The Analysis of Vascular Plant Species Composition in Dok-do Island (독도 유관속 식물상과 종조성 분석)

  • The investigated vascular plants of Dok-do island were confirmed that consisted of 46 species, 1 subspecies, 1 varieties in total of 48 taxa growing spontaneously. 1 taxa of Pteridophyta, 36 taxa of Dicotyledon and 11 taxa of Monocotyledon identified. In 48 identified taxa in this study, total 13 taxa included Orobanche coerulescens Stephan, Fallopia sachalinensis (F. Schmidt) Rouse Decr., Lonicera insularis Nakai identified according to the Korean plant taxa for environmental assessment. Planting species were about 10 taxa included Pinus thunbergii Parl., Hibiscus syriacus L. and etc. Four species confirmed in this study such as between Chenopodium virgatum Thunb. and Chenopodium album L., Echinochloa crusgalli (L.) P.Beauv. and Echinochloa crusgalli var. oryzicola (Vasinger) Ohwi. were doubt on identification in taxonomic entity due to morphological characteristics. The dispersion type of Do-kdo plants were anemochore 31 taxa, zoodchore 8 taxa, hydrochore 2 taxa and the others by artificial means.

The Distribution of Vascular Plants Recorded in the Hangul Tripitaka (한글대장경에 기록된 관속식물의 분포)

  • The purpose of this research is to classify various plants written in the Korean Buddhist scriptures and assign scientific name to them in accordance with the Hangul Tripitaca as standard scriptures, thereby establishing the foundation of the plants in Buddhist scriptures. As a natural resource, we also assessed the value of the plants written in the Buddhist scriptures that distributed from other countries. To this end, we inferred the route of introduction to Korea of the plants and investigated their function and usage. Taxonomic classification of the plants written in Buddhist scriptures identified a total of 331 taxa belonging to 107 families, 244 genera, 313 species, 1 subspecies, 16 varieties, 1 forms. Species composition of the 331 taxa of plants consisted of two taxa of pteridophyte, 15 taxa of gymnosperm, 261 taxa of dicotyledon, and 53 taxa of monocotyledon. Among them, 183 and 148 were woody and herbaceous plants, respectively. Analysis of the distribution of 331 kinds of plants that India's portion of origin of place is 8.9% which holds a low rank compared with those of Korea (10.5%), China (10.9%), and Japan (10.0%). This explains why many taxa of plants distributed from the three countries appeared in the Hangul Tripitaca. Accordingly, we hypothesized that the majority of the plants were added and replaced at some point in past while Buddhist scriptures were propagated from India to China and from China to Korea. Our analysis indicated that 119 out of the 331 taxa (36 %) were distributed from Korea.

  • This study has been focused on improving transformation efficiency of rice using Agrobacterium tumefaciens. We have demonstrated the effect of this system when the GPAT gene related to the cold-resistance was transferred by Agrobacterium tumefaciens in rice. Transformation conditions were modified using intron $\beta$-glucuronidase (GUS) expression as a reporter gene in the rice. In this study, mature seed-derived calli of rice (Oruza sativa L. cv. Dongjin) were pre-cultured for 3 days and then infected with Agrobacterium. When this infected calli were cultured in the dark for 10 days on co-cu]lure medium containing 50 mg/L of CaCl$_2$, 30 mg/L of acetosyringone, 2 mg/L of 2,4-D, 120 mg/L of betaine, high GUS expression was observed. In the present transformation system, the efficiency of transformation of GPAT gene was about 54%. Stable integration of GPAT gene into chromosomal DNA was proven by southern blot analysis of genomic DNA isolated from T$_{0}$ progenies. The progenies (T1 generation) derived from primary transformant of 5 lines were segregated with a 3 (resistant) : 1 (sensitive ratio) in medium containing hygromycin. This high frequency transformation system can be used as a useful tool in transformation of another monocotyledon.n.

