Effects of Some Environmental Factors on Mycerial Growth, Sporulation and Sclerotial Formation of Botrytis elliptica (Berk.) Cooke (백합(百合) 잎마름병균(病菌)의 균사생장(菌絲生長), 포자형성(胞子形成) 및 균핵형성(菌核形成)에 미치는 몇가지 환경요인(環境要因)의 영향(影響))

  • This experiment was conducted to study the effect of some environmental factors on mycerial growth, sporulation and sclerotial formation of Botrytis elliptica cultured on artificial media. Mycerial growth of B. elliptica was the most favorable on V-8 juice agar among the seven different media tested and sporulation of the fungus was favorable on the medium under NUV light irradiation. Abundant conidia could be obtained from V-8 juice agar medium by NUV light irradiation after 3 days of incubation at $23^{\circ}C$ under darkness. The optimum temperature for mycerial growth and conidial germination was $23^{\circ}C$ and the mycerial growth was favorable at relatively lower temperature ranged $19^{\circ}C$ to $23^{\circ}C$. The optimum pH of the medium for mycerial growth of this fungus ranged 4.5 to 5.0 and that was inhibited at higher pH of the media. Mycerial growth of the fungus was not highly influenced by irradiation of fluorescent light, however sporulation was stimulated under NUV light irradiation. Sclerotia of B. elliptica were formed when it was cultured at lower temperature below $19^{\circ}C$.

  • This study was done to find out the antifungal activity of two antibacterial antibiotics, Chlorampenicol and Streptomycin sulfate, against Phytophthora n. var. Parasitica, the causal agent of Phytophthora blight of sesame, growing on artificial media. On PDA medium, Chlorampenicol at 10 ppm, Streptomycin sulfate at 25ppm highly inhibited mycerial growth and completely inhibited zoosporangial formation of Phytophthora n. var. parasitica, and Chlorampenicol at 5 ppm, Streptomycin sulfate at 10 ppm slightly inhibited the mycerial growth and zoosporangial formation of the fungus. These antibiotics showed considerably increased inhibitory effect on the fungal growth when they were mixed with other chemical. Protein content in myceria of the fungus was decreased and abnormal growth of mycerial apex was observed by treatment of these antibiotics. Phytotoxicity on sesame seedlings was not observed by application of them.

  • The effects of amino acids on mycerial growth and sporulation of the decreased and increased pathogenicity variants of potato dry rot Fusarium roseum ‘Sambucinum' were as following. The increased pathogenicity variants Ic52 and Ic116 showed the trend of greater mycerial growth, marking up to 67mg on leucine, than the decreased pathogenicity variants Dc14 and Dc91, and produced markedly low conidia below $25{\times}10^{4}/ml$ or no sporulation. On methionine all the variants and the wild type showed the lowest growth recording 14mg to 20mg with no sporulation. Cystine similarly as glysine supported the poor growth 30mg to 19mg of the variants Dc14 and Dc91, respectively, and did not support the sporulation. The former amino acid like asparagine supported the growth of the variants Ic52 and Ic1l6 marking 47mg and 36mg, respectively. On all the eleven amino acids the variant Dc14 showed the trend of more mycerial growth and sporulation than the variant Dc91 which marked the exceptionally high sporulation $195{\times}10^{4}/ml$ on glutamic acid.

  • In this study, th effective extraction of 5 vegetables, which have been shown to inhibit the growth of the aflatoxin production of Aspergillus parasiticus R-716, was investigated and their effects were examined. Radish, cabbage, garlic and zinger were effectively with water-chloroform, but crown daisy with n-hexane. Among them, water-chloroform extract of radish was remarkably effective, and garlic extract only inhibited the growth strongly. The growth and the aflatoxin production of the strain were showed 0.758g/25ml, 763ug/25ml with the addition of water-chloroform extract equivalent 15g of raw radish, and an increase in the level of radish extract resulted in a decrease both the growth and the aflatoxin production per mycerial weight.

  • The effects of fermentor types on biopolymer production with Cordyceps militaris has been studied in this articles. Since the shear stresses of stirred tank ferementor and bubble column fermentor were different, the mycelium growth and biopolymer production were different. The mycerial growth was high with the bubble column fermentor for the early stage but biopolymer production was lower than that with stirred tank fermentor. The production of biopolymer was closely related to shear stress to the cells and biopolymer production was increased with increased shear stress.

