Physiological Activities of Mulberry Leaf and Fruit Extracts with Different Extraction Conditions (용매에 따른 뽕잎과 오디의 생리활성 효과)

  • Physiological activities in mulberry leaf and fruit were examined. Electron-donating ability (EDA), tyrosinase activity, SOD-like action (SOD), angiotensin I-converting enzyme-(ACE) inhibitory activity,and nitrite-scavenging ability of mulberry leaf and fruitextracted with water, with 50% (v/v) or with 100% ethanol, were measured. The EDA of mulberry leaf and fruit extracted with water or 50% (v/v) ethanol were greater (by 65.72-81.30%) than that with the 100% ethanol extract, whereas the activities of both former extracts were lower than those with 1.0% and 0.1% (both w/v) L-ascorbate solutions. The SOD-like activities of water, 50% (v/v) and 100% ethanol extracts of all samples were 24.13.26.80% lower than those of 1.0% and 0.1% (both w/v) L-ascorbate solutions. Nitrite-scavenging activity at pH 1.2 was observed in all extracts. The results further our understanding of the physiological activities of mulberry leaf and fruit extracts.

Transmission and Histochemical Detection of Mulberry Dwarf Mycoplasma in Several Herbaceous Plants (뽕나무 오갈병 마이코플라스마의 몇가지 초본식물에의 전염과 조직화학적 검정)

  • Transmission of mulberry dwarf mycoplasma (MDM) from diseased mulberry to 5 herbaceous plants (periwinkle, white clover, Ladino clover, red clover, and Chinese milk vetch) through insect vector, Hishimonus sellatus, was confirmed by symptom expression and microscopic evidences. The earliest symptom appearance was noticed on periwinkle in which incubation period was 25-30 days, while it ranged 35-40 days in the other plant species. The common symptoms of MDM infected plants were characterized by poor plant growth with accompanying discolorations of leaves (chlorosis with vein clearing in periwinkle, reddish in red clover, brownish in white and Ladino clovers, and yellowish in Chinese milk vetch). Mycoplasmal infections were diagnosed light microscopically by Dienes' and toluidine blue staining of hand-cut and Epon-embedded sections, respectively. In Dienes' stain, all the plants infected with MDM showed specific staining reaction in phloems. In toluidine blue stain, mycoplasmal existence was noted by granular appearance in sieve tubes which were confirmed to be mycoplasma-like organisms under an electron microscope.

Reduction of Phosphorus Fertilizer in Mulberry Fields, High in Phosphate (인산축적 뽕밭의 인산 감량시비에 관한 연구)

  • Field experiment and silkworm rearing were conducted for 4years in an attempt to reduce phosphorus fertilizer in mulberry field with high levels of phosphate. Experiments consisted of four treatments; normal recommended P application(13kg P2O5/10a), half, null and no fertilizer on each of 3 mulberry fields, two fields with 300ppm and one filed begining with 450ppm of aviable P2O5 content in soil. One of 300ppm fields was used for leaf quantity and one for leaf quality test by silkworm rearing. Leaf yield in the 300ppm field decreased after the 3rd spring in the no phosphorus treatment and in the 4th fall in the half phosphorus treatment. No yield decrease occurred in 450ppm field for 4 years. Yield decrease did occur in the next rearing season after soil phosphate content in the 10~20cm zone dropped below 150ppm. Phosphorus fertilizer should be applied at this time. Though P2O5 concentration in leaves of no phosphorus treatment was 0.18% lower that that of conventional treatment, there were no difference in cocoon quality.

Effects of Barley Noodles Contained Mulberry Leave Extracts on Blood Glucose Regulation in Diabetic Mice (당뇨 쥐에서 상엽추출물을 첨가한 보리면의 혈당조절효과)

  • This study aimed to investigate the blood glucose regulatory effect of barley noodles contained mulberry leaves water extract (MLWE) in diabetic mice in order to provide fundamental data as a functional food. ICR mice were divided into six groups as follows: normal control, positive control, barley control, mulberry control, barley+MLWE 2.5% and barley+MLWE 5% groups. High fat/high carbohydrate diet was fed for 4 weeks and diabetes mellitus was induced by i.p. injection of streptozotocin and nicotinamide. After diabetes induction, experimental diet was supplemented for 2 weeks and simultaneous measurement of body weight, blood glucose (including glucose tolerance test), liver function and blood lipid profile was conducted. As a result, there was less decrease in body weight compared to the positive control group in the groups that supplemented barley and MLWE together than the group ingesting barley and MLWE alone. In addition, regulatory effects on blood glucose, liver function and blood lipid concentration were also stronger in barley and MLWE concurrently supplemented groups. Consequently, combination of barley and MLWE was effective in the regulation of body weight, blood glucose, liver function, and blood lipid in diabetic mice. Therefore, this recipe could be a promising strategy for beneficial effects in the patients of diabetes mellitus.

Evaluation of In Vivo Safety of Inhibitory Compounds from Cheongmoknosang Mulberry Leaves against Helicobacter pylori (청목노상 뽕잎으로부터 분리한 Helicobacter pylori 저해물질에 대한 In Vivo 안전성 평가)

  • Biological compounds (caffeic acid, rosemarinic acid, and chlorogenic acid) from mulberry leaf extracts were administered to mice in order to confirm their stability. All male and female mice survived upon a 4,000 mg/g dose in an acute toxicity test, and they also survived after injection of 2,000 mg/kg for 13 weeks repeatedly. Therefore, the level of toxicity was not high. In a comparison of the control and test groups, there were no significant differences upon naked eye inspection, and the weights of stomachs infected by Helicobacter pylori were not significantly different. Regarding the effects on immune cells, NO of macrophages decreased more than that of control when medicine was administered. The spleens of the female mice group proliferated slightly in LPS and Con A within 48 hr, whereas the other test group showed a similar level and the cell toxicity of natural killing cells decreased. Therefore, we concluded that caffeic acid, rosemarinic acid, and chlorogenic acid from mulberry leaf extracts are not harmful for the treatment of infected patients the development as a healthy functional food.

