Regulation of the Phagocyte Respiratory Burst Oxidase by Protein Interactions

  • The activity of the phagocyte respiratory burst oxidase is regulated by complex and dynamic alterations in protein-protein interactions that result in the rapid assembly of an active multicomponent NADPH oxidase enzyme on the plasma membrane. While the enzymatic activity has been studied for the past 20 years, the past decade has seen remarkable progress in our understanding of the enzyme and its activation at the molecular level. This article describes the current state of knowledge, and proposes a model for the mechanism by which protein-protein interactions regulate enzyme activity in this system.

  • The development of high resolution capillary columns and a large variety of different detectors led to a strong position of gas chromatography in instrumental analysis. Every effort has been made to solve sophisticated separation problems by column switching. Nowadays, several systems are commercially available for this purpose. The principle and the capabilities of multidimensional gas chromatography(MDGC) are illustrated by different applications in the field of modern flavor and essential oil research.

  • Breakthrough curves for separation of cesium and strontium binary components from aqueous solutions in fixed bed by chabazite and 13X were experimentally obtained and simulated the breakthrough curves for binary component adsorption. It is shown that agreements between model predictions and experimental data are relatively good although some deviations are observed. From engineering point of view, the simple model used here can be applied to simulate adsorption breakthrough curves satisfactorily.

  • The study presents the results of the research of heat and heat exchange processes on the heat-stressed elements of the structure of an advanced TsAGI descent vehicle. The studies were carried out using a mathematical model based on solving discrete analogs of continuum mechanics equations. Conclusions were drawn about the correctness of the model and the dependence of the intensity of heat and mass transfer processes on the most heat-stressed sections of the apparatus surface on its geometry and the catalytic activity of the surface.

Sulfate and Freeze-thaw Resistance Characteristic of Multi-component Cement Concrete Considering Marine Environment (해양환경을 고려한 다성분계 시멘트 콘크리트의 황산염 및 동결융해 저항 특성)

  • Recently, concrete using multicomponent blended cement has been required to increase the freeze-thaw and sulfate resistances of concrete structures exposed to a marine environment. Thus, the purpose of this study was to propose the use of concrete containing multicomponent blended cement as one of the alternatives for concrete structures exposed to a marine environment. For this purpose, batches of concrete containing ordinary portland cement (OPC), binary blended cement (OPC-G, G: ground granulated blast slag), ternary blended cement (OPC-GF, F: fly ash), and quaternary blended cement (OPC-GFM, M: mata-kaolin) were made using a water-binder ratio of 50%. Then, the durability levels, including thesulfate and freeze-thaw resistances, were estimated for concrete samples containing OPC, OPC-G, OPC-GF, and OPC-GFM. It was observed from the tests that the durability levels of the concrete samples containing OPC-G and OPC-GF were found to be much better than that of the concrete containing OPC. The optimum mixing proportions were a40% replacement ratio of ground granulated blast slag for the binary blended cement and a30% replacement ratio of ground granulated blast slag and 10% fly ash for the ternary blended cement.

Synthesis of Monolithic Gel to Bulk glass-Ceramic in Multicomponent Li2O-Al2O3-SiO2 System (Sol-Gel법에 의한 Li2O-Al2O3-SiO2계 괴상겔 및 결정화유리의 합성)

  • The purpose of this investigation was to prepare multicomponent monolithic Li-Al-Si gels of composition(mol%) 16.67 Li2O-16.67 Al2O3-66.67 SiO2 and to convert the gels to monolithic glass-ceramic at low temperature without melting. The hydrolysis, DTA, TGA, TMA, SEM, pore distribution, density and the activation energy for crystallization of the glass-ceramic formation with rawmaterials of which tetraethl orhosilicate of networkforming cation(Si) is partially hydrolyzed, aluminum isoproxide and lithium methoxide prepared by Li-metal react with methanol were studied. The results were as follows : 1) Monolithic gels which were added with additional water, resulting in a total water content 2.5 to 3.0 times the stoichiometric amount required to fully hydrolyze the alkoxides. 2) Specimens were dried to form crylinders 60mm in length and 40mm in diameter in about 800 hrs at 5$0^{\circ}C$. 3) $\beta$-eucryptite crystals and $\beta$-spodumene crystals were detected in samples heated above 75$0^{\circ}C$. 4) Within the temperature and range of 25-50$0^{\circ}C$ and 1,00$0^{\circ}C$ the thermal expansion coefficient for crystallized samples were shown as 2.6-5.7$\times$10-7/$^{\circ}C$ and 7.4-12.5$\times$10-7/$^{\circ}C$, respectively. 5) The activation energy for the crystal growth was 11.01kcal/mol at 794$^{\circ}C$ to 85$0^{\circ}C$.

