느타리버섯, 표고버섯 및 양송이의 유리당(遊離糖)과 당(糖)알코올 조성 (Contents of Free-Sugars & Free-Sugaralcohols in Pleurotus ostreatus, Lentinus edods & Agaricus bisporus)

  • 국내에서 널리 인공재배되고 있는 식용버섯인 느타리버섯(Pleurotus ostreatus), 표고버섯(Lentinus edodes) 및 양송이(Agaricus bisporus)의 유리당 및 당알코올을 GCL 법으로 분석하여 버섯의 종류. 부유 및 조기별 차이점을 비교 정토한 결과는 다음과 같다. 3종의 버섯에서 trehalose, glucose, fructose, mannitol, arabitol 및 glycerol등이 확인되었다. 유리당 및 당알코올의 충 함량은 표고버섯 $12.7{\sim}18.9%$, 양송이 $9.2{\sim}16.1%$, 느타리버섯 $8.2{\sim}10.6%$ 순 이었고 표고버섯에서는 mannitol과 arabitol, 양송이에서는 mannitol, 느타리버섯에서는 tehalose와 mannitol이 특징적으로 많이 함유되어 있었다. 부위별로 비교했을때 느타리버섯과 양송이는 자루보다 갓부위에 많았으나 표고버섯은 자루부위에 많았고 크기별로는 크기가 클수록 대체로 유리당 및 당알코올 함량이 감소하는 경향이었다.

느타리버섯, 양송이버섯, 팽이버섯 추출물의 핵산 관련 물질 함량 분석 (Contents of Nucleic Acids(Nucleosides and Mono-Nucleotides) in Extracts of Pleurotus ostreatus, Agaricus bisporus and Flammulina velutipes)

  • 본 연구는 버섯에 정미 성분 및 기능성 성분으로 작용하는 핵산 관련 성분의 분석을 위하여 느타리, 양송이, 팽이버섯을 동결 건조한 뒤 열수 추출 및 효소적 가수분해과정을 거친뒤 HPLC로 정성 및 정량하였다. 연구 결과, 8종류의 핵산 관련 성분의 정성 및 정량 분석을 위한 효과적인 분석 조건을 확립하였으며, 버섯에 함유되어 있는 핵산 관련 성분은 cytidine, uridine, inosine, guanosine, 5'-UMP, 5'-GMP 및 5'-IMP 7종으로 분석되었다. 각각의 핵산 관련 성분 함량 분석 결과는 양송이 ($26.06{\pm}0.01\;mg/g$)>느타리($24.13{\pm}1.01\;mg/g$)>팽이버섯($18.84{\pm}0.03\;mg/g$)의 순으로 나타났으며, 그 중 cytidine 및 inosine 성분이 가장 많이 함유되어 있는 것으로 나타났다.

The effect of mushroom components on the proliferation of HeLa cell line in vitro

  • In order to find out nutritional effects of Korean edible mushrooms on the multiplication of tissue cells, the alcohol extracts and acid bydrolysates of eleven species of mushrooms were added to Earle's ESS. A comparison of the respective multiplications of HeLa cells in this solution and in control solution of TC=199. Yielded the following result: The alcohol extracts and acid hydrolysates of a Coprinus comatur, Agaricus campestris, Agaricus bisporus, Lentinus edodes, Tricholoma matsutake, Pleurotus ostretus, Ramaria botrytis and Pholiota nameko influenced favorably the maintenance of the normal form and monolayer of HeLa cells. The growth curves of HeLa cells in the cultures containing, respectively, the alcohol extracts and acid hydrolysates of these eight mushrooms showed that five species, i.e., Coprinus comatus, Agaricus compestris, Agaricus bisporus, Lentinus edodes and Tricholoma matsutake effected an excellent multiplication and that the other three species were less effective than those five species. As to the effects on the cell multiplication, no marked difference was observed between the alcohol extracts and the acid hydrolysates of the mushrooms tested.

양송이 마이코곤병(病)의 방제약제(防除藥劑)에 관한 연구(硏究) (Studies on the Chemical Control of Mycogone perniciosa Magn in Cultivation of Mushroom Agaricus bisporus(Lang) Sing)

  • Prochloraz of fungicide was applied on Mycogone perniciosa causing wet bubble in cultivated mushroom, Agaricus bisporus. In vitro, Prochloraz was an excellent fungicide on two strains of Mycogone, tolerant and non-tolerant to Benomyl, respectively. At the low dosage, Prochloraz more inhibited mycelial growth of mushrooms than Benomyl. At the higher dosage, Benomyl more inhibited the mycelial growth than prochloraz. The higher yield of sporophore of the mushroom with low inferction rate was abtained from several trial of Prochloraz. Prochloroz was concluded to be effective fungicide on Mycogone perniciosa on Agaricus cultivation.

