  • In crustacea the molting cycle is the most important physiological process affecting growth, behaviour, reproduction and population dynamics through their life span. Particularly molt cycle is closely related to reproductive cycle, and it is important to determine the successive stages of molt cycle to understand reproductive phenomena including ovarian cycle and the development of the embryos in the marsupium.   • Ovarian cycle, marsupial development and molt cycle of Archaeomysis kokuboi were studied to understand these processes as a whole event. Based on morphological characters the marsupial development is divided into 3 stages, Embryonic stage (duration time of 4 days), Nauplioid stage (5 days) and Postnauplioid stage (4 days). Morphological description was ma de for the 3 stages, and each stage was examined in relation to the corresponding stages of ovarian cycle and molt cycle.

초등예비교사들의 곤충의 탈피와 변태에 관한 개념 (Elementary Preservice Teachers' Conceptions on Molt and Metamorphosis of Insect)

  • Elementary preservice teachers' understanding about the insect molt and metamorphosis was examined. Data were collected through the paper-pencil test of 448 junior students at a National University of Education. The instrument used was developed by Son(2003) and modified for the test. The results are as follows: First, Only 3.1$\%$ of students had a scientific concept on the molt periods in the insect life cycle. Students had an alternative conception that molt occurs either only in the larval period (29.9$\%$) or only in the period from pupa to adult (22.3$\%$). The percentage of the students thinking that molt occurs in the period from larva to pupa was low. Second, 98$\%$ of students stated that molt has to do with the larval growth, but 39.1$\%$ of students had an alternative conception that molt occurs after the larval growth at each instar. 25.1$\%$ had a conception that molt is related with the larval growth, but did not show any further understanding. Third, most students understood the correct meaning of metamorphosis. 34..2$\%$ of students had an alternative conception that metamorphosis occurs only in the period changing from pupa to adult, and only a few thought that it occurs in the period from larva to pupa. 24.8$\%$ of students had a scientific concept on the periods in which metamorphosis occurs. Fourth, some students understood the hatching process as molt in the sense that the egg shell is taken off (21.0$\%$), and as metamorphosis in terms of the appearance change from egg to larva (25.0$\%$). Fifth, 35.5$\%$ of students selected bees as an insect of incomplete metamorphosis, and 35.3$\%$ responded 'I have no idea', showing that they had poor understanding about the insect life cycle.

Induction of Intrinsic and Extrinsic Apoptosis Pathways in the Human Leukemic MOLT-4 Cell Line by Terpinen-4-ol

  • Terpinen-4-ol is a terpene found in the rhizome of Plai (Zingiber montanum (Koenig) Link ex Dietr.). In this study apoptogenic activity and mechanisms of cell death induced by terpinen-4-ol were investigated in the human leukemic MOLT-4 cell line. Terpinen-4-ol exhibited cytotoxicity in MOLT-4 cells, with characteristic morphological features of apoptosis by Wright's staining. The mode of cell death was confirmed to be apoptosis by flow cytometric analysis after staining with annexin V-FITC and propidium iodide. A sub-G1 peak in DNA histograms of cell cycle assays was observed. Terpinen-4-ol induced-MOLT-4 cell apoptosis mediated through an intrinsic pathway involving the loss of mitochondrial transmembrane potential (MTP) and release of cytochrome c into the cytosol. In addition, terpinen-4-ol also induced apoptosis via an extrinsic pathway by caspase-8 activation resulting in the cleavage of cytosolic Bid. Truncated-Bid (tBid) translocated to mitochondria and activated the mitochondrial pathway in conjunction with down-regulation of Bcl-2 protein expression. Caspase-3 activity also increased. In conclusion, terpinen-4-ol can induce human leukemic MOLT-4 cell apoptosis via both intrinsic and extrinsic pathways.

징거미 새우, Macrobrachium nipponense(De Haan) 유생의 탈피주기와 관련한 생리적 변화 (Physiological Changes in Related to Molt Cycle of Macrobrachium nipponense(De Haan))

