산뽕잎 및 은행잎 차의 항산화 활성과 플라보노이드 함량 (Antioxidant Capacities and Flavonoid Contents of Wild Mulberry and Ginko Leaves Teas)

  • 산뽕잎, 은행잎 및 혼합차의 침출시간에 따른 항산화 활성을 DPPH 라디칼 소거 활성과 FRAP 방법으로 조사하였으며, 총 페놀 화합물의 함량은 Folin-Ciocalteu법, 플라보노이드 함량은 HPLC로 분석하였다. 침출시간에 따라서 DPPH 라디칼 소거활성과 FRAP 활성은 은행잎차, 혼합차, 산뽕잎차 순으로 높게 나타났다. 총 페놀 성분 함량은 은행잎차, 혼합차, 산뽕잎차 순으로 높았으며 산뽕잎에 은행잎을 2:1로 혼합하였을 때 항산화 활성이 증가하였다. 침출시간 3분 정도에서 산뽕잎차 및 혼합차는 비교적 항산화 활성이 높았으며 은행잎차는 6분 까지 활성이 증가하였다. HPLC로 분석한 플라보노이드 성분 함량은 quercetin이 모든 차에서 가장 함량이 높았으며, 은행잎차, 혼합차, 산뽕잎차 순으로 플라보노이드의 총 함량이 높게 나타났다. 제조한 침출차 들의 항산화 성분 함량과 항산화 활성의 상관계수는 0.87 이상으로 상관성이 상당히 높았다.

뽕나무 유전자원의 항산화능 비교 (Anti-oxidative capacity of mulberry genetic resources)

  • Much attention has been focused on the activity of the natural antioxidants present in fruits and vegetables, because potentially these components may reduce the level of oxidative stress. Especially, mulberry leaves containing many natural components are considerable resource for natural antioxidants. The antioxidant capacity of mulberry leaves was investigated with minilum L-100 device and ARAW-KIT (anti-radical ability of water-soluble substance), in comparison to the ascorbic acid. The antioxidant capacity of 16 varieties was 3303.4 nmol at opening stage of five leaves in spring. The highest stage of antioxidant capacity (3708.0 nmol) and yield rate was just before the coloration stage with anthocyanin in fruits, whereas the lowest stage was middle of June (2231.6 nmol) and about two months growing stage after summer pruning (2064.6 nmol). But after summer pruning, the antioxidant capacity of mulberry leaves increased gradually until just before fallen leaves stage. Even if samples were same variety, antioxidant effect of those showed different results according to collected regions. Also, antioxidant effect of mulberry leaves were higher than that of branches. The antioxidant capacity of yield-type mulberry leaves and fruits (Morus alba L., M. bombycis Koidz, and M. Lhou (Ser.) Koidz) collected from In-je, Won-ju and Yang-yang regions, Kang-won province, Korea, was investigated. The results indicated that total antioxidant capacity of yield-type mulberry leaves was 2711.2 nmol. In the antioxidant capacity analysis of Jeollabuk-Do genetic resources, autumn's mulberry leaves showed higher antioxidant capacity than that of spring's it. To investigate the effect of tea on antioxidative capacity, five kinds of tea(coffee mix, green tea added brown rice, mulberry leaf tea, Polygonatum odoratum tea and black tea added lemon) were selected and analyzed. Their's anti-oxidative capacity were 2,531.01 nmol, 1,867.42 nmol, 1,053.72 nmol, 292.71 nmol and 188.91 nmol, respectively. The antioxidative capacity of drinking water soaked with mulberry leaf showed 891.96 nmol.

