The partial nucleotide sequences of the genomic dsRNA mycoviruses infecting Pleurotus ostreatus (isolates ASI2596, ASI2597, and Bupyungbokhoe) and Agaricus blazei Murrill were determined and compared with those of the other dsRNA mycoviruses. Partial nucleotide sequences of the purified dsRNA from ASI2596 and ASI2597 revealed RNA-dependent RNA polymerase sequences that are closely related to Oyster mushroom isometric virus 2, while nucleotide sequences and the deduced amino acid sequence from dsRNA mycovirus infecting Agaricus blazei did not show any significant homology to the other dsRNA mycoviruses. Specific primers were designed for RT-PCR detection of these dsRNA viruses and were found to specifically detect each dsRNA virus. Northern blot analysis confirmed the homogeneity of RT-PCR products to each purified dsRNA. Altogether, our results suggest that these virus-specific primer sets can be employed for the specific detection of each dsRNA mycovirus in infected mushrooms.

The primary objective of the present study is the characterization of the hybrids of monokaryon- monokaryon (mono-mono) crosses in mushroom breeding. We employed this technique for developing superior strains from Pleurotus species strains with 85 mono-mono intraspecific hybrids of 7 combinations between six Pleurotus ostreatus strains and one Pleurotus florida strain. In this study, the results of analysis on hybridization rate, nuclear DNA patterns, and colors and yields of fruit-bodies, are presented as follows. The crossability between mono-mono crossing ranges between 50 and 93.75%. The results of the analysis on the nuclear DNA patterns of 85 hybrid strains of mono-mono crosses share the nuclei of both parents, but their genetic similarities were predominated by either parent. The hybrid strain between P. florida and P. ostreatus showed patterns more similar to P. florida, while the hybrid strain between P. ostreatus and P. ostreatus either had patterns predominated by either parent strain. The fruiting body colors of the mono-mono crosses mostly had combined colors of both parents but showed the tendency of being more similar to that of either parent. 82% of the hybrid strain indicated similar fruiting body yields compared to both parent strains, while 0% was higher and 18% were lower than both parents. The present study was able to find out and suggest superior hybrid trains by identifying the nuclear DNA patterns of hybrids between Pleurotus species as well as the characteristics of their fruiting bodies. This study expects that the advantages of the mono-mono crossing are needs to be fully utilized in mushroom breeding and it is better to develop superior strains of Pleurotus species strains together with the mono-mono crossing.

The primary objective of the present study is the characterization of the somatic hybrids of dikaryon-monokaryon (di-mono) crosses in mushroom breeding. We employed this technique for developing superior strains from Pleurotus ostreatus strains with 48 intraspecific hybrids of 12 combinations between six P. ostreatus strains and one P. florida strain. The results on the experiments of hybridization rate, nuclear DNA patterns, and colors and morphology of fruit-bodies, are presented. In di-mono crosses, somatic hybrids among Pleurotus strains showed 100% of crossability as seen in those between P. ostreatus and P. florida strains indicating that the nuclei of a dikaryon is inferred to be migrated to a recipient. 87.5% of the somatic hybrids among Pleurotus strains were similar to the donor dikaryons, and 12.5% of the somatic hybrids presented DNA patterns of both parents. In 16.6% of di-mono crosses between P. ostreatus and P. florida, the nuclear DNA patterns of all hybrids showed the same or similar patterns compared to the donor dikaryons. 70.9% of the hybrids between P. ostreatus and P. ostreatus were similar to the donor dikaryons and 12.5% of them presented the DNA patterns of both parents. 79.2% of fruiting body morphology of the hybrids among Pleurotus strains were similar to the dikaryons and 20.8% of them were similar to both parents. Interestingly, the morphology of all dikaryons were dissimilar each other. All hybrid strains between dikaryon P. florida and monokaryon P. ostreatus showed the fruiting body of which colors were similar to those of the dikaryon, while the hybrids between dikaryon P. ostreatus and monokaryon P. florida were showed the combined colors of both parents. Therefore, the fruiting body color of P. florida tends to be generally dominant. In conclusion, the present study provides a way to find out and suggest superior hybrid strains using the nuclear DNA patterns of hybrids between Pleurotus strains as well as the characteristics of their fruiting bodies. The advantages of the di-mono crossing are needs to be fully utilized in mushroom breeding because it is an ideal way to develop the superior strains of Pleurotus.

