Helicobacter pylori 감염과 관련된 철 결핍성 빈혈에서 Lactoferrin Sequestration의 역할 (Lactoferrin Sequestration and Its Contribution to Iron Deficiency Anemia in Helicobacter pylori Infected Gastric Mucosa)

  • 배경: Lactoferrin은 위장 점막에서 H. pylori의 철분의 공급원으로 알려져 있다. 본 연구는 H. pylori에 감염된 위장 점막의 조직검체에서 lactoferrin 값을 측정하고 철 결핍성 빈혈의 유무에 따라 lactoferrin 발현의 주요 위치를 정하는 방법을 이용하여 lactoferrin과 철 결핍성 빈혈을 동반한 H. pylori감염 사이의 연관성을 밝히기 위해 시행하였다. 방법: 상부 위장관 내시경을 시행한 55명의 환아를 세 군으로 분류하였다: 정상군(NL, n=19), H. pylori 감염군(HP, n=15), H. pylori 감염이 동반된 철 결핍성 빈혈군(IDA, n=21). 조직 병리학 소견은 Updated Sydney System을 이용하여 정도를 나누었다. 위장 점막의 lactoferrin 값은 면역 측정법으로 측정하였다. lactoferrin의 발현 위치와 양을 측정하는 것은 면역조직화학적 염색방법을 이용하였다. 결과: 위 전정부에서의 lactoferrin의 값은 조직 검체의 H. pylori 밀도나 다형핵세포의 침습정도, 만성 염증에 비례하여 의미있게 증가되었다. HP와 IDA군에서는 정상군에 비하여 lactoferrin의 수치가 통계학적으로 유의하게 증가되었다(p=0.0001). IDA군은 HP군에 비하여 lactoferrin의 수치가 높은 경향이 있었다(p=0.2614). 면역조직화학적 방법에 의한 lactoferrin 발현의 주요부위는 상피세포내의 분비샘과 호중구에 있었다. Lactoferrin은 정상군에서는 약하게 염색되었고 HP와 IDA군에서는 강하게 염색되었다. 결론: IDA군에서 위장 점막내 lactoferrin 격리가 현저하였으며 이 소견은 H. pylori 감염이 철 결핍성 빈혈에 영향을 미치는 기전을 밝히는데 중요한 단서로 작용할 것이다.

Alcaligenes sp.에 의한 Poly-$\beta$-Hydroxybutyric Acid의 축적 (Accumulation of Poly-$\beta$-Hydroxybutyrie Acid by Alcaligenes sp.)

  • 토양으로부터 PHB 생산능이 우수한 균주 FL-027을 분리하여 분류학적 제 특성을 검토한 결과 Alcaligenes속으로 동정되었다. Alcaligenes sp.의 최적 생육 조건은 과당 8.0$g/\ell$, $(NH_4)_2S0_4$ 3.0$g/\ell$ (즉 C/N molar ratio가 5.04) 및 pH7.0과 $30^{\circ}C$였으며, PHB 축적은 과당 8.0$g/\ell$, (NH4)2SO4 0.25g/l(즉 C/N molar ratio가 60) 및 pH6.5와 30'C에서 가장 양호하였다. $NH_4^+$, $Ca^{2+}$, $SO_4^{2+}$의 결핍이 PHB축적을 촉진하였으나 이들 중 $NH_4^+$가 가장 효과적으로 PHB축적을 유도하였다. 고농도배양을 위해 과당을 간헐적으로 첨가하여 최적농도를 유지하면서 유가배양을 실시한 결과 균체량은 25.1$g/\ell$, PHB 축적량은 10.84$g/\ell$로 건조균체량의 43까지 축적되었다. 분리정제된 PHB를 IR 및 $^1H-NMR$로 분석한결과 3-hydroxybutyric acid의 homopolymer임을 알 수 있었다.