  • This study was carried out a field survey and review articles to investigate a characteristic of the distribution pattern in Korean endemic Corylopsis coreana. Distribution of Corylopsis coreana was found from Boseung-gun, Gwangyang-si, Suncheon- si, Jeumanl-myeon Goheung-gun, Ganjeon-myeon Gurye-gun, Mt. Cheongwan Jangheung-gun, Nogodan Mt. Jiri in Jeollanam-do, Mt. Cheonhwang Namwon-si and Beamsagol and Banyabong Mt. Jiri, in Jeollabuk-do, Sancheong-gun, hadong-gun and Namhea-gun in Gyeongsangnam-do and Mt. Backun, Pocheon-si in Gyeonggi-do. Total flora in the distribution region of Coryloplsis coreana were recorded as 242 taxa; 70 families, 159 genus, 216 species, 22 varieties, 4 forma. Ratio of taxonomic categories was consisted of Pteridophyte $5.8\%$, Gymnosperm $3.7\%$, Dicotyledon $18.6\%$ and Monocotyledon $71.9\%$. Ratio of Life cycle styles was Annual $1.7\%$ and Perennial $98.3\%$. Ratio of growth habit was appeared to Herb $44.2\%$, Vine $12.4\%$, Shrub $17.8\%$ and Tree $25.6\%$. The distribution region of Corylopsis coreana found to typical type which distribute in N, NE or NE slope. The native Corylopsis coreana of Sinjeonri Sungju-eup in Jellanam-do appeared to a declining tendency which caused by influence of tree lager.

Establishment of a regeneration system for the production of Calla plants (Zantedeschia spp.) via embryogenic callus culture (배발생캘러스 배양에 의한 칼라 식물체 재분화 체계 확립)

  • Calla lilies (Zantedeschia spp.) are monocotyledonous ornamental plants which belongs to the Araceae family. After the release of elite calla cultivar, an efficient propagation system is needed for commercial use. Despite the use of conventional propagation methods such as splitting of tubers and rhizomes of calla, rapid and efficient propagation system should be developed. In order to achieve this goal, stem segments contained apical meristems derived from calla lily cultivar (cv. Gag-si) were cultured on Murashige and Skoog (MS) medium supplemented with various concentrations of cytokinin and auxin. This was aimed at inducing embryogenic calluses, shoots and multiple shoots. As a result, about 25% of induction rates of yellow embryogenic calluses were observed with MS medium containing both $0.5mg{\cdot}L^{-1}\;NAA$ and $1.5mg{\cdot}L^{-1}\;BA$ as growth regulators. In the experiments involving the regeneration from embryogenic calluses through shoot formation, MS medium supplemented with $0.5mg{\cdot}L^{-1}\;IAA$ and $2.0mg{\cdot}L^{-1}\;BA$ showed the highest rates at approximately 85 ~ 90% with regard to the formation of shoots in calla. Moreover, multiple shoots needed for rapid propagation were generated when explants were cultured on MS medium supplemented with $0.5mg{\cdot}L^{-1}\;IAA$ and $2.0mg{\cdot}L^{-1}\;BA$ with 40% of formation rate. In this study, the combination of auxin and cytokinin showed positive effects on both the induction of embryogenic calluses, the formation of shoots as well as multiple shoots in calla. The regeneration system described here can contribute to the development of breeding programs of calla in the future.

The Vegetation and Flora of Village Groves in Paengseong-eup, Pyongtaek City, Gyonggi-Do, Korea (경기도 평택시 팽성읍 마을숲의 식물상 및 식생)

  • A village grove is the one fostered and protected for a district's traditional community life near the vicinity of a village. This study is done by surveying the vegetation and flora at the village grove situated at the former location for the US military camp. Through such data collected from this study, this researcher could have access to preliminary data needed for restoration project & preservation of not only the village grove surveyed but also the village grove of its neighboring district. According to the survey, there appeared an outgrowth flora of Pteridophyta consisting of 2 families, 2 genuses, 1 species, and 1 variety, totalling to 2 taxa; a Gymnosperm of 2 families, 2 genuses, 3 species, totalling to 3 taxa; in the realm of an Angiosperm, a Monocotyledon consisting of 5 families, 10 genuses, and 11 species, totalling to 11 taxa; Dicotyledoneal consisting of 24 families, 39 genuses, 40 species, and 3 varieties, totalling to 43 taxa, thus revealing 33 families, 53 genuses, 55 species, 4 varieties and 59 taxa in total. Urbanization index(4.2%) and Naturalization ratio(20.3%) showed a relatively higher numerical value than those of other districts. The vegetation unit comprising a village grove is classified Into the four community units, such as Pinus densiflora community, Pinus densiflora-Quercus serrata community, Quercus serrata community, and Rosa multiflora community. Quercus serrata community is of a community type dominated by Quercus serrata, in place of Pinus densiflora selected by natural succession, which shows relatively superior lighting adaptability and was surveyed to grow up on not only a tree layer but also a sub-tree layer. With the recent appearance of industrialization, the use of a village grove is reducing due to villagers' leaving, decrease in the number of households, and urbanization; consequently, there came out a pattern of succession to Quercus serrata community, which could be assumed to be potential natural vegetation of this district, after passing through the process of Pinus densiflora-Quercus serrata community.

