  • The behavior of continuous flow culture of Lartobacillus bulgricus was investigated by application of Monod's kinetic model. The parameters obtained from Monod's chemostat theory successfully predicted the behavior of the chemostat. Then, it was found that Monod's kinetics were applicable to the growth rate dependence on glucose concentration. Under steady-state condition, the maximum growth rate, saturation constant, and wash out were found to be 0.62/hr, 7.69 g/1, 0.51/hr of continuous culture. And the optimum condition for the highest cell production was 0.41/hr in dilution rate, and at that point the cell production rate was 0.178g/1 hr.

  • Monod kinetics에 관련된 주요 생분해 파라미터를 도출하기 위하여 microcosm 규모의 배치실험에서 BTEX 화합물에 대해 분해능이 우수한 Pseudomonas aeruginosa을 이용해 다양한 농도의 벤젠에 대한 분해기작을 고찰하였다. 벤젠의 생분해율(D)과 Maximumspecific growth rate ($\mu$$_{max}$)는 기질의 농도가 증가할수록 높아지다가 최고점에 도달 후에 점차적으로 감소하였으며 이것은 어느 한계점 이상의 벤젠 농도가 미생물의 생분해에 방해 요소로 작용한다는 것을 나타낸다. 그러나 미생물에 의한 벤젠 분해의 상관관계를 나타내는 yield coefficient(Y)는 벤젠의 초기 농도가 낮을수록 높은 값을 나타내었다. Microbial decay constant( b)와 half-saturation constant(K$_{c}$)는 각각 0.21~0.48day$^{-1}$와 218mg/$\ell$로서 문헌값 보다 높은 수치를 나타내었다. 실험으로부터 결정된 생분해 파라미터들은 초기 벤젠 농도에 따라 큰 차이를 보이므로 생분해 모델링에 사용할 파라미터는 기질농도에 따라 적절하게 선택되어야 한다고 사료된다.

  • Palm Oil Mill Effluent (POME) is the mixed organic wastewater generated from palm oil industry. In this study, kinetic analysis with treating POME in an anaerobic hybrid reactor (AHR) was performed. Therefore, the AHR was monitored for its performances with respect to the changes of COD concentrations and hydraulic retention time (HRT). Batch tests were performed to find out the substrate removal kinetics by granular sludge from POME. Modified Stover Kincannon, First-order, Monod, Grau second-order kinetic models were used to analyze the performance of reactor. The results from the batch test indicate that the substrate removal kinetics of granular sludge is corresponds to follow Monod's theory. However, Grau second-order model were the most appropriate models for the continuous test in the AHR. The second order kinetic constant, saturation value constant, maximum substrate removal rate, and first-order kinetic constant were 2.60/day, 41.905 g/L-day, 39.683 g/L-day, and 1.25/day respectively. And the most appropriate model was Grau second-order kinetic model comparing the model prediction values and measured COD concentrations of effluent, whereas modified Stover-Kincannon model showed the lowest correlation.

Batch and Continuous Culture Kinetics for Production of Carotenoids by ${\beta}$-Ionone-Resistant Mutant of Xanthophyllomyces dendrorhous

  • A ${\beta}$-ionone-resistant mutant strain isolated from the red yeast Xanthophyllomyces dendrorhous KCTC 7704 was used for batch and continuous fermentation kinetic studies with glucose media in a 2.5-1 jar fermentor at $22^{\circ}C$ and pH 4.5. The kinetic pattern of growth and carotenoid concentration in the batch fermentations exhibited a so-called mixed-growth-associated product formation, possibly due to the fact that the content of intracellular carotenoids depends on the degree of physical maturation toward adulthood. To determine the maximum specific growth rate constant (${\mu}_m$) and Monod constant ($K_s$) for the mutant, glucose-limited continuous culture studies were performed at different dilution rates within a range of $0.02-0.10\;h^{-1}$. A reciprocal plot of the steady-state data (viz., reciprocal of glucose concentration versus residence time) obtained from continuous culture experiments was used to estimate a ${\mu}_m$ of $0.15\;h^{-1}$ and $k_s$ of 1.19 g/l. The carotenoid content related to the residence time appeared to assume a typical form of saturation kinetics. The maximum carotenoid content ($X_m$) for the mutant was estimated to be $1.04\;{\mu}g/mg$ dry cell weight, and the Lee constant ($k_m$), which was tentatively defined in this work, was found to be 3.0 h.

