Role of E2F1 in Endoplasmic Reticulum Stress Signaling

  • The transcription factor E2F1 coordinates cell cycle progression and induces apoptosis in response to DNA damage stress. Aside from DNA damage, the role of E2F1 in the endoplasmic reticulum (ER) stress signaling pathways is unclear. We found that $E2F1^{-/-}$ murine embryonic fibroblasts (MEFs) are resistant to apoptosis triggered by the ER stress inducer thapsigargin. In addition, E2F1 deficiency results in enhanced phosphorylation of eukaryotic translation initiation factor $2{\alpha}$ ($elF2{\alpha}$). These results therefore indicate that E2F1 deficiency increases phosphorylation of $elF2{\alpha}$ in response to ER stress triggered by thapsigargin, and suggest that the reduction in ER stress-induced apoptosis in E2F1-deficient cells is related to the high level of $elF2{\alpha}$ phosphorylation.

The effects of royal jelly protein on bone mineral density and strength in ovariectomized female rats

  • [Purpose] Sex hormones deficiency leads to dramatically bone loss in particular postmenopausal women. Royal jelly has anti-osteoporosis effect due to maintain bone volume in that condition. We hypothesized that royal jelly protein (RJP, a latent residue after extracting royal jelly) also prevents bone deficient in ovariectomized (OVX) female rats, the animal model of postmenopausal women. [Methods] Female Sprague-Dawley rats (n = 30, 6 weeks age old) were sham operated (Sham; sham operated group, n = 7), OVX control group (OC, n = 7), OVX with low RJP intake group (ORL, n = 8), and OVX with high RJP intake group (ORH, n = 8) during 8 weeks experimental periods. In the end point of this experiment, the bone samples (lumbar spine, tibia, and femur) were surgically removed under anesthesia. These bone samples were evaluated bone mineral density (BMD) and bone strength. [Results] BMD of lumbar spine in RJP intake groups (ORL, ORH) were higher than that in OC group (p < 0.05 and p < 0.01) in RJP intake volume dependent manner. BMD of tibial proximal metaphysis and diaphysis in RJP intake groups were also higher than these in OC group (p < 0.01 and p < 0.01 / p < 0.05 and p < 0.001). In addition, breaking force of femur in RJP intake groups were significantly increase compared with that in OC group (p < 0.001 respectively). [Conclusion] These findings indicate that RJP contribute to prevent sex hormone related bone abnormality.

중.노년기 제2형 당뇨병 여성의 아연 및 구리 영양상태 (Zinc and Copper Status of Middle- and Old-Aged Women in Type 2 Diabetes)

  • The purpose of this study was to assess the zinc and copper status of type 2 diabetic women and to analyse the relation-ship among zinc and copper status, and diabetic control indices of diabetes. The mean age of diabetes was 57.9 years old. The mean duration of diabetes was 8.0 $\pm$ 6.5 years. The mean daily energy intake of diabetes was 1562 kcal. There were no significant differences of age, BMI, %body fat, nutritional intakes, total energy intake, and energy composition from carbohydrate, protein, and fat between diabetes (n = 50) and control group (n = 68). However, both zinc intake density (4.15 mg/1000 kcal) and zinc %RDA (62.0%) of diabetes were significantly lower than control group (p <0.05, P < 0.01, respectively). The plasma zinc level was not significantly different between diabetes and control group (90$\mu$g/dl, 91$\mu$g/dl respectively). The proportion of diabetic women whose plasma zinc levels were lower than 76 $\mu$g/dl(borderline zinc deficiency) was 18.8%. This was about 38% higher than control group. It has been suggested that insulin secretion might decrease in borderline plasma zinc and copper deficiency and increase in normal plasma zinc and copper status. The urinary zinc excretion was twice higher in diabetes than in control group (p < 0.001). The urinary zinc loss was positively correlated with the duration of diabetes (p < 0.05), hyperglycemia (p < 0.001) and insulin resistance (p < 0.05). These results lead us to conclude that normal blood glucose level controlled by diet therapy could improve the hyperzincuria in diabetic women.