In order to breed a new P. eryngii cultivar with high temperature tolerance trait to cope with climate change, strains and cultivars were characterized at $20^{\circ}C$ that is $5^{\circ}C$ higher than normal condition followed by screening for the characteristics such as required days to harvest, quality and yield. Monokaryons from the selected strains were crossed. $Da-32{\times}KNR2322-15$ derived from the crosses between KNR2322 having characteristics of short growing day and Da($Ga5Na5-4{\times}KNR2312-7$) having charateristics of high guality and yield at $20^{\circ}C$, showed 14.9 days for harvest, 120.6 g yield, and 7.0 quality in the first trial. The strains were named as Taeyangsongi and cultivated on a large scale to compare with Kenneutari No. 2 at a mushroom farm. Yield of Taeyangsongi (109 g) was significantly different(P=0.001) from Kenneutari No.2. Quality of the new (6.6) and the reference cultivar (3.5) was also statistically different (P=0.001) The brightness of pileus of Taeyangsongi (59.5) was 10 points less than the reference cultivar due to which it has an inability to bear high temperature stress. Thus, PCR reactions with URP2 discriminated between Taeyansongi and reference cultivars (Keneutari No. 2 and Aeryni).
For the breeding purpose, in this study we attempted to select a new Lentinula edodes strain by comparing mycelial growth rate, fruit-body quality and mushroom productivity per substrate weight of hybrid strains with parent strains. Nineteen hybrid strains were made by cross of di-karyotic and mono-karyotic to develop strains suitable for sawdust cultivation of L. edodes. Among the nineteen strains, ten were selected by comparing mycelial growth rate on PDA media and wood rot on sawdust-based substrates. The most mushroom harvesting was achieved by 100 days incubation from KFRI 960, KFRI 961, KFRI 962, KFRI 963, KFRI 964, KFRI 971, KFRI 973 and 125 days from KFRI 968, 970. Experiment of incubation periods with the ten strains showed that there were productivity differences between 100 and 150 days incubations. Through comparison of hybrid strains' mushroom productivity it was found that there was no difference in mushroom productivity through crossing high temperature types with high temperature types, crossing low temperature types with high temperature types and crossing unknown temperature types with high temperature types, but there was difference through crossing middle temperature types with high temperature types. Hybrid strains showed better productivity compared with parent strains. Fruiting temperature type analysis of hybrid strains confirmed that strains of high temperature types have a dominant character.

Worldwide production of mushrooms has been increasing by 10-20% every year. Recently, Pleurotus eryngii and P. nebrodensis have become popular mushroom species for cultivation. In particular, China exceeded 8.7 million tons in 2002, which accounted for 71.5% of total world output. A similar trend was also observed in Korea. Two kinds of mushrooms-Gumji (金芝; Ganoderma) and Seoji-are described in the ancient book 'Samguksagi' (History of the three kingdoms; B.C 57~A.D 668; written by Bu Sik Kim in 1145) during the Korea-dynasty. Many kinds of mushrooms are also described in more than 17 ancient books during the Chosun-dynasty (1392~1910) in Korea. Approximately 200 commercial strains of 38 species of mushrooms were developed and distributed to cultivators. The somatic hybrid variety of oyster mushroom, 'Wonhyeong-neutari,' was developed by protoplast fusion, and distributed to growers in 1989. Further, the production of mushrooms as food was 199,829 metric tons, valued at 850 billion Korean Won (one trillion won if mushroom factory products are included) in 2015. In Korea, the major cultivated species are P. ostreatus, P. eryngii, Flammulina velutipes, Lentinula edodes, Agaricus bisporus, and Ganoderma lucidum, which account for 90% of the total production. Since mushroom export was initiated in 1960, the export and import of mushrooms have increased in Korea. Technology was developed for liquid spawn production, and automatic cultivation systems led to the reduction of production cost, resulting in the increase in mushroom export. However, some species were imported owing to high production costs for effective cultivation methods. In academia, RDA scientists have conducted mushroom genome projects since 1997. One of the main outcomes is the whole genome sequencing of Flammulina velutipes for molecular breeding. With regard to medicinal mushrooms, we have been conducting genome research on Cordyceps and its related species for developing functional foods. There are various kinds of beneficial substances in mushrooms; mushroom products, including pharmaceuticals, tonics, healthy beverages, functional biotransformants, and processed foods have also became available on the market. In addition, compost and feed can likewise be made from mushroom substrates after harvest.

