in the high-pressure extraction than the reflux and ultrasonic extractions, although there were no significant differences in pH and UV maxima absorption spectra among three extraction techniques. These results indicate that the high-pressure extraction technology is a simple and effective extraction for preparation of a high quality of safflower seed extract containing CS and FS with anti-wrinkle activity.

Study on the hypochlolesterolemic and antioxidative effects of tyramine derivatives from the root bark of Lycium chenese Miller

  • The aim of the present study was to investigate the hypocholesterolemic effect and potential of tyramine derivatives from Lycii Cortex Radicis (LCR), the root bark of lycium (Lycium chenese Miller) in reducing lipid peroxidation. The activities of enzymes, hepatic 3-hydroxy 3-methylglutaryl (HMG) CoA reductase and acyl-CoA:cholesterol acyltransferase (ACAT) and LDL oxidation were measured in vitro and animal experiments were also performed by feeding LCR extracts to rats. The test compounds employed for in vitro study were trans-N-p-coumaroyltyramine (CT) and trans-N-feruloyltyramine (FT), LCR components, N-(p-coumaroyl)serotonin (CS) and N-feruloylserotonin (FS) from safflower seeds, ferulic acid (FA) and 10-gingerol. It was observed that FT and FS at the concentration of 1.2 mg/mL inhibited liver microsomal HMG CoA reductase activity by ~40%, but no inhibition of activity was seen in the cases of CT, CS, FA and 10-gingerol. Whereas, ACAT activity was inhibited ~50% by FT and CT, 34-43% by FS and CS and ~80% by 10-gingerol at the concentration of 1 mg/mL. A significant delay in LDL oxidation was induced by CT, FT, and 10-gingerol. For the animal experiment, five groups of Sprague-Dawley male rats were fed high fat diets containing no test material (HF-control), 1 and 2% of LCR ethanol extract (LCR1 and LCR2), and 1% of extracts from safflower seed (Sat) and ginger (Gin). The results indicated that total cholesterol level was significantly lower in Saf, LCR2 and Gin groups, and HDL cholesterol level was lower only in Gin group when compared with HF-control group; while there was no difference in the serum triglyceride levels among the five experimental groups. The level of liver cholesterol was significantly lower in LCR1 and LCR2 groups than HF-control Serum levels of TBARS were significantly lower only in LCR2 group when compared with HF-control group. From the observed results, we concluded that LCR can be utilized as a hypocholesterolemic ingredient in combination with ginger, especially for functional foods.
