Isolation and Partial Purification of the Steroid 9${\alpha}$-Hydroxylase from Mycobacterium fortuitum (Mycobacterium fortuitum의 스테로이드 9${\alpha}$-하이드록실라제의 분리 및 부분정제)

  • The steroid 9${alpha}$-hydroxylase activity has been detected in cytosol fraction, $100,00{\times}g$ supernatant of cell free extract of Mycobacterium fortuitum. The activity was not linear with protein concentration in the assay suggesting 9${alpha}$-hydroxylase is a multicomponent enzyme. The 9${alpha}$-hydroxylase system was partially purified through fractional saturation of ammonium sulfate, strong anion exchange (Mono Q) column chromatography, gel filtration (Superose 12) column chromatography, and testosterone affinity gel chromatography. Ammonium sulfate 50~60% saturated fraction of the cytosol gave 9${alpha}$-hydroxylase activity. For further purification, the half-saturated ammonium sulfate fraction was applied to Mono Q, Superose 12, or affinity gel column. The purification factors of 9${alpha}$-hydroxylase containing fraction after Mono Q, Superose 12, and affinity gel chromatography was 13, 11, and 17 respectively.

  • (1) The durability of fifteen species of hard woods grown in Kangwon-do in relation to Polystictus Versecolor L. Fr. and Irpex consors Berk was studied. (2) The least affected hard wood species by Polystutus Versecolor L. Fr. were Q. serrata, C. crenata, M. amurensis, F. rhynchophylla and J. mandschrica; the moderatly affected species were Q. Variabilis, U. Davidiana var japonica, B. latiforia, C. controversa, P. padus and A. mono; and the most highly affected species were P. serrulata var. spondanea, S. obassia, T. amurensis and B. schmidtii. The least affected hard wood species by Irpex consors Berk, were U. Davidiana var japonica, C. crenata, M. amurensis, F. rhynchophylla, and J. mandshurica; the moderatly affected species were Q. Valiabilis, Q. serrata, P. serrulata var. spondanea, C. controversa, and A. mono; and the most highly affected species were B. latiforia, P. padus, S. obassia, T. amurensis, and J-mandshurica. (3) No difference was obscured on the durabilities on effected by kind of fungus. (4) Correlationship between specific gravity and durability of the woods was not significant.

Vegetation Structures and Ecological Niche of Quercus serrata Forests (졸참나무림의 식생구조와 생태적지)

  • The aim of this study was to characterize the forest vegetation structure and site of Quercus serrata forest for ecological forest management and ecological niche. The results are as follows : The chemical properties of Q. serrata forest soil were 0.24% of total nitrogen, 8.27 of organic matter, 74ppm of available phosphorous, 1.64(me/100g) of Ca, 0.22(me/100g) of Mg, 0.74(me/100g) of K and 9.3(me/100g) of cation exchangeable capacity. The dominant species in Quercus serrata forest were Quercus serrata, Quercus acutissima, Quercus variabilis, Quercus mongolica, Styrax obassia, Fraxinus rhynchophylla and Styrax japonica. DBH analysis showed that Quercus serrata seems to remain as a dominant species for the present because they had random distribution based on few of big individuality, many of small and middle individuality. But the Q. serrata community is competing with Q. mongolica and F. rhynchophylla, whose density of small individuality has increased. With the classification of TWINSPAN, Q. serrata forest was classified three groups, such as Q. serrata-Acer mono, Q. serrata, Q. serrata-Q. acutissima communities. The results of the correlation analysis of Q. serrata major communities and environment factors are as follows; Q. serrata-A. mono community was found relatively in high elevated and eastern and northern area that has relatively high percentage organic matter. Also Q. serrata community was found in high elevated and eastern and northern area that has high percentage organic matter. Q. serrata-Q. acutissima community was found in low elevated and southern and western area that has low percentage organic matter.

