Spore Inoculum Optimization to Maximize Cyclosporin A Production in Tolypocladium niveum

  • The cyclic undecapeptide, cyclosporin A (CyA), is one of the most commonly prescribed immunosuppressive drugs. It is generated nonribosomally from a multifunctional cyclosporin synthetase enzyme complex by the filamentous fungus Tolypocladium niveum. In order to maximize the production of CyA by wild-type T. niveum (ATCC 34921), each of three culture stages (sporulation culture, growth culture, and production culture) were sequentially optimized. Among the three potential sporulation media, the SSMA medium generated the highest numbers of T. niveum spores. The SSM and SM media were then selected as the optimal growth and production culture media, respectively. The addition of valine and fructose to the SM production medium was also determined to be crucial for CyA biosynthesis. In this optimized three-stage culture system, 3% of the spore inoculum generated the highest level of CyA productivity in a 15-day T. niveum production culture, thereby implying that the determination of an appropriate size of T. niveum spore inoculum plays a critical role in the maximization of CyA production.

  • Polymer nanocomposite is considered a great alternative to solve the limitations that exist in a simple combination material, as well as to produce multifunctional and high-performance results. In this research, PVA/bentonite nanocomposite films were prepared based on the presence or absence of modification of nano-clay(bentonite) a SUPERGEL® product, modification conditions and content, and the structural variation of the prepared PVA/bentonite nanocomposite films were examined. The effect of variations in the internal structure of the nanocomposite on mechanical and thermal properties was investigated. As a result of evaluating the thermal characteristics of the PVA/bentonite nanocomposite film based on the concentration of the modified bentonite, it was verified that the thermal characteristics and stability were improved because of interaction between the polymer and the modified nano-clay.

  • At present, many machine leaning and data mining methods are used for analyzing and predicting structural response characteristics. However, the platform that combines big data analysis methods with online and offline analysis modules has not been used in actual projects. This work is dedicated to developing a multifunctional Hadoop-Spark big data platform for bridges to monitor and evaluate the serviceability based on structural health monitoring system. It realizes rapid processing, analysis and storage of collected health monitoring data. The platform contains offline computing and online analysis modules, using Hadoop-Spark environment. Hadoop provides the overall framework and storage subsystem for big data platform, while Spark is used for online computing. Finally, the big data Hadoop-Spark platform computational performance is verified through several actual analysis tasks. Experiments show the Hadoop-Spark big data platform has good fault tolerance, scalability and online analysis performance. It can meet the daily analysis requirements of 5s/time for one bridge and 40s/time for 100 bridges.

  • Nowadays, the complexity of object tracking models among hardware applications has become a more in-demand duty to complete in various indeterminable environment tracking situations with multifunctional algorithm skills. In this paper, we propose a virtual city environment using AirSim (Aerial Informatics and Robotics Simulation - AirSim, CityEnvironment) and use the DQN (Deep Q-Learning) model of deep reinforcement learning model in the virtual environment. The proposed object tracking DQN network observes the environment using a deep reinforcement learning model that receives continuous images taken by a virtual environment simulation system as input to control the operation of a virtual drone. The deep reinforcement learning model is pre-trained using various existing continuous image sets. Since the existing various continuous image sets are image data of real environments and objects, it is implemented in 3D to track virtual environments and moving objects in them.

  • 코로나 19로 인하여 다중이용시설에 출입 시 정부 지침에 따라 QR코드 스캔, 출입 명부 작성, 체온 측정 등 방역절차를 지켜야한다. 본 연구에서는 방역 절차를 간편화하고 동합한 방역 시스템을 제안한다. QR코드 스캐너를 통하여 출입자의 신상 정보를 확인하며 체온 측정 모듈을 통하여 출입자의 체온을 측정한다. 추가적으로 워터펌프를 통하여 소독제를 분사하며 서보모터를 통하여 출입문을 열고 닫는다. 또한, 산업 현장에서는 알코올 측정 센서를 통하여 작업자의 알코올 수치를 측정하여 음주로 인한 산업사고도 예방한다.

