Physiological and Molecular Characterization of Cephaleuros virescens Occurring in Mango Trees

  • The objective of this work was to accomplish the isolation, molecular identification and characterizing the physiology of the causal agent of the algal spot in mango trees. For this purpose, the pathogen growth was assessed in different culture media, with subsequent observation and measurements of the filamentous cells. The molecular identification was made using mycelium obtained from leaf lesions and pure algae colonies grown in culture medium. Descriptions based on DNA sequencing indicated that the algae is Cephaleuros virescens. The algae must be isolated primarily in liquid medium for further pricking into agar medium. The highest mycelial growth average in Petri dishes occurred when the algae were grown in Trebouxia and BBM. Trebouxia enabled larger cells in the filamentous cells when compared to other culture media.

Proteomic Analysis of Coprinopsis cinerea under Conditions of Horizontal and Perpendicular Gravity

  • Coprinopsis cinerea was employed to investigate the fungal response to gravity. Mycelium growth revealed a consistent growth pattern, irrespective of the direction of gravity (i.e., horizontal vs. perpendicular). However, the fruiting body grew in the direction opposite to that of gravity once the primordia had formed. For the proteomic analysis, only curved-stem samples were used. Fifty-one proteins were identified and classified into 13 groups according to function. The major functional groups were hydrolases and transferases (16%), signal transduction (15%), oxidoreductases and isomerases (11%), carbohydrate metabolism (9%), and transport (5%). To the best of our knowledge, this is the first report on a proteomic approach to evaluate the molecular response of C. cinerea to gravity.

Antifungal Activity of an Endophytic Fungus Aspergillus versicolor DYSJ3 from Aphanamixis grandifolia Blume against Colletotrichum musae

  • An endophytic fungus strain DYSJ3 was isolated from a stem of Aphanamixis grandifolia Blume, which was identified as Aspergillus versicolor based on the morphological characteristics, internal transcribed spacer (ITS) and calmodulin gene sequences analyses. A. versicolor DYSJ3 exhibited strong antagonistic activity against Colletotrichum musae, C. gloeosporioides and Fusarium oxysporum f. sp. cubense with the inhibition rates of 61.9, 51.2 and 55.3% respectively. The antifungal metabolites mainly existed in the mycelium of A. versicolor DYSJ3, and its mycelial crude extract (CE) had broad-spectrum antifungal activities against plant pathogenic fungi. The CE had a good thermal stability, and the inhibition rate of 100 mg/mL CE against C. musae was above 70.0% after disposing at 120 ℃ for 1 h. Five secondary metabolites were isolated from the CE and identified as averufanin, ergosterol peroxide, versicolorin B, averythrin and sterigmatocystin. Activity evaluation showed versicolorin B exhibited inhibitory effects on the mycelial growth and conidial germination of C. musae, and sterigmatocystin had a weak inhibitory effect on the mycelial growth of C. musae.

콩나물 부산물 첨가량에 따른 표고의 생장특성 (Growth characteristics of Lentinula edodes treated with bean sprout waste)

  • 본 연구는 미강의 대체 원료로 콩나물 부산물(BW)을 활용하여 표고의 생장특성을 확인하였다. 참나무톱밥과 미강을 8:2(v/v) 섞은 배지와 미강을 대체하기 위해 미강과 각 BW 50%, 100%로 BW를 섞은 혼합배지에 표고의 주요 재배 품종인 산조 701의 균사생장을 비교한 결과 대조구와 BW 50%의 균사생장량은 13.5 cm였으며, BW 100%의 균사생장량은 12.2 cm인 것으로 확인되었다. 자실체 생장을 비교한 결과 대조구에 비해 BW 50%는 갓의 직경은 1.6 cm 증가하였으며, 대길이는 0.4 cm 감소되었다. 반면에 BW 100%는 갓의 직경은 9.6 cm 감소하였으며, 대의 길이는 1.4 cm 감소하였다. 구성아미노산의 분석 값에 의하면 영양적인 측면에서 BW 50%는 대조구에 비해 전반적으로 낮은 함량을 보인 반면 BW 100%는 대조구에 비해 Met 함량은 낮았지만 감칠맛을 더해주는 성분을 가진 Glu와 필수아미노산 중 하나인 Val이 각 3.95 mg/g, 2.30 mg/g 높은 것을 확인하였다. 따라서 콩나물 부산물을 효율적으로 처리 및 활용한다면 미강 대체제로서 활용이 가능할 것으로 보여진다.