Selection of Desirable Species and Estimation of Composition Ratio in a Natural Deciduous Forest (천연활엽수림(天然闊葉樹林)의 경영대상(經營對象) 수종(樹種) 선정(選定) 및 구성비율(構成比率) 추정(推定))

  • Based on the community structural attributes, such as species composition, diameter and height distribution, topographic position, and species diversity in the natural deciduous forest of Mt. Gari area, this study suggested desirable species and composition ratio to achieve ecological management of forests so as to maintain forest stability and enhance economical values. The results are as follows : 1. Twenty-five tree species were growing in the study forest. Of these Quercus mongolica, Pinus densiflora, Juglans mandshurica, Quercus serrata, Cornus controversa, Acer mono, Fraxinus rhynchophylla, and Tilia mandshurica were selected for desirable species through the evaluation of dominant and dominant potential. Kalopanax pictus, considered to be highly valuable species, was also included. 2. Taking account of different species composition pattern by topographic positions, we select as desirable species of J. mandshurica, C. controversa, Q. mongolica, A. mono, T. mandshurica, and F. rhynchophylla in the valley area, Q. mongolica, Q. serrata, A. mono, T. mandshurica, F. rhynchophylla, and K. pictus in the mid-slope area, and Q. mongolica, P. densiflora, Q. serrata, and Fraxinus rhynchophylla in the ridge area. 3. Based on the estimation of species diversity index for the overstory components, the reasonable forest stability levels of the indices were estimated at 1.96, 1.68, 1.94, and 1.27 for whole forest, valley, midslope, and ridge, respectively. 4. The recommended species composition ratios in the study forest were suggested Q. mongolica to be 30%, A. mono, F. rhynchophylla, Q. serrata, and T. mandshurica to be 10%~15%, J. mandshurica, P. densiflora, and C. controversa to be 5%~10%, and K. pictus to be 5%.

  • The purposes of this study are to improve the ultrasound resolution of various nylon and metallic mono-filament wires, therefore, it was tested that it analyze on nylon mono-filament wire of 0.1 mm in A Co.'s ultrasonic phantom and synthesis of C15 g tissue mimicking materials(TMM), analyze resolution of nylon and metallic mono-filament wires in water and TMM. The results obtained were summarized as follows: 1. Metallic mono-filament wire of 0.1 mm and nylon mono-filament wire of 0.12 mm, 180 denier showed that it cleared dot echo pattern. 2. Metallic and nylon mono-filament wire of 0.2 mm showed that it cleared comet tail echo by reverberation artifact. 3. Nylon and metallic mono-filament wire of 0.1 mm showed that it can used for dead zone and axial resolution test. 4. Nylon mono-filament wire compared with metallic mono-filament wire showed that it satisfy elasticity and construction. 5. Degree of hardness of na not changed mono-filament's echo textures.

Structure-Property Relationship of PVA-SbQ Water Soluble Photosensitive Polymer and its Application to Screening Process of Color Monitor (PVA-SbQ 수용성 감광성 고분자의 구조와 감도관계 및 칼라 수상관 스크린 공정에의 응용)

  • Photosensitive compound, 1-methyl-4-[2-(4-diethylacetylphenyl)ethenyl] pridinium methosulfate(SbQ-A salt), was synthesized from dimethyl sulfate, terephthalaldehyde mono-(diethylacetal) and 4-picoline. SbQ-A salts were reacted with poly(vinyl alcohol)s, (PVA) in aqueous solution with phosphoric acid as catalyst to give photosensitive PVA-SbQ with different SbQ content and molecular weight. Relative photosensitivity of PVA-SbQ was determined by gray scale(GS) method. The rotative sensitivity of PVA-SbQ increased with increasing amount of bound SbQ in the case of high molecular weight(MW=77,000-79,000g/mol) as substrate and decreased with decreasing molecular weight of PVA with about constant(1.3mol%) amount of bound SbQ. The most sensitive polymer was obtained when SbQ group content in PVA-SbQ reached about 2.63mol% in the case of high molecular weight(77,000-79,000g/mol) PVA. This sample showed 90 times greater sensitivity than dichromated PVA as reference photosensitive system. PVA-SbQ photosensitive polymer synthesized was applied to the photolithographic screening process of phosphor on the panel of cathode ray tube(CRT). Phosphor slurry was made with PVA-SbQ, phosphor, a small amount of surfactant and other additives using water as medium. The slurry was coated onto panel, dried by heater, exposed to UV light and then developed by distilled water. When a small amount of cationic surfactant such as cetyltrimethylammonium chloride was used in the slurry formulation, the sharpness of phosphor pattern was equal to or better than that of dichromated PVA photosensitive polymer system used currently.