  • 담수산 징거미새우, Macrobrachium nipponense의 유생을 부화후부터 후기유생단계까지의 전 유생기간동안 수온 $25^{\circ}C$, 염분 $7\%0$의 일정한 조건하에서 사육하고 섭이율, 호흡율, 배설율 및 성장율을 측정하였다. 성장은 건중, 탄소, 질소, 수소 및 단백질, 지질의 함량변화로서 측정하였는데 단백질보다는 지질이 더 높은 비율로 축적되었다. 그리고 모든 생리적, 생화학적 특성은 각 유생단계간에 그리고 개체의 탈피주기에 따라서 상당한 변화를 나타내었다. 평균섭이율은 탈피간기에 가장 높았으며 bell-shaped 양상을 나타내었다. 호흡율은 부화후부터 후기유생 단계까지 계속 증가한 반면, 배설율은 탈피간기에서 증가하여 bell-shaped 양상을 나타내었다. 이러한 생리적, 생화학적 자료로서 시간의 경과에 따른 발생변화의 양상과 생물에너지학적 효율을 분석하기 위하여 섭이율, 성장율, 호흡율 및 배설율등을 회귀방정식으로 나타내었다. 탈피주기동안 탄소는 초기에 증가하는 경향을 나타내었으며, 암모니아 질소는 탈피전기까지 꾸준히 증가하는 경향을 나타내었다. 단백질은 탈피전기의 중기에 다소 높은 축적을 보이면서 탈피전기의 말기까지 일정량을 유지하면서 체물질을 구성하는 우점성분으로 일정하게 유지되었다.

  • 담수산 징거미새우, Macrobrachium nipponeue의 유생을 수온 $25^{\circ}$, 염분 $7\%o$의 일정한 조건하에서 사육하면서 섭이율, 호흡률 및 성장률을 측정하였으며, 이를 토대로 하여 생물에너지학적 효율을 분석하기 위하여 모든 유생기간 동안 탄소의 흡수 및 분배에 대한 탄소수지를 구명하였다. 생리적, 생화학적 자료로서 시간의 경과에 따른 발생변화의 양상과 섭이율, 성장률 및 호흡률 등을 회귀방정식으로 나타내었다. 일일 섭이율(F)과 비탄소섭이율(F/C)은 대체로 탈피후기에서 최소를 나타내었다. 그리고 성장율(G, G/C)은 탈피주기 동안 감소하는 경향을 나타내었다. 탈피주기 동안 탄소는 초기에 증가하는 경향을 나타내었으며, 질소는 탈피전기까지 꾸준히 증가하는 경향을 나타내었다. 단백질은 탈피전기의 중기에 다소 높은 축적을 보이면서 탈피전기의 말기까지 일정량을 유지하면서 체물질을 구성하는 우점성분으로 일정하게 유지되었다. 유생의 섭이(F), 성장(G), 호흡(R) 및 동화량(A)의 절대값은 탈피주기동안 뚜렷한 변화를 나타내었다. 탈피주기 가운데 탈피전기로 진행하면서, 성장률보다 호흡율이 더 커졌으며, 탈피주기 동안 변화하고 있는 호흡을 대사계수, 즉 호흡이 탄소로 전환되어 이화되고 있는 부분으로 나타내었다. 탈피후기에서 R/G<1, (R1, (R>G)로 변동하였다. 부화후부터 후기 유생단계까지의 누적탄소수지를 보면, 총섭이량은 $491.54{\mu}g\;C/ind$였으며, 동화효율은 $85.3\%$ 호흡율은 $47.7\%$ 그리고 성장율은 $37.6\%$였다. 그리고 총섭이량중 동화되지 않고 소실된 양은 $72.19{\mu}g\;C/ind$로서 $14.6\%$를 차지하였다.

  • This study has been conducted to investigate the molting behavior and the effects of rearing temperature and day length on molting in the behavior and the effects of rearing temperature and day length on molting in the giant freshwater prawn, Macrobrachium rosenbergii reared in the laboratory. The results obtained were summarized as follow: 1. After pre-spawning molting, the protopodites of 1st, 2nd, 3rd, and 4th except 5th pleopod bore new breeding setae which conserve eggs in the brooding chamber and the basis of 3rd, 4th, and 5th pereiopods bore new breeding dresses which transport the ovulated eggs into the brooding chamber. 2. Adult females reared in 27.5-$29.4^{\circ}C$ molted 10-12 times per year at interval of 27-35 days, of which four or six moltings were common molting for growth and another four or five moltings were pre-spawning molting for spwaning and brooding. In winter season, pre-spawning molting did not happen to most of adult females in spite of the same temperature. 3. Duration of intermolt cycle was 31-38 days and 26-30 days at 25.3- $26.5^{\circ}C$ and 28.7- $30.4^{\circ}C$ of rearing temperature, respectively.