급성기 뇌경색 환자의 변증군간 호전도 차이에 대한 연구 (Differences of Symptom Improvement Between Different Diagnosis Classification Groups in Acute Cerebral Infarction Patient)

  • Object : The aim of this study was to examine the differences of symptom improvement between different diagnosis classification groups in acute cerebral infarction patients. Methods : We studied inpatients within a month after the onset of cerebral infarction who were admitted at Kyunghee University Oriental Medical Center from May 2011 to October 2012. We compared the improvement of Motricity Index and Scandinavian Stroke Scale score between Fire-heat group(n=20), Yin deficiency group(n=31), Dampness-phlegm group(n=30), and Qi deficiency group(n=13). Results : Yin deficiency group patients with cerebral infarction showed the most improvement in MI and SSS scores, and patients in Fire-heat group showed the poorest improvement in MI and SSS scores. There was a significant difference between the two groups, but there were no significant differences between all four diagnosis classification groups. Conclusions : This study provides evidence that diagnosis classification could be considered as an important factor in predicting the prognosis of acute cerebral infarction.

Interaction between Maternal Serum Folate and the Methylenetetrahydrofolate Reductase (MTHFR) Polymorphisms on Infant Birthweight

  • The purpose of this study was to evaluate whether the interactions between maternal folate deficiency and methylenetetrahydrofolate reductase (MTHFR) polymorphism increase the risk of elevated maternal serum homocysteine, short gestation and reduced infant birthweight. Healthy pregnant (n = 170; 24-28 gestational weeks; 20-40 years old) women were analyzed for the MTHFR genotype and serum levels of folate and homocysteine, and were then followed for gestational age and infant birthweight. The mean infant birthweight was highest in mothers carrying MTHFR CC and with a normal folate range, and they were followed by mothers carrying MTHFR CT or TT and a normal range of folate or a folate deficiency. Birthweight was the lowest in mothers whose carrying MTHFR CC with folate deficiency. Using two way ANOVA, we found that folate level and the MTHFR polymorphism interacted to affect birth-weight of infants (p=0.05). Among those mothers carrying MTHFR CC, those with folate deficiency showed a 543 g reduction in infant birthweight compared with those with normal folate levels. However, infant birthweight was no different for mothers, those who with folate deficiency compared to those with normal range of folate among mothers carrying the MTHFR CT or TT genotypes. This study suggests an interaction between maternal serum folate and the MTHFR polymorphisms of the mother on the risk of delivering reduced birthweight offspring. Folate supplementation of folate deficient pregnant women with the MTHFR wild type is suggested to reduce the risk of low birthweight.

가족의 기능적 결손 정도가 청소년의 자아존중감, 우울, 자살생각에 미치는 영향 (Effect of Functional Deficiency in Family on Self-esteem, Depression, and Suicidal Inclination of Adolescents)

  • Purpose: The aim of this study was to investigate the structural relationships between functional deficiency of Family and self-esteem, depression, suicidal inclination of adolescents. Methods: The data in this study came from the Korea Youth Panel Survey Data (N=3,449) for 2005~2007. To investigate the causal relationship between each variable presented in the research model, structural equation modeling (SEM) was carried out by using AMOS 7.0. And all data were analyzed by using SPSS/WIN 12.0. Results: The main analytical results of this study were as follows; First, the family function made a positive impact on the self-esteem of adolescents. Second, the severe functional deficiency in family appeared to have a negative effect on the suicidal inclination and depression of adolescents. Third, the higher the adolescents' self-esteem, the lower depression and suicidal ideation. Fourth, the more severe the depression, the higher occurrence of suicidal inclination. Conclusion: In conclusion, functional deficiency in adolescents' family impacted negatively on self-esteem, and this contributed to increased depression and anxiety, and to suicidal inclination finally. These findings confirmed that the strength of family function, the closeness with parents, forming a stable relationship and strength of the emotional bond are very important factors to form self-esteem and reduce depression and suicidal inclination.

The evaluation of Spectral Vegetation Indices for Classification of Nutritional Deficiency in Rice Using Machine Learning Method

  • Detection of stress responses in crops is important to diagnose crop growth and evaluate yield. Also, the multi-spectral sensor is effectively known to evaluate stress caused by nutrient and moisture in crops or biological agents such as weeds or diseases. Therefore, in this experiment, multispectral images were taken by an unmanned aerial vehicle(UAV) under field condition. The experiment was conducted in the long-term fertilizer field in the National Institute of Crop Science, and experiment area was divided into different status of NPK(Control, N-deficiency, P-deficiency, K-deficiency, Non-fertilizer). Total 11 vegetation indices were created with RGB and NIR reflectance values using python. Variations in nutrient content in plants affect the amount of light reflected or absorbed for each wavelength band. Therefore, the objective of this experiment was to evaluate vegetation indices derived from multispectral reflectance data as input into machine learning algorithm for the classification of nutritional deficiency in rice. RandomForest model was used as a representative ensemble model, and parameters were adjusted through hyperparameter tuning such as RandomSearchCV. As a result, training accuracy was 0.95 and test accuracy was 0.80, and IPCA, NDRE, and EVI were included in the top three indices for feature importance. Also, precision, recall, and f1-score, which are indicators for evaluating the performance of the classification model, showed a distribution of 0.7-0.9 for each class.

