재래흑염소와 교잡종 염소의 Monomorphic SNP 분석을 통한 유전적 다양성과 집단구조의 비교 및 검증 (Comparison and Validation of Genetic Diversity and Population Structure Using Monomorphic SNP Data of the Korean Native Black Goat and Crossbred Goat)

  • 본 연구는 우리나라 고유의 재래흑염소 집단인 당진, 장수, 통영 및 경상대 계통과 교잡종 염소 계통 또는 해외품종의 개체 식별을 위한 유전적 다양성과 관계 조사 및 검증을 위해 수행하였다. 각 염소 집단에 존재하는 Monomorphic SNP를 수집한 이후 공통적으로 존재하는 SNP 133개를 선발하여 분석에 이용하였다. Monomorphic SNP 133개를 통한 재래흑염소와 교잡종 염소의 유전적 구조 차이를 나타냈으며, 주성분 분석 결과 재래흑염소와 교잡종 염소가 명확히 구분되는 것으로 나타났다. 또한, 참조집단 이외의 70두(Native Korean goat = 24, Cross breed = 46)로 구성된 검증집단을 분석한 결과 국내 재래흑염소 계통의 참조집단과 동일한 유전적 구조를 나타냈으며, 교잡종 염소의 경우 참조집단의 일부 유전적 구성을 공유하는 것으로 나타났다. 국내 재래흑염소의 경우는 하나의 군집을 형성한 반면 해외 품종 및 교잡종 계통의 경우 재래흑염소 계통에 비해 넓게 퍼져 군집을 형성하는 것으로 나타났다. 따라서, 본 연구 결과는 국내 재래흑염소 유전자원 집단을 보존을 위한 기초자료로 활용하고 추후 유전적 다양성을 고려한 염소의 개량을 위한 기초자료로 유용하게 활용 할 수 있을 것으로 판단된다.

  • Let R be a commutative ring with total quotient ring K. Each monomorphic R-module endomorphism of a cyclic R-module is an isomorphism if and only if R has Krull dimension 0. Each monomorphic R-module endomorphism of R is an isomorphism if and only if R = K. We say that R has property (${\star}$) if for each nonzero element $a{\in}R$, each monomorphic R-module endomorphism of R/Ra is an isomorphism. If R has property (${\star}$), then each nonzero principal prime ideal of R is a maximal ideal, but the converse is false, even for integral domains of Krull dimension 2. An integral domain R has property (${\star}$) if and only if R has no R-sequence of length 2; the "if" assertion fails in general for non-domain rings R. Each treed domain has property (${\star}$), but the converse is false.

Phase Transitions and Phase Diagram of the Island Model with Migration

  • We investigate the evolutionary dynamics and the phase transitions of the island model which consists of subdivided populations of individuals confined to two islands. In the island model, the population is subdivided so that migration acts to determine the evolutionary dynamics along with selection and genetic drift. The individuals are assumed to be haploid and to be one of two species, X or Y. They reproduce according to their fitness values, die at random, and migrate between the islands. The evolutionary dynamics of an individual based model is formulated in terms of a master equation and is approximated by using the diffusion method as the multidimensional Fokker-Planck equation (FPE) and the coupled non-linear stochastic differential equations (SDEs) with multiplicative noise. We analyze the infinite population limit to find the phase transitions from the monomorphic state of one type to the polymorphic state to the monomorphic state of the other type as we vary the ratio of the fitness values in two islands and complete the phase diagram of our island model.

한국산 또아리물달팽이과 ( family Planorbidae ) 3종에서의 동위 효소 변이 (Isozyme Variability in Three Species of Freshwater Planorbid Snails in Korea : Gyraulus convexiusculus , Hippeutis cantori and Segmentina hemisphaerula)

  • A horizontal starch gal electrophoresis for enzyme proteins extracted from 3 species of Korean planorbid snails; Gyraulus convexiusculus, Hippeutis cantori and Segmentina hemisphaerula was carried out in order to elucidate their genetic relationships.The results from 12 enzymes employed in three different kinds of buffer systems are summarized as follows:1) Two loci from each enzyme of aldehyde oxidase, esterase, glucose phosphate isomerase. isocitrate dehydrogenase, leucine aminopeptidase, malate dehyogenase, peptidase and xanthine oxidase were detected, and only one locus was observed from each of the following four enzymes: 6-phosphogluconate dehydrogenase, glyceraldehyde-phosphate dehydrogenase, glutamate oxaloacetate transaminase and glycerol-3-phosphate dehydrogenase.2) Most of loci in 3 species of planorbid snails employed showed homozygous and monomorphic banding patterns and some of them were specifis as genetic markers among different species. However, a few of loci (EST-1. EST-2 and GPI-2)showed polymorphic banding patterns. 3)Hippeutis cantori and Segmentina hemisphaerula were more closely clustered in a dendrogram with the genetic iddentity value of 0.431, and these two species were lineated with Gyraulus convexiusculus as another cluster at the value of 0.294.In summarizing the above results, three species of Korean planorbid snails employed in this study mostly showed monomorphic enzyme protein banding patterns and genetic differences specific among 3 species.