The hepatoprotective effects of silkworm: Insights into molecular mechanisms and implications

  • The liver, a multifunctional organ, plays a vital role in maintaining overall health and well-being by regulating metabolism, detoxification, nutrient storage, hormone balance, and immune function. Liver diseases, such as hepatitis, cirrhosis, fatty liver disease, and liver cancer, have significant clinical implications and remain a global health concern. This article reviews the therapeutic potential of silkworm larvae (Bombyx mori) and explores their underlying molecular mechanisms in protecting against liver diseases. Silkworm larvae are rich in proteins, vitamins, minerals, and n-3 fatty acids, making them a promising candidate for therapeutic applications. The anti-inflammatory mechanisms of silkworm larvae involve modulating the production of cytokine such as TNF-α and interleukins, inflammatory enzymes including cyclooxygenase-2 and macrophage polarization, thereby attenuating liver inflammation. Silkworm larvae also exhibit anti-oxidative effects by scavenging free radicals, reducing intracellular reactive oxygen species and enhancing the liver's antioxidant defense system. Moreover, silkworms have been reported to decrease the serum alcohol concentration and lipid accumulation. Understanding the therapeutic properties of silkworm larvae contributes to the development of innovative strategies for liver injury prevention and treatment. Further research is warranted to elucidate the precise signaling pathways involved in the anti-inflammatory and anti-oxidative effects of silkworm larvae, paving the way for potential therapeutic interventions in liver diseases.

  • Recently subscribers in mobile communication services increase gradually, and the new multifunctional portable terminals can provide various functionalities. However this trend may not be desirable in Green IT respect because the emission of carbon dioxides emitted during operations of base station and mobile stations can cause significant effect on the emissions of carbon dioxides. In this paper the power saving scheme which can adjust slot index while the system is in a paging phase has been proposed and its performance has been evaluated. The slot cycle index can be adjusted according to individual user's property, its effect on the system performance has been investigated. In order to mitigate its adverse effect, power ramping scheme is considered and evaluated thereafter.

  • Ubiquitous city which was appeared since 2006 is defined as "The city provided the ubiquitous city service anytime and anywhere using ubiquitous urban infrastructure to improve the city's competitiveness and quality of life" under ubiquitous city construction law. To realize the ubiquitous city, the city has to be constructed efficiently based on the spatial information. And connection of information achieve effective synergy. In this study, applied multilateral analysis to spatial information and administrative information to implement u-service focused on MACCA(Multifunctional Administrative City Construction Agency). Study about main function and product data with connection process of Korean Land Spatialization System and administrative information System was performed. Also, details of u-service and method of acquiring necessary data were derived. In Addition, the information connection possibility for efficient u-service realization was suggested by linkage analysis method to utilize the related information infrastructure jointly.

Nebulin C-terminus Interacts with NCBP51, a New Isoform of RING Finger Protein 125 (RNF125)

  • Nebulin, a giant modular protein from muscle, is thought to act as molecular ruler in sarcomere assembly. In skeletal muscle, the C-terminal ${\sim}50 kDa$ region of nebulin extends into the Z-line lattice. The most recent studies implicated highlighting its extensive isoform diversity and exciting reports revealed its expression in cardiac and non-muscle tissues containing brain. Also these novel findings are indicating that nebulin is actually a multifunctional filament system, perhaps playing roles in signal transduction, contractile regulation, and myofibril force generation, as well as other not yet defined functions. However the binding protein of nebulin and function in brain is still unknown. A novel binding partner of nebulin C-terminal region was identified by screening a human brain cDNA library using yeast two-hybrid system. Nebulin C-terminus binding protein 51 (NCBP51) was contained a RING-finger domain and identified a new isoform of RING finger protein 125 (RNF125). The interaction was confirmed using the GST pull-down assay. NCBP51 belongs to a family of the RING finger proteins and its function remains to be identified in brain. The role of nebulin and NCBP51 will be studied by loss-of-function using siRNA technique in brain.

  • TOD (Transit-Oriented Development) is one of the urban structure concentrated on the multifunctional space/district with public transportation system, which is introduced for maintaining sustainable future cities. With such trends, the project of building complex transferring centers located at a urban railway station has widely been spreaded and a comprehensive and systematic analytical framework is required to clarify and readily understand the complicated procedure of estimation with the large scale of the project. By doing so, this study is to develop a comprehensive analytical framework for estimating a pedestrian OD matrix using a spatial information and an integrated smart card data, which is so called a data depository and it has been applied to the Samseong station for the model validation. The proposed analytical framework contributes on providing a chance to possibly extend with digitalized and automated data collection technologies and a BigData mining methods.