Temporal Changes in N Assimilation and Metabolite Composition of Nitrate-Affected Tomato Plants

  • The role of inorganic nitrogen assimilation in the production of amino acids, organic acids and soluble sugars is one of the most important biochemical processes in plants, and, in order to achieve normally, nitrate uptake and assimilation is essential. For this reason, the characterization of nitrate assimilation and metabolite composition from leaves, roots and xylem sap of tomato (Solanum lycopersicum) was investigated under different nitrate levels in media. Tomato plants were grown hydroponically in liquid culture under five different nitrate regimes: deficient (0.25 and 0.75 mM $NO_3{^-}$), normal (2.5 mM $NO_3{^-}$) and excessive (5.0 and 10.0 mM $NO_3{^-}$). All samples, leaves, roots and xylem sap, were collected after 7 and 14 days after treatment. The levels of amino acids, soluble sugars and organic acids were significantly decreased by N-deficiency whereas, interestingly, they remained higher in xylem sap as compared with N-normal and -surplus. The N-excessive condition did not exert any significant changes in metabolites composition, and thus their levels were similar with N-normal. The gene expression and enzyme activity of nitrate reductase (NR), nitrite reductase (NIR) and glutamine synthetase (GS) were greatly influenced by nitrate. The data presented here suggest that metabolites, as a signal messenger, existed in xylem sap seem to play a crucial role to acquire nitrate, and, in addition, an increase in ${\alpha}$-ketoglutarate pathway-derived amino acids under N-deficiency may help to better understand plant C/N metabolism.

소아청소년기 성장호르몬결핍증의 성인기 지속에 영향을 미치는 요인 (Factors for persistent growth hormone deficiency in young adults with childhood onset growth hormone deficiency)

  • 목 적 : 소아청소년기 GHD 환자들은 성인기에 진단된 환자보다 체성분과 심혈관계의 변화가 심한 것으로 알려져 있어 성인기에도 GHD가 지속되면 성장호르몬 치료를 꼭 받아야 한다. 본 연구에서는 소아청소년기에 GHD로 진단 받은 성인 환자들을 대상으로 ITT를 시행하여 성인기에도 GHD가 지속되는 요인을 살펴보고, 성인기에도 GHD가 지속될 가능성이 높은 대상을 알아보고자 한다. 방 법 : 성인키에 도달한 55명(남 39명, 여 16명)을 대상으로 ITT의 최대 성장호르몬 값이 $3{\mu}g/L$ 미만이면 성인기 GHD로 진단하였다. 특발성 GHD 환자(n=12)는 뇌하수체 구조적인 이상이 있는 2명과 둔위 분만 또는 가사의 병력이 있는 10명이었으며, 기질성 GHD 환자(n=43)는 시상하부-뇌하수체를 침범하는 뇌종양(n=33), 그 외의 뇌종양(n=3), 뇌수막염(n=3), 백혈병(n= 2), 지주막낭종(n=1), 뇌하수체염(n=1)에 해당하였다. 결 과 : 성인기에도 GHD가 지속된 환자는 49명(89.1%)이었다. 최대 성장호르몬 값의 log값은 기저 IGF-I 농도와 양의 상관관계를 보였다(r=0.57, P<0.001). 특발성 GHD 환자의 91.7%가 성인기에도 GHD가 지속되어, 성인기 GHD 여부는 소아청소년기 특발성, 기질성 GHD 여부에 따른 차이가 없었다. 동반된 뇌하수체호르몬 결핍 개수가 많을수록(단독 GHD 40%, 1개의 결핍 동반 80%, 2개 이상 동반 95.6%, P=0.002), 기질성 GHD 환자 중에서 병변이 시상하부-뇌하수체를 일차적으로 침범하는 경우(P=0.003)에 성인기에도 GHD가 지속될 가능성이 높았다. 두개 내 방사선 치료를 받은 18명 중에서 15명이 성인기에도 GHD가 지속되었는데, 시상하부-뇌하수체를 침범하는 종양으로 방사선 치료를 받은 환자들 14명은 모두 방사선조사량에 관계없이 성인기에도 GHD가 지속되었다. 동반된 뇌하수체호르몬 결핍이 있는 경우(P=0.005), 소아청소년기 완전 GHD인 경우(P=0.005)에 성인기에도 GHD가 지속되는 비율이 높았다. 결 론 : 소아청소년기에 GHD로 진단 받은 환자들 중에서 동반 뇌하수체호르몬 결핍이 2개 이상이거나, 기저 질환의 병변이 시상하부-뇌하수체를 포함하면 성인기에도 GHD가 지속될 가능성이 높다.