  • This study was carried out to obtain the basic data on artificial culture of Lampteromyces japonicus. Favorable media for mycerial growth were glucose peptone medium, malt yeast extract, yeast malt peptone, potato dextrose medium. The optimum conditions for the mycelial growth were $30^{\circ}C$ and pH 6.0. Dextrose as a carbon source was favorable to mycelial growth. The optimal dextrose concentration was 1.2%. As nitrogen sources, yeast extract, $(NH_4)_2HPO_4$ and 0.2% for glutamine. The mineral salts of $Al_2(SO_4)_3{\cdot}14H_2O$ were effective and the optimal concentration was 0.1 M.

  • Pholiota nameko is one of the four major profitable mushrooms along with oak mushroom, winter mushroom, and oyster mushroom. It contains abundant proteins, carbohydrates, organic acids and vitamins. Its unique taste and flavor as well as its nutritional features make it widely favoured. Mushroom complete medium was the optimal medium for mycelial growth of Pholiota nameko. The optimal temperature and pH for the mycelial growth were $25^{\circ}C$ and 5.0, respectively. The best carbon sources for mycelial growth were glucose and mannose, and the best nitrogen sources were yeast extract, peptone, asparagine, etc. The 8:2 ratio mix of oak sawdust and wheat bran was the best for the bottle cultivation. The best mushroom was yielded after 30 days incubation. The best yield was produced with 850g of medium weight in a PP bag and bottle.

  • This experiments were carried out to development low-priced mushroom culture media, and the results were as follows : Mycerial growth and density of Pleurotus ostreatus in the culture media of 40% of yellow soil, upland soil and lowland soil(each and all) added 60% sawdust and rice bran(80:20, %, v/v) were 89, 88, 68mm/10days in that order. Lentinus edodes were in the media of $40{\sim}50%$ of that added 60% sawdust and rice bran(80:20, %, v/v) were 74, 71, 68mm/10days. Ganoderma lucidum were in the media of $40{\sim}50%$ of that added 60% sawdust and rice bran(80:20, %, v/v) were 98, 94, 89mm/10days in that order.

Control Effect of Stenotrophomonas maltophilia BW-13 strain to the lettuce Bottom rot

  • An antagonistic bacteria, Stenotrophomonas maitophilia BW-13 strain which was effectively inhibited mycerial growth of Bottom rot pathogen, Rhizoctonia solani PY-1 strain was isolated from the rhizosphere of the lettuce in Uiryeong-Gun, Gyeongsangnam-Do from 2002 to 2003. For the biological control, the most suitable inoculum and its density of pathogen, PY-1 strain ware tested prior biological control test, For the pathogenicity test, A inoculum (wheat bran)sawdust+rice bran+PDB) showing disease incidence of 100% was selected as the most suitable inoculum, which showed more effective than B inoculum (sawdust+rice bran+DW) and mycelial disc. also, In selection of the amount of inoculum (40g, 50g, 60g, 70g, 80g), most suitable amount of inoculum of pathogen determined as 40g showing disease incidence of 80%. For the selection of effective microorganism to control bottom rot on lettuce, about 200 isolates were isolated from the diseased soil and lettuce leaves, and examined their antifungal activity to the pathogen on PDA. As the pots assay, BW-13 strain showed the highest control value as 90%, and followed by R-13 and R-26 strain as 80% and 60%, respectively. Selected BW-13 isolates identified as 5. maltophilia (GeneBank accession no. AJ293473.1, 99%) by 16S rRNA sequencing. This is the first report on the biological control using by S. maltophilia to the bottom rot pathogen, Rhizoctonia solani PY-1 strain.

  • A group of Pinus densiflora trees attacked by Rhizina root rot were observed at Kangnung. Diseased roots are characterized by rot patches, radial rot traces or formation of adhesive soil masses. The damage has proceeded about 6m per annum, and the pathogen in the infected soil was detected by trap logs. Ascospores of Rhizina undulata was germinated by heat shock at $37^{\circ}C$ for 24 hours or at $40^{\circ}C$ for 17 hours. The mycerial growth was optimum on PDA medium at $25-30^{\circ}C$and pH 5.6-6.3. Coniferous trees were more susceptible than non-coniferous trees in inoculation test in vitro.
