부식질의 광산화 및 오존산화에 있어서의 분자량 크기분포 변화 특성에 관한 연구 (A Study of Molecular Size Distributions of Humic Acid by Photo-Oxidation and Ozonation)

  • 고급산화공정 (AOP)인 UV시스템과 오존시스템을 이용하여 부식산의 광산화 및 오존 산화를 실시한 후, 용존유기탄소 (DOC)의 제거 효율에 따른 분자량 분포 특성을 한외여과법을 이용하여 조사하였다. 반응 전의 부식산의 분자량 분포는 30,000 daltons 이상의 고분자 물질이 41.5%로 가장 큰 부분을 차지하고 있었으며, 500 dalton 이하의 저분자 물질은 15.2%로 상대적으로 낮은 분포율을 보였다. UV 조사 시간이 증가함에 따라 고분자에서 저분자로의 전환율이 증가하였다. 특히, 30,000 daltons 이상의 고분자물질이 생물학적으로 처리 효율이 높은 500 daltons 이하의 저분자물질로 전환되는 비율은 UV 단독조사 (35.3%)에 비교해 촉매가 첨가된 경우인 $UV/TiO_2$$UV/H_2O_2$ 시스템에서 각각 58.9%와 87.7%으로 증가하였다. 오존 시스템에서는 500 daltons 이하의 저분자로의 전환율보다는 3,000~30,000 daltons의 중간크기 분자량 분포율이 증가하였다. 오존 단독 시스템에서는 10,000~30,000 daltons 크기의 분포율이 최종 60분 처리시 41.5%로 가장 높게 나타났으며, $O_3/H_2O_2$ 시스템에서는 10,000~30,000 daltons과 3,000~10,000 daltons이 각각 38.9%, 26.2%으로 높은 분포율을 나타냈다. 이상에서 얻어진 결과를 토대로 수중 부식산의 보다 효과적인 제거를 위하여, $UV/H_2O_2$, $UV/TiO_2$$O_3$, $O_3/H_2O_2$ 시스템 등과 연계하여 처리할 수 있는 단위공정을 제안하였다.

Effect of γ-Irradiation on the Molecular Properties of Bovine Serum Albumin and β-Lcatoglobulin

  • To elucidate the effect of oxygen radicals on the molecular properties of proteins, the secondary and tertiary structure and molecular weight size of BSA and ${\beta}$-lactoglobulin were examined after irradiation of proteins at various doses. Gamma-irradiation of protein solutions caused the disruption of the ordered structure of protein molecules as well as degradation, cross-linking, and aggregation of the polypeptide chains. As a model system, BSA and ${\beta}$-lactoglobulin were used as a typical ${\alpha}$-helical and a ${\beta}$-sheet structure protein, respectively. A circular dichroism study showed that the increase of radiation decreased the ordered structure of proteins with a concurrent increase of aperiodic structure content. Fluorescence spectroscopy indicated that irradiation quenched the emission intensity excited at 280 nm. SDS-PAGE and a gel permeation chromatography study indicated that radiation caused initial fragmentation of proteins resulting in a subsequent aggregation due to cross-linking of protein molecules.

Effect of γ-Irradiation on the Molecular Properties of Myoglobin

  • To elucidate the effect of gamma-irradiation on the molecular properties of myoglobin, the secondary and tertiary structures, as well as the molecular weight size of the protein, were examined after irradiation at various irradiation doses. Gamma-irradiation of myoglobin solutions caused the disruption of the ordered structure of the protein molecules, as well as degradation, cross-linking, and aggregation of the polypeptide chains. A SDS-PAGE study indicated that irradiation caused initial fragmentation of the proteins and subsequent aggregation, due to cross-linking of the protein molecules. The effect of irradiation on the protein was more significant at lower protein concentrations. Ascorbic acid protected against the degradation and aggregation of proteins by scavenging oxygen radicals that are produced by irradiation. A circular dichroism study showed that an increase of the irradiation decreased the a-helical content of myoglobin with a concurrent increase of the aperiodic structure content. Fluorescence spectroscopy indicated that irradiation increased the emission intensity that was excited at 280 nm.