Genetic Diversity and Relationships of Korean Chicken Breeds Based on 30 Microsatellite Markers

  • The effective management of endangered animal genetic resources is one of the most important concerns of modern breeding. Evaluation of genetic diversity and relationship of local breeds is an important factor towards the identification of unique and valuable genetic resources. This study aimed to analyze the genetic diversity and population structure of six Korean native chicken breeds (n = 300), which were compared with three imported breeds in Korea (n = 150). For the analysis of genetic diversity, 30 microsatellite markers from FAO/ISAG recommended diversity panel or previously reported microsatellite markers were used. The number of alleles ranged from 2 to 15 per locus, with a mean of 8.13. The average observed heterozygosity within native breeds varied between 0.46 and 0.59. The overall heterozygote deficiency ($F_{IT}$) in native chicken was $0.234{\pm}0.025$. Over 30.7% of $F_{IT}$ was contributed by within-population deficiency ($F_{IS}$). Bayesian clustering analysis, using the STRUCTURE software suggested 9 clusters. This study may provide the background for future studies to identify the genetic uniqueness of the Korean native chicken breeds.

  • 저자들은 비교적 젊은 연령이며 호모시스테인혈증이 있는 환자들에서 발생한 폐혈전색전증 및 심부정맥혈전증 2예를 문헌과 함께 고찰하였다. 폐혈전색전증이 연령이 증가할수록 발생률이 증가한다는 점과 다른 위험요소가 존재하지 않는다는 점으로 보아 증례들에서 호모시스테인혈증이 폐혈전색전증을 일으킨 원인으로 보인다. 위험요소가 없으며 젊은 연령에서 생긴 폐혈전색전증에 있어 호모시스테인혈증을 원인의 하나로 고려하여야 할 것이다.

한국 남해 마산만에서 수질환경의 계절적 변동과 기초생산 제한인자 (Variability of Water Quality and Limiting Factor for Primary Production in Semi-enclosed Masan Bay, South Sea of Korea)

  • Seasonal variations of various physicochemical components (temperature, salinity, pH, DO, COD, DOC, nutrients-silicate, DIN, DIP) and potential limiting factor for phytoplankton primary production were studied in the surface water of semi-enclosed Masan Bay. Seasonal variations of nutrient concentrations, with lower values in summer and winter, and higher in fall, are probably controlled by freshwater loadings to the bay, benthic flux and magnitude of occurrence of phytoplankton communities. Their spatial distributional patterns are primarily dependent on physical mixing process between freshwater and coastal seawater, which result in a decreasing spatial gradient from inner to outer part of the bay. In the fall season of strong wave action, the major part of nutrient inputs (silicate, ammonium, dissolved inorganic phosphorus) comes from regeneration (benthic flux) at sediment-water interface. During the summer period, high Si:DIN and Si:DIP and low DIN:DIP relative to Redfield ratios suggest a N- and secondarily P-deficiency. During other seasons, however, silicate is the potential limiting factor for primary production, although the Si-deficiency is less pronounced in the outer region of the bay. Indeed, phytoplankton communities in Masan Bay are largely affected by the seasonal variability of limiting nutrients. On the other hand, the severe depletion of DIN (relatively higher silicate level) during summer with high freshwater discharge probably can be explained by N-uptake of temporary nanoflagellate blooms, which responds rapidly to pulsed nutrient loading events. In Masan Bay, this rapid nutrient consumption is considerably important as it can modify the phytoplankton community structures.

바다부유물질로 만든 퇴비를 시용한 토양에서 발생한 딸기의 질소결핍증 해결 사례연구 (Case Study of N Deficiency Symptom of Strawberry in the Soil Applied with Sea Deposit Compost)

  • pH가 높은 바다부유물질로 만든 퇴비를 시용한 토양은 암모니아 휘산이 야기되어 토양 중 질산태 질소함량이 낮고, 딸기는 질소를 흡수하지 못하여 생육이상증상이 유발되었다. 이 토양의 pH를 질산을 처리하여 낮추어 주고 부족한 질소와 인산 및 칼륨을 공급한 결과 회복되었다. 이 결과로부터 pH가 높은 토양에서 발생하는 질소결핍증은 토양의 pH를 낮추어 주어야 회복시킬 수 있다고 판단되었다.

