Improvement of Virus Safety of an Antihemophilc Factor IX by Virus Filtration Process

  • Viral safety is an important prerequisite for clinical preparations of plasma-derived pharmaceuticals. One potential way to increase the safety of therapeutic biological products is the use of a virus-retentive filter. In order to increase the viral safety of human antihemophilic factor IX, particularly in regard to non-enveloped viruses, a virus removal process using a polyvinylidene fluoride membrane filter (Viresolve NFP) has been optimized. The most critical factor affecting the filtration efficiency was operating pH and the optimum pH was 6 or 7. Flow rate increased with increasing operating pressure and temperature. Recovery yield in the optimized production-scale process was 96%. No substantial changes were observed in the physical and biochemical characteristics of the filtered factor IX in comparison with those before filtration. A 47-mm disk membrane filter was used to simulate the process performance of the production-scale cartridges and to test if it could remove several experimental model viruses for human pathogenic viruses, including human hepatitis A virus (HAV), porcine parvovirus (PPV), murine encephalomyocarditis virus (EMCV), human immunodeficiency virus type 1 (HIV), bovine viral diarrhea virus (BVDV), and bovine herpes virus (BHV). Non-enveloped viruses (HAV, PPV, and EMCV) as well as enveloped viruses (HIV, BVDV, and BHV) were completely removed during filtration. The log reduction factors achieved were $\geq$6.12 for HAV, $\geq$4.28 for PPV, $\geq$5.33 for EMCV, $\geq$5.51 for HIV, $\geq$5.17 for BVDV, and $\geq$5.75 for BHV. These results indicate that the virus filtration process successfully improved the viral safety of factor IX.

A Study of Air Stripping and Ozonation Characteristics of Methyl Tert-butyl Ether (MTBE) (Methyl Tert-butyl Ether (MTBE)의 탈기와 오존산화 특성에 관한 연구)

  • In recent years, there has been considerable concern over the release of methyl tert-butyl ether (MTBE), a gasoline additive, into the aquifers used as potable water sources. MTBE readily dissolves in water and has entered the environment via gasoline spills and leaking storage tanks. In this study air stripping and ozonation of MTBE in aqueous solution were performed in a laboratory scale batch reacter. The mass transfer rate (N) was evaluated and a values about $1.24{\times}10^{-6}\;mol{\cdot}sec^{-1}$ was found. In the ozonation of MTBE a 8.3% decrease of the COD and a 6.5% decrease of the TOC lead to BOD/COD = 0.03. The peudo first-order rate constants of the ozonation of MTBE was $3.75{\times}10^{-5}\;sec^{-1}$. The resulting Ea of 4.80 kcal;mol-1 was observed for molecular ozone reactions.

  • Three classes of proteinase inhibitors are known in soybean; the Kunitz trypsin inhibitor (SKTI), the Bowman-Birk proteinase inhibitor and its isoinhibitors. To study the molecular structure and expression characteristics of the SKTI, antibody was obtained by immunizing rabbit with the SKTI purified from soybean by preparative electrophoresis. Anti-SKTI antibody was not only specific for mature SKTI in soybean seed but also recognized the precursor which was synthesized in vitro. Translation in vitro was carried out in wheat germ extract with polyadenylated mRNA isolated from developing soybean seeds. One of the seed specific translation products, MW 24K, was identified to be the precursor for the SKTI by immunoprecipitation with anti-SKTI antibody. Mature SKTI of MW 20K, however, was not detected in the translates in vitro. These results suggest that the precursor polypeptide is synthesized from the mRNA and is cleaved to yield mature SKTI in soybean seed. The SKTI gene was expressed with the maturation of soybean seed in a tissue-specific and development stage-specific manner.

Genetic Identification on Natural Population of Triploid Crucian Carp, Carassius auraus in Korea (자연산 3배체 붕어 (Carassius auratus) 클론 집단에 대한 유전학적 동정)

  • Natural clonal stock of triploid crucian carp, Carassius aurahs was identified and its cytogenetic, molecular genetic and morphological traits were studied. Cytogenetic analysis of the clonal crucian carp revealed that they were natural triploidy, evidenced by 1.5-fold increases of cell size, DNA content, and chromosome number. Multi-locus DNA fingerprinting using $(GATA)_4$ probe showed that they had an identical fingerprint profile, indicating the clonal propagation of the population. External morphology and morphometric characteristics of triploid individuals were much uniform compared to those of diploids. Natural triploid crucian carp was proven to be all-female in this study.

Production properties of pyrolytic matter of PP and PS plastics in n low temperature pyrolysis condition (저온 열분해조건에서 PP 및 PS계 플라스틱의 열분해물 생성특성 연구)

  • Thermal degradation characteristics of polypropylene and polystyrene have been studied in the thermogravimetric(TG) reactor and batch-type microreactor. The dynamic thermogravimetric curve of TG provided a valuable information about pyrolysis temperature. It was found that PS was thermally degraded at lower temperature of $30{\sim}50^{\circ}C$ than PP. It was found that the yield and molecular weight of liquid product in the microreactor were decreased with the increase of reaction temperature and time in the case of PP. The production of styrene monomer was significantly increased by the promotion of depolymerization with the increase of temperature and time. The chain-end scission rate parameters were determined to be 50.0 kcal/mole of PP, 45.2 kcal/mole of PS by the Arrhenius plot.

