Pharmacognostical Study on the Dioscoreae Rhizoma in Korea (한국산(韓國産) 산약류(山藥類)의 생약학적(生藥學的) 연구(硏究))

  • Yam, Dioscorea batatas Decne (Dioscoreaceae) is a plant growing in Korea. Dioscoreae Rhizoma in the markets are classified into natural and cultural ones. These studies were conducted to investigate the differencies between natural and cultural Yams by characteristics of morphology, cell contents, chemical components and physical properties. More mucilage cells which contain calcium-oxalate raphides were contained in cultural Yam than natural Yam. However, the amount of crude protein, crude lipid and mucilage viscosity, swelling, gelatinization of polysaccharides were greater in natural Yam than the other. It is suggested that natural Yam is excellent as nutrition agent in quality and worthwhile as a resource of crude drugs.

Physicochemical and Antioxidant Properties of Red Ginseng Marc Fermented by Bacillus subtilis HA with Mugwort Powder Addition (고초균 발효에 의한 홍삼박 발효물에 쑥 분말 첨가에 따른 물리화학적 및 항산화적 특성)

  • Red ginseng marc (RGM) was fermented by the solid-state fermentation using Bacillus subtilis HA to produce biologically active compounds. The red ginseng marc fermented without mugwort possessed higher mucilage content (11.5%) and proteolytic activity (277.5 unit/g). The RGM fermented with 3% mugwort showed lower production of mucilage and protease activity whereas higher tyrosine content (581.3 mg%) and consistency index ($8.8\;Pa{\cdot}s^n$). The mucilage produced from fermented RGM contained $\gamma$-PGA with 1,100 kDa of molecular weight, and its yield was 15.9 g/kg. 70% ethanol extract from the RGM fermented with 3% mugwort had the highest DPPH radical scavenging effect ($IC_{50}$ value of 0.57 mg/mL), and the water extract showed the highest ABTS radical scavenging effect, indicating $IC_{50}$ value of 1.24 mg/mL. Overall, the RGM fermented by B. subtilis HA with mugwort contained various biologically active compounds having antioxidant effects.

Physicochemical Characteristics of Cheonnyuncho Fruit (Opuntia humifusa) Fermented by Leuconostoc mesenteroides SM (Leuconostoc mesenteroides SM을 이용한 천년초(Opuntia humifusa) 열매 발효물의 물리·화학적 특성)

  • Functional properties of Cheonnyuncho fruit paste (CFP) were enhanced by fermentation, using Leuconostoc mesenteroides SM. The stability of CFP pigment was maintained and microorganisms in crude CFP were sterilized by heating at $80^{\circ}C$ for 30 min. The production of dextran in CFP was increased by increasing the fermentation time and sugar content, resulting in an increase of consistency. The CFP fermented with 20% sucrose contained 4.8% mucilage after fermentation for 2 days. In particular, the production of mucilage was greatly enhanced by fortification with 3% sodium citrate in CFP, resulting in mucilage production of 7%. In addition, the viscous modulus of fermented CFP showed a greater increase than that of the elastic modulus. Thus, it was concluded that CFP fermented by L. mesenteroides SM can be fortified with functional ingredients, such as mucilage and probiotics, with enhanced rheological properties. Therefore fermented CFP can be utilized as the functional ingredients for functional foods.

Evaluation of Rheological and Functional Properties of Roasted Soybean Flour and Mixed Cereals Fermented by Bacillus sp. (고초균을 이용한 볶은 콩과 곡류 혼합 발효물의 물성 및 기능성 평가)

  • Roasted soybean flour (RSF) and mixed cereals were fermented by the solid state fermentation using Bacillus subtilis HA to optimize the production of biologically active compounds. The RFS fermented with 52.7% moisture content showed higher production with protease activity of 42.6 unit/g and 10% mucilage content after fermentation for 24 hr. Tyrosine content and protease activity after 48 hr fermentation time were the highest values with 445.5 mg% and 55.1 unit/g, respectively. However, the wholesome fermented RSF (FRSF) was obtained by fermentation for 24 hr because of the production of unpleasant flavors after fermentation for 48 hr. The RSF fermented with various types and contents of cereals has no effects on tyrosine content and protease activity. However, the addition of brown rice significantly increased mucilage content, especially indicating 24.55% at the addition of 80% (w/w). For addition of barley, fibrinolytic activity was increased to 11.82 unit/g by the fortification of 60% barley. It is concluded that biologically active compounds including fibrinolytic activity and mucilage content in FRSF were dependent upon the type and content of various cereals.

  • This research provides insight on the laboratory investigation of the engineering properties of a lateritic soil modified with the mucilage of Opuntia ficus-indica cladodes (MOFIC), which has a history of being used as an earthen plaster. The soil is classified, according to AASHTO classification system, as A-2-6(1). The Atterberg limits, compaction, permeability, California bearing ratio (CBR) and unconfined compressive strength of the soil were determined for each of 0, 4, 8 and 12% addition of the MOFIC, by dry weight of the soil. The plasticity index, optimum moisture content, swell potential, unconfined compressive strength and permeability decreased while the soaked and unsoaked CBR increased, with increasing MOFIC contents. The engineering properties of the natural soil, which only satisfies standard requirements for use as subgrade material, became improved by the application of MOFIC such that it meets the standard requirements for use as sub-base material for road construction. The effects of MOFIC on the engineering properties of the soil resulted from bioclogging and biocementation processes. MOFIC is recommended for use as a modifier of the engineering properties of soils, especially those with similar characteristics to that of the soil used in this study, to be used as a pavement layer material. It is more economical and environment-friendly than conventional soil stabilizers or modifiers.

