Mutagenicity of Lithium Carbonate Assessed by Bacterial Reverse Mutation(Ames) Test (미생물복귀돌연변이(Ames)시험을 통한 탄산리튬의 변이원성 고찰)

  • Objectives: To evaluate the mutagenicity of lithium carbonate, a bacterial reverse mutation(Ames) test was carried out using four strains of S. typhimurium(TA1535; TA1537; TA98; and TA100) and one strain of E. coli(WP2uvrA). Materials: This was carried out in a dose range from 312.5 to $5,000{\mu}g/plate$ in triplicate with and without S9 activation, which is the most commonly used metabolic activation system supplemented by a post-mitochondrial fraction prepared from the livers of rodents treated with enzyme-inducing agents such as Aroclor 1254 or a combination of phenobarbitone and ${\beta}$-naphthoflavone. Results: No significant increases in the number of revertants were observed under the conditions examined in this study. Conclusions: Based on the above observations, it can be concluded that lithium carbonate has no mutagenic activity. Despite the results, it can have an effect by inducing acute oral toxicity, eye irritation and acute aquatic toxicity. Based on this study, we suggest that future studies should be directed toward chronic, carcinogenic testing and other related areas.

Purification and the Catalytic Site Residues of Pseudonomas fragil Lipase Expressed in Escherichia coli

  • The P. fragi lipase overexpressed in E. coli as a fusion protein of 57 kilodalton (kDa) has been purified through glutathione-agarose affinity chromatography by elution with free glutathione. The general properties of the purified GST-fusion protein were characterized by observing absorbance of released p-nitrophenoxide at 400 nm which was hydrolyzed from the substrate p-nitrophenyl palmitate. The optimum condition was observed at 25 $^{\circ}C$, pH 7.8 with 0.4 ${\mu}g$ of protein and 1.0 mM substrate in 0.6% (v/v) TritonX-100 solution. Also the lipase was activated by Ca+2, Mg+2, Ba+2 and Na+ but it was inhibited by Co+2 and Ni+2. pGEX-2T containing P. fragi lipase gene as expression vector was named pGL191 and used as a template for the site-directed mutagenesis by sequential PCR steps. A Ser-His-Asp catalytic triad similar to that present in serine proteases may be present in Pseudomonas lipase. Therefore, the PCR fragments replacing Asp217 to Arg and His260 to Arg were synthesized, and substituted for original fragment in pGL19. The ligated products were transformed into E. coli NM522, and pGEX-2T harboring mutant lipase genes were screened through digestion with XbaI and StuI sites created by mutagenic primers, respectively. No activity of mutant lipases was observed on the plate containing tributyrin. The purified mutant lipases were not activated on the substrate and affected at pH variation. These results demonstrate that Asp217 and His260 are involved in the catalytic site of Pseudomonas lipase.

The Possible Protective Role of Korean Ginseng on Ochratoxicosis: with Special References on Chromosomal Aberrations in Rats.

  • Ochratoxin A (OA) is a potent mycotoxin causing considerable health hazard and economic loss- e,i. OA is of concern as it is hepato-nephrotoxic, mutagenic, and carcinogenic to a great variety of animals. LDso of crude OA was 8.5 mgf kg.b.w., i.p. The clinical symptoms, mortalities and necropsy were recorded in rats injected with OA (LD5o, i.p.) during 10 days of daily treatment. Ginseng treatments (20 mg 1 kg. b.w., i.p.) : before, mixed with, or after OA dose, completely prevented the mortality in rats. OA-treated animals showed microcytic normochromic anaemia, lucocytosis, hypoproteinaemia and elevation of serum ALT, AST, AP, urea, and creatinine values. These findings were declined near the normal levels when ginseng injected with OA. OA (115 LDso) induced chromosomal aberrations (65.66%) compared to the control. When ginseng given 10 min before OA injection, chromosomal aberrations were reduced to be 31.66% compared to OA-treated animals. In conclusion: ginseng has a protective effect against ochratoxicosis, it has anti-genotoxic activity and it can repair the chromosomal damage induced by ochratoxin A. Biological Activities of Roasted Chicory Root (볶음 치커리의 생리활성)