Flavonoids analysis in leaves and fruits of Korean mulberry cultivar, Baekokwang having white fruits

  • Morus alba has white and/or purple fruits with a very sweet taste and low acidity. Most Korean mulberry trees have purple fruits. However, Baekokwang is a unique mulberry genetic resource in Korea with white fruits. In this study, flavonoids contents of Baekokwang mulberry leaf and fruit were analyzed using ultrahigh performance liquid chromatography coupled with diode array detection and quadrupole time-of-flight mass spectrometry (UPLC-DAD-QTOF/MS) technique. UPLC-DAD-QTOF/MS chromatogram showed that 15 flavonoids and 9 flavonoids were isolated and identified from the mulberry leaf and fruit. Total flavonoids contents of Baekokwang leaves and fruits were 812.7 mg and 35.0 mg, respectively. Baekokwang leaves had 4 major flavonoids including quercetin 3-O-(6"-O-malonyl) glucoside, 235.3 ppm, kaempferol 3-O-(6"-O-malonyl) glucoside, 132.3 ppm, kaempferol 3-O-rutinoside (nicotiflorin), 108.1 ppm, and quercetin 3-O-rutinoside (rutin), 103.8 ppm. Baekokwang fruits had 3 major flavonoids including quercetin 3-O-(6"-O-malonyl) glucoside, 13.0 ppm, quercetin 3-O-rutinoside (rutin), 7.8 ppm, and kaempferol 3-O-rutinoside (nicotiflorin), 5.7 ppm. From the above results, mulberry leaves have rich flavonoids compared to its fruits.

뽕잎추출액코팅농도에 따른 뽕잎쌀밥의 품질에 관한 연구 (The Study on the Characteristic of Cooked Rice According to the Different Coating Ratio of Mulberry Leaves Extracts)

  • The purpose of this study was to assess the optimum coating ratio for rice, using various ratios of mulberry leaves extract, 1.0, 1.5, and 2.0%, and to determine the optimum ratio of added water, in proportion to the total weight of mulberry rice. The moisture content of the soaked rice, and the optimum water uptake rate, moisture content of the cooked rice, as well as its blue and color values, mechanical characteristics, internal structure and sensory evaluation, were analyzed. The statistical data analyses were completed using the SAS program. The results are summarized as follows: The moisture content of mulberry rice was less than that of raw rice. The average optimum water uptake of the soaked mulberry rice at the different water temperatures, 10, 20 and 30, was 20% of the total weight of the raw mulberry rice. As for the results of the sensory evaluation,; 140% water, in proportion to the total weight of raw mulberry rice, was judged to be the optimum. The average moisture content of the cooked mulberry rice was 45∼50%, but there was no significant difference in the various coating ratios. The blue value of the cooked mulberry rice awas highest on the first day of cooking. The L- and a-values decreased with increasing coating ratio, but the b-value increased under the same conditions. As for the mechanical characteristics,; the adhesiveness, hardness and springiness decreased during 2 days of storage. The internal structure of the mulberry rice, observed by SEM, showed a close structure on increasing the coating ratios of mulberry leaves extracts. It was concluded that the optimum coating ratio of mulberry rice and ratio of added water for cooking wereas 1.5 and 140%, respectively, in proportion to the total weight of raw mulberry rice.

뽕잎과 민들레 추출물을 급여한 계육의 저장성에 미치는 영향 (Effects of Dietary Supplementation of Mulberry Leaves and Dandelion Extracts on Storage of Chicken Meat)