식용 버섯류의 도파민 베타 수산화효소에 대한 저해활성 검색 (Screening of Inhibitory Activity of Edible Mushrooms on Dopamine ${\beta}-Hydroxylase$)

  • 식용버섯류 중 국내에서 재배되고 있는 식용버섯류에 대한 생리활성 검색의 일환으로 영지, 양송이, 표고 등 3종의 버섯을 대상으로 dopamine ${\beta}-hydroxylase(DBH)$에 대한 저해활성을 tyramine을 기질로 하여 검색하였다. 각 버섯들의 DBH에 대한 저해활성은 영지버섯은 클로로포름 분획에서 100% 이상의 현저한 저해활성을 나타내었으며 표고와 양송이는 에틸아세테이트 분획에서 각각 79.7%라 64.7%의 저해활성을 나타내었다. 한편, 이들 버섯류의 각 용매분획에 대하여 DBH 저해활성을 검색하여 각 분획의 $IC_{50}$을 구하고 용매분획 총량의 specific activity를 비교한 결과, 영지버섯의 클로로포름분획의 $IC_{50}$$1.60{\times}10^{-4}\;g$으로 DBH에 대한 저해활성을 나타내었고 양송이와 표고버섯의 경우 에틸아세테이트 분획의 $IC_{50}$$5.50{\times}10^{-4}\;g,\;2.35{\times}10^{-4}\;g$으로 각각 검색되었다.

Occurrence of Internal Stipe Necrosis of Cultivated Mushrooms (Agaricus bisporus) Caused by Ewingella americana in Korea

  • The internal stipe necrosis of cultivated mushrooms (Agaricus bisporus) is caused by the bacterium Ewingella americana, a species of the Enterobacteriaceae. Recently, Ewingella americana was isolated from cultivated white button mushrooms in Korea evidencing symptoms of internal stipe browning. Its symptoms are visible only at harvest, and appear as a variable browning reaction in the center of the stipes. From these lesions, we isolated one bacterial strain (designated CH4). Inoculation of the bacterial isolate into mushroom sporocarps yielded the characteristic browning symptoms that were distinguishable from those of the bacterial soft rot that is well known to mushroom growers. The results of Gram stain, flagellal staining, and biochemical tests identified these isolates as E. americana. This was verified by pathogenicity, physiological and biochemical characteristics, and the results of an analysis of the 16S rRNA gene sequences and the fatty acids profile. This is the first report of the isolation of E. americana from cultivated white button mushrooms in Korea.

버섯 피클(Fickle) 제조 및 저장 중의 품질변화 (The Preparation of Mushroom Pieties and Change in Quality during Storage)

  • The mushroom pickles were prepared as a processed mushroom product. The changes in pH, total acidity, brix. color and texture of the mushroom pickles (Lentinus edodes, Pleurotus ostreatus, Agaricus bisporus and Pleurotus eryngii) were measured during storage at 4$^{\circ}C$. In addition, sensory evaluations on the mushroom pickles were performed. While the pH of Lentinus edodes and Pleurotus ostreatus pickles declined slowly, the pH of Agaricus bisporus and Pleurotus eryngii pickles declined rapidly during storage. The total acidity of Lentinus edodes pickles rose slowly, while the acidity Pleurotus ostreatus, Agaricus bisporus and Pleurotus eryngii pickles rose rapidly. As for the color of mushroom pickles, "L" , "a" and "b" values changed slightly. The changes in texture were determined by a rheometer Hardness, cohesiveness, springiness, gumminess and brittleness of Lentinus edodes pickles were lower than those of the other mushroom pickles. In the sensory evaluation of mushroom pickles, Lentinus edodes showed higher acceptability ratings than the other mushrooms in taste, color, clearness, texture and overall acceptability. This study showed that among the mushrooms tested, Lentinus edodes was the most acceptable mushroom pickle for storage.

