SRD5A2 Gene Polymorphisms and the Risk of Benign Prostatic Hyperplasia but not Prostate Cancer

  • Background: Testosterone, a primary androgen in males, is converted into its most active form, dihydrotestosterone (DHT), by $5{\alpha}$-reductase type 2 (encoded by the SRD5A2 gene) in the prostate. DHT is necessary for prostatic growth and has five times higher binding affinity than testosterone for androgen receptors. We hypothesized that polymorphic variations in the SRD5A2 gene may affect the risk of benign prostatic hyperplasia and prostate cancer. Materials and Methods: We analyzed SRD5A2 gene polymorphisms in 217 BPH patients, 192 PCa cases, and 171 controls. Genotyping was undertaken using direct DNA sequencing. Genotype data were compared between cases and controls using a Chi square statistical tool. Results: We found that the A49T locus was monomorphic with 'AA' genotype in all subjects. At V89L locus, the presence of 'VV' showed a marginally significant correlation with increased BPH risk (p=0.047). At the $(TA)_n$ locus, longer TA repeats were found to be protective against BPH (p=0.003). However, neither of these polymoprhisms correlated with the risk of PCa. Conclusions: We conclude that A49T is monomorphic in the study population, VV marginally correlates with BPH risk, and longer $(TA)_n$ repeats are protective against BPH. None of these polymorphisms affect the risk of PCa.

Classification of Capsicum annuum Germplasm Using Random Amplified Polymorphic DNA (RAPD를 이용한 고추(Capsicum annuum) 유전자원의 분류)

  • This study was initiated to evaluate genetic relationship among various domestic and exotic pepper accessions using random amplified polymorphic DNA(RAPD) markers. The results suggested that the optimum conditions for PCR with random primers in Capsicum spp. could be obtained with 3mM of $MgCl_2$, 1.5U of Taq. DNA polymerase, 10ng of template DNA, $200{\mu}M$ of dNTPs, 200nM of random primer, and $42^{\circ}C$ of annealing temperature. Sixteen random primers showing high band intensity and reproducibility were selected from 80 random primers. Primers having 70% GC content were more effective in DNA amplification than primers having 60% GC content. The total 93 DNA bands including 71 polymorphic bands and 22 monomorphic bands were obtained with selected 16 random primers for 31 pepper cultivars and lines. About 4.4 polymorphic bands per primer were produced. Similarity coefficients were calculated by using 71 polymorphic bands and dendrogram based on the similarity coefficient showed clear classification of 31 peppers into three Capsicum species of Capsicum annuum, Capsicum chinense and Capsicum chacoense.

Comparison of SNP Variation and Distribution in Indigenous Ethiopian and Korean Cattle (Hanwoo) Populations

  • Although a large number of single nucleotide polymorphisms (SNPs) have been identified from the bovine genome-sequencing project, few of these have been validated at large in Bos indicus breeds. We have genotyped 192 animals, representing 5 cattle populations of Ethiopia, with the Illumina Bovine 8K SNP BeadChip. These include 1 Sanga (Danakil), 3 zebu (Borana, Arsi and Ambo), and 1 zebu ${\times}$ Sanga intermediate (Horro) breeds. The Hanwoo (Bos taurus) was included for comparison purposes. Analysis of 7,045 SNP markers revealed that the mean minor allele frequency (MAF) was 0.23, 0.22, 0.21, 0.21, 0.23, and 0.29 for Ambo, Arsi, Borana, Danakil, Horro, and Hanwoo, respectively. Significant differences of MAF were observed between the indigenous Ethiopian cattle populations and Hanwoo breed (p < 0.001). Across the Ethiopian cattle populations, a common variant MAF (${\geq}0.10$ and ${\leq}0.5$) accounted for an overall estimated 73.79% of the 7,045 SNPs. The Hanwoo displayed a higher proportion of common variant SNPs (90%). Investigation within Ethiopian cattle populations showed that on average, 16.64% of the markers were monomorphic, but in the Hanwoo breed, only 6% of the markers were monomorphic. Across the sampled Ethiopian cattle populations, the mean observed and expected heterozygosities were 0.314 and 0.313, respectively. The level of SNP variation identified in this particular study highlights that these markers can be potentially used for genetic studies in African cattle breeds.

