홍화씨 추출물 유래 세로토닌 유도체의 지방전구세포 분화억제 효능에 대한 연구 (Inhibitory Effects of Serotonin Derivatives on Adipogenesis)

  • N-(p-Coumaroyl)serotonin과 N-Feruloylserotonin은 홍화씨에서 유래하는 특이적인 세로토닌유도체로 주로 식물이 병원균을 방어하기 위해서 만들어내는 hydroxycinnamic acid amides계 물질이다. 본 연구에서는 N-(p-Coumaroyl) serotonin과 N-Feruloylserotonin이 지방전구세포의 지방분화에 미치는 영향을 oil-red O염색과 triglyceride 양 측정, GPDH 활성 측정 등을 통해 알아보았고, 그 결과 두 물질 모두 유의적으로 지방세포분화를 억제함을 관찰하였다. 효능 비교물질로 세로토닌을 처리했을 때 세로토닌 자체로는 지방분화에 유의미한 효과는 관찰되지 않았다. N-(p-Coumaroyl) serotonin과 N-Feruloylserotonin은 또한 우수한 항산화능을 보여주었다. 이러한 결과를 통해, N-(p-Coumaroyl)serotonin과 N-Feruloylserotonin이 지방세포분화억제를 통한 항비만 소재로서의 가능성을 확인하였다.

  • 홍화종자의 메탄올 추출물에서 preparative HPLC($\mu$-Bondapak $C_{18}$ column($7.8{\times}300mm$) 분석을 통하여 DPPH radical 소거 활성도가 높은 두 종류의 항산화 물질(CA와 CB)을 확인하였다. 확인된 항산화물질의 대량 추출을 위하여 hexane, chloroform, ethyl acetate 그리고 butanol 등의 유기용매를 이용하였으며 이 중 ethyl acetate 분획에서 DPPH radical 소거 활성이 높은 물질이 추출되었다. Ethyl acetate 분획물을 silica gel chlomatography하여 CA와 CB를 분리하였고, $^1H$-NMR과 $^{13}C$-NMR 분석에 의하여 CA는 N-(p-Coumaroyl) serotonin, 그리고 CB는 N-feruoylserotonin으로 동정되었다. 홍화종자에서 N-(p-Coumaroyl) serotonin과 N-feruoylserotonin의 함량을 $\mu$-Bondapak $C_{18}$ column($3.9{\times}300\;mm$)으로 300nm에서 acetonitrile의 용매로 10%에서 분석을 시작하여 30분 동안 50%까지 기울기 모드로 분석한 결과 N-(p-Coumaroyl)serotonin의 함량은 4.11 mg/gDW, N-feruoylserotonin의 함량은 7.29 mg/gDW이었으며, 이 두 가지 항산화 물질은 모두 종자껍질에서만 추출되었다.

Carthamus tinctorius seeds-Taraxacum coreanum combination attenuates scopolamine-induced memory deficit through regulation of inflammatory response and cholinergic function

  • BACKGROUND/OBJECTIVES: There is growing interest in herbal medicines for managing age-related diseases, such as Alzheimer's and Parkinson's. Safflower seeds (Carthamus tinctorius L. seeds, CTS) and dandelions (Taraxacum coreanum, TC) are widely used to treat bone- or inflammation-related diseases in Oriental countries. This study investigated the protective effect of the CTS-TC combination on scopolamine (Sco)-induced memory deficits through inflammatory response and cholinergic function. Moreover, marker components such as serotonin, N-(p-coumaroyl) serotonin, N-feruloylserotonin, chlorogenic acid, and chicoric acid in the CTS-TC combination were analyzed for their potential benefits on memory function. MATERIALS/METHODS: Water extracts of CTS, TC, and the CTS-TC combination at various ratios (4:1, 1:1, and 1:4) (100 mg/kg) were orally administered to mice for 14 days. Sco (1 mg/kg) was intraperitoneally injected into the mice before each behavioral test. T-maze and novel object recognition tests were conducted to monitor behavioral changes after the treatment. Western blotting was performed to detect protein expression. In addition, the presence of 5 biomarkers, serotonin, N-(p-coumaroyl) serotonin, N-feruloylserotonin, chlorogenic acid, and chicoric acid, was analyzed using high-performance liquid chromatography (HPLC). RESULTS: Behavioral tests showed that the CTS-TC combination enhanced memory function in Sco-injected mice. Inflammation-related proteins (inducible nitric oxide synthase, cyclooxygenase-2, and glial fibrillary acidic protein) were downregulated after treatment with the CTS-TC combination. The acetylcholinesterase protein expression was also downregulated. HPLC analysis revealed that N-feruloylserotonin and chicoric acid were the predominant components, followed by N-(p-coumaroyl) serotonin, chlorogenic acid, and serotonin. CONCLUSION: These findings suggest that the CTS-TC combination protects against Sco-induced memory deficits by inhibiting inflammatory responses and cholinergic dysfunction. N-feruloylserotonin and chicoric acid, along with N-(p-coumaroyl) serotonin, chlorogenic acid, and serotonin, might be biomarkers for the CTS-TC combination, and their effects on memory protection warrant further study.