양송이 버섯의 MAP및 CA저장 효과 (Effect of M.A.P. and C.A. storage on quality of Mushrooms(Agaricus bisporus) during storage)

  • In order to study the effect of modified atmosphere packaging(M.A.P.) and controlled atmosphere(C. A.) storage on keeping freshness of mushrooms (Agricus-bisporus). Mushrooms was packaged with polyethylene(P.E.) film (40${\mu}{\textrm}{m}$, 60${\mu}{\textrm}{m}$, 80${\mu}{\textrm}{m}$) and C.A. conditions(CO2 concentration of 2%, O2 concentration of 2%) and storage at 0$\pm$1$^{\circ}C$, RH(relative hummidity) 92$\pm$1%. Gas composition in film was changed rapidly at early of storage, but it kept a constant level after 14 days of storage, and then kept at the level of 6~9% CO2 and 2~5% O2. Weight loss was 5% in non-packed mushrooms after 3 days of storage, but P.E. film, CO2 treated, C.A. conditions were 5% after 21 days of storage. Flesh firmness of the mushrooms was continuously decreased throughout storage period and the lowest of flesh firmness changed was 80${\mu}{\textrm}{m}$-P.E. film packing. Discoloration of the piteous of mushrooms appears to be the most Important factors to determine its marketability, L value of it appears to be of the high values at P.E. film packing and C.A. conditions during period. Large amount of ethanol and acetaldehyde were produced from the 7 days during storage, large contents of mannitol and trehalose were at the 14 days and 7 days during storage.

양송이버섯의 포장실태조사 및 예냉처리가 품질에 미치는 효과 (Survey on Packaging Status and Effects of Precooling on the Quality of $Agaricus$ $bisporus$)

  • 양송이버섯의 유통 시 문제점 및 포장실태를 파악하기 위하여 2011년 5월~9월에 양송이농가, 백화점, 대형할인마트 및 슈퍼마켓 등을 직접 방문하여 유통 중 포장실태를 조사하였다. 양송이버섯 농가에서는 수확 당일 약 2 kg씩 EPS 및 골판지 상자를 이용하여 유통하였으며 백화점, 대형할인마트 및 슈퍼마켓에서는 필름 및 용기를 사용하여 자체 소포장을 하여 슈퍼마켓을 제외한 곳에서 모두 상온에서 판매되고 있었다. 이러한 실태조사 내용을 참고하여 양송이버섯의 예냉실험을 실시한 결과 예냉처리 하지 않은 경우보다 예냉처리를 한 양송이버섯의 중량감소율 및 색도의 변화가 적었으나 예냉처리에 따른 경도의 차이는 크게 없었다. 예냉처리에 대하여 포장과 예냉의 순서를 달리한 경우 선 포장, 후 예냉처리를 한 양송이버섯에서 품질변화가 더욱 적어 효과적이었으며 앞으로 양송이버섯의 장기선도유지를 위하여 새로운 소포장기술 등의 연구가 추가로 진행될 필요가 있다고 사료된다.

Genotyping of Agaricus bisporus Strains by PCR Fingerprints

  • Agaricus bisporus, commonly known as the button mushroom, is the most widely cultivated species of edible fungi. Low frequency of recombination ratio and homokaryotic or monokaryotic spore on meiotic basidia form obstacles for breeding programs. Since the first hybrid varieties for white button mushrooms were released in Europe, new varieties released afterwards were either identical of very similar to these first hybrids on morphologies. Therefore, different DNA markers have been used to define unique varieties of A. bisporus strains. Aim of this study is to assess the genetic diversity of different A. bisporus strains in Korea. Twelve UFP (Universal fungal primer, JK BioTech. Ltd), 12 simple sequence repeat (ISSR) and 30 SSR primers were used to assess genetic diversity of monokaryotic and dikaryotic Agaricus bisporus strains including other 19 Agaricus spp. Of them, four UFP, four SSR primers, $(GA)_8T$, $(AG)_8YC$, $(GA)_8C$ and $(CTC)_6$ and seven SSR markers produced PCR polymorphic bands between the Agaricus species or within A. bisporus strains. PCR polymorphic bands were inputted for UPGMA cluster analysis. Forty five strains of A. bisporus are genetically clustered into 6 groups, showing coefficient similarity from 0.75 to 0.9 among them. In addition, genetic variations of monokaryotic and dikaryotic Agaricus bisporus strains were partially detected by PCR technologies of this study. The varieties, Saea, saedo, Saejeong and Saeyeon that have recently been developed in Korea were involved in the same group with closely genetic relationship of coefficient similarity over 0.96, whereas, other strains were genetically related to A. bisporus strains that were introduced from USA, Eroupe and Chinese.