Deficiency or activation of peroxisome proliferator-activated receptor α reduces the tissue concentrations of endogenously synthesized docosahexaenoic acid in C57BL/6J mice

  • BACKGROUND/OBJECTIVES: Docosahexaenoic acid (DHA), an n-3 long chain polyunsaturated fatty acid (LCPUFA), is acquired by dietary intake or the in vivo conversion of ${\alpha}$-linolenic acid. Many enzymes participating in LCPUFA synthesis are regulated by peroxisome proliferator-activated receptor alpha ($PPAR{\alpha}$). Therefore, it was hypothesized that the tissue accretion of endogenously synthesized DHA could be modified by $PPAR{\alpha}$. MATERIALS/METHODS: The tissue DHA concentrations and mRNA levels of genes participating in DHA biosynthesis were compared among $PPAR{\alpha}$ homozygous (KO), heterozygous (HZ), and wild type (WT) mice (Exp I), and between WT mice treated with clofibrate ($PPAR{\alpha}$ agonist) or those not treated (Exp II). In ExpII, the expression levels of the proteins associated with DHA function in the brain cortex and retina were also measured. An n3-PUFA depleted/replenished regimen was applied to mitigate the confounding effects of maternal DHA. RESULTS: $PPAR{\alpha}$ ablation reduced the hepatic Acox, Fads1, and Fads2 mRNA levels, as well as the DHA concentration in the liver, but not in the brain cortex. In contrast, $PPAR{\alpha}$ activation increased hepatic Acox, Fads1, Fads2, and Elovl5 mRNA levels, but reduced the DHA concentrations in the liver, retina, and phospholipid of brain cortex, and decreased mRNA and protein levels of the brain-derived neurotrophic factor in brain cortex. CONCLUSIONS: LCPUFA enzyme expression was altered by $PPAR{\alpha}$. Either $PPAR{\alpha}$ deficiency or activation-decreased tissue DHA concentration is a stimulus for further studies to determine the functional significance.

Genetic Variation and Population Structure of Crepidiastrum lanceolatum (Compositae)

  • Crepidiastrum lanceolatum ($H_{OUTT}$) $N_{AKAI}$ (Compositae) is distributed in East Asia including Korea. Genetic diversity and population structure of six C. lanceolatum populations in Korea and two populations in Japan were determined using genetic variation at 19 allozyme loci. The percent of polymorphic loci within the enzymes was 42.1%. Genetic diversity at the species level and at the population level was low (Hes : 0.077; Hep : 0.068, respectively), where-as the extent of the population divergence was relatively low ($G_{ST}$ : 0.093). One of the most striking features of this study was the more significant difference within populations than among populations. An indirect estimate of the number of migrants per generation (Nm : 2.44) indicated that gene flow was moderate among eight populations of the species. In addition, analysis of fixation indices revealed a slight heterozygosity deficiency in some populations and at some loci. Narrow geographic ranges, short-lived perennial herbaceous, and small population sizes are mainly associated with the low level of genetic variation.n.

MEROMORPHIC SOLUTIONS OF A COMPLEX DIFFERENCE EQUATION OF MALMQUIST TYPE

  • In this paper, we investigate the finite order transcendental meromorphic solutions of complex difference equation of Malmquist type $$\prod_{i=1}^{n}f(z+c_i)=R(z,f)$$, where $c_1,{\ldots},c_n{\in}\mathbb{C}{\backslash}\{0\}$, and R(z, f) is an irreducible rational function in f(z) with meromorphic coefficients. We obtain some results on deficiencies of the solutions. Using these results, we prove that the growth order of the finite order solution f(z) is 1, if f(z) has Borel exceptional values $a({\in}\mathbb{C})$ and ${\infty}$. Moreover, we give the forms of f(z).