  • The mathematical model of specific growth rate of Saccharomyces cerevisiae ATCC 24858 is proposed as a function of sugar and ethanol concentrations by the combination of Andrew's equation and Aiba's equation. The maximum concentration of sugar Sm, which was the highest concentration of sugar not having any effect on the growth inhibition, was 150 g/L and the substrate inhibition was expressed as a function of (S-Sm). The maximum specific growth inhibition, was 150 g/L and the substrate inhibition was expressed as a function of (S-Sm). The maximum specific growth rate ${\mu}m$, Monod's constant Ks, and Andrew's inhibition constant KI were 0.49 hr-1, 19 g/L, and 139 g/L, respectively. The maximum ethanol concentration, Pm, which did not show any inhibition effect on the specific growth rate was found to be 2 g/L. Therefore, the ethanol inhibition was represented as a function of (P-Pm). The final mathematical model for the specific growth rate of the microorganism in this work is proposed as the following. And the average percent of errors between the calculated specific growth rate and the experimental values was 5.96%.

A Kinetic Modeling for the Dynamics of Hybridoma Cells in Suspension Culture (현탁배양 하이브리도마 세포의 속도론적 모델링)

  • Batch suspension cultures of hybridoma cell were performed with various initial glutamine concentrations to investigate the effects of glutamine on cell growth and death, monoclonal antibody production, glucose and glutamine consumption, and the production of lactate and ammonium ion. An mathematical kinetic model was formulated to describe the kinetics of cell growth, the consumption of nutrients (glucose and glutamine), and the production of monoclonal antibody and waste metabolites (lactate and ammonium ion) based on experimental data. An equation for the specific growth rate was developed such that superimposed Monod equation in glucose and glutamine, with non-competitive type inhibition relations in ammonium ion and lactate. The inhibition constant for lactate was inversely proportional to the lactate concentration. The specific death rate was considered to be a function of glucose, glutamine, ammonium ion and lactate concentration.

Optimal Cultur Conditions for the Production of Insecticidal Toxin by Xenorhabdus nematophilus Isolated from Steinernema carpocapsae (Steinernema carpocapsae로부터 분리된 Xenorhabdus nematophilus에 의한 살충물질 생산을 위한 최적 배양조건)

  • Optimal medium composition, culture conditions, characteristics of phase variation and activity of insecticidal toxin by Xenorhabdus nematophilus isolated and identified from Korean entomopathogenic nematode Steinernema carpocapsae were examined. Optimal medium composition of this strain was 50-70 g/L yeast extract, 3 g/L $K_{2}HPO_{4}$, 1g/L $NH_{4}H_{2}PO_{4}$, 2g/L ${MgSO}_4$$\cdot$${7H}_{2}O$, 10g/L NaCl and, these, yeast extract was found as a limiting nutrient for cell growth. When Monod equation was applied, maxmum specific growth rate and Monod constant were estimated as 0.13 $hr^{-1}$ and 20g/L, respectively. The pH of culture medium increased up to 8.5-9.5 regardless of initial pH 6-7 as the cells continued to grow. The specific growth rate in a 7 L fermentor was 0.18 $hr^{-1}$, which was enhancement 1.4 fold compared to a flask culture. In case of phase variation, phase I fraction was maintained above 90% at the stationary phase for both flask and fermentor cultures. According to oral toxicity test of Gallena mellonella by Xenorhabdus nematophilus, the addition of cell pellets into feed inhibited normal growth of insect larvae and killed completely then after 20 days cultivation. When culture supernatant of this strain was injected into hemolymph of insect larva, the toxicity was strongest at 24hr cultivation in the early exponential phase and gradually decreased as the culture time proceeded.

