Morphological Anomaly of Primordial Follicle in $\gamma$-Irradiated Mice

  • Ovarian follicles are faced with one of two fates, atresia or development. Up to 99% of follicles become degenerated rather than ovulated in female life span. Thus, atresia occurs at all stages of follicle development in mammalian ovaries. In the present experiment, the effect of ${\gamma}$-radiation on primordial follicles was morphologically analyzed in a mouse ovary. Thirty-seven percent of the primordial follicles in the non-irradiated control mice ovaries were abnormal. At day 8 post irradiation, most of primordial follicles became atretic. They lost their integrity of architecture in the follicular shape. Then, all the oocytes disappeared from the follicles. And only 3 to 4 granulosa cells lay down onto the basement membrane. Disappearance of granulosa cells or oocytes resulted from the radiation-induced apoptotic process. It is definitely clear that ${\gamma}$-radiation induces rapid apoptotic degeneration of the primordial follicles. The morphological degeneration induced by radiation in the primordial follicles can be used as an experimental model to draw out a deeper insight for radioprotectant researches.

Genetic Analysis of Morphological Traits of Rice Grain and Their Inter-relationships

  • Two rice varieties, 'Oochikara' with large grain and 'Hwayeongbyeo' and their progenies (F$_1$, F$_2$, B$_1$ and B$_2$) were tested to understand gene action of morphological traits of rice grain and their relationships. The evaluated traits were 1,000-grain weight, grain length, width, thickness, length-width ratio and chalkiness of brown rice. Correlation between grain weight and chalkiness was highly significant in the all progenies, and grain length were not associated with width and thickness in an F$_2$ population. Scaling test and jonit scaling test revealed that inheritance of grain traits were fitted to additive-dominance model without epistasis. Additive effects for the traits were much greater than the dominance effects.

  • Certain bacterial species possess the capability of differentiation through several morphogenetic changes which enable them to adapt to certain internal and external stimuli(Losick and Shapiro 1984). Upon induction, cells of A. vinelandii undergo a morphological process which leads to the production of one cyst per cell (Sadoff, 1975). The cysts are considerably resistant to desiccation, which confers a survival advantages upon the organism(Socolofsky and Wyss 1962). Like other prokaryotic differentiations encystment provides a relatively simple model of cellular differentiation. Like in other differentiating bacteria, vegetative growth can be separated from differentiation. Furthermore, the differentiation cycle can be synchronized by specific inducer. There have been a great deal of morphological and physiological studies on this process. However, the mechanisms used to regulate cell differentiation can be clearly defined by careful genetic analysis of the process. Unfortunately, A. vinelandii has proven to be difficult for genetic analysis (Sadoff 1975). For example, it has been shown that a variety of metabolic mutants of Azotobacter speicies are difficult to isolate after mutagenesis with chemical mutagens or UV irradiation. Nevertheless recent advances in molecular genetics in Azotobacter species, especially in the nitrogen fixation research area, appear to be able to overcome this difficulty (Robinson et al. 1986; Kennedy et al. 1986).

A Comprehensive and Practical Image Enhancement Method

  • Image enhancement is a challenging problem in the field of image processing, especially low-light color images enhancement. This paper proposed a robust and comprehensive enhancement method based several points. First, the idea of bright channel is introduced to estimate the illumination map which is used to attain the enhancing result with Retinex model, and the color constancy is keep as well. Second, in order eliminate the illumination offsets wrongly estimated, morphological closing operation is used to modify the initial estimating illumination. Furthermore, in order to avoid fabricating edges, enlarged noises and over-smoothed visual features appearing in enhancing result, a multi-scale closing operation is used. At last, in order to avoiding the haloes and artifacts presented in enhancing result caused by gradient information lost in previous step, guided filtering is introduced to deal with previous result with guided image is initial bright channel. The proposed method can get good illumination map, and attain very effective enhancing results, including dark area is enhanced with more visual features, color natural and constancy, avoiding artifacts and over-enhanced, and eliminating Incorrect light offsets.

  • The purpose of the research is to track the morphological transformation of apartment complex sites in Busan, especially focusing on building arrangement on apartment complex sites. Though there have been numerous studies regarding apartment housing, only few studies have been performed about the building arrangement. The arrangement has been poorly treated and not well disclosed as compared with other apartment-based research such as form of residential towers, floor plan, etc. The building arrangement is one of the most critical decisions at the early design stage and is affected by numerous social aspects such as building/zoning code, market demand, cultural preference, etc. Thus, the transformation of the arrangement may provide the hint for the change of socio-cultural demands of apartment housing. The research has been done on the existing 269 apartment complexes which have been built through 2016 in Busan. There are three categories set for analysis: relationship between buildings, relation between road and buildings, and the building arrangement along site perimeter. The result shows that significant change has occurred since 2000s; the arrangement has become diverse from three types to seven types. The early model of parallel or lattice types have been dominant until 2000, but the skewed one replaced the parallel or lattice types in Period V and Period VI, which is from 2001 through 2016.