$3{\beta}$[L-Lysinamide-Carbamoyl] Cholesterol Cationic Lipid as a Biocompatible Vector for Efficient Gene Transfer

  • In this paper, we report a new cationic lipid composed of L-lysinamide and cholesterol as a potent gene delivery vector. $3{\beta}$[L-Lysinamide-carbamoyl] cholesterol could self-assemble with plasmid DNA forming discrete lipoplexes. From atomic force microscopic images of the complexes, the size distribution was observed to range from 100 to 150 nm in diameter. The transfection efficiency of this amphiphile on different cell lines was evaluated as a micellar solution in the absence of the fusogenic helper lipid, dioleoyl phosphatidyletbanolamine (DOPE). Transfection experiments were performed as a function of charge ratio (lipid/DNA) and transfection time. Cytotoxicity and in vitro transfection efficiency of the amphiphile was demonstrated and compared with those of commercially available Lipofectin and polyethylenimine (PEI).

  • Some of the benzopyrene (BP)-DNA adduct are known to build intercalated motif between flanking base pairs in damaged DNA depending on the structural condition. The size of benzopyrene itself is definitely not comparable with any of the DNA bases and thus the question whether the lesion of some base pair by insertion of benzopyrene can happen with or without a dramatic distortion of the helical structure is a highly interesting theme. In this work we used a molecular dynamics simulation based on the theory of molecular mechanics. The specific consequences about the structural properties of the intercalated structures and benzopyrene motif in minor groove of the double helix are deduced after 5 ns simulation time.

Grain Size Effect on Mechanical Properties of Polycrystalline Graphene

  • Characteristics of nanocrystalline materials are known substantially dependent on the microstructure such as grain size, crystal orientation, and grain boundary. Thus it is desired to have systematic characterization methods on the various nanomaterials with complex geometries, especially in low dimensional nature. One of the interested nanomaterials would be a pure two-dimensional material, graphene, with superior mechanical, thermal, and electrical properties. In this study, mechanical properties of "polycrystalline" graphene were numerically investigated by molecular dynamics simulations. Subdomains with various sizes would be generated in the polycrystalline graphene during the fabrication such as chemical vapor deposition process. The atomic models of polycrystalline graphene were generated using Voronoi tessellation method. Stress strain curves for tensile deformation were obtained for various grain sizes (5~40 nm) and their mechanical properties were determined. It was found that, as the grain size increases, Young's modulus increases showing the reverse Hall-Petch effect. However, the fracture strain decreases in the same region, while the ultimate tensile strength (UTS) rather shows slight increasing behavior. We found that the polycrystalline graphene shows the reverse Hall-Petch effect over the simulated domain of grain size (< 40 nm).

폴리우레탄 폼을 이용한 친유성 유체의 흡착 (Adsorption of Hydrophobic Fluid by Polyurethane Foam)

  • 본 연구에서는 폴리우레탄 폼의 셀 내부구조 및 크기가 친유성 유체의 흡유능에 미치는 영향을 체계적으로 연구하였다. 우선 다양한 분자량의 폴리올 (GP-1000, GP-3000, GP-4000, GP-5000)을 이용하여 폴리우레탄 폼을 제조하여 그 기본적인 물성을 조사하였다. 폴리올의 분자량이 증가함에 따라 셀 크기가 감소하고 흡유량이 2000% 이상 증가하였다. Surfactant량이 증가하고 흡착되는 유체의 점도가 증가함에 따라 흡유량이 현저하게 감소함을 알 수가 있었다. 또한 폴리올의 분자량이 증가함에 따라 표면강도가 상승하였는데 이는 분자량 증가에 따른 유체점도의 상승이 폼밀도를 증대시킨데 기인한 것으로 해석할 수 있었다. 폼생성시 교반속도가 증가하여도 밀도가 증가하였다.