Vegetation Pattern and Successional Sere in the Forest of Mt. Odae (오대산 삼림식생의 패턴과 천이계열)

  • The vegetation pattern of Mt. Odae based on the soil humidity gradient showed 3 types: (1) the forest of Pinus densiflora under the mesic or xeric conditions of the low altitudinal area, (2) the forest of Acer including A. mono, A. pseudo-sieboldianum and Tilia amurensis under the submesic or subxeric conditions and (3) the forest of Quercus including Q. mongolica of the higher elevational area and Q. variabilis of the lower elevational area under the xeric condition. Water content, organic matter and total nitrogen of soil were relatively low in Pinus densiflora and Quercus variabilis communities while they were relatively high in Betula platyphylla var. japonica and Quercus mongolica communities. According to the result of cluster analysis based on similarity indices of the communities, the proposed successional sere in the forest vegetation of Mt. Odae was as follows. P. densiflora community $\longrightarrow$ P. densiflore + Q. mongolica community $\longrightarrow$ Q. mongolica + A. pseudo-sieboldianum community. P. densiflora community $\longrightarrow$ P. densiflora + Q. variabilis community $\longrightarrow$ Q. variabilis community $\longrightarrow$ Q. mongolica + Q. variabilis community $\longrightarrow$ Q. mongolica + A. pseudo-sieboldianum community.

  • To investigate the forest structure in Ijontong Cholto area, twenty four plots were set up (each l0$\times$10m ) and surveyed. According to the analysis of ordination by DCA, community divided into 5 groups. The divided groups are Quercus serrata -Q. valiabilis community(I), Q. serrata - Pinus densiflora community(II), Q. mongolica - Lindera obtusiloba community(III), Carpinus laxiflora - Acer pseudo-sieboldianum community(IV) and Acer mono community(V). So the successional trends seems to be from P. densiflora through Q. serrata, Q. valiabilis, Q. mongolica to C. laxiflora, C. cordata, A. mono in canopy layer. And in the subtree and shrub layer, it was expected that from Rhododendron mucronulatum, Rh. schlippenbachii to A. pseudo-sieboldianum, L. obtusiloba. But some sites were likely to become Q. mongolica edaphic climax.

Isolation and Characterization of Exogenously Expressed Calmodulin from Endogenous Tobacco Calmodulin by Anion-exchange Fast Protein Liquid Chromatography

  • A Mono Q HR 5/5 anion-exchange column with a FPLC system was used to separate exogenously expressed calmodulin from endogenous tobacco calmodulins. Transgenic tobacco calmodulins were purified by fractionation with ammonium sulfate, precipitation with sulfuric acid and hydrophobic chromatography on phenyl-Sepharose CL-4B. The purified calmodulins were chromatographed in the FPLC using the column. This method was selected because of the slight differences in the net charge of foreign and endogenous plant calmodulins due to amino acid sequence differences. By this approach, the exogenously expressed calmodulin was isolated from endogenous tobacco calmodulins. The isolated calmodulin was characterized by amino acid composition analysis as well as methylation analysis.

Isolation of calcium-binding peptides from porcine meat and bone meal and mussel protein hydrolysates (돼지 육골분 및 진주담치 단백질의 가수분해물 제조 및 칼슘 결합 물질의 분리)

  • Calcium is one of the essential mineral for the humans due to its crucial physiological functions in the body. Calcium deficiency results in many diseases, such as osteoporosis. Therefore, calcium supplements are available as a functional food. However, most calcium supplements in the market have a limitation due to poor absorption and low bioavailability. Thus, calcium-chelated peptides for improving the absorption rate of calcium have been isolated from foods including porcine meat and bone meal (MBM), and mussel using the enzymatic hydrolysis of their protein. The hydrolysates of food were ultra-filtered in order to obtain small peptides less than 3 kDa and the Ca-binding peptides were isolated via the anion exchange chromatography. The binding activity and concentration of Ca-binding pepetides were determined. In particular, the MBM and mussel protein hydrolysates were fractionated by mono Q and Q-Sepharose, respectively. As a result, among the fractions, the fractions of MBM F2 and mussel F3 showed the highest Ca-binding activity. These results suggest that MBM and mussel protein hydrolysates can be used as calcium supplements.
