외부 자극에 의하여 세포 내 $Ca^2$$^{+}$이 증가하면 세포 내 $K^{+}$이 유출되어 세포 외 $K^{+}$ 농도는 수 mM 범위에서 증가할 수 있다. 이러한 세포 외 $K^{+}$의 증가가 혈관 수축성에 미치는 영향을 규명하고자, 세포 외 $K^{+}$가 혈관평활근 수축성, 내피세포 의존성 이완과 혈관내피세포 $Ca^2$$^{+}$ 농도에 미치는 영향을 알아보고자 하였다. 토끼에서 분리한 경동맥, 상장간막동맥 분지, 기저동맥과 쥐의 대동맥에서 등장성 수축을 기록하였으며 배양한 쥐의 대동맥 혈관내피세포와 인간 제대정맥 내피세포에서 세포 내 $Ca^2$$^{+}$ 변화를 측정하였다. 세포 외 $K^{+}$ 농도를 6에서 12 mM로 증가하는 경우 도관동맥인 토끼 경동맥은 수축성에 변화가 없는 반면 저항혈관인 기저동맥과 상장간막동맥분지는 이완하였다. 이러한 $K^{+}$ 유발 이완은 혈관 종류에 따라 차이가 있었는데 기저동맥에서는 세포 외 $K^{+}$ 농도를 6에서 12 mM로 증가하였을 때보다 세포 외 $K^{+}$ 농도를 1에서 3 mM로 증가하였을 때 더 크게 이완하였으며 상장간막동맥의 분지에서는 반대로 세포 외 $K^{+}$ 농도를 6에서 12 mM로 증가하였을 때 더 크게 이완하였다. 그리고 세포 외 $K^{+}$ 농도를 6에서 12 mM로 증가하였을 때의 이완은 $Ba^2$$^{+}$에 의하여 억제되는 반면 1에서 3 mM로 증가에 의한 이완은 억제되지 않았다. 쥐 대동맥에서도 토끼 경동맥과 동일한 효과가 관찰되었는데 세포 외 $K^{+}$ 농도를 6 mM에서 12 mM로 변화시켜도 norepinephrine혹은 prostaglandin $F_2$$_{\alpha}$에 의한 수축력은 유의한 변화가 없었다. 또한 세포 외 $K^{+}$ 농도를 점차 증가시키는 경우 12 mM 이상 증가가 되면 혈관평활근이 수축하기 시작하였지만 12 mM 이하의 증가에 의해서는 혈관평활근의 수축력은 증가하지 않았다. 한편 쥐 대동맥에서 acetylcholine에 의하여 유발된 내피세포 의존성 이완은 세포 외 $K^{+}$ 농도를 정상 6 mM에서 12 mM로 증가시키면 억제되었다. 한편 배양한 쥐 대동맥 내피세포에서는 acetylcholine 혹은 ATP에 의하여 세포 내 $Ca^2$$^{+}$이 증가하였다. 증가한 세포 내 $Ca^2$$^{+}$은 세포 외 $K^{+}$농도를 6 mM에서 12 mM로 증가시키면 가역적 및 농도 의존적으로 감소하였다. 세포 외 $K^{+}$ 증가에 의한 세포 내 $Ca^2$$^{+}$ 억제 효과는 인간 제대정맥 내피세포에서도 관찰되었다. 그리고 세포 외 $K^{+}$ 증가에 의한 내피세포 의존성 이완의 억제효과는 $Na^{+}$- $K^{+}$ pump 억제제인 ouabain과 $Na^{+}$-C $a^2$$^{+}$exchanger 억제제인 N $i^2$$^{+}$에 의하여 억제되었다. 이러한 실험 결과로 미루어 세포 외 $K^+$의 증가는 저항혈관 평활근을 이완시키는데 그 기전은 혈관 종류에 따라 차이가 있었다. 그리고 세포 외 $K^{+}$의 증가는 혈관내피세포 $Ca^2$$^{+}$을 감소시켜 내피세포 의존성 이완을 억제하는데 이는 $Na^2$$^{+}$- $K^2$$^{+}$pump를 활성화시켜 일어나는 것으로 생각된다.