  • An enzyme analysis of the liver fluke, Clonorchis sinensis from Kimhae, Korea and from Shenyang, China was conducted using a horizontal. starch gel electrophoresis in order to elucidate their genetic relationships. A total of eight enzymes was employed from two different kinds of buffer systems. Two loci from each enzyme of aconitase and esterase (${\alpha}-Na{\;}and{\;}{\beta}-Na$) : and only one locus each from six enzymes, gluucose-6-phosphate dehydrogenase (G6PD), ${\alpha}-glycerophosphate$ dehydrogenase (GPD), 3-hydroxybutyrate dehydrogenase (HBDH), malate dehydrogenase (MDH), phosphoglucose isomerase (PGI), and phosphoglucomutase (PGM) were detected. Most of loci in two populations of C. sinensis showed homozygous monomorphic banding patterns and one of them, GPD was specific as genetic markers between two different populations. However, esterase (${\alpha}-Na$), GPD, HBDH and PGI loci showed polymorphic banding patterns. Two populations of C. sinensis were more closely clustered within the range of genetic identity value of 0.998-1.0. In summarizing the above results, two populations of C. sinensis employed in this study showed mostly monomorphic enzyme protein banding patterns, and genetic differences specific between two populations.

갑상샘 섬암종의 세침흡인 세포학적 소견 - 온전한 섬모양 구조를 가진 1예 보고 - (Fine Needle Aspiration Cytology of Insular Carcinoma of the Thyroid - Report of A Case with Intact Insulae -)

  • Insular carcinoma of the thyroid(ICT) is an uncommon thyroglobulin-producing neoplasm, intermediate between well differentiated and anaplastic carcinoma. Only a few publications have addressed the fine needle aspiration cytologic(FNAC) findings from ICT. We experienced a case histologically diagnosed as ICT and with preoperative FNAC in a 52 - year - old woman. The FNAC displayed scanty colloid and abundant monomorphic follicular cells presented singly, in small loose aggregates, and in cohesive trabecular or acinar clusters. Intact insulae of tumor cells were also identified. Necrosis and mitosis were rare. Tumor cells showed round and monomorphic nuclei, finely granular chromatin, and inconspicuous nucleoli. When insular structure is identified in thyroid FNAC specimen, ICT should be included in the differential diagnosis. Herein we discuss and review the cytologic criteria for separation of ICT from other thyroid neoplasms.

  • A horizontal starch gel electrophoresis for enzyme proteins extracted from 2 species of Korean viviparid snails; Cipangopaludina chinensis malleata and C. japonica was carried out in order to elucidate their genetic relationships. A total of 10 enzymes were employed in three different kinds of buffer systems. Two loci from each enzyme of alcohol dehydrogenase, esterase, glucose phosphate isomerase, isocitrate dehydrogenase, iditol dehydrogenase, malate dehydrogenase and peptidase(VL); and only one locus dach from two enzymes, glycerlo-3-phosphate dehydrogenase and phosphoglucomutase were detected; but, four loci from peptidase(LGG) were observed. Most of loci in two viviparid species showed homozygous monomorphic banding patterns and some of them were specific as genetic markers between two different species. However, EST-1, MDH-1, PEP(VL)-1loci showed polymorphic banding patterns. Foru populations of C. chinensis malleata were more closely clustered in a dendrogram within the range of genetic identity values of 0.928-1.00, and these clusters were lineated with C. japonica at the value of 0.355. In summarizing the above results, two viviparid snail species dmployed in this study mostly showed monomorphic enzyme protein banding patterns, and genetic differences specific between two species.