Antioxidant Properties and Quantification of Phenolic Compounds from Safflower (Carthamus tinctorius L.) Seeds

  • The antioxidant properties of twelve phenolic compounds, including matairesinol 4'-O-$\beta$-D-glucoside, 8'-hydroxyarctigenin 4'-O-$\beta$-D-glucoside, matairesinol, 8'-hydroxyarctigenin, N-feruloylserotonin 5-O-$\beta$-D-glucoside, N-(p-coumaroyl)-serotonin-5-O-$\beta$-D-glucoside, N-feruloylserotonin, N-(p-coumaroyl)serotonin, luteolin 7-O-$\beta$-D-glucoside, luteolin, acacetin 7-O-$\beta$-glucuronide, and acacetin, isolated from defatted safflower (Carthamus tinctorius L.) seeds were evaluated with regard to the DPPH, superoxide and hydroxyl radicals. Additionally, levels of phenolic compounds were determined by HPLC in two cultivars of safflower seeds. Among them, four serotonin derivatives showed potent DPPH ($IC_{50}=10.83-21.75\;{\mu}M$) and hydroxyl ($IC_{50}=75.93-374.63\;{\mu}M$) radical scavenging activities, and their activities were significantly stronger than that of ${\alpha}-tocopherol$. Four flavonoids ($IC_{50}=170.65-275.83\;{\mu}M$) and four lignans ($IC_{50}=114.22-406.10\;{\mu}M$) exhibited significant superoxide and hydroxyl radical scavenging activities, respectively, whereas these compounds contained less activity toward the DPPH and hydroxyl radicals than serotonin derivatives. The levels of serotonin derivatives, lignans and flavonoids in safflower seeds of two cultivars ranged from 49.30 to 260.40, 3.72 to 158.90, and 11.72 to 214.97 mg% (dry base), respectively. Of the two cultivars, 'Cheongsu' had somewthat higher concentrations of phenolic compounds than 'Uisan'. These results suggest that phenolic compounds in safflower seeds may playa role as protective phytochemical antioxidants against reactive oxygen-mediated pathological diseases.

Phenolic Glycosides Isolated from Safflower (Carthamus tinctorius L.) Seeds Increase the Alkaline Phosphatase (ALP) Activity of Human Osteoblast-like Cells

  • The chemical compositions of the seeds of the safflower (Carthamus tinctorius L.) plant were evaluated to determine possible compound having proliferative effects on human osteoblast cells. Three-[4,5-dimethylthiazole-2-yl]-2,5-diphenyltetrazolium bromide (MTT) test and alkaline phosphatase (ALP) activity were used to assess the effects of the isolates on the human osteoblast-like line (Saos-2). Activity guided fractionation led to the isolation of ALP activating lignin and alkaloid glycosides through the extraction of the seeds, solvent partitioning and repeated silica gel and octadecyl silica (ODS) column chromatographic separations. The data from Nuclear Magnetic Resonance (NMR), Mass (MS), and Infrared (IR) analyses enabled the determination of the chemical structure and characterization of two compounds as a tracheloside and an N-(p-coumaroyl)-serotonin mono-${\beta}$-D-glucopyranoside. These two compounds showed respectively $149.2{\pm}4.2$ and $138.9{\pm}3.5%$ ALP activity compared to the control when evaluated at a concentration of $100\;{\mu}g/mL$.