Protective Effects of Auraptene against Free Radical-Induced Erythrocytes Damage

  • Objectives: Auraptene is the most abundant natural prenyloxycoumarin. Recent studies have shown that it has multiple biological and therapeutic properties, including antioxidant properties. Erythrocytes are constantly subjected to oxidative damage that can affect proteins and lipids within the erythrocyte membrane and lead to some hemoglobinopathies. Due to the lack of sufficient information about the antioxidant effects of auraptene on erythrocytes, this study intended to evaluate the potential of this compound in protecting radical-induced erythrocytes damages. Methods: The antioxidant activity of auraptene was measured based on DPPH and FRAP assays. Notably, oxidative hemolysis of human erythrocytes was used as a model to study the ability of auraptene to protect biological membranes from free radical-induced damage. Also, the effects of auraptene in different concentrations (25-400 µM) on AAPH-induced lipid/protein peroxidation, glutathione (GSH) content and morphological changes of erythrocytes were determined. Results: Oxidative hemolysis and lipid/protein peroxidation of erythrocytes were significantly suppressed by auraptene in a time and concentration-dependent manner. Auraptene prevented the depletion of the cytosolic antioxidant GSH in erythrocytes. Furthermore, it inhibited lipid and protein peroxidation in a time and concentration-dependent manner. Likewise, FESEM results demonstrated that auraptene reduced AAPH-induced morphological changes in erythrocytes. Conclusion: Auraptene efficiently protects human erythrocytes against free radicals. Therefore, it can be a potent candidate for treating oxidative stress-related diseases.

Field Observation and Quasi-3D Numerical Modeling of Coastal Hydrodynamic Response to Submerged Structures

  • Even though submerged breakwater reduces incident wave energy, it redistributes the coastal area's wave-induced current, sediment transport, and morphological change. This study examines the coastal hydrodynamics and the morphological response of a wave-dominated beach with submerged breakwaters installed through field observation and quasi-3D numerical modeling. The pre-and post-storm bathymetry, water level, and offshore wave under storm forcing were collected in Bongpo Beach on the East coast of Korea and used to analyze the coastal hydrodynamic response. Four vertically equidistant layers were used in the numerical simulation, and the wave-induced current was examined using quasi-3D numerical modeling. The shore normal incident wave (east-northeast) generated strong cross-shore and longshore currents toward the hinterland of the submerged breakwater. However, the oblique incident wave (east-southeast) induced the southeastward longshore current and the sedimentation in the northeast area of the beach. The results suggested that the incident wave direction is a significant factor in determining the current and sediment transport patterns in the presence of the submerged breakwaters. Moreover, the quasi-3D numerical modeling is more appropriate for estimating the wave transformation, current, and sediment transport pattern in the coastal area with the submerged breakwater.

  • This paper proposes a method of improving deep learning based numbers and characters recognition performance on parts of drawing through image preprocessing. The proposed character recognition system consists of image preprocessing and 7 layer deep learning model. Mathematical morphological filtering is used as preprocessing to remove the lines and shapes which causes false recognition of numbers and characters on parts drawing. Further.. Further, the used deep learning model is a 7 layer deep learning model instead of VGG-16 model. As a result of the proposed OCR method, the recognition rate of characters is 92.57% and the precision is 92.82%.

Proposal of Parameter Range that Offered Optimal Performance in the Coastal Morphodynamic Model (XBeach) Through GLUE

  • The process-based XBeach model has numerous empirical parameters because of insufficient understanding of hydrodynamics and sediment transport on the nearshore; hence, it is necessary to calibrate parameters to apply to various study areas and wave conditions. Therefore, the calibration process of parameters is essential for the improvement of model performance. Generally, the trial-and-error method is widely used; however, this method is passive and limited to various and comprehensive parameter ranges. In this study, the Generalized Likelihood Uncertainty Estimation (GLUE) method was used to estimate the optimal range of three parameters (gamma, facua, and gamma2) using morphological field data collected in Maengbang beach during the four typhoons that struck from September to October 2019. The model performance and optimal range of empirical parameters were evaluated using Brier Skill Score (BSS) along with the baseline profiles, sensitivity, and likelihood density analysis of BSS in the GLUE tools. Accordingly, the optimal parameter combinations were derived when facua was less than 0.15 and simulated well the shifting shape, from crescentic sand bar to alongshore uniform sand bars in the surf zone of Maengbang beach after storm impact. However, the erosion and accretion patterns nearby in the surf zone and shoreline remain challenges in the XBeach model.

Ant-Inflammatory Effect of Prunus serrulata var. spontanea Extract in OVA-Induced Asthma Animal Model (벚나무 추출물의 OVA 유도 천식동물모델에서 항염증 효능)

  • The objective of this study was to determine the efficacy of a natural product of cherry tree (Prunus serrulata var. spontanea: PS) as a test substance for improving cytokine and ovalbumin-specific IgE using an ovalbumin-induced asthma disease model of 5-week-old male BALB/c mice. Lung tissue pathology was analyzed to confirm anti-inflammatory and asthmatic effects. As a result of examining the effect on changes in inflammatory cells in bronchoalveolar lavage fluid in an ovalbumin-induced asthma disease model by administering the PS sample, total cells, eosinophil, neutrophil, lymphocyte, and monocytes were significantly decreased. Concentrations of cytokine-based TNF-alpha and IL-4 and immunoglobulin E in serum were significantly increased in the asthma-inducing negative control group than in the normal group. However, high concentrations of PS decreased them. In histopathological examination of the lung tissue, it was confirmed that inflammatory cells infiltrated around the alveoli and bronchioles were increased in ovalbumin-induced asthma disease model. After administration of cherry tree extract, bronchiolar morphological changes such as mucosal thickening were slightly improved. From the above results, it was confirmed that extract of cherry tree significantly reduced inflammation expression and tissue damage in alveolar tissues. It was also confirmed that the cherry tree extract had an excellent efficacy in improving asthma inflammation.