  • 본 시험은 육계에 뽕잎과 민들레 추출물을 각각 1%와 2%를 급여하여 5주간 사육한 계육의 pH, 육색, TBARS 및 VBN을 조사하였다. 실험구는 뽕잎과 민들레 추출물을 급여하지 않은 대조구, 뽕잎 추출물 1% 급여구는 T1, 뽕잎 추출물 2% 급여구는 T2, 민들레 추출물 1% 급여구는 T3 그리고 민들레 추출물 2% 급여구를 T4 등 5개 처리구로 나누어 사육한 후 도계하여 4주간 냉장 온도($4{\pm}1^{\circ}C$)에서 저장하면서 실험하였다. 계육의 pH는 저장 기간이 경과하면서 감소하였고(P<0.05), 대조구에 비해 뽕잎과 민들레 추출물 급여구에서 낮았으며, 특히 민들레 2% 급여구인 T4에서 가장 낮았다(P<0.05). 가슴살의 육색 중 $L^*$값과 $a^*$값은 저장 기간이 지남에 따라 일정한 변화가 없었고 $b^*$값은 증가하였으며, 다리살의 $L^*$값은 감소하였고, $a^*$값은 증가하였다. TBARS는 저장 기간이 지나면서 모든 처리구에서 증가하였고(P<0.05), 가슴살과 다리살에서 대조구보다 뽕잎과 민들레 추출물 급여구에서 낮았으며, 뽕잎과 민들레 추출물 급여에 의한 차이는 없었다. VBN 함량은 계육을 저장하는 동안 증가하였고, 대조구에 비해 뽕잎과 민들레 추출물 급여구에서 낮았으며(P<0.05), 뽕잎과 민들레 추출물 급여에 의한 차이는 없었다. 이상의 결과를 종합적으로 고찰해 보면 뽕잎과 민들레 추출물을 계육에 급여하면 pH, TBARS 및 VBN 함량이 낮아져 계육의 저장성 개선에 도움이 되리라 사료된다.

산란계 사료에 뽕잎 분말 첨가 급여가 산란율, 계란 품질 및 혈액 성상에 미치는 영향 (Effects of Dietary Supplementation of Powdered Mulberry Leaves on Egg Production, Egg Quality and Blood Characteristics in Laying Hens)

  • 본 시험은 산란계에 뽕잎 분말의 급여량(0, 1, 2 및 5%)과 급여 기간에 따라 사육한 산란계의 계란 품질, 혈액 생화화적 분석 및 혈액학적 지표 분석를 조사하였다. 실험구는 뽕잎 분말을 첨가 급여하지 않은 처리구를 대조구, 뽕잎 분말 1% 급여구는 T1, 뽕잎 분말 2% 급여구는 T2 그리고 뽕잎 분말 5% 급여구를 T3 등 4개 처리구로 나누어 실시하였다. 산란계의 산란율과 계란의 난황, 난각 무게는 뽕잎 분말의 급여량과 급여 기간에 의한 유의적인 변화는 없었다. 그러나 난백 무게는 뽕잎 분말의 급여량과 급여 기간이 지남에 따라 감소하였다(P<0.05). 그리고 난백 높이, 난황 색, 난각 두께 및 난각 강도는 뽕잎 분말의 급여에 의한 차이는 없었다. 산란계의 혈액 콜레스테롤 성상 중 HDL-cholesterol은 뽕잎 급여량이 많고 급여기간이 길어질수록 증가하였고, 총콜레스테롤과 triglyceride는 낮아지는 결과였으며(P<0.05), 뽕잎분말을 5% 급여할 경우 유의한 변화를 보였다. 또한, 혈액 내 WBC 함량이 뽕잎 분말 급여구에서 높아지는 결과이었다. 결론적으로 뽕잎 분말 5%를 급여하면 총콜레스테롤과 triglyceride는 감소하고, HDL-cholesterol, glucose 및 WBC 함량이 증가되어 혈액 성상을 개선할 가능성이 있는 것으로 판단된다.

뽕나무의 생육특성과 엽질과의 관계 (Relation Between the Growth Habitat and the Leaf Quality of Cultivated Mulberry Plants)

  • Mulberry plants cultivated in mid-Korea were studied on the growth habitat, canopy type, productive structure and vertical light intensity in relation to the quality of mulberry leaves for sericulture. The growth in length of new branch of summer cut mulberry in spring was vigorous on the uppermost of old shoot. Numbers of leaves on a branch were barely 4 leaves on lower than 20th branch from base, but those were 13 leaves on higher than 24th branch. Area of leaves completely expanded was broad on high branch, but narrow on low one. The form of canopy, the type of productive structure and the vertical light distribution depend upon varieties of cultivated mulberry plants were classified according to characteristic of each variety in two groups, respectively; globular and columnar form in canopy, broad leaves and narrow leaves types in productive structure, and seriously obstructured the light and fairly transmitted the light into lower leaf stratum invertical light distribution. Out of varieties of mulberry investigated, Suwon No. 4 and Rosang were belong to the former characters, but Illchiroe and Kaeryangsubun were to the latters. The values of leaf dry matter index and dry matter production produced by leaves were increased in direct proportion to the height of leaf stratum; the leaf dry matter index of the uppermost leaf was twice as much that of the lowest, and the dry matter production by the highest leaf was 17 times compared with that by the lowest leaf. It is assumed that the meagre leaves occurred in the lower leaf stratum of Suwon No. 4 and Rosang were caused to diminished the dry matter index and dry matter production in the under leaves of plant, which were affected by receiving the weak light.