Serotonins of safflower seeds play a key role in anti-inflammatory effect in lipopolysaccharide-stimulated RAW 264.7 macrophages

  • Safflower (Carthamus tinctorius) seeds are wellknown traditional oriental medicines that have long been used for the remedies of blood stasis and bone formation in east Asia. In this study, ethyl acetate (EtOAc) was used for extraction of the main chemical compounds from C. tinctorius seeds. Four major compounds were identified, acacetin, cosmosiin, N-feruloyl serotonin and N-(p-coumaroyl) serotonin. Each compound was evaluated for its inhibitory activity against the inflammatory process of macrophages. All compounds significantly inhibited production of lipopolysaccharide (LPS)-stimulated nitric oxide (NO) and pro-inflammatory cytokines. The protein levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) were dramatically decreased by serotonins in a dose-dependent manner in LPS-stimulated RAW 264.7 macrophages. These results suggest that serotonin derivatives from safflower seeds may reduce inflammation-related diseases.

Preparation of High Quality Safflower (Carthamus tinctorius L.) Seed Extract by High-Pressure Extraction Process

  • Safflower seed extract was prepared by a high-pressure extraction technology and its quality characteristics were compared to that of other conventional extraction techniques, such ultrasonic and reflux extractions. Safflower seeds were extracted with 80% aqueous ethanol by three above extraction methods, and further fractionated with Diaion HP-20 column chromatography to obtain a partially purified safflower seed extract (PPSSE). Among the three extraction techniques examined, the reflux extraction showed the higher yields of EtOH extract and PPE than the ultrasonic and high-pressure extractions. Levels of most phenolic compounds in the EtOH extract of safflower seed are higher in reflux and ultrasonic extractions than the high pressure extraction, but levels of two serotonin aglycones, N-(p-coumaroyl)serotonin (CS) and N-feruloylserotonin (FS), in PPSSE were higher in the high pressure extraction than the reflux and ultrasonic extractions. In addition, color values (L and a) of the PPSSE were higher in the high-pressure extraction than the reflux and ultrasonic extractions, although there were no significant differences in pH and UV maxima absorption spectra among three extraction techniques. These results indicate that the high-pressure extraction technology is a simple and effective extraction for preparation of a high quality of safflower seed extract containing CS and FS with anti-wrinkle activity.

국외 수집 홍화 유전자원의 항산화 활성 및 세로토닌 유도체 함량 분석 (Analysis of Antioxidant Activity and Serotonin Derivatives in Safflower (Carthamus tinctorius L.) Germplasm Collected from Five Countries)

  • 본 연구는 이란 등 5개국에서 수집한 홍화자원을 대상으로하여 천연항산화제로써 홍화종자의 항산화 활성을 평가하고, 세로토닌 유도체인N-(p-Coumaroyl) serotonin과N-feruloylserotonin의 함량을 측정하여 항산화 활성과의 상관관계를 조사하였다. 총폴리페놀함량은 $28.25{\sim}90.53{\mu}g\;GAE/mg\;DE$의 범위를 나타내었고, 색소화합물인 DPPH 라디칼과$ABTS^{+}$을 이용한 유리라디칼소거능을 측정하는 DPPH assay와 ABTS assay의 값은 18.76 ~ 93.98 및 $48.91{\sim}163.73{\mu}g\;ASCE/mg\;DE$의 범위를 나타내었다. 시료의 환원능을 측정하는 방법인 FRAP assay과 RP assay는 3.80 ~ 132.29 및 $26.32{\sim}80.08{\mu}g\;ASCE/mg\;DE$의 범위로 나타내었다. UHPLC를 이용하여 CS와 FS를 분석한 결과, CS 함량은 2.56 ~ 64.99 mg/g DE이고, FS 함량값은 1.92 ~ 65.36 mg/g DE이었다. 5개국 중 이란 원산종은 다른 나라에 비해 세로토닌 유도체 함량과 항산화 활성의 평균값이 높게 측정되었다. 항산화 활성과 세로토닌 유도체 함량간의 상관관계 분석 결과, CS는 TPC와 ABTS, DPPH와 높은 상관성을 보였고(r = 0.673, 0.727, 0.820), FS는 DPPH와 높은 상관성을 보였다(r = 0.740). RACI를 통하여 선발된 IT321214과 IT321215는 항산화 활성이 높은 자원으로 천연항산화제 소재 탐색 및 개발에 기초자료로 활용할 수 있을 것으로 생각된다.
