Study on Dormancy Mechanisms of American Ginseng Seed II - Germination Inhibition of Seed Coat

  • This paper gives a description about the germination inhibition of American ginseng (Panax quinquefolium L.) seed coat. The existence of seed coat is one of the inhibitory factors which inhibit the embryo growth, particularly during the morphological after-ripening stage. The seed coat can obstruct the water absorption at the beginning of seed stratification, but it can not threaten seed germination. The inhibition of seed coat is not caused by the mechanical fetter neither. However, before splitting the seed coat, the inhibition of seed coat comes from both air-tight character and inhibitors, and after splitting the seed coat, the inhibition may come mainly from the inhibitors.

Seed Dormancy Type and Germination Characteristics in Tiarella polyphylla D. Don Native to Korea (한반도 자생식물 헐떡이풀 종자의 휴면유형과 발아특성)

  • Tiarella polyphylla D. Don is a native plant distributed only in Ulleung Island in Korea and has been traditionally used for medicinal purposes, although it is also used ornamentally. This study was conducted to determine the requirements for dormancy break and germination and to classify the type of seed dormancy. The experiments were performed with cold stratification (0 or 12 weeks at $5^{\circ}C$), warm stratification (0, 4, 8, or 12 weeks at $23^{\circ}C$, followed by 8 weeks at $5^{\circ}C$, and then incubation at $23^{\circ}C$), and $GA_3$ treatments (0, 10, 100, or 1000 mg/L). The treated seeds were incubated on aseptic media at room chamber ($23^{\circ}C$, a 16h photoperiod of fluorescent lamps with $40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$). The seeds were dispersed in nature as underdeveloped embryos with no physical barrier to absorb water to prevent water absorption. However, the seeds did not germinate for 30 days after sowing without any pre-treatments. Thus, the seeds had morphological dormancy (MD) and physiological dormancy (PD). The final germination percentage following cold stratification (0 or 12 weeks) was 66.7% and 45.6%, respectively. The cold stratification delayed seed germination by about 3 weeks. In the warm stratification experiment (0, 4, 8, or 12 weeks), the final germination percentage was 21.1%, 27.8%, 41.1%, and 57.8%, respectively, 20 weeks after sowing. The embryos of the T. polyphylla seed grew in relatively warm temperatures ($23^{\circ}C$). $GA_3$ application overcame seed dormancy and promoted germination. Following $GA_3$ treatment (0, 10, 100, or 1000 mg/L), the final germination percentage was 33.3%, 45.0%, 42.5%, and 72.5%, respectively. These results suggest that the T. polyphylla seeds had non-deep simple morphophysiological dormancy (MPD) and $GA_3$ treatment could be used as a substitute for warm stratification for breaking seed dormancy. To our knowledge, this is the first report of seed dormancy characteristics of the genus Tiarella native to Korea.

Effect of Alternating Temperature Conditions, and GA3 Treatment on Seed Germination of Trollius macropetalus (Regel) F.Schmidt (변온 조건 및 GA3 처리가 큰금매화의 종자 발아에 미치는 영향)

  • This study was conducted to determine the conditions for breaking dormancy in the seeds of the North Korean plant resource, Trollius macropetalus, and to investigate its germination characteristics. The morphological characteristics and viability of T. macropetalus seeds were examined. Germination characteristics were investigated based on temperature conditions (15/6℃ & 25/15℃), cold stratification treatment, and GA3 treatment. The results showed that cold stratification treatment had no effect on breaking the dormancy of T. macropetalus. GA3 treatment significantly increased the germination rate of T. macropetalus and also improved the mean germination days and germination speed. However, compared to the 15/6℃ condition, the effectiveness of GA3 treatment for T. macropetalus decreased under the 25/15℃ condition. It was determined that T. macropetalus have a nondeep-type morpho-physiological dormancy. The most effective conditions for breaking dormancy were found to be the 15/6℃ temperature condition and GA3 treatment at 500 mg·L-1. The findings of this study are expected to provide valuable information for the mass propagation of T. macropetalus, which has potential applications in landscaping and pharmaceutical materials.

Physiological and Morphological Aspects of Bipolaris sorokiniana Conidia Surviving on Wheat Straw