Comparative study on the effects of micro- and nano-sized zinc oxide supplementation on zinc-deficient mice

  • Background: Zinc (Zn) is an essential cofactor for physiological homeostasis in the body. Zn oxide (ZnO), an inorganic compound that supplies Zn, exists in various sizes, and its bioavailability may vary depending on the size in vivo. However, comparative studies on the nutritional effects of micro-sized ZnO (M-ZnO) and nano-sized ZnO (N-ZnO) supplementation on Zn deficiency (ZnD) animal models have not been reported. Objectives: This study investigated the nutritional bioavailability of N-ZnO and M-ZnO particles in dietary-induced ZnD mice. Methods: Animals were divided into six experimental groups: normal group, ZnD control group, and four ZnO treatment groups (Nano-Low, Nano-High, Micro-Low, and MicroHigh). After ZnD induction, N-ZnO or M-ZnO was administered orally every day for 4 weeks. Results: ZnD-associated clinical signs almost disappeared 7 days after N-ZnO or M-ZnO administration. Serum Zn concentrations were higher in the Nano-High group than in the ZnD and M-ZnO groups on day 7 of ZnO treatment. In the liver and testis, Nano-Low and Nano-High groups showed significantly higher Zn concentrations than the other groups after 14-day treatment. ZnO supplementation increased Mt-1 mRNA expression in the liver and testis and Mt-2 mRNA expression in the liver. Based on hematoxylin-and-eosin staining results, N-ZnO supplementation alleviated histological damage induced by ZnD in the testis and liver. Conclusions: This study suggested that N-ZnO can be utilized faster than M-ZnO for nutritional restoration at the early stage of ZnD condition and presented Mt-1 as an indicator of Zn status in the serum, liver, and testis.

Schisandrol A and gomisin N from Schisandra chinensis extract improve hypogonadism via anti-oxidative stress in TM3 Leydig cells

  • BACKGROUND/OBJECTIVES: Male hypogonadism is a condition where the body does not produce enough testosterone and significantly impacts health. Age, obesity, genetics, and oxidative stress are some physiological factors that may contribute to testosterone deficiency. Previous studies have shown many pharmacological benefits of Schisandra chinensis (S. chinensis) Baillon as an anti-inflammatory and antioxidant. However, the molecular mechanism of attenuating hypogonadism is yet to be well established. This research was undertaken to study the effects of S. chinensis extract (SCE) on testosterone deficiency. MATERIALS/METHODS: S. chinensis fruit was pulverized and extracted using 60% aqueous ethanol. HPLC analysis was performed to analyze and quantify the lignans of the SCE. RESULTS: The 2,2-diphenyl-2-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) scavenging assays confirmed that the SCE and its major lignans (schisandrol A and gomisin N) inhibit oxidative stress. Effects of SCE analysis on the testosterone level under oxidative stress conditions revealed that both schisandrol A and gomisin N were able to recover the lowered testosterone levels. Through mRNA expression of TM3 Leydig cell, we observed that the SCE lignans were able to induce the enzymes involved in testosterone biosynthesis-related genes such as 3β-HSD4 (P < 0.01 for SCE, and P < 0.001 for schisandrol A and gomisin N), 17β-HSD3 (P < 0.001 for SCE, schisandrol A and gomisin N), and 17, 20-desmolase (P < 0.01 for schisandrol A, and P < 0.001 for SCE and gomisin N). CONCLUSIONS: These results support that SCE and its active components could be potential therapeutic agents for regulating and increasing testosterone production.