  • This study was carried out to investigate physiological activities of chicory root (Cichorium intybus L. var. sativus). The anti-hepatotoxic activity of roasted chicory was studied using primary cultured rat hepatocytes where cytotoxicity was induced by galactosamine. The water extract of roasted chicory did not induced of cytotoxicity in primary cultured rat hepatocytes. Treatment with 5 mM galactosamin for 5.0 hr showed maximum increase in activity of lactic dehydrogenase (LDH) released in the medium. The water extract of roasted chicory inhibited significantly and dose-dependently the release of LDH activity increased by galactosamine-induced cytotoxity. The antidiabetic activity of water extract of roasted chicory was examined in streptozotocin induced diabetic rats. The increased blood glucose level in the streptozotocin induced diabetic rats was significantly decreased by the administration of chicory extract (800 mg/kg). Chicory water extract (800 mg/kg) prevented weight losses in streptozotocin induced diabetic rats. The antimutagenic activities of chicory water extract were tested using Salmonella thyphimurium YG 1024 as tester strains and 2-aminofluorence as a potent carcinogen in the presence of S-9 mix. No mutagenic activities of the water extracts of roasted chicory were observed on all the tested strains at dose $10{\sim}5,000$ ${\mu}/g$ per plate. Water extract of roasted chicory did not inhibit the mutagencities of Salmonella thyphimurium YG 1024 induced by 2-aminofluorene.

Antimutagenic Activities of Cell Wall and Cytosol Fractions of Lactic Acid Bacteria Isolated from Kimchi

  • Cell wall (lactic acid bacteria-sonicated precipitate ; LAB-SP) and cytosoll(lactic acid bacteria-sonicated supernatant ; LAB-SS) fractions were prepared from kimchi fermenting lactic acid bacteria such as Leuconostoc mesenteroides, Lactobacillus brevis, Lactobacillus fermentum , Lactobacillus plantarum and Pediococcus acidilactici, with Lactobacillus acidophillus isolated from yogurt. Using the Ames mutagenicity test and SOS chormotest system, the antimutagenic acitivity of those cell fractions was studied . One hundered eighty $\mu$l of LAB-SP from lactic acid bacteria isolated from kimchi, excepting Pediococcus acidilactici, supressed the mutagenicity of 4-nitroquinoline-1-oxide(4-NQO) in Ames mutagenicity test and SOS chromotes system , by above 90% and 60% , respectively. LAB-SP from lactic acid bacteria also inhibited the mutagenicity mediated by 3-amino-1-methyl-5H-pyrido [4,3-b]indole (Trp-P-2). Lactobacillus fermentum, Lactobacillus plantarum, and Lactobacillus acidphillus had higher antimutagenicity against Trp-P-2). Lactobacillus fermentum , Lactobacillus plantarum , and Lactobacillus acidphillus had higher antimutagenicity against Trp-P-2 than the other lactic acid bacteria. However, LAB-SS of lactic acid bacteria did not show any mutagenic activity against 4-NQO in Ames mutagenicity test and SOS chromotest systems. On the mutagenicity of MEIQ and Trp-P-2 , LAB-SS of lactic acid bacteria from kimchi or dairy products exhibited a weaker inhibitory effect than LAB-SP of those bacteria. These results represent that, whether the lactic acid bacteria from kimchi are viable or nonviable, antimutagenic acitivity was still effective. We suggest that the strong, antimutaganic activity of lactic acid bacteria might be found in the cell wall fraction , rather than in the cytosol fraction.

Studies on the Anti-apoptotic Effect of the Mudanpi (목단피가 세포고사의 억제에 미치는 영향에 관한 연구)

  • Mudanpi (Cortex Moutan Radicis; the root cortex of Paeonia suffruticosa Andrews) is an important Chinese crude drug used in many oriental prescriptions. 1,2,3,4,6-Penta-O-galloyl-beta-D-glucose (PGG), a major component of this crude drug, has been shown to possess potent antioxidant, anti-mutagenic and anti-proliferative effects. In this study, I examined whether PGG could protect Neuro 2A cells, a kind of neuronal cell lines, from oxidative damage through the induction of HO-1 expression and HO activity. Exposure of Neuro 2A cells to PGG (10-50μM) resulted in a concentration- and time-dependent induction of HO-1 mRNA, and protein expressions and heme oxygenase activity. PGG protected the cells from hydrogen peroxide-induced cell death. The protective effect of PGG on hydrogen peroxide-induced cell death was abrogated by zinc protoporphyrin IX (ZnPP IX), a HO inhibitor. These results indicate that PGG is a potent inducer of HO-1 and HO-1 induction is responsible for the PGG-mediated cytoprotection against oxidative damage.

