Microtensile bond strength and micromorphologic analysis of surface-treated resin nanoceramics

  • PURPOSE. The aim of this study was to evaluate the influence of different surface treatment methods on the microtensile bond strength of resin cement to resin nanoceramic (RNC). MATERIALS AND METHODS. RNC onlays (Lava Ultimate) (n=30) were treated using air abrasion with and without a universal adhesive, or HF etching followed by a universal adhesive with and without a silane coupling agent, or tribological silica coating with and without a universal adhesive, and divided into 6 groups. Onlays were luted with resin cement to dentin surfaces. A microtensile bond strength test was performed and evaluated by one-way ANOVA and Tukey HSD test (α=.05). A nanoscratch test, field emission scanning electron microscopy, and energy dispersive X-ray spectroscopy were used for micromorphologic analysis (α=.05). The roughness and elemental proportion were evaluated by Kruskal-Wallis test and Mann-Whitney U test. RESULTS. Tribological silica coating showed the highest roughness, followed by air abrasion and HF etching. After HF etching, the RNC surface presented a decrease in oxygen, silicon, and zirconium ratio with increasing carbon ratio. Air abrasion with universal adhesive showed the highest bond strength followed by tribological silica coating with universal adhesive. HF etching with universal adhesive showed the lowest bond strength. CONCLUSION. An improved understanding of the effect of surface treatment of RNC could enhance the durability of resin bonding when used for indirect restorations. When using RNC for restoration, effective and systemic surface roughening methods and an appropriate adhesive are required.

EVALUATING TWO METHODS FOR FINGERPRINTING GENOMES FOR STREPTOCOCCUS MUTANS IN CHILDREN : A COMPARISON WITH AP-PCR AND SOUTHERN BLOT RFLP (유전자형에 따른 Streptococcus mutans의 subtyping: Southern blot RFLP와 AP-PCR을 이용한 비교)

  • The arbitrary primer polymerase chain reaction(AP-PCR) and Southern blot restriction fragment length polymorphism(RFLP) were used to genotype the cariogenic pathogen S. mutans in children. Following the morphologic chracteristics of colony on selective medium for S. mutans, total genomic DNA from 155 strains was extracted by conventional methods. Among 155 strains, 143 strains (92.3%) were confirmed S. mutans by PCR with dexA gene and 114 strains were used in this study. Three random sequence 10-base oligonucleotide primers were chosen for AP-PCR. The amplified DNA products were separated electrophoretically in a 2% agarose gel containing ethidium bromide and the banding patterns were compared among different strains. For RFLP analysis, DNA was digested with EcoRI and BamHI, separated on a 0.7 % agarose gel and transferred to a nylon membrane. The membrane was probed with a previously characterised 1.6 kilobases (kb) DNA fragment cloned from gtf B gene of S. mutans. The probe was labeled with isotope[32PαCTP], and hybridized fragments were detected with intensifying screen. AP-PCR produced 4-8 DNA bands in the 0.25-10 kb regions and distinguished 9, 10 or 12 genotypes, depending on the specific primer used. Southern blot RFLP analysis revealed 2 hybridization patterns consisting of 1 DNA fragments 450, 500 bp. These results indicate that AP-PCR is more discriminative method for genotyping of S. mutans.

Therapeutic Effect of the Impatiens balsamina Linne Extract on the Membranous Nephropathy (봉선화(鳳仙花) 추출물의 막성신증(膜性腎症)에 대한 치료효과(治療效果))

  • Objective : Membranous nephropathy(MN) is one of the most common causes of nephrotic syndrome in adults. MN has been defined as granular subepithelial deposition of IgG immune complexes along the glomerular basement membrane(GBM). However, there is not a satisfactory treatment for MN. We aimed to identify the effect of Impatiens balsamina Linne(IBL) treatment on cationic bovine serum albumin(cBSA)-induced MN in a mouse model. Methods : Mice were divided into 4 groups. The normal group was injected with saline. The Control group was treated with cBSA(50 mg/kg i.p) only. The third group IBL-100, was treated with cBSA(50 mg/kg, i.p) and IBL(100 mg/kg, p.o). The fourth group IBL-400, was treated with cBSA(50 mg/kg, i.p) and IBL (400 mg/kg, p.o). After cBSA and IBL treatment for 6 weeks, we measured change of body weight, proteinuria, serum albumin, total cholesterol, triglyceride, BUN, creatinine, IgA, IgM, IgG, TNF-α, IL-6 and IL-1β levels. The morphologic changes of renal glomeruli were also observed with a light microscope. Results : The level of proteinuria significantly decreased and serum albumin increased in groups treated with cBSA and IBL extract compared with the control. The levels of serum triglyceride, BUN, IgG, TNF-α, IL-1β significantly decreased in both IBL groups. In histological findings of kidney tissue, thickening of GBM decreased in both IBL groups. Conclusions : This study shows that IBL might be effective for treatment of acute stage MN. More clinical data and studies are to be done for efficient application.