  • We attempted to fabricate a dendrimer Langmuir-Blodgett(LB) films containing 48 pyridinepropanol functional end group. As the pyridinepropanol functional group could form a complex structure with metal ions. In this study the samples for electrical measurement were fabricated to two types metal complexes with $Pt^{4+}$ and $Fe^{2+}$ ions by LB method. And we have investigated the surface activity at the air-water interface as well as the electrical properties for the monolayers of pure G4-48PyP dendrimer and its complex with metal ions($Pt^{4+}$ and $Fe^{2+}$ ions). In the surface pressure-area($\pi-A$) isotherms of the dendrimers, the stable condensed films formed at the air-water interface and the metal ions effect showed the difference on molecular behavior. We have studied the electrical properties of the ultra thin dendrimer LB films investigated by the current-voltage(I-V) characteristics of metal/dendrimer LB films/metal(MIM) structure. In conclusion, it is demonstrated that the metal ion around G4-48PyP dendrimer can contribute to make formation of network structure among dendrimers and it result from the change of electrical properties.

  • The determination of DNA hybridization can apply the molecular biology research. clinic diagnostics. bioengineering, environment monitoring, food science and other application area. So, The determination of hybridization is very important for the improvement of DNA detection system. In this study, we report the characterization of the DNA hybridization by the electricalchemical methods. The probe oligonucleotide was used to determine the amount of target oligonucleotide in solution using Methylen Blue(MB) as the electrochemical indicators. The cathodic peak currents($I_{peak}$) of MB were linearly related to the concentration of the target oligonucleotide sequence in the range $1[{\mu}M]{\sim}0.1[{\mu}M]$. The detection limit of this approach was 0.01[nM]. As a result, the match oligonucleotide(CR-1) was most stable state and the peak of redox current measured by DNA hybridization detection sensors by using electrochemical method seem to be similar to 1-mer terminal mismatch oligonucleotide(MR-3). The MR-2, MR-3, MR-22 and MR-33 have each mismatching sequence of central and terminal. With this set the role of point mutations was to be investigated. Terminal mismatch oligonucleotide (MR-3, 33) is shown more stable state than central mismatch oligonucleotide(MR-2, 22). And 1-mer mismatch oligonucleotide(MR-2 or 3) is shown more stable state than 2-mer mismatch oligonucleotide(MR-22 or 33).

  • The calculated and measured dielectric constants of (1-x)(A $l_{1}$2/ T $a_{1}$2/) $O_2$-x(M $g_{1}$3/ T $a_{2}$3/) $O_2$ (0$\leq$x$\leq$1.0) solid solutions were investigated by variations of ionic polarizability and crystal structure. (A $l_{1}$2/ T $a_{1}$2/) $O_2$ and (M $g_{1}$3/ T $a_{2}$3/) $O_2$ were orthorhombic and tetragonal trirutile structure, respectively. When (A $l_{1}$2/ T $a_{1}$2/) $O_2$ was substituted by (M $g_{1}$3/ T $a_{2}$3/) $O_2$, the phase transformed to tetragonal structure over 60 mole. Because the total ionic radius of [(Mg+2Ta)/3]$^{4+}$ was slightly bigger than one of [(Al+Ta)/2]$^{4+}$, the lattice parameters increased with an increase of (M $g_{1}$3/ T $a_{2}$3/) $O_2$ substitution. The measured dielectric constant increased with an increase of (M $g_{1}$3/ T $a_{2}$3/) $O_2$ substitution and coincided with dielectric mixing rule and the calculated dielectric constant with the molecular additivity rule. There were some differences between the measured and the calculated dielectric constant. The reason of the lowered dielectric constant comparing with the calculated one was compressed stress due to the electronic structure of tantalum.

Estimation of Rutting Property of Artificially-Aged Asphalt Mixtures Based on LMS(%) and $S_D$ (LMS(%)와 $S_D$에 의한 노화 아스팔트 혼합물의 소성번형 추정)

  • This study was carried out for evaluating correlation of aging level and deformation resistance of asphalt mixtures conditioned by artificial aging treatment. The specimens were made of the short-term aged loose asphalt mixture which was kept in a convection oven at $154^{\circ}C$ for 2 hours, and then were artificially aged at $110^{\circ}C$ for 24, 48, 72 and 96 hours for long-term aging. Deformation strength ($S_D$) and large molecular size ratio (LMS(%)) were measured after short-term and each long-term aging treatment to evaluate aging extents of each mixture with aging time using GPC(Gel-permeation chromatograph) test. It is shown that the $S_D$ and LMS(%) were increased with aging time increment, and that LMS(%) quantity and $S_D$ have relatively good correlation each other. This study found that the aged asphalt mixture had better resistance against rutting and it was possible to estimate rutting characteristics of asphalt mixtures by using LMS(%) without a binder recovery.

Characterization of Monilinia fructicola Associated with Brown Rot of Cherry Fruit in Korea (체리 잿빛무늬병에 관여하는 Monilinia fructicola의 특성)

  • In August 2013, brown rot was observed on cherry fruits (Prunus avium L.) in Hwaseong city, Korea. Fruit rot first appears as small, circular brown spots that increase rapidly in size causing the entire fruit to rot. Grayish spores appear in tufts on rotted areas. Based on these morphological characteristics, the two isolates were identified as Monilinia fructicola. Molecular analysis of 5.8S subunit and flanking internal transcribed spacers (ITS1 and ITS2) was performed to confirm the identification. The ITS sequences had 100% identity with those of other reference M. fructicola isolates of NCBI GenBank. Pathogenicity was tested using spore suspension inoculation on wounded or unwounded cherry fruits. The lesions were observed on wounded and unwounded fruits after inoculation for 7 days. This is the first report on M. fructicola causing brown rot on cherry fruits in Korea.