High frequency somatic embryogenesis through leaf explant-derived callus culture in Muscari armeniacum cv. 'Early Giant' (무스카리 'Early Giant' 잎 절편 유래 캘러스 배양을 통한 고빈도 체세포배 발생)

  • Using calli of $Muscari$ $armeniacum$ cv. 'Early Giant' that is monocotyledonous ornamental bulb crop with increasing demand in Korea, we carried out current studies to establish an in vitro multiple propagation protocol via somatic embryogenesis. We found that soft pale yellow green calli were induced from leaf explants cultured on all media containing 0.1~3.0 $mg{\cdot}L^{-1}$ auxins such as 1-naphthalene acetic acid (NAA) and 2,4-dichlorophenoxyacetic acid (2,4-D). However, induced calli showed vigorous growth only when they further transferred on same media containing 2,4-D, 4-amino-3,5,6-tri-chloropicolinic acid (picloram), or 3,6-dichloro-o-anisic acid (dicamba). Although frequency of somatic embryo induction depended on callus source and PGR composition in somatic embryo induction media, somatic embryogenesis was initiated on surface of proliferated calli after transferring on media with no PGR or 0.01 $mg{\cdot}L^{-1}$ NAA co-supplemented with various cytokinins such as $N^6$-benzylaminopurine (BAP). Highest number of embryo at 9.3 per callus clump was obtained when calli which were grown under 0.1 $mg{\cdot}L^{-1}$ picloram supplementation were sub-cultured on medium with 0.01 $mg{\cdot}L^{-1}$ NAA and 0.5 $mg{\cdot}L^{-1}$ BAP. In addition, morphological characteristics of somatic embryo were categorized into following nine phases: globular, biased heart, biased torpedo, early cotyledonary, middle cotyledonary, late cotyledonary, early sprouting, middle sprouting, and late sprouting embryos.

  • Camelina sativa that belongs to Brassicaceae family is an emerging oilseed crop. Camelina seeds contain approximately 40% storage oils per seed dry weight, which are useful for human and animal diets and industrial applications. Microsomal delta-12 fatty acid desaturase2 (FAD2) catalyzes the conversion of oleic acid to linoleic acid. The polymorphisms of FAD2 genes are correlated with the levels of oleic acids in seed oils. In this study, three CsFAD2 genes (CsFAD2-1, CsFAD2-2 and CsFAD2-3.1) were isolated from developing seeds of Camelina sativa (L.) cv. CAME. The nucleotide and deduced amino acid sequences of three CsFAD2 genes were compared with those from dicotyledon and monocotyledon plants including Camelina cultivars Sunesone and SRS933. Three histidine motifs (HECGH, HRRHH, and HVAHH) required for FAD activity and a hydrophobic valine or isoleucine residue, which is a SNP (single nucleotide polymorphism) marker related with enzyme activity are well conserved in three CsFAD2s. The expressions of CsFAD2-1 and CsFAD2-3.1 were ubiquitously detected in various Camelina organs, whereas the CsFAD2-2 transcripts were predominantly detected in flowers and developing seeds. The contents of oleic acids decreased, whereas the amounts of linoleic acid increased in dry seeds of transgenic fad2-2 lines expressing each CsFAD2 gene compared with fad2-2 mutant, indicating that three CsFAD2 genes are functionally active. The isolated CsFAD2 genes might be applicable in metabolic engineering of storage oils with high oleic acids in oilseed crops.