프라이머 중합체를 이용한 원위치 중합효소 연쇄반응 In situ PCR 방법의 개발 (Development of In situ PCR Method Using Primer Polymers)

  • 효과적인 원위치 중합효소 연쇄반응 (In situ PCR)을 위해서는 증폭된 PCR 산물의 세포외 유출을 감소시켜야 한다. 이를 위한 한 방법으로 거대분자 PCR 산물을 합성시키기 위한 5'쪽에 서로 상보적인 꼬리서열을 가진 프라이머(꼬리 프라이머; tailed primer)가 사용되었으나 많은 PCR 횟수로 인해 시간의 낭비와 세포조직의 형태보존성이 저하되는 문제가 발생하였다. 따라서 PCR 조건을 가능한 최적화시키고, 최소의 PCR 횟수로써 세포외 유출을 막을 수 없는 방법이 필요하게 되었다. 이러한 방법의 일환으로 꼬리 프라이머를 이용하여 PCR 튜브 속에서 목표 핵산없이 프라이머 중합체(primer polymers)의 형성을 유도하였고, 이를 유리 슬라이드위에 고정시킨 Molt/LAV 세포들에 처리하여 20 회의 짧은 시간에서도 적절한 탐침을 할 수 있게 되었다. 이로 인해 프라이머 중합체의 원위치 중합효소 연쇄반응에서의 사용가능성을 타진하였다.

보중익기탕가미방의 면역기능 증진 효과 (Immune Function-enhancing Effects of Bojungikkitanggami-bang)

  • The immune system acts to protect the host from infectious agents that exist in the environment and from other noxious insults. The immune system has two functional divisions: the innate and the acquired. Both components involve various factors such as cytokines. A number of methodologies exist to assess aspects of immune function. There are large inter-individual variations in many immune functions even among the healthy. Genetics, age, gender, smoking habits, habitual levels of exercise, alcohol consumption, diet, stage in the female menstrual cycle, stress, history of infections and vaccinations, and early life experiences are likely to be important contributors to the observed variation. While it is clear that individuals with immune responses significantly below 'normal' are more susceptible to infectious agents and exhibit increased infectious morbidity and mortality, it is not clear how the variation in immune function among healthy individuals relates to variation in susceptibility to infection. Oriental medicine is an important factor contributing to immune competence. The author investigated the immune enhancement effects of Bojungikkitanggami-bang (BITB). The forced swimming test (FST) has been used as a screening model for new immune enhancement agents. In the present study, the author investigated the effects of BITB on FST and blood biochemical parameters related to fatigue, glucose (Glc); blood urea nitrogen (BUN); lactate dehydrogenase (LDH); creatinine; and total protein (TP). The author found that BITB (1 g/kg) significantly reduced the immobility time in the FST compared to the control. In addition, the contents of Glc, LDH, BUN, TP in the blood serum were increased in BITB (1g/kg)-fed group. Also, the author investigated the effects of BITB on the production of cytokines in human T-cell line, MOLT-4 cells. BITB (1 mg/ml) significantly increased the interferon (IFN)-vproduction compared with media control (about 2.2-fold for IFN-γ) at 24 h. However, BITB has not affect the production of IL-2 and IL-4. In addition, BITB increased the protein expression level of IFN-γ in MOLT-4 cells. Thus, BITB may have therapeutic value in generating or enhancing immune function in a clinical setting.

Endocrine Disruptors in Developing Embryo on Daphnia magna

  • In crustaceans, as in other arthropods, the molt cycle and the physiological process of growth are controlled by molting hormones (MH) which are steroid hormones, the ecdysteroids. Ecdysteroids are major arthropod hormones which control both development (embryonic and larval molts, metamorphosis) and reproduction. The purpose of the present study was to evaluate both fenarimol and methoprene for embryotoxicity to daphnids. The embryotoxicity associated with each compound was assessed to discern whether the embryotoxicity of methoprene might be due to ecdysone agonist and the ecdysone antagonistic effects of fenarimol on Daphnia embryo. Exposure of daphnids for three weeks to 50 M methoprene resulted in a significantly high incidence of offspring that exhibited general toxicity. This exposure concentration had significant effects on the overall number of embryo death. However, exposure to 3 or 1 $\mu$M fenarimol were no significant effects on the embryo toxicity. The incidence of both of these toxicity increased with methoprene exposure. This observation suggest that methoprene showed embryonic general toxicity during embryo development, while, only fenarimol showed weak general toxicity with early stages of embryonic development.