  • Wheat samples showing typical spot blotch symptoms on stems and sheaths were collected from the field after physiological maturity, and were sealed in paper bags and stored in the laboratory at room temperature to study the survival of Bipolaris sorokiniana conidia on wheat straw. The materials were observed at monthly intervals to assess the conidia viability during storage. After 4 months, the frequency of individual conidia already present on wheat straw at the time of sampling was reduced and appeared to be progressively replaced by the formation of round structures consist-ing of conidia aggregates. After 5 months, distinct, individual conidia were no longer detected, and only 'clumps of conidia' were observed. These dark black aggregates or 'clumps of conidia’measured 157-170$\mu\textrm{m}$ in diameter and were grouped into boat-shaped olivacious conidia showing thick wall and measuring 50-82$\times$20-30$\mu\textrm{m}$. The germination was unipolar and below 0.5%, suggesting the occurrence of dormancy, In contrast, individual conidium produced on wheat during the growing season were 96-130$\times$16-20$\mu\textrm{m}$, slightly curved, hyaline to light pale, and euseptate with a bipolar germination reaching 98-100%. Bipolaris sorokiniana conidia produced on PDA were 55-82$\times$20-27$\mu\textrm{m}$, tapered at both ends, dark brown to olivacious, distoseptate, showed up to 1% germination, and were predominantly unipolar. Results of the present study suggest that B. sorokiniana conidia belonged to two different physiological categories corresponding to the pathogen's infection phase and its survival, respectively. The infection phase is characterized by a high germination percentage as opposed to the survival phase harboring apparent dormancy.

The Effects of Environmental Conditions and Chemical Treatments on Seed Germination in Astilboides tabularis (Hemsl.) Engl.

  • Tabulis [Astilboides tabularis (Hemsl.) Engl.] has the potential for becoming a useful horticultural crop. This study was conducted to classify the seed dormancy types and to identify the germination conditions that improve the horticultural usefulness of this plant. We found that A. tabularis (Hemsl.) Engl. produced undifferentiated embryos and had a low germination rate of under 50%. In addition, water submersion led to moisture absorption. Therefore, A. tabularis seeds were designated as morphologically dormant. The germination rate was highest (86.3%) at $30^{\circ}C$ under light conditions. Higher temperatures generally led to an acceleration of the germination process, regardless of light condition. To improve the germination rate, seeds were submerged in various concentrations of growth regulators, such as $GA_3$ and kinetin, as well as minerals such as $KNO_3$ and KCl. An analysis of germination characteristics at $30^{\circ}C$ under light conditions revealed that treatment with 200 and $500mg{\cdot}L^{-1}GA_3$ led to an excellent germination rate of 97.0%. Treatment with $100mg{\cdot}L^{-1}GA_3$ led to a better germination rate than the control as well. However, treatment with kinetin, $KNO_3$, and KCl had no effect on germination, regardless of concentration.

Germination Characteristics and Seed Dormancy of Iris dichotoma Pall., an Endangered Species Native to Korea

  • Iris dichotoma Pall. found on Daechung Island in Korea has been designated as an endangered species. To aid in conservation efforts of this species, this study investigated its germination characteristics and seed dormancy type. Four sets of seeds were incubated at different temperatures (4/1℃, 15/6℃, 20/10℃, and 25/15℃). One set of seeds was cold stratified (4 weeks at 4/1℃). The final germination rate and mean germination time showed that the optimal germination temperature was 25/15℃. Final germination rates were ~70%, showing no significant difference among temperature treatments. However, mean germination time were significantly different among all temperature treatments except for 4/1℃. Mean germination time for seeds with temperature treatments of 15/6℃, 20/10℃, and 25/15℃ were 3.2, 2.1, and 1.5 weeks, respectively. At 25/15℃, the mean germination time was half of that at 15/6℃. Seeds of I. dichotoma had fully developed embryos at the time of dispersal. No additional growth of the embryo was observed. Cold stratification did not affect the final germination rate or the mean germination time. This study shows that seeds of I. dichotoma have no physiological or morphological dormancy, unlike other members of the Iris genus known to have seed dormancy that needs a relatively high incubation temperature (≥25/15℃) for mass propagation to occur. These results will be useful for understanding ecophysiological mechanisms related to the species' habitat. They are also useful for mass propagation of I. dichotoma for the purpose of conserving this endangered species.

  • Dynamic parameters of physiology including embryo ratio (ER), embryo length to endosperm length, dry weight ratio between embryo and endosperm (DWR) and respiration rate (RR) in American send (Panax quinquefolium L.) seed were investigated. According to the changes of ER during seed stratification, the duration of embryo afterripening could be divided into three stages as \circled1 embryo slow growth stage (ESGS), ER increased from 7.31% to 20.48% (0.16% day-1): \circled2 embryo rapid growth stage (ERGS), ER increased to 80.98% (0.61% day-1) (75G5+ ERGS=morphological afterripening stage (MP,5)) and \circled3pysiological afterripening stage (PAS), ER Increased to 88.50% (0.094 day-1) only. DWR Increased slowly from 0.20% to 2.76clp (0.016% day-1) in MAS and rapidly to 8.81% (0.061% day-1) in PAS. The RR correlated significantly with ER as well (r=0.8934 > rot, 0.6610). The steep increment of both DWR and RR in PAS indicated that the PAS was not a static stage although the ER was not changed too much. All of these may provide some information for understanding the dormancy mechanisms of American sting seed.