Bolus intravenous injection of adenosine resulted the temporal decrease of systemic blood pressure and heart rate in the anesthetized rats. Adenosine also resulted the persistent decrease of contractility and heart rate in the isolated spontaneously beating rat right atria. Both of the above inhibition effets of adenosine were increased by the pretreatment of NBI (nitrobenzylthioinosine), whitch is an adenosine transport inhibitor, but decreased by the pretreatment of 8- phenyltheophy1line, which is an adenosine antagonist. In isolated thoracic aorta ring segment of normotensive rats, intact rings were relaxed by adenosine ($42.3{\pm}8.7%$) and ATP ($85.9{\pm}15.8%$) in the concentration of $10^{-4}M$, but rubbed rings were relaxed by adenosine ($35.2{\pm}1.9%$) and ATP ($11.3{\pm}9.0%$) in $10^{-4}M$. After pretreatment of L-NAME (N-Nitro-Larginine methyl ester), which is an NO inhibitor, adenosine-induced relaxation was not affected, but ATP-induced relax ation was significantly inhibited (P<0.01). Meanwhile, adenosine resulted almost same as vasorelaxation in isolated thoracic aorta of SHR comparing to those of normotensive rats. But, vasodilation responses of ATP in intact rings of SHR are significantly inhibited comparing to those of normotensive rats. Adenosine-induced relaxation is attenuated after 8-phenyltheophylline pretreatment, but increased after NBI pretreatment. However, ATP-induced relaxations are not affected by 8-phenyltheophylline or NBI pretreatment. These results suggested that the hypotensive effects of adenosine was due to the decrease of contractile force and heart rate through the A1 receptor and vasodilation are mediated by A2 receptor of the vascular smooth muscle. And, the heart protective and vasodilation effects of adenosine might suggest that it would be useful in the acute treatment of coronary artery disease.
The general pharmacological effects of a new anthracycline anticancer agent, DA-125 $[7-0-(2,\;6-dideoxy-2-fluoro-{\alpha}-L-talopyranosyl)-adriamycinone-14-{\beta}-alaninate{\cdot}HCI]$ were investigated in mice, rats, guinea pigs, rabbits and dogs. Intravenous administration of DA-125 presented no significant effects on the central and peripheral nervous systems of ICR mice except a decrease in the numbers of acetic acid-induced writhing response at a dose of 10 mg/kg. In anesthetized rats and dogs, DA-125 produced a transient depression of blood pressure and an increase in heart rate, but did not affect the peripheral blood flow in the isolated ear vessels of rabbits and the mechanical functions of the isolated hearts of guinea pigs. No significant effects were observed on the gastrointestinal functions and the contractilities of smooth muscle preparations obtained from guinea pig trachea, rabbit ileum, pregnant and non-pregnant uterus and vas deferens of rats. DA-125 Increased the contractility of the isolated ileum of guinea pigs in a dose range of $10^{-6}{\sim}10^{-9}g/ml$, and also increased, but weaker than adriamycin, the vascular permeability in rat skin. DA-125 had no effect on the kallikrein-induced increase in permeability and the permeability of the visceral organs. DA-125 did not adversely affect the liver function and the blood coagulation system, and did not induce hemolysis in vitro. It is concluded from the results that the general pharmachological effects of DA-125 are similar to or weaker than those of adriamycin, and that little adverse effects are anticipated with a therapeutic dose range.