Genetic Variations and Phylogenetic Relationship of and Pueraia lobata Ohwi (Fabaceae) and Related Taxa by RAPD Makers (RAPD분자마커를 이용한 칡(콩과) 및 근연분류군의 유전적 변이 및 유연관계)

  • RAPD analyses were performed to investigate genetic relationships and useful molecular maker for 3 species and their 17 regional populations of the Pueraria lobata and related taxa. The length of amplified DNA fragments ranged from 200 to, 2,800 bp. Two hundred and eight scorable polymorphic makers and three scorable monomorphic makers were found from the PCR reactions with 15 random oligo primers, and those were analyzed by Nei's genetic distance coefficient. Based on the UPGMA phenogram from RAPD analyses, two major groups (9 populations from Korea; 3 populations from foreign countries) were recognized. And it showed distinct genetic differences from related taxa. The RAPD results was very useful to define the samples by geographical distribution and to discuss the relationships among the populations and their related taxa of the Pueraria lobata.

Analyticity and Completeness in Intuitionistic Type Theory (직관주의적 유형론에서의 분석성과 완전성)

  • Based on his analysis of judgement forms in intuitionistic type theory, Martin-L$\ddot{o}$f claims that the usual logical laws and interesting mathematical judgements are synthetic, not analytic. He further claims that the logic of analytic judgements is decidable and complete, while the logic of synthetic judgements is undecidable and incomplete. The aim of this article is to clarify and examine his claims. In section 1, I explain and give some comments on the monomorphic version of intuitionistic type theory. In section 2, after clarifying Martin-L$\ddot{o}$f's distinction between analytic and synthetic judgements, I examine some possible objections to it and evaluate the thesis that the usual logical laws and interesting mathematical judgements are synthetic. In section 3, I clarify and examine the thesis that the logic of analytic judgements is decidable and complete, while the logic of synthetic judgements is undecidable and incomplete.

Allozyme Variation of 6-Phosphogluconate Dehydrogenase in the Freshwater Snail Genus Gyraulus (Pulmonata : Planorbidae)

  • The electrophoretic banding patterns of 6-phosphogluconate dehydrogenase (PGD) in the two different chromosomal ploidy groups of Gyraulus were compared. The monomeric or dimeric banding patterns or allelic variations in a locus of PGD were observed in four diploid populations (Osan, Sohre, Kimpo and Kangwha) of G. convexiusculus occurring in Korea, whereas the isozyme banding patterns encoded by at least 3 different loci were shown in the tetraploid populations of G. (Torquis) groups collected from Michigan, the U.S.A. Of 3 different tetraploid groups, G. (T.) circumstriatus group showed 3 monomorphic isozyme banding patterns, and the other 2 populations showed some allelic variations. Such results provided good evidence to differentiate tetraploid subgenus Torquis group from the diploid Gyraulus populations.

  • We experienced a case of well-differentiated acinic cell carcinoma of the parotid gland in a 65 year-old woman, which was correctly diagnosed preoperatively by fine needle aspiration(FNA) cytology. FNA cytology smears showed clusters or sheets of monomorphic acinic cells having reticulated or finely vacuolated basophilic or acidophilic cytoplasm. The cellular population was homogeneous or slightly polymorphic, having centrally located, round nuclei with finely reticular chromatin and inconspicuous nucleoli. Herein we report this case with its histologic features and review of literatures.

  • Acinic cell carcinoma is a rare salivary gland tumor of low-grade malignancy. It comprises only about 2.5% of all salivary gland tumors. We recently experienced a case of fine needle aspiration cytology of acinic cell carcinoma of the parotid gland. The characteristic cytopathologic features were 1) cellular aspirate consisting of monomorphic cells in large sheets or singly, 2) formation of acini and/or microcysts, 3) abundant granular cytoplasm with sharp cytoplasmic borders, 4) bland nuclei with micronucleoli, and 5) clear background.

  • DNA restriction fragment length polymorphisms(RFLPs) were used for the classification of 22 leading silkworm races and wild silkworm, Bombyx mandarina. A genomic DNA library from silkworm was partially constructed and was prescreened to evaluate the selected DNA probes. Three DNA probes (SP1-13, SP1-28, 10-42) were selected to determine the polymorphism between silkworm races. As a result, high polymorphism with the probe SP1-28, moderate polymorphism with SP1-13 and monomorphism with 10-42 were obse-rbed.

  • Secretory carcinoma is a very rare tumor of the breast, having characteristic histologic findings. A cytologic study of a secretory carcinoma is presented. The smears were abundant in tumor cells which were arranged in loosely adherent or large tight clusters in clear background. The individual cells were round or oval, monomorphic, and had abundant eosinophilic cytoplasm. The cytoplasm showed distinct borders and single or fine vacuolization. The nuclei were peripherally located, oval, and vesicular with small prominent nucleoli. As in histology, the cytologic features were so characteristic that a diagnosis of secretory carcinoma could be made by ctyologic study only.