  • An acephalic budding Cysticercus of 1.2 cm long was removed surgically at the abdominal wall of a Korean man. The worm revealed abnormal buds on the bladder wall and absence of suckers and hooklets in the scolex body. The buds were of two histologic types; branching bud covered with normal tegumentum and with subtegumental cells of normal density, and buds of proliferated subtegumental cells with lacunae formation. On the bases of the morphologic features, it was identified as a racemose cysticercus. This case confirms that its extracranial location is possible.

  • 1. Purpose : This research is a study about constitutional diagnosis through the external appearances. especialy it is for finding shape differences of the upper-face pictures of moire, according to the Sasang constitution. 2. Method : We have collected 113 cases of patients of the Sasang Constitutional Department, including employees of the Kyung-Hee Medical Center and took pictures of moire of upper face and measured 50 points. We analyzed shape differences of the upper face according to the Sasang constitution. 3. Results : Analysing the moire of upper face, tile characteristics according to sasang constitution presented. 4. Conclusion : Soyangin had a projected part of left upper face compared with taeumin and soeumin, and taeumin had a projected part of right upper face. With the image of moire, the characteristics of upper face according to sasang constitution was found.

  • The responses of human pulp fibroblastic cells to Ga-As Semi-Conductor-Dens-Bio Laser (Frequency: 5 Hz~10,000 Hz Model: SD-101A RCA, U.SA)) were examined in vitro using pulp fibroblastic cells obtained from the pulp tissue of human tooth. The mitogenic effect of soft laser was assessed by measuring the MTT assay. The morphologic effect for soft laser showed under the scanning and transmission electron microscopy. The results as follows; 1. The mitogenic response of the soft laser was not observed until 4th time of radiation, while the mitogenic response at 4th time increased mitogenic effect by as much as 1.7 fold compared to the control value. 2. The mitogenic response of the soft laser on pulp fibroblast differ from the mitogenic response on other fibroblasts. 3. In scanning electron microscopic study, The microvilli of cell surface increased gradually with width and length after laser radiation, it demonstrate that development of microvilli have close connection with differentiation of cells. 4. Under the transmission electron microscope, The laser-treated cells maintained their elongated shape and a high degree of cellular polarization. The large cell body containing a well developed Golgi complex, a large number of profiles of rough endoplasmic reticulum, and great numbers of mitochondria. 5. The laser-treated cells maintained the long straight bundles of closely apposed microfilaments or individual filaments forming a cross-linked network. These findings suggest that the laser may have important roles in promotion of pulp healing and consequently may be useful for clinical application in pulp regenerative procedures.

Nucleus Recognition of Uterine Cervical Pap-Smears using Fuzzy Reasoning Rule (퍼지 추론 규칙을 이용한 자궁 경부진 핵 인식)

  • In this paper, we apply a set of algorithms to classily normal and cancer nucleus from uterine cervical pap-smear images. First, we use lightening compensation algorithm to restore color images that have defamation through the process of obtaining 1×400 microscope magnification. Then, we remove the background from images with the histogram distributions of RGB regions. We extract nucleus areas from candidates by applying histogram brightness, Kapur method, and our own 8-direction contour tracing algorithm. Various binarization, cumulative entropy, masking algorithms are used in that process. Then, we are able to recognize normal and cancer nucleus from those areas by using three morphological features - directional information, the size of nucleus, and area ratio - with fuzzy membership functions and deciding rules we devised. The experimental result shows our method has low false recognition rate.

