해양 패류 렉틴성분 연구 (V) - 반지락조개의 렉틴성분 분리정제에 관한 연구 (Studies on Lectins From Marine Shells (V) - Isolation and Purification of Letin from Tapes philippinarum.)

  • The result of the screening of lectins on 28 species of marine shell fishes(mollusks) showed that 10 species (Tapes philippinarum, Cyclosunetta menstrualis, Neptunea arthritica cumingii, Omphalius pfeifferi carpenteri, Chlorostoma argyrostoma turbinatum, Chiorostoma argyrostoma lischkei, Semisulcosira corea, Neptunea polycosta, Babylonica japonica, Noverita didyma) were present hemagglutinating properties to human A, B, and O group, and animal blood erythrocytes. A new lectin from Tapes philippinarum. was isolated and partially characterized. The lectin was purified by (NH$_4$)$_2$SO$_4$ precipitation and ion exchange chromatography on DE 53 column. Six fractions were obtained from DE 53 column by salt gradient elution but only 0.3M, and 0.4M NaCl fraction had strong lectin activity. On its 0.3M NaCl fraction, purity was identified by polyacrylamide gel electrophoresis. This lectin was inhibited by N-acetyl-D-galactosamine. It seems that two kinds of lectin react as antigen by immunochemical studies.

  • This study was to investigate the effects of maternal diabetes on the lung tissue of the fetal rat using lectin histochemistry and electron microscope technique. Maternal diabetes was induced by intraperitoneal injection of streptozotocin (75 mg/kg the body weight) into pregnant Sprague-Dawley rats on the 7th day of gestation. Fetuses of streptozotocin induced diabetic rats exhibited delayed lung maturation and reduced air space. In lectin histochemistry, the binding of Maclura pomifera (MPA) to fetal lungs from diabetic mothers was reduced, but no significant changes in the bindings of Concanavalin A (Con A), Wheat germ agglutinin (WGA), Ricinus communis I (RCA I) and Griffonia simplicifolia (GSI-$B_4$) were noted. Because the MPA has affinity to terminal N-acetyl-D-galactosamine residues constantly linked O-glycosidically to serine or threonine, the present findings may indicates that maternal diabetes interfere with the processes of O-linked glycosylation in fetal rat lung.

  • The aqueous extract of European mistletoe (Viscum album, L.) has been used in cancer therapy. The purified mistletoe lectins, main components of mistletoe, have demonstrated cytotoxic and immune-system-stimulating activities. Korean mistletoe (Viscum album L. coloratum), a subspecies of European mistletoe, has also been reported to possess anticancer and immunological activities. A galactose- and N-acetyl-D-galactosamine- specific lectin (Viscum album L. coloratum agglutinin, VCA) with Mr 60 kDa was isolated from Korean mistletoe. (omitted)

별불가사리 렉틴의 특성 및 암 세포 성장저해 효과 (Characteristics and Cancerostatic Activity of the Starfish Lectin)

  • A new lectin was partially purified from starfish,Asterina pectinifera by means of physiological saline extraction, salt fractionation, ion exchange chromatography and hy droxyapatite chromatography, and it was named APL. The biochemical properties of the APL were characterized. In addition, its effects on lymphocyte mitogenicity and cancer cell agglutinability were tested. The APL agglutinated nonspecifically human erythrocytes and rabbit blood cells. Agglutinability was decreased to 30% of control activity below pH 5 and above pH 9 and was relatively unstable at increasing temperatures above 60$^{\circ}C$. The activity was reduced by addition of two kinds of metal ions, $Ba^{2+},\;Mn^{2+}$ and chelating agent, EDTA. APL was proved to be glycoproteins containing 9% sugars. For carbohydrate specificity, it was found that the activity of APL was inhibited by D(+)-glucosamine, D(+)-galactosamine, stachyose, N-acetyl-galactosamine and methyl-${\alpha}$-D-galactopyranoside among 35 sugars tested. In amino acid composition, the contents of acidic amino acids such as aspartic acid and glutamic acid were relatively high. This result suggest that the isoelectric point would be in a lower range. APL was found that it promotes the division of human lymphocytes. APL was proved to be a potent agglutinin for cancer cells such as HeLa, L929 and L1210 cells. Significant changes on the HeLa cell surfaces affected by APL were observed under the electron microscope.