We bred a new strain of Pleurotus eryngii having few number of fruit body per bottle. It's name is 'Saegonji' and it was bred by mating monokaryotic strain isolated from E12-176 and monokaryotic strain 'aerini No.3' in Mushroom Research Institute, Gyonggi province A.R.E.S. The characteristics of a new strain 'Saegonji' are as follows. The optimum temperature for mycelium growth was from 23 to $26^{\circ}C$ on PDA medium and those for the primodium formation and the growth of fruit body were about $15^{\circ}C$. The period from spawn innoculation to harvesting required around 51 days at $20^{\circ}C$. The number of fruit body per bottle was 12.8 and it was 9.3 less than 'Keunneutari No.2'. The length was 135.8 mm. It was longer than 'Keunneutari No.2', The color of cap was white grey, while that of 'Keunneutari No.2' was grey. The yield was about 159 g per bottle(1100cc) and it was same as Keunneutari No.2.
The button mushroom 'A. bisporus' is the most commonly grown mushroom worldwide. The color of 'Agaricus bisporus' is white and brown. Recently, the consumption of brown button mushrooms has been increasing steadily. 'Danseok-1ho' was bred using the multi-spore random mating method and named after Danseok Mountain in Gyeongju, the region where 'A. bisporus' was first grown in Korea. The optimum temperature for mycelial growth was between $20^{\circ}C$ and $30^{\circ}C$, and the mushroom production temperature was between $15^{\circ}C$ and $20^{\circ}C$. The color and shape of 'Danseok-1ho' are medium brown and spherical, respectively. The width and thickness of the pileus are 45.1 mm and 23.5 mm, respectively. The hardness and L of chromaticity of 'Danseok-1ho' are $8,937g/{\Phi}5mm$ and 50.4, respectively. The incubation period is around 20 days and mushroom production takes about 18 days after casing. A typical characteristic of 'Danseok-1ho' is that the individual occurrence rate is high, so mushroom losses are low during harvest.

A new variety of Pleurotus eryngii which is named 'Gonji No. 8' was bred by mating monokaryotic strain isolated from E085D2 and a monokaryotic strain 'aerini No.3' obtained from the Mushroom Research Institute, Gyeonggi-Do A.R.E.S. The characteristics of the new variety 'Gonji No.8' is as follows. The optimum temperature for mycelium growth was from $23^{\circ}C$ to $26^{\circ}C$ on potato dextrose agar (PDA) medium. For the primodia formation and the growth of fruit bodies, the optimum temperature was from $14^{\circ}C$ to $16^{\circ}C$. The period of spawn running was around 30 days at $22^{\circ}C$ and the days taken after the removal of the spawn layer to initiate primodia was seven days. The hardness value of fruit body was $8,432{\pm}2,193$$g/cm^2$, which was two times more than that of 'Keunneutari No.2'. The yield of 'Gonji No.8' was about 133 g per bottle(900cc) and it was same as 'Keunneutari No.2'.

The purpose of this study was to breed a variety of stable productivity and high storage characteristics of white Hypsizygus marmoreus, which has high demand at domestic and global market due to a low bitter taste and the preference for white species. Accordingly, 'Baekmaru' was bred by hybridizing brown and white cap color species and backcrossing with white species. The 'Baekmaru' variety was bred by crossbreeding the brown and white species and backcrossing the white species. Through repeated cultivation of 'Baekmaru', a variety with a low contamination rate was selected when culturing the spawn for stable cultivation. As a results of demonstration test, the yield of 'Baekmaru' was 14% higher than that of the commercial variety. In addition, among the fruiting body characteristics of 'Baekmaru', diameter and thickness of the pileus were 16.43±15.27mm and 6.46±0.58mm, which were slightly higher than the commercial variety, and the hardness was 2.69±0.89N for the pileus and 3.09±0.89N for the stipe. The shelf life showed less change in thickness and color of pileus compared to commercial variety. The hardness of pileus of 'Baekmaru' was maintained in the range of 3.5 to 4.0N in the 4℃ and 4℃ and 20℃ mixed treatment until 30 days of storage, and was higher than that of the commercial variety. Therefore, it was suggested to be excellent in storability.