  • 3-Monochloro-1,2-propanediol (3-MCPD) and 1,3-dichloro-2-propanol (1,3-DCP) are not only produced in the manufacturing process of foodstuffs such as hydrolyzed vegetable proteins and soy sauce but are also formed by heat processing in the presence of fat and low water activity. 3-MCPD exists both in free and ester forms, and the ester form has been also detected in various foods. Free 3-MCPD and 1,3-DCP are classified as Group 2B by the International Agency for Research on Cancer. Although there is no data confirming the toxicity of either compound in humans, their toxicity was evidenced in animal experimentation or in vitro. Although few studies have been conducted, free 3-MCPD has been shown to have neurotoxicity, reproductive toxicity, and carcinogenicity. In contrast, 1,3-DCP only has mutagenic activity. The purpose of this study was to analyze 3-MCPD and 1,3-DCP in various foods using gas chromatography-mass spectrometry. 3-MCPD and 1,3-DCP were analyzed using phenyl boronic acid derivatization and the liquid-liquid extraction method, respectively. The analytical method for 3-MCPD and 1,3-DCP was validated in terms of linearity, limit of detection (LOD), limit of quantitation, accuracy and precision. Consequently, the LODs of 3-MCPD and 1,3-DCP in various matrices were identified to be in the ranges of 4.18~10.56 ng/g and 1.06~3.15 ng/g, respectively.

Detoxification of Aflatoxin B1 Contaminated Maize Using Human CYP3A4

  • Aflatoxin B1 (AFB1) is a mycotoxin produced by Aspergillus flavus (A. flavus). AFB1 is reported to have high thermal stability and is not decomposed by heat treatment during food processing. Therefore, in this study, knowing that AFB1 is metabolized by cytochrome P450 (CYP), our aim was to develop a method to detoxify A. flavus-contaminated maize, under normal temperature and pressure, using Escherichia coli expressing human CYP3A4. First, the metabolic activity of AFB1 by recombinant human CYP3A4 was evaluated. As a result, we confirmed that recombinant human CYP3A4 metabolizes 98% of AFB1. Next, we found that aflatoxin Q1, a metabolite of AFB1 was no longer mutagenic. Furthermore, we revealed that about 50% of the AFB1 metabolic activity can be maintained for 3 months when E. coli expressing human CYP3A4 is freeze-dried in the presence of trehalose. Finally, we found that 80% of AFB1 in A. flavus-contaminated maize was metabolized by E. coli expressing human CYP3A4 in the presence of surfactant triton X-405 at a final concentration of 10% (v/v). From these results, we conclude that AFB1 in A. flavus-contaminated maize can be detoxified under normal temperature and pressure by using E. coli expressing human CYP3A4.

  • 1. Penicillium sp. C 13-13 strain was obtained with the treatment of mutagenic agents(N.T.G.) and by single spore isolation method from the Penicillium sp. C8-14 strain, which was reported in the previous paper. 2. The above strain had a few spores and to obtain seed culture, it was cultured at $30^{\circ}C$ and initial pH $4.5{\sim}5.0$, with air rate 6l/min., and agitation 600 rpm for 48 hours in 10% wheat bran medium in 20l- Jar fermenter. When the broth that had above 70ml of mycelium was inoculated into wheat bran medium and incubated at $29{\sim}33^{\circ}C$ for 72 hrs, the cellulase activity of the koji was higher. 3. Adding calcium chloride and magnecium sulfate to the wheat bran medium to 1.5% and 0.015% respectively, the cellulase activity of the koji was higher than that of the control.

The Antimutagenic Activity of Sulfated Alginate and Its Effect on the Growth of Lactobacillus acidophillus (황 흡착 알긴산의 Lactobacillus acidophillus 성장에 미치는 영향 및 항 돌연변이 작용)

  • The alginate extracted from Hizikia fusiform by the acid alkali soluble alginate (AASA) extraction method was reacted with Na$_2$SO$_4$. The amount of sulfate absorbed in the 3% alginate solution was determined about 18,435 ppm by inductively coupled plasma-atomic emission spectrometry. Both alginate and S-alginate stimulated the growth of Lactobacillus acidophilus in MRS broth and peptone water medium. The addition of S-alginate to the media showed the higher bacterial numbers than alginate supplement. There was no mutagenic activity of S-alginate in the Ames test using the Salmonella Typhimurium TA98 and TA100 strains. The S-alginate showed suppressive effect against 2-NF and MMS of mutagens.