Characteristics and suitability of various cereal grains in spawn production of button mushroom

  • Spawn is the vegetative growth of the mycelium and serves as the inoculum or seed for cultivating mushrooms. Various cereal grains, such as wheat, millet, barley, sorghum, brown rice, rye, and oat were assessed to compare their characteristics and suitability for spawn production of button mushroom (Agaricus bisporus). The mycelial growth rates, density, and the number of completely colonized grains were measured from the twentieth day of inoculation. Wheat grains showed fastest mycelial growth with 8.4 cm followed by rye, oat, barley with 8.2, 7.5 and 7.3 cm, respectively. In the mycelial density, foxtail millet, barley, and sorghum were best compared with that of wheat grains. Especially, the number of grains which were completely colonized by mycelia were greatest in foxtail millet with 5,123 grains followed by proso millet, and wheat with 3,052 and 914, respectively. Based on the results obtained, barley, foxtail millet, and sorghum grains would be appropriate substituting for wheat grain in spawn production of button mushroom.

커피박에 의한 구름버섯 균사체의 생장 촉진 (Acceleration of the Mycelial Growth of Trametes veriscolor by Spent Coffee Ground)

  • 구름버섯은 온대지역에서 흔히 발견되는 목재 부후균으로서 중요한 약용버섯의 일종이다. 본 연구에서는 커피박, 과립커피 및 과립 디카페인 커피의 구름버섯 균사체 생장에 미치는 영향을 조사하였다. 그 결과, 커피박은 10% 농도에서까지 그리고 과립커피의 경우 1% 미만에서 균사체 생육을 촉진시키는 것으로 확인되었다. 1% 커피박의 경우 무처리에 비해 10배 정도의 건조중량 증가가 측정됨으로써 구름균사체의 생육촉진제로서의 커피 효능을 확인할 수 있었다. 또한 커피는 균사체의 페놀화합물 및 항산화 효능 증대를 위한 유용 물질로 판단되었다.

배지에 따른 Genus Monascus의 증식도와 색소함량 (Growth and Pigment Formation of Genus Monascus on Medium Compositions)

  • Genus Monascus(M. pilosus and M purpureus)의 배지(Lin, SP, YM YE, GMIN, SMO, MY, GY, Nishikawa, sucrose, stock culture, Mizutani, modified Lit Toya 및 쌀배지)에 따른 균의 증식도와 색소생성량을 조사하였다. M. pilosus 및 M purpureus의 증식도가 높은 배지는 Mizutani 배지와 Lin 배지 였으며 3$0^{\circ}C$에서 10일간 배양하였을 때 생균체 무게는 각각 24.5~26.9 g/100 mL 및 15.9~17.2 g/100 mL이였고 M. pilosus의 성장률이 높았다. 색소생성량이 높은 배지는 두 균주 다같이 Lin 및 Mizutani 배지였으며 각각 흡광도는 1.2~1.6 및 0.8~l.0이었다. 균주 중에서는 M. pilosus에서 높았다.

맛버섯 균사체의 배양 특성에 관한 연구 (A Study on the Cultural Characteristics of Pholiota nameko Mycelium)

  • 맛버섯( Pholiota nameko KACC50453)의 균사체 배양 특성에 관한 연구 결과 맛버섯 균사체의 배양에 적합한 기본 배지는 ME였으며, 최적 배양 온도는 $25^{\circ}C$, 최적 초기 pH는 5.5였다. 맛버섯 균사체 생장을 위한 최적 배지 조성은 glucose 3%(w/v),malt extrac 0.25%(w/v), yeast extract 0.25%(w/v),$KH_2PO_4$ 0.046%(w/v),$K_2HPO_4$ 0.1%(w/v),$MgSO_4$ $.$$7H_2O$0.05% (w/v)로 나타났다.기본배지(ME)와 최적배지로 액체 배양한 결과 최적 배지에서 균사생장이 양호하였다.