  • Effects of Foliar spray of Kinetin on the growth and metabolism of mulberry tree was investigated in this studies. The results are summarized as follows; 1. Appropriate level of Kinetin for the optimal growth of mulberry tree was 100ppm. 2. In the non-fertilizer, N, P, K, and N+P+K treated plots supplemented with Kinetin, the growth of mulberry tree was generally promoted, especially in the roots, in comparison to non-supplemented plots. The effect was notably outstanding in the N, and N+P+K treated plots. 3. The mechanism by which the growth of root is stimulated was fundamentally attributed to the hypertrophy of unit cells. 4. The chlorophyll contents of the leaves in the Kinetin supplemented plots were higher than that of the non-supplemented, especially in the N, and N+P+K treated ones. 5. Likewise, total sugar contents of Kinetin supplemented plots were higher than that of the non-supplemented. Particularly the N+P+K treated plots showed higher level of sugar contents. In other plots, there were no significant differences in the level of sugar contents. 6. The activity of GOT and GPT was higher in the Kinetin supplemented plots, particularly in the N, and N+P+K treateated. 7. The contents of ascorbic acid were increased in plots with kinetin supplement in the order of N+P+K>potassium>Nitrogen>phosphorus$\geq$non-fertilizer. 8. Difference between Kinetin treatment and non-treatment was not recognized in the contents of inorganic and organic compounds.

Accumulating Pattern of ${\alpha}-glycosidase$ Inhibitor in Various Silkworm Varities (누에품종별 혈당강하물질 축적양상 구명)

  • [ ${\alpha}-Glycosidase$ ] inhibitors slows the velocity of the uptake of monosaccharides in the small intestine by retarding the speed of degradation of disaccharides to monosaccharides, which made it possible to develop ${\alpha}-glycosidase$ inhibitors as the antihyperglycemic ('antihyperglycemic' means 'blood-glucose-level-lowering') reagent for the diabetic patients such as acarbose and miglitol. Twenty kinds of ${\alpha}-glycosidase$ inhibitors have been reported to exist in mulberry, Morns alba, and some of them are also found in the silkworm, Bombyx mari, as the result of its daily feeding of mulberry leaves as the sole diet. 1-Deoxynojirimycin (DNJ), one of the most potent ${\alpha}-glycosidase$ inhibitor, is the most abundant among polyhydroxylated alkaloids with ${\alpha}-glycosidase$ inhibiting activity in both M alba and B. mari, therefore considered the antihyperglycemic criterion of the mulberry- or silkworm-based neutroceutical products. DNJ is thought to be accumulated in the body of silkworm because the its concentration in the silkworm body is two to three times as much as that in the mulberry leaves. Eighteen silkworm F1 varieties have been recommended for industrial rearing in Korea by some standards such as pathological strength. DNJ concentration in 18 recommendedwere measured at 3rd day in the 5th instar after lyophilization to determine the varieties that accumulates DNJ in its body most. GeumOk-Jam was the highest in the DNJ concentration of 5.45 mg/gDW among the recommended F1 varieties.

Evaluation of Biological Activity and Analysis of Functional Constituents from Different Parts of Mulberry (Morus alba L.) Tree (뽕나무(Morus alba L.) 부위별 생리활성 측정 및 기능성 물질 분석)

  • Evaluation of biological activity and analysis of functional constituents from water and ethanol extracts of four different parts of mulberry (Morus alba L.) tree were carried out to develop functional ingredients and foods using extracts of mulberry tree. The water and ethanol extracts of four different parts of mulberry tree were prepared and their biological activities and functional constituents determined by in vitro assays and HPLC, respectively. In general, ethanol extracts showed stronger biological activities and higher functional constituents than water extracts. Ethanol extracts of mulberry fruit, root bark, and twig showed stronger antioxidant ($IC_{50}=128.4{\mu}g/mL$), ${\alpha}$-glucosidase ($IC_{50}=12.0{\mu}g/mL$), and lipoxygenase ($IC_{50}=36.3{\mu}g/mL$) and tyrosinase ($IC_{50}=410.3{\mu}g/mL$) inhibitory activities, respectively, than those of other parts. Mulberry fruit and leaf showed the highest contents of anthocyanin (cyanidin 3-glucoside: 213.20 mg/100 g) and chlorogenic acid (514.97 mg/100 g), and especially ethanol extract of mulberry leaf contained higher quercetin 3-O-(6-O-malonyl)glucoside (143.25 mg/100 g) and kaempferol 3-O-(6-O-malonyl)glucoside (30.25 mg/100 g) contents without water extract of mulberry leaf. Meanwhile, mulberry twig contained both oxyresveratrol glycoside (48.90 mg/100 g) and its aglycone (21.88 mg/100 g), whereas mulberry root bark contained mostly oxyresveratrol glycoside (724.05 mg/100 g). Additionally, mulberry root bark and leaf contained much higher ${\gamma}$-aminobutyric acid (223.90 mg/100 g) and 1-deoxynojirimycin (86.07 mg/100 g) contents, respectively, than other parts of mulberry tree. These results suggest that high quality processed foods and functional foods using mixtures of mulberry fruits, leaves, twigs, and root barks should be developed for prevention and inhibition of several pathological disorders.