The purified mistletoe lectins, main components of mistletoe, have demonstrated cytotoxic and immune-system-stimulating activities. Korean mistletoe (Viscum album L. coloratum), a subspecies of European mistletoe, has also been reported to possess anticancer and immunological activities. A galactose- and N-acetyl-D-galactosamine-specific lectin (Viscum album L. coloratum agglutinin, VCA) with Mr 60 kDa was isolated from Korean mistletoe. Mistletoe preparations have been given subcutaneously due to the low stability of lectin in the gastrointestinal (GI) tract. In the present study, we investigated the possibility of alginate/chitosan microcapsules as a tool for oral delivery of mistletoe lectin. In addition, our strategy has been to develop a system composed of stabilizing cores (granules), which contain mistletoe lectin, extract or powder, coated by a biodegradable polymer wall. Our results indicated that successful incorporation of VCA into alginate/chitosan microcapsules has been achieved and that the alginate/chitosan microcapsule protected the VCA from degradation at acidic pH values. And coating the VCA with polyacrylic polymers, Eudragit, produced outstanding results with ideal release profiles and only minimal losses of cytotoxicity after manufacturing step. The granules prepared with extract or whole plant produced the best results due to the stability in the extract or whole plant during manufacturing process.

바지락(Tapes japonica)으로부터 분리정제된 새로운 렉틴의 생물물리학적 특성 (Purification and Biophysical Characterization of New Lectin from Baby Clam, Tapes japonica)

  • 한국산 바지락으로부터 새로운 렉틴을 아세톤파우더, 황산암모늄 침전, 친화력 크로마토그라피 및 FPLC의 이온교환 크로마토그라피법으로 분리 정제하였다. 이 렉틴은 사람의 적혈구를 비특이적으로 응집시켰으며, 생쥐와 토끼의 적혈구 및 생쥐의 복수 Sarcoma 180 세포를 응집시키지 않았고 사람의 말초혈관 임파구도 분열 촉진시키지 못하였다. 전기영동상에서 하나의 주된 띠로 나타났으며 분자량은 Biogel P-300겔 여과에서 131,000, SDS 전기영동상에서는 125,000으로 나타났다. Subunit는 33,000과 30,000의 다른 폴리펩타이드로 tetramer로 추정된다. EDTA에 의해서 활성이 저해된 바지락 렉틴은 $Ca^{++}$$Mn^{++}$에 의하여 적혈구 응집력이 회복되었다. 또한 이 렉틴은 약 4.2% 중성당을 함유한 당단백질임이 확인되었다.

겨우살이 Lectin의 정제 및 생화학적 특성 (Purification and Biochemical Characterization of Lectin from Viscum album)

  • 겨우살이가 가지고 있는 lectin의 약리작용의 가능성에 대한 연구의 일환으로, lectin을 분리 . 정제하였으며, 분자량, subunit수, 탄수화물의 조성, 단백질의 아미노산 조성, 적혈구 응집력, 탄수화물 특이성, pH 및 열 안정성등의 생화학적 특성을 연구하였다. Lectin의 분리는 0.15 M NaCl에 의한 추출, $(NH_4)_2SO_4$ 침전, sepharose 4B affinity chromatography 및 sephadex G-150 gel filtration에 의해 11.64배 정제에 0.14%의 수율을 획득하였으며, HPLC와 전기영동을 이용하여 순도를 검정하였다. Gel filtration에 의해 분석된 lectin의 분자량은 124,000 Da이였고, SDS-PAGE에 의해 분자량이 30,000과 32,000 Da인 2개의 band로 나타나서 각각 2개의 subunit를 갖는 tetramer로 확인되었다. 겨우살이 lectin의 탄수화물은 glucose, fructose, arabinose 및 xylose를 함유하고 있다. 또한 phenlylalanine, lysine, tyrosine, aspartic acid가 비교적 풍부하였고 methionine, alanine, arginine은 비교적 적었다. Lectin은 적혈구에 종 특이성을 나타내는데 겨우살이의 혈구응집반응에서는 사람혈액형간의 차이는 없었으며, 쥐에서 가장 높았고 소에서 가장 낮았으므로 동물 종간에 대한 특이성은 뚜렸하였다. Lectin은 당 특이성을 가지는데, D-galactose, D-mannose, D-lactose 및 D-raffinose에 대해 특이성을 나타내었다. 겨우살이 lectin은 pH4~7에서 안정하였으며, 5$0^{\circ}C$까지는 10~30분 열처리해도 활성이 유지되는 열 안정성 단백질이었다.