Synergistic Effect of Carboxymethyl Chitosan and Adipose-Derived Mesenchymal Stem Cells on Osteoarthritis Model in Rabbits

  • The purpose of this study is to investigate the clinical effects of carboxymethyl chitosan (CMC) and adipose-derived mesenchymal stem cells (MSCs) on osteoarthritis (OA). Thirty New Zealand white rabbits were used as cranial cruciate ligament transection and partial meniscectomy models. The rabbits were divided into five groups (n = 6) according to the intra-articular injection materials: the control group with PBS, the HA group with hyaluronic acid, the CMC group with CMC, the MSC group with MSCs emerged in PBS, and the MSC+CMC group with CMC and MSCs. Knee thickness, extension angle, gross morphology, histopathology and immunohistochemistry were performed to evaluate the effects of CMC and MSCs on rabbit OA. On the morphologic and histologic examination, the articular surfaces of the femur and tibia were markedly damaged in control group with higher Mankin score and lower cartilage surface thickness. However, OA related cartilage defects were alleviated by the treatment of MSC and/or CMC. The expressions of apoptotic and inflammatory cytokines were decreased and cartilage extracellular matrix (ECM) related collagens I and II were enhanced by the treatment of MSC and/or CMC. In conclusion, this study showed that CMC and MSC treatments have a beneficial effects on OA via the protection of cartilage damage, the stimulation of ECM, and the inhibition of inflammatory and apoptotic reaction.

Histopathological classification of the bovine lymphoma caused by bovine leukemia virus in Korea (소 백혈병 바이러스 감염에 의한 소 림프종의 병리조직학적 분류)

  • Several studies have been performed on the bovine leukemia since bovine leukemia virus (BLV) had been detected in 1982 in Korea. We have conducted histopathological study on the bovine lymphoma caused by BLV because only limited results were reported on the pathological characterization of lymphoma. Lymphoma tissues were obtained from cattle necropsied and slaughtered during a designated period. Lymphoma was classified histopathologically according to the National Cancer Institute Working Formulation. Leukotic tissues consisted of fairly uniform sheets of closely packed medium to large lymphocytes without any architectural arrangement in all 30 cases. Twenty five cases belong to diffuse large cell type, while three cases were diffuse mixed cell type, and two cases were immunoblastic large cell type among 30 cases. Follicular type lymphoma was not detected in this study. The mitotic index of tumor cells showed average 2.5 in the field of 400X. Nuclear cleavage was detected in 53% of cases. Multi-nucleated cells were detected among tumor cells in 30% of lymphoma cases. In conclusion, the most common morphologic cell type of bovine lymphoma in Korea was a diffuse large cell type with multinucleated cells and nuclear cleavages.

The effect of selenium on renal lesions induced by adriamycin in rats (Selenium이 adriamycin에 의해서 유발되는 랫드 신장병변에 미치는 영향)

  • This study was carried out to investigate the effect of selenium on the adriamycininduced renal lesions in male Sprague Dawley rats. A total of 60 Sprague-Dawley male rats were divided into 2 control groups(C1: saline, C2: selenium) and 2 treatment groups(T1: adriamycin, T2: adriamycin+selenium). The rats of the C1 and T1 groups were given normal saline(0.15ml/rat), the rats of the C2 and T2 groups were given sodium selenite(0.5mg/kg) intraperitoneally three days a week for 4 weeks. The treatment groups were dosed intraperitoneally with adriamycin(2mg/kg/day) five days at the second week. Animals were sacrificed at the 1st week, 2nd week and 3rd week after dosing with adriamycin. The morphologic abnormalities of the glomeruli and tubules in the kidney of male rats were examined histopathologically and electron microscopically.The results obtained were as follows : The mean body weight of adriamycin dosed group was significantly decreased as compared with that of control group at 4th week(p<0.05). In adriamycin and selenium dosed group, the mean body weight was decreased until the end of 2nd week but gradually increased from 3rd to 4th week. The histopathological findings of the renal corpuscle in adriamycin dosed group were parietal epithelial cell proliferation, vacuolization of glomerulus, and thickened basement membrane of the parietal epithelium. Proximal convoluted tubules were significantly dilated and the lumens were filled with renal cast. These lesions were generally not very significant in the rats given adriamycin and selenium. The electron microscopical findings of the renal glomerulus in the adriamycin dosed group were focal loss and fusion of the pedicels of the podocyte, and some vacuoles in the cytoplasm of the podocytes. There were numerous cytoplasmic vacuoles in the proximal and distal convoluted tubular cells. However, these ultrastructural changes were not significantly observed in the renal tubules of the rats of adriamycin and selenium dosed group. These results suggest that selenium may act as an inhibitor of the renal lesions induced by adriamycin in male rats.