상엽 추출물을 이용한 항당뇨 음료의 개발 (I) - 상엽 추출물의 제제화 탐색 - (Development of An Anti-Diabetic Functional Drink (I) - Screening of the Manufacturing of Mulberry Leaf Extract -)

  • 본 연구는 천연 자원인 뽕나무에서 혈당강하 효능을 가진 뽕잎(상엽)의 열수 추출법을 이용하여 상엽 추출물의 공정화 확립과 Formulation 개발을 위해 상엽 추출물과 silk peptide 및 한약재를 첨가한 시제품을 동물실험을 통하여 Formulation을 최종 결정하여 제품화하여 기능성 음료를 개발하고자 하였다. 연구 결과, 상엽 추출의 최적 조건은 상엽 크기는 3mm 이하로 하고, 2/3 정도 packing된 면포자루로 포장된 상엽을 사용하며, 상엽 사용량은 50~100 g/L, 추출 온도는 9$0^{\circ}C$, 추출 시간은 2 hr으로 하였으며 이때 수율은 20~25%였다. 또한 시료 배합에 따른 혈당강하 실험을 통하여 상엽 추출물과 silk peptide 의 농도는 각각 1%, 0.1%일 때 혈당강하 작용이 가장 우수하였으며 부재료로 사용된 둥글레는 상엽추출물의 10% 첨가한 경우가 혈당강하작용이 가장 컸으며, polydextrose도 효과가 있으므로 첨가하는 것이 바람직하다고 사료된다.

상엽 유래 퀘세틴과 아스트라갈린의 항염증에 대한 상승효과 (Synergic Effect of Quercetin and Astragalin from Mulberry Leaves on Ani-inflammation)

  • The leaf of mulberry (Morus alba L) has long been used in Oriental medicine for the prevention or treatment of several diseases. However, little is known about the inhibitory effects of a single compound isolated from the mulberry leaves on inflammatory response. We are isolate a single compound of quercetin (3,3',4',5,7-pentahydroxyflavone) and astragalin (kaempferol-3-O-glucopyranoside) from the mulberry leaves, and then investigate the anti-inflammatory effects of quercetin, astragalin or quercetin plus astragalin in lipopolysaccharide (LPS)-stimulated murine peritoneal macrophages. Each compound suppressed the production of inflammatory mediators (NO, $PGE_2$ and IL-6) in LPS-stimulated murine peritoneal macrophages in a dose-dependent manner. Especially, the cotreatment of quercetin (2.5 ${\mu}M$) and astragalin (2.5 ${\mu}M$) markedly suppressed the production and the expression of inflammatory mediators. These suppressive effects were synergistically increased by their combination. These results suggest that the combination of quercetin and astragalin from the mulberry leaves may be useful for therapeutic drugs against inflammatory immune diseases, probably by suppressing the production of inflammatory mediators.

  • CCC의 엽면살포가 상수의 생장 및 생체내 대사에 미치는 영향을 분석 검토하여 얻어진 결과를 요약하면 다음과 같다. 1. 상엽의 생건중, 엽면적, 지조장, 지조중은 감소되었으나 지조경장은 약간 증대되었다. 2. 상엽의 엽록소 함량 증대에 효과가 있었다. 3. 상엽의 GOT, GDP 활성에 영향을 주지 않았다. 4. 상엽내 분해능의 활성은 낮았으나 합성능의 활성은 높았다.