In recent years, experimental evidence have been suggested that melatonin has either contractive or relaxing effects on the vascular smooth muscle in vitro. But the effect of melatonin on the cardiovascular system in vivo had been emphasized about the hypotensive effect. In this work, we found not only hypotensive effect but also hypertensive effect of melatonin in rats and attempted to determine the mechanism of these effects elicited by melatonin. Regadless of concentration, melatonin(0.002~5 mg/kg) produced increase in mean blood pressure (MBP) in 36% (54/150 cases) and decrease in mean blood pressure in 64%(96/150 cases). As a whole melatonin caused an increase or a decrease in MBP without compensatory decrease or increase in heart rate. The melatonin-induced hypertension was abolished by the pretreatment of phenoxybenzamine, a ${\alpha}$-adrenoceptor antagoninst. The melatonin-induced hypotension was abolished by the pretreatment of propranolol, a ${\beta}$-adrenoceptor antagonist, ODQ, a NO-sensitive guanylate cyclase inhibitor, or nifedipine, a L-type $Ca^{2+}$ channel blocker, but not by bilateral cervical vagotomy. The results indicate that melatonin-induced hypertension may be related to ${\alpha}$-adrenoceptor stimulation and melatonin-indued hypotension may be related to ${\beta}$-adrenoceptor stimulation, inhibition of $Ca^{2+}$ channel and/or activation of guanylate cyclase.
Purpose: We investigate the possibility of vessel preservation with glycerol and evaluate the potential of preserved gastroepiploic artery as a tool for the microsurgical practice. Methods: In 5 early gastric cancer patients, IRB(No. C-0603-040-170), we gained gastroepiploic artery specimens(5 segments, about 10 cm) after gastrectomy. Each segment was rinsed in a normal saline and subsequently placed in a bottle, containing 50 mL anhydrous glycerol (87%). The bottles were refrigerated at $4^{\circ}C$, the longest preservation time being 5 months. Results: At first glance, no fragmentation was detected and the surface of vessel seemed mild sclerotic. In histological examination, vascular structures remained intact though preservation with glycerol led to a mild atrophy of the smooth muscle in the tunica media. Especially, we found out the elastic fibers and endothelial lining of the intima were preserved until 5 months. Adequate status for using microsurgical practice was also maintained and leakage test was performed successfully with gentian violet ink. Conclusion: Based on the results obtained in this study, the gastroepiploic artery preservation with glycerol as a training model for microvascular anastomosis is a technically very simple procedure and useful for the novice microsurgeon.
Higenamine은 부자의 활성 성분의 하나로서 아드레나린성 beta-수용체를 통하여 강심효과를 나타내는 것으로 알려졌다. 본 연구는 norepinephrine 또는 potassium에 의한 토끼 페동맥의 수축에 미치는 Higenamine의 영향을 관찰한 것이다. 1. Norepinephrine으로 수축을 이르킨 폐동맥편은 higenamine의 전처치 또는 후처치로 용량의존적으로 이완되었다. 이같은 higenamine의 효과는 propranolol 전처치로 억제되었다. Higenamine의 propranolol에 대한 $pA_2$ 값은 8.25였다. 2. Higenamine의 페동맥편에 대한 효과는 phentolamine 전처치로 억제되지 않았다. 3. Isoproterenol도 norepinephrine에 의한 페동맥편 수축을 이완시켰으며 효력은 higenamine 보다 10배 켰다. 그러나 고농도$(3.3{\times}10^{-6})$의 isoproterenol은 내인성 활성을 보였다. 4. Higenamine과 isoproterenol은 potassium으로 유도된 페동맥편 수축에 대하여 이완효과를 나타내지 않았다. 이상의 결과로 미루어 higenamine은 혈관평활근에 대하여도 아드레나린성 beta-수용체를 통하여 이완효과를 나타낼 것으로 생각된다.