Exopolysaccharide Production and Mycelial Growth in an Air-Lift Bioreactor Using Fomitopsis pinicola

  • For effective exopolysaccharide production and mycelial growth by a liquid culture of Fomitopsis pinicola in an air-lift bioreactor, the culture temperature, pH, carbon source, nitrogen source, and mineral source were initially investigated in a flask. The optimal temperature and pH for mycelial growth and exopolysaccharide production were $25^{\circ}C$ and 6.0, respectively. Among the various carbon sources tested, glucose was found to be the most suitable carbon source. In particular, the maximum mycelial growth and exopolysaccharide production were achieved in 4% glucose. The best nitrogen sources were yeast extract and malt extract. The optimal concentrations of yeast extract and malt extract were 0.5 and 0.1%, respectively. $K_2HPO_4\;and\;MgSO_4{\cdot}7H_2O$ were found to be the best mineral sources for mycelial growth and exopolysaccharide production. In order to investigate the effect of aeration on mycelial growth and exopolysaccharide production in an air-lift bioreactor, various aerations were tested for 8 days. The maximum mycelial growth and exopolysaccharide production were 7.9 g/l and 2.6 g/l, respectively, at 1.5 vvm of aeration. In addition, a batch culture in an air-lift bioreactor was carried out for 11 days under the optimal conditions. The maximum mycelial growth was 10.4 g/l, which was approximately 1.7-fold higher than that of basal medium. The exopolysaccharide production was increased with increased culture time. The maximum concentration of exopolysaccharide was 4.4 g/l, which was about 3.3-fold higher than that of basal medium. These results indicate that exopolysaccharide production increased in parallel with the growth of mycelium, and also show that product formation is associated with mycelial growth. The developed model in an air-lift bioreactor showed good agreement with experimental data and simulated results on mycelial growth and exopolysaccharide production in the culture of F. pinicola.

Effects of Varying Nutritional and Cultural Conditions on Growth of the Ectomycorrhizal Fungus Pisolithus tinctorius SMF

  • The culture conditions and nutritional requirements for enhanced mycelial growth of the ectomycorrhizal fungus P. tinctorius SMF were determined in flask scale experiments. Optimum culture conditions for growth of P. tinctorius SMF in a further modified Melin-Norkrans broth were as follows; temperature 25~$27^{\circ}C$, agitation 120 rpm, and pH 4.0. P. tinctorius SMF utilized various carbon sources including monosaccharides, disaccharides, and polysaccharides. D-Glucose and mannitol were respectively the first and second most suitable carbon sources for mycelial growth. With D-Glucose as the principal carbon source, supplementation of modified Melin-Norkrans (MMN) broth with Lysine (800 mg/l), Glutamic Acid (500 mg/l), or Proline (50 mg/l) enhanced mycelial yields 63%, 34%, and 22% respectively as compared to growth in medium lacking amino acids. ThiaminㆍHCl+biotin+pyridoxine supplementation also enhanced growth. As compared to mycelial growth in the MMN medium, growth of P. tinctorius SMF was enhanced 120% in MMN broth when the carbon/nitrogen ratio was 25/1 in citrate buffer at pH 4.5, and growth was 50% greater in MMN broth of carbon/nitrogen ratio with a 10/1~20/1 without using the buffer. Standard conditions established for growth of P. tinctorius SMF in MMN broth were 25~$27^{\circ}C$, agitation 120 rpm, buffered to pH 4.0 with citrate, in MMN medium containing 10 g/l D-glucose supplemented with 800 mg/l lysine. In this medium the carbon/nitrogen ratio was 20/1~25/1, and the maximal mycelial yield ($Y_{x/s}$ ) was 0.472 (4.72 mg/ml) after 7 days of incubation, as compared to 0.214 (2.14 mg/ml), when the fungus was grown in standard MMN broth.
