Effect of Plant Regulators on Direct Shoot Formation and Bulblet Formation from Flower Stalk Culture of Muscari armeniacum 'Early Giant' (무스카리(Muscari armeniacum 'Early Giant')의 화경 조직으로부터 신초형성과 구형성에 미치는 생장조절물질의 영향)

  • This study was carried out to produce multiple shoots and bulblets from flower stalk tissue cultures of Muscari armeniacum LEICHTLIN ex BAKER 'Early Giant', which were cultured in the half strength Murashige & Skoog's (MS) medium supplemented with auxin, NAA in combination with kinetin, BA, and TDZ, alone and/or. In flower stalk tissue culture, upper part explant was the most suitable as a source of culture material. Direct shoot formation was much more favorable in half strength MS medium supplemented with $0.1mg{\cdot}L^{-1}$ NAA and $1.0mg{\cdot}L^{-1}$ TDZ. On the other hand, bulblet formation was increased when cultured in half strength MS medium added with $0.01mg{\cdot}L^{-1}$ NAA and $0.1mg{\cdot}L^{-1}$, $0.2mg{\cdot}L^{-1}$ kinetin. Acclimatized plant flowered during the second year of the growing period without any phenotypic variations and formed average 1.5 bulblets per mother bulb.

  • This experiment was carried out in order to determine proper plant growth regulators (PGR) and their concentrations for direct shoot induction and somatic embryogenesis from leaf tissue cultures of Muscari armeniacum 'Early Giant'. Direct shoot formation from the leaf explant culture was effective only on a half-strength MS medium containing $0.1mg{\cdot}L^{-1}$ 2,4-D, while embryogenesis was occurred on a half-strength MS medium containing $0.1{\sim}1.0mg{\cdot}L^{-1}$ IPA or without PGR. The regenerated bulblets derived from embryos or shoots were harvested and transplanted into a greenhouse. The sprouting percentage of bulblets obtained from different culture media ranged from 80 to 100% and growth of quality bulblets was enhanced when the bulblets were harvested from the medium containing $0.1mg{\cdot}L^{-1}$ NAA and $1.0{\sim}3.0mg{\cdot}L^{-1}$ IPA.

High frequency somatic embryogenesis through leaf explant-derived callus culture in Muscari armeniacum cv. 'Early Giant' (무스카리 'Early Giant' 잎 절편 유래 캘러스 배양을 통한 고빈도 체세포배 발생)

  • Using calli of $Muscari$ $armeniacum$ cv. 'Early Giant' that is monocotyledonous ornamental bulb crop with increasing demand in Korea, we carried out current studies to establish an in vitro multiple propagation protocol via somatic embryogenesis. We found that soft pale yellow green calli were induced from leaf explants cultured on all media containing 0.1~3.0 $mg{\cdot}L^{-1}$ auxins such as 1-naphthalene acetic acid (NAA) and 2,4-dichlorophenoxyacetic acid (2,4-D). However, induced calli showed vigorous growth only when they further transferred on same media containing 2,4-D, 4-amino-3,5,6-tri-chloropicolinic acid (picloram), or 3,6-dichloro-o-anisic acid (dicamba). Although frequency of somatic embryo induction depended on callus source and PGR composition in somatic embryo induction media, somatic embryogenesis was initiated on surface of proliferated calli after transferring on media with no PGR or 0.01 $mg{\cdot}L^{-1}$ NAA co-supplemented with various cytokinins such as $N^6$-benzylaminopurine (BAP). Highest number of embryo at 9.3 per callus clump was obtained when calli which were grown under 0.1 $mg{\cdot}L^{-1}$ picloram supplementation were sub-cultured on medium with 0.01 $mg{\cdot}L^{-1}$ NAA and 0.5 $mg{\cdot}L^{-1}$ BAP. In addition, morphological characteristics of somatic embryo were categorized into following nine phases: globular, biased heart, biased torpedo, early cotyledonary, middle cotyledonary, late cotyledonary, early sprouting, middle sprouting, and late sprouting embryos.

Selecting Plants for the Extensive Rooftop Greening Based on Herbal Plants (초화류를 중심으로한 관리조방적 옥상녹화용 식물 소재 선정)

  • The purpose of this study was to select herbaceous plants that can be used for presenting various views and biodiversity on the extensive rooftop greening. Experiment plots were constructed in July 2005 on the rooftop of the Administration Building in the Seoul Women's University. For this experiment, planters were used to design rooftop greening. The size of a planter is 500mm${\times}$500mm${\times}$100mm and each planter has the water storage plate in the lower part of it. The soil was constructed by mixing pearlite, vermiculite, cocopeat, and leaf mold in the ratio of 6 : 2 : 1 : 1. The plot was divided into the watered plot and the dry plot. Since each plot was constructed 2 times, finally 4 planters were constructed in total. One hundred species were used for the experiment and 9 plants per species were planted in each planter. Plants were organized according to types of plants and the experiment used 86 native herbaceous plants, 6 herbs, and 8 foreign plants. The plots were monitored once a month, from July to November 2005. The length and width of plants were tape-measured and covering rate was calculated by CAD program. "SPSS 10.1" was used for a statistical analysis. The result showed no significant difference between the watered plots and the dry plots. In cases of some plants, there were statistically significant differences between the watered planter and dry planter such as follows : Astilbe chinensis and Polygonatum odoratum which are shade plants were measured as the highest value on the watered pots, and Aquilegia buergeriana, Chrysanthemum zawadskii, Calendula arvensis and Gypsophila cerastioides D.Don which are sunny plants were measured as the highest value on the dry plots. According to the final analysis of the data collected and observed for growth condition during the first year of the research, 51 species including Prunella vulgaris var. lilacina and Veronica linaiaefolia in native herbaceous plant, 5 species with Lavandula angustifolia in herbs, and 3 species with Lantana camara and Muscari armeniacum in foreign plants showed the highest growth condition. In conclusion, it is suggested that various plants including sedums could be effectively used for extensive rooftop greening to improve landscape(a view) of the rooftop and increase ecological values.