본 연구는 ethanol의 reactive metabolite인 acetaldehyde의 일차 배양혈관 평활근 세포에 대한 독성 발현 양식을 규명하기 위한 연구의 일환으로, prostaglandin 합성과 세포독성과의 관련성 여부를 확인하기 위하여 수행되었다. Acetaldehyde는, 일차 배양 혈관 평활근 세포에서의 prostaglandin 합성을 현저히 증가 시켰으며, 이때, cyclooxygenase activity 는 큰 변화없거나 오히려 감소시키는 경향을 보였다. Cyclooxygenase inhibitor 인 indometancin은 acetaldehyde에 의한 LDH release를 현저히 차단시켰으며, aspirin 및 salicylic acid는 전혀 영향을 주지 못했다. Phospholipase $A_2\;(PLA_2)$ inhibitor로 알려져 있는 dexamethasone은 유의적인 세포 독성 경감 작용을 보이지 못하였으며, Lipoxygenase inhibitor 들인 NDGA, propyl gallate 등은 현저한 독성 경감 효과를 보였다. 이상의 결과로부터 acetaldehyde에 의한 일차 배양 혈과 평활근 세포에서의 prostaglandin 합성 증가는 Cell death에 대한 직접적인 원인이 아님을 추론할 수 있었으며, $PLA_2/lipoxygenase$ inhibitor들의 강력한 세포독성 경감 작용으로 미루어 볼 때, acetaldehyde 에 의한 세포독성은 lipoxygenase 대사 산물 혹은 $PLA_2$의 직접 작용에 기인할 가능성을 확인할 수 있었다.
Diltiazem inhibits calcium channels and Iεads to vascular smooth muscle rεlaxation and negative inotropic and chronotropic effects in the hεart. Diltiazem is almost completely absorbεd after oral administration, but its extent of absolute oral bioavailability is reduced because of considerable first-pass hepatic metabolism. Diltiazem is able to dilate renal vasculature and can increase the glomerular filtration rate and renal sodium excretion. The purpose of this study was to report the pharmacokinetic changes of diltiazem after oral administration of diltiazem, 20 mg/kg, in rabbits coadministered with cimetidine, 20 mg/kg and pretreated twice per day for 3 days at cimetidine dose of 20 mg/kg. The area under the plasma concentration-time curve (AUC) of diltiazem was significantly higher in rabbits pretreated with cimetidine than that in control rabbits (p<0.01), showing about 149% increased relative bioavailability. The peak plasma concentration $(C_{max})$ and elimination half-life of diltiazem were increased significantly (p<0.05) in rabbits pretreated with cimetidine compared with those in control rabbits. This findings could be due to significant reduction of elimination rate constant by pretreated with cimetidine. The effects of cimetidine on the pharmacokinetics of oral diltiazem were more considerable in rabbits pretreated with cimetidine compared with those in control rabbits. The results suggest that the dosage of diltiazem should be adjusted when the drug would be co-administerεd chronically with cimetidine in a clinical situation.
Cationic liposomes have been actively used as gene delivery vehicle because of their minimal toxicity, but their relatively low efficiency of gene delivery is the major disadvantage of these vectors. Recently, cysteine residue incorporation to HIV-1 Tat peptide increased liposomemediated transfection compared with unmodified Tat peptide. Therefore, we designed a novel modified Tat peptide having a homodimeric (Tat-CTHD, Tat-NTHD) and closed structure (cyclic Tat) simply by using the disulfide bond between cysteines to develop a more efficient and safe nonviral gene delivery system. The mixing of Tat-CTHD and Tat-NTHD with DNA before mixing with lipofectamine increased the transfection efficiency compared with unmodified Tat peptide and lipofectamine only in MCF-7 breast cancer cells and rat vascular smooth muscle cells. However, cyclic Tat did not show any improvement in the transfection efficiency. In the gel retardation assay, Tat-CTHD and Tat-NTHD showed more strong binding with DNA than unmodified Tat and cyclic Tat peptide. This enhancement was only shown when Tat-CTHD and Tat-NTHD were mixed with DNA before mixing with lipofectamine. The effects of Tat- CTHD and Tat-NTHD were also valid in the experiment using DOTAP and DMRIE instead of lipofectamine. We could not find any significant cytotoxicity in the working concentration and more usage of these peptides. In conclusion, we have designed a novel transfection-enhancing peptide by easy homodimerization of Tat peptide, and the simple mix of these novel peptides with DNA increased the gene transfer of cationic lipids more efficiently with no additional cytotoxicity.