Brassica rapa Sec14-like protein gene BrPATL4 determines the genetic architecture of seed size and shape

  • Seed size traits are controlled by multiple genes in crops and determine grain yield, quality and appearance. However, the molecular mechanisms controlling the size of plant seeds remain unclear. We performed functional analysis of BrPATL4 encoding Sec14-like protein to determine the genetic architecture of seed size, shape and their association analyses. We used 60 $T_3$ transgenic rice lines to evaluate seed length, seed width and seed height as seed size traits, and the ratios of these values as seed shape traits. Pleiotropic effects on general architecture included small seed size, erect panicles, decreased grain weight, reduced plant height and increased sterility, which are common to other mutants deficient in gibberellic acid (GA) biosynthesis. To test whether BrPATL4 overexpression is deleterious for GA signal transduction, we compared the relative expression of GA related gene and the growth rate of second leaf sheath supplied with exogenous $GA_3$. Overexpression of BrPATL4 did not affect GA biosynthesis or signaling pathway, with the same response shown under GA treatment compared to the wild type. However, the causal genes for the small seed phenotype (D1, SRS1, and SRS5) and the erection of panicles showed significantly decreased levels in mRNA accumulation compared to the wild type. These results suggest that the overexpression of BrPATL4 can control seed size through the suppression of those genes related to seed size regulation. Although the molecular function of BrPATL4 is not clear for small seed and erect panicles of BrPALT4 overexpression line, this study provides some clues about the genetic engineering of rice seed architecture.

Association of Polymorphisms in Fecundity Genes of GDF9, BMP15 and BMP15-1B with Litter Size in Iranian Baluchi Sheep

  • The incidence of mutation in three loci of GDF9, BMP15 and BMP15-1B and their effects on litter sizes was evaluated in Baluchi sheep. Wild-type alleles were detected for BMP15 and BMP15-1B loci and all individuals were found to be as non-carriers for FecB and $FecX^G$ mutations but, a G to A nucleotide substitution was found in GDF9 locus. The frequency of $FecG^+$ (0.82) wild type allele was higher than the frequency of $FecG^l$ (0.18) mutant allele and the frequencies of $FecG^+/FecG^+$, $FecG^+/FecG^1$ and $FecG^1/FecG^1$ genotypes were 0.72, 0.20 and 0.08, respectively in GDF9 locus. The heterozygous ($FecG^+/FecG^1$) and homozygous ($FecG^+/FecG^+$) non-carrier ewes had 0.35 and 0.21 more lambs than the homozygous ($FecG^1/FecG^1$) carrier ewes, respectively (p<0.05). In addition to the finding of segregation of non-additive gene effect on litter size in the previous study in Baluchi sheep, these findings for the first time shows that the $FecG^1$ gene has a major effect on litter size in this breed.

Comparison Between Phylogenetic Relationships Based on 18S rDNA Sequences and Growth by Salinity of Chlorella-like Species (Chlorophyta)

  • This study was carried out to understand the correlation between phylogenetic relationships based on 18S rDNA sequences and growth by salinity of Chlorella-like species. The 18S rDNA sequences of 71 Chlorella-like species which were mainly collected from Korean waters were analyzed. The 18S rDNA sequences of Chlorella-like species were divided into three groups (group A, B and C) and group B was further divided into three subgroups (subgroup B-1, B-2 and B-3). Thirty-seven Chlorella-like species in group A grew well at high salinity (32 psu) but the other groups grew well in freshwater. The sequence identities of the species in group A and B were 97.2-99.5%, but those of 6 species in group C ("Chlorella" saccharophila), which contained group I intron sequences region were 75.0-75.4%. Two representative species of each group were cultured at different salinities (0, 16 and 32 psu) to examine the correlation between the molecular phylogenetic groups and the phenotypic characteristics on cell growth and size by different salinities. The size of cell cultured at different salinities varied according to the species of each molecular phylogenetic group. The size of "Chlorella" saccharophila in group C was bigger and more obviously elliptical rather than that of the other Chlorella-like species. Considering the results on molecular and phenotypic characteristics, the group A and B belonged to Chlorellaceae, but group C was distinctly different from them.