RAPD를 이용한 용담의 유전적 유사도 분석 (Analysis Genetic Similarity of Gentiana scabra var. buergeri by Randomly Amplified Polymorphic DNA)

  • 서철재래종, 내장산 수집종, 대둔산 수집종, 거제도 수집종, 진부재래종 및 일본 도입종 등 계통의 용담을 대상으로 RAPD 분석을 통한 유사도 분석을 수행한 결과 적용한 20가지의 10-merprimer 중 8가지에서 54개의 증폭된 DNA 절편을 얻었다. 그 중 53.7%에 해당하는 29개의 band가 다형현상을 보였으며, 9.3%에 해당하는 5개의 band는 모든 계통에서 공통적으로 나타났고, 37%인 20개의 band가 특이성을 보였다. 특이성을 보이는 band들은 내장산 수집종에서 2개, 거제도 수집종에서 1개, 진부재래종에서 11개, 일본 도입종에서 6개가 관찰되었다. 유전적 유사도의 분석결과 서천재래종, 내장산 수집종, 대둔산 수집종 및 거제도 수집종이 0.76-0.87의 유사도를 보여 한 집단으로 구분되었으며, 이 집단과 진부재래종 및 일본 도입종 사이의 유사도는 0.23-0.34로 각 각 다른 집단으로 구별되었다. 또한 진부재래 종과 일본 도입 종 사이의 유사도는 0.41로 나타나 서로 구분되었다.

타액선 종양에 있어서 S-100 단백과 Lactoferrin에 관한 면역조직화학적 연구 (Immunohistochemical Studies on S-100 Protein and Lactoferrin in Salivary Gland Tumors)

  • Immunohistochemical studies on S-100 protein and lactoferrin were carried out to evaluate the existence and distribution pattern of S-100 protein and lactoferrin positive cells in salivary gland tumors. The specimens used were 25 cases of pleomorphic adenoma, 2 cases of monomorphic adenoma, 2 cases of mucoepidermoid tumor, 2 cases of acinic cell tumor, 3 cases of adenoid cystic carcinoma and 2 cases of adenocarcinoma occured in parotid and submandibular salivary gland. ABC kits(Dako corp. Copenhagen. Denmark) for S-100 protein and lactoferrin were used. The results obtained were summarized as follows: In the normal salivary gland. positive immunoreaction for S-100 protein was observed in myoepithelial cells of acini and intercalated ducts. Positive immunoreaction for lactoferrin was observed in serous acinic cells, epithelial cells of intercalated ducts, and excretory material in the ductal lumina. In the pleomorphic and monomorphic adenomas. most of tumor cells were positive for S-100 protein, while luminal tumor cells in gland-like or duct-like structures were rarely positive for lactoferrin. In mucoepidermoid tumor, most of squamous cells and a few of intermediate cells were positive for S-100 protein, but all of tumor cells were negative for lactoferrin. In acinic cell tumor, most of tumor cells were positive for lactoferrin, but all of tumor cells were negative for S-100 protein. In adenoid cystic carcinoma, basaloid tumor cells in trabecular structure were focally positive for S-100 protein. and in adenocarcinoma, many of tumor cells were posivive for both S-100 protein and lactoferrin. Thus, according to the embryonic stage of the development of the tumor cell origin, it was possible to classify the salivary gland tumor as followings: mucoepidermoid carcinoma which originated from the earliest stage, acinic cell tumor which originated from the end stage. Between these two extremes, there were pleomorphic adenoma, adenoid cystic carcinoma and adenocarcinoma which originated in the middle stage of the development of .the salivary glands. Based on the above results, it can be stated that S-100 protein is demonstrated in tumor cells orginated from myoepithelial cells and lactoferrin in glandular differentiated tumor cells.

RAPD Analysis of Three Deer Species in Malaysia

  • The genetic variability within and among three deer species in Malaysia, namely Cervus nippon (sika), Cervus timorensis (rusa) and Cervus unicolor (sambar), were evaluated using the RAPD technique. The DNA extracted from the buffy coat of 34 sika, 38 rusa and 9 sambar were analysed using ten primers that gave bands which showed good resolution. The primers generated 164 RAPD markers in total, and these ranged in size from 150 to 900 bp. The percent of polymorphism of the bands generated per primer ranged from 66.66-93.33% for rusa, 36.84-61.14% for sambar and 52.38-100% for sika. The overall percent polymorphism observed for the 164 RAPD markers was 99.39%. The results revealed five exclusive, monomorphic markers for sambar and one exclusive, monomorphic marker for sika; none was observed for rusa. However, these cannot be declared as markers for the identification of the species without analysis of more samples, populations and species. The means of within population genetic distances, based on Dice's and Jaccard's similarity indices, were similar for the rusa (0.383 and 0.542, respectively) and sika (0.397 and 0.558, respectively) populations with the sambar population being the least variable (0.194 and 0.323, respectively). The Dice based genetic distances within the species ranged from 0.194 to 0.397 and the genetic distances among the species were 0.791-0.911. The genetic distances based on Dice's and Jaccard's similarity indices between the rusa and sambar were 0.556 and 0.713, between the rusa and sika populations were 0.552 and 0.710, and between sambar and sika were 0.622 and 0.766, respectively.