Mucilage Separation of Korean Yam Using Microparticulation/Air Classification Process (초미세분쇄/공기분급을 이용한 마의 점질물 분리)

  • To separate and concentrate mucilage from yam(Dioscorea batatas DECNE), yam was dried, microparticulated using impact mill and air-classified at different air classifying wheel speed(ACWS) in classifier. As ACWS increased from 5,000 rpm to 22,500 rpm, the contents of dietary fiber, protein and lipid of air classified microparticles(ACM) increased remarkably. Especially the ACM with ACWS over 15,000 rpm showed 36.41% dietary fiber and 16.66% protein. The dietary fiber and protein components were concentrated to $2.5{\sim}9.0$ times as compared with whole yam powder. Concomitantly the non-fibrous carbohydrate decreased from 88.31% to 16.84. The damaged starch(%), WSI and WAI of ACM of ACWS over 15,000 rpm were $1.5{\sim}3.0$ times higher than those of ACM under ACWS 15,000 rpm. The apparent viscosity of ACM was 0.0800 Pa s over ACWS 15,000 rpm and 0.0080 Pa s under ACWS 15,000 rpm. Judging from viscosity of ACM, the mucilage component of yam was concentrated to 10 times. In conclusion, the optimum process to separate and concentrate the mucilage from yam consisted of the microparticulation to $5{\sim}30{\mu}m$ and the air-classification at ACWS over 15,000 rpm.

Skin Hydration Effect of Brasenia schreberi Mucilage Polysaccharide Extract (순채 점액질 다당체 추출물의 피부 보습 효과 연구)

  • In this study, we evaluated the skin moisturizing effect of Brasenia schreberi (B. schreberi) mucilage polysaccharide on human skin and in vitro and the potent cosmetic ingredient for skin. To protect skin from various environmental stresses and aging, we should increase moisture content of skin and prevent water loss. We have found that polysaccharides extracted from mucilage of B. schreberi improved the roughness of skin with its lubricating behavior. In vitro, the expression of transglutaminase 1 (TGM1) gene, which plays a role in cross-linking the skin barrier, was increased when the keratinocytes were treated with B. schreberi polysaccharides. In addition, the expression of hyaluronan synthase 3 (HAS3) gene, an enzyme that synthesizes water-binding matrix hyaluronic acid, aquaporin 3 (AQP3), which regulates the movement of water and glycerol were also increased. In addition, an experiment to evaluate its potential as a cosmetic ingredient has shown anti-inflammatory and collagen synthesis-promoting effects. As a result, the mucilaginous polysaccharide from natural products which has not existed before, showed moisturizing effect, anti-inflammation and collagen synthesis-promoting effects for skin protection and hydration.

  • It is well known that Dioscoreae Rhizoma tonifies and benefits the Spleen and Stomach, benefits the Lung and nourishes the Kidney. A major component isolated from this herb consist of dioscin, diosgenin, allantoin, starch, alanin, mucilage, choline, amylase, and glycoprotein. In this study, we aimed to measure the content of allantoin, one of major component, to determine the quality of Dioscoreae Rhizoma.

A Study on a Morphological Identification of Pinelliae Tuber, Typhonii Flagelliformis Rhizoma and Arisaematis Rhizoma (반하(半夏), 수반하(水半夏) 및 천남성(天南星)의 형태(形態) 감별에 관한 연구)

  • Objectives : A morphological classification among Pinelliae Tuber, Typhonii Flagelliformis Rhizoma and Arisaematis Rhizoma was made through microscopic observation. Method : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Result : 1. Pinelliae Tuber is like a ball with a $1{\sim}1.5cm$ diameter, with a concave apex and pockmarked root trace around. 2. Typhonii Flagelliformis Rhizoma is somewhat oval or like a cone, with a $4{\sim}8mm$ diameter, a $7{\sim}15mm$ height, a projected round apex, and a pin-pointed back. 3. Arisaematis Rhizoma is somewhat flat round with a $2{\sim}2.5cm$ diameter, a little than Pinelliae Tuber, and its apex has incomplete lobopodium and root trace. 4. Fundamental tissues of Pinelliae Tuber, Typhonii Flagelliformis Rhizoma, and Arisaematis Rhizoma have starch grains and mucilage cells, and their mucilage cells include the raphides of calcium oxalate. Conclusion : Internal forms of Pinelliae Tuber, Typhonii Flagelliformis Rhizoma, and Arisaematis Rhizoma are difficult to distinguish from one another, but Pinelliae Tuber and Arisaematis Rhizoma can be distinguished by their external forms and sizes, and Pinelliae Tuber and Typhonii Flagelliformis Rhizoma were distinguished by the forms of apex.

  • There have been many problems like plasmolysis in the ultrastructural studies with kelps. The present study was conducted with series of different fixatives and buffer solutions and several resins to solve the plasmolysis phenomena. The fixative of 1470 mosmol($3\%$ glutaraldehyde, $2.4\%$ paraformaldehyde in 0.05M cacodylate buffer with 0.2M sucrose and $0.5\%$ caffeine at pH of 7.2) showed the least frequency of plasmolysis. The relationship between the concentration of primary fixatives and the frequency of plasmolysis was not found. With above fixative the possibility of plasmolysis during infiltration period was not identified using ultra-low viscosity resin. Generally, the kelp(Laminaria religiosa) contains a lot of mucilage, which interupts the infiltration of fixative. The possible physiological mechanisms related with mucilage secretion may be unable to fix the region of plasmalemma properly.