Quantitative Analysis of High-Temperature Mullitization from a Multicomponent Oxide System (다성분 산화물 요업체의 고온 물라이트화 반응 정량분석)

  • Mullitization in a multicomponent oxide system(alumina-kaolin-quartz-feldspar-talc) was studied as a function of sintering temperature from 1200 to 1500$^{\circ}C$ based upon a quantitative X-ray diffraction analysis. In the present study mullite grew as wiskers and its formation reaction showed characteristic there stages as follows In the first stage(1255-1295$^{\circ}C$) an appreciable mullitization(nucleation) occurred while corun-dum dissolution into glass (increasing glass content ) limited the rate of the reaction. At 1295-1335$^{\circ}C$ (second state) the reaction was significantly enhanced with a considerable glass consumption and with no appreciable change in corundum content. Finally (above 1335$^{\circ}C$) the reaction rate was attenuated re-markably with an apparent decrease in glass consumption rate. The impingement of mullite whiskers by oth-er whiskers and crystals was speculated to cause mullite growth in thickness direction with a slow growth rate resulting in the diminished reaction rate in the final stage.

Electron Transfer to Hydroxylase through Component Interactions in Soluble Methane Monooxygenase

  • The hydroxylation of methane (CH4) is crucial to the field of environmental microbiology, owing to the heat capacity of methane, which is much higher than that of carbon dioxide (CO2). Soluble methane monooxygenase (sMMO), a member of the bacterial multicomponent monooxygenase (BMM) superfamily, is essential for the hydroxylation of specific substrates, including hydroxylase (MMOH), regulatory component (MMOB), and reductase (MMOR). The diiron active site positioned in the MMOH α-subunit is reduced through the interaction of MMOR in the catalytic cycle. The electron transfer pathway, however, is not yet fully understood due to the absence of complex structures with reductases. A type II methanotroph, Methylosinus sporium 5, successfully expressed sMMO and hydroxylase, which were purified for the study of the mechanisms. Studies on the MMOH-MMOB interaction have demonstrated that Tyr76 and Trp78 induce hydrophobic interactions through π-π stacking. Structural analysis and sequencing of the ferredoxin domain in MMOR (MMOR-Fd) suggested that Tyr93 and Tyr95 could be key residues for electron transfer. Mutational studies of these residues have shown that the concentrations of flavin adenine dinucleotide (FAD) and iron ions are changed. The measurements of dissociation constants (Kds) between hydroxylase and mutated reductases confirmed that the binding affinities were not significantly changed, although the specific enzyme activities were significantly reduced by MMOR-Y93A. This result shows that Tyr93 could be a crucial residue for the electron transfer route at the interface between hydroxylase and reductase.

Establishment of a BaTiO3-based Computational Science Platform to Predict Multi-component Properties (다성분계 물성을 예측하기 위한 BaTiO3기반 계산과학 플랫폼 구축)

  • Barium titanate (BaTiO3) is considered to be a beneficial ceramic material for multilayer ceramic capacitor (MLCC) applications because of its high dielectric constant and low dielectric loss. Numerous attempts have been made to improve the physical properties of BaTiO3 in response to recent market trends by employing multicomponent alloying strategies. However, owing to its significant number of atomic combinations and unpredictable physical properties, finding a traditional experimental approach to develop multicomponent systems is difficult; the development of such systems is also time-consuming. In this study, 168 new structures were fabricated using special quasi-random structures (SQSs) of Ba1-xCaxTi1-yZryO3, and 1680 physical properties were extracted from first-principles calculations. In addition, we built an integrated database to manage the computational results, and will provide big data solutions by performing data analysis combined with AI modeling. We believe that our research will enable the global materials market to realize digital transformation through datalization and intelligence of the material development process.