Improved Production of Live Cells of Lactobacillus rhamnosus by Continuous Cultivation using Glucose-yeast Extract Medium

  • In this study, the growth kinetics of Lactobacillus rhamnosus and lactic acid production in continuous culture were assessed at a range of dilution rates $(0.05 h^{-1}\;to\;0.40h^{-1})$ using a 2L stirred tank fermenter with a working volume of 600ml. Unstructured models, predicated on the Monod and Luedeking-Piret equations, were employed to simulate the growth of the bacterium, glucose consumption, and lactic acid production at different dilution rates in continuous cultures. The maximum specific growth rate of L. rhamnosus, ${\mu}_{max}$, was estimated at $0.40h^{-1}$I, and the Monod cell growth saturation constant, Ks, at approximately 0.25g/L. Maximum cell viability $(1.3{\times}10^{10}CFU/ml)$ was achieved in the dilution rate range of $D=0.28h^{-1}\;to\;0.35h^{-1}$. Both maximum viable cell yield and productivity were achieved at $D=0.35h^{-1}$. The continuous cultivation of L. rhamnosus at $D=0.35h^{-1}$ resulted in substantial improvements in cell productivity, of 267% (viable cell count) that achieved via batch cultivation.

Bio-kinetic and Design Analysis for Box-mill Wastewater Treatment Using Anoxic Activated Sludge Process (무산소 활성오니공정을 이용한 판지공장 폐수처리의 동력학적 해석 및 설계분석)

  • The anoxic activated sludge process was applied to the treatment of industrial box-mill wastewater, which exhibited the high removal efficiencies of $90{\sim}94%$$ TCOD_{Mn}$ and $58{\sim}81%$ Color. For the design of industrial anoxic activated sludge process, Monod bio-kinetic coefficients of box-mill wastewater were estimated as follows: $K_{max}$(maximum specific substrate removal rate)=0.52 $day^{-1}$, $K_s$(half saturation constant)=314 mg/L, $K_d$(decay coefficient)=0.274 $day^{-1}$, y(microbial yield coefficient)=0.908 mg/mg, and ${\mu}_{max}$(maximum specific growth rate)=0.472 $day^{-1}$. Space loading factors for the design analysis were practically determined as the values of F/M ratio=$0.043{\sim}0.07$ kg-$TCOD_{Mn}$/kg-SS-day, BOD space loading=$0.18{\sim}0.3$ kg-$TCOD_{Mn}/m^3-day$, and ${\theta}_x=6.8{\sim}26.4$ day when considering the relationship of these loading factors with growth dynamics of microorganisms, the F/M ratio that is inversely proportional to ${\theta}_x$ should be equivalent to ${\mu}_{max}$ in units, but exhibited the significant difference between theses two values. Therefore, it is considered that high safety factors are requested in the design of anoxic activated sludge process that is based on Monod bio-kinetics of microorganism.

광합성세균 미생물막반응기에 의한 유기성폐수의 처리특성

  • An efficient packed-bed type biofilm reactor charged with immobilized phototrophs was developed to treat organic wastewater at an extremely high volumetric loading rate. The packed bed reactor (PBR) charged with porous ceramic beads was superior to a fluidized-bed reactor suspended with activated carbon powders in terms of many aspects such as BOD removal efficiency, operational stability, and overall economics. For wastewater with BOD concentration as high as 20, 000mg/l, the BOD removal efficiency was maintained above 90% when the hydraulic retention time (HRT) was longer than 1 day. The allowable volumetric BOD loading rate of this reactor (20gBOD/l day) is more than ten-folds higher than that of an ordinary activated sludge method. The behaviour of the reactor was represented well by a Monod type kinetic equation with a maximum specific BOD loading rate(P) of 22.2gBOD/l day and a half saturation constant(K$_{s}$) of 1, 750 mgBOD/l.