Germinability, Morphological Development during Germination and Anatomical Observation Gentiana scabra Bunge var. buergeri Max. (자생 용담의 발아성 및 발아과정 중의 형태 형성과 조직의 해부학적 관찰)

  • Gentiana scabra Bunge var buergeri Max, can be widely used such as cut-flower, ornamental and medicinal plant among the native plants. Microscopic and Scanning electron microscopic observation plant were carried out to estimates the characteristics correlated with germinability and germination process, and the results were as follows. The seeds were germinated just after harvest, and it means that there is no or little dormancy of the seeds. Matured seeds showed higher germinability than those of immatured. As the storage period was longer, immatured seeds showed shorter longevity than those of matured. The average longevity was about 190 days. According to the observation results of morphological changes during the germination, seed coat was expanded with water absorption immediately after seeding, and radicals were observed 2 days after seeding. Root hairs were formed behind the growing tip of the root on 4 days after seeding and cotyledon were unfolded on 6 days after seeding. In the microscopic observation of leaf tissues, it could be easily distinguished between midrib, and leaf blade, and also observed upper epidermis, palisade parenchyma and spongy parenchyma consisted as a sing1e layer. Palisade parenchyma was consisted of a single layer of cell. Stomata were restricted lower surface, arranged as anamocytic type, and surrounded by guard cells. In the microscopic anatomy of stem were consisted of cortical layer, forming a single layer of epidermis and parenchyma, and vascular bundle which was consisted of continuous cylinder of vascular tissues.

Variation of the Morphological Characteristics in the Accessions of Zoysia Species and Their Hybrid Lines (Zoysiagrass 수집계통들과 종간교배계통들의 형태적 특성들의 변이)

  • Fifty accessions in Zoysia species and forty-one hybrid lines were evaluated in order to detect analyze the genectic variation of Zoysiagrasses and to obtain the basic information for breeding. 1.Plant height ranged from 13 to 34.5cm, leaf length from 7 to 26cm, leaf width from 2 to 7mm, and first leaf height from 1.1 to 8.5cm 2. The correlations among plant height, leaf length and first-leaf height were high, but leaf Width was correlated only With leaf length. 3. Stolon length was varied from 10 to 108cm, number of stolons from 12 to 53, internode length from 2 to 6.1cm and coverage percentage from 10 to 90%. 4.There were correlation among stolen length, number of stolons, internode length and coverage percentage except between stolen and internode length. 5.M1J and MC2 showed the highest level in stolen length, number of stolons and coverage percentage. Therefore they will be the best breeding materials for the purpose of fast covering. 6.According to the analysis on October 26, 1995, yellow color-change rate of leaves ranged from 5 to 85%, red color-change from 0 to 75% and sum of yellow and red change from 20 to 95%. Speciallty M2M1 and FL-41 kept green color untill early December, indicating that they had the late dormancy. 7. Visual assessment of stolon length and number of stolons, showed similar tendency With actually measured data. 8. Shoot density was very high in M2M1 and S2, and showed negative correlation with leaf width. 9. It is known in Korea that the darker green color of lawngrass is, the better it looks. There were 11 lines with dark green color leaves such as J 85-10, Meyer in Z. japonira, one asacession in Z. matrella and M2M1, M2S8, JM2, 55C2 in hybrid lines.

Analysis on Morphological Characters and Germination Conditions of Seeds in Ledum palustre var. diversipilosum (백산차(Ledum palustre var. diversipilosum) 종자의 형태적 특성 및 발아조건 분석)

  • The Hairy labrador tea (Ledum palustre L. var. diversipilosum Nakai) is a evergreen small shrub, belonging to the Ericaceae and mainly distributed along the alpine areas of Hamgyeong-do in North Korea. Through seed physiological research on North Korean plants, we intend to obtain basic data for the development of mass propagation methods and use them for vegetation restoration. The internal and external morphological characters of the seed were observed using scanning electron microscopy or stereoscopy, and full seeds were selected through X-ray test. Seeds were cultured on a growth condition at 15/6℃ and 25/15℃ to examine the germination response according to temperature, and the number of germinated individuals was examined every 24 hours to calculate germination percentage and mean germination time. Low-temperature treatment (4℃) and hormone treatment (GA3) were performed to investigate the effect of dormancy breaking, and analysis of variance (ANOVA) was performed on the experimental results using SAS 9.4. The seeds of the Hairy labrador tea were elliptical-narrow in shape, and the epidermal cell wall pattern was reticulated. The final germination rate (FGP) was the highest at 58.9% under the condition of low temperature treatment at 25/15℃. The average number of days to germination (MGT) was the highest at 25.1 days without treatment at 15/6℃, and the average of the last germination days was confirmed to be about 13 days. As a result of low temperature treatment, it was confirmed that the final germination rate (FGP) increased by 43.3%. The significance of the final germination rate (FGP) was verified according to the pretreatment at 25/15℃.
