담도암에서 $^{18}F-FDG$ PET의 임상 이용 (Clinical Application of $^{18}F-FDG$ PET in Bile Duct Cancer)

  • Reports about FDG PET in biliary tumor are limited and there are almost no reports regarding its efficacy. Biliary tumor is divided to intrahepatic and extrahepatic bile duct cancer, and intrahepatic bile duct cancer can be further divided to peripheral type which occurs at lobular duct and hilar type which occurs at hepatic hilum. Surgical resection is the only curative method for bile duct tumor, and accurate staging plays an important role in deciding treatment modality. Among intrahepatic bile duct tumors, peripheral type and hilar type have the same histological characteristics, but different clinical manifestations and tumor growth pattern. On PET image, FDG uptake is also different between peripheral type and hilar type. Most of the former shows high FDG uptake at primary and metastasis site so it is very useful for determining stage and changing treatment plans. However, the later is diversified among low uptake and very high uptake. The FDG uptake pattern of hilar type is similar to that of extrahepatic bile duct cancer, and mucinous component is an important factor, which affects FOG uptake. When tumor cells are scattered in desmoplatsic stroma, then FDG uptake is low as well. In contrast, when FDG uptake is high, it is likely to be tubular type which has high tumor density. Tumor growth pattern also affects FDG uptake. Nodular type mostly takes higher FDG compared to infiltrative type. There are many cases where benign inflammatory diseases take high FDG that PET alone can not distinguish malignant lesion from benign lesion. In conclusion, studies about PET using FDG are still limited. Thus, it is hard to make accurate conclusion about the roles of PET or PET/CT in biliary cancers, but peripheral type intrahepatic bile duct cancers and mass forming hilar and extrahepatic bile duct cancers appear to be good indications performing FDG PET or PET/CT.

Aeromonas sp. MN44의 특성과 망간 산화에 관한 연구 (A Study on the Manganese Oxidation and Characteristics of Aeromonas sp)

  • 충청남도 목천과 충청북도 오창 근교의 토양으로부터 망간을 산화하는 64 집락을 분리하고 이 중 망간 산화능이 가장 우수한 한 균주를 최종 선별하여 생리, 생화학적 특성을 조사하고, 16S rRNA 염기 서열분석 등을 통하여 동정한 결과 최종 선별된 균주는 Aeromonas sp. MN44로 확인되었다. 최종 선별된 Aeromenas sp. MN44는 lactose를 제외한 여러 당들은 이용하지 못하였으며, 중금속내성은 lithium과 manganese에 대해서는 mg/ml 단위의 높은 농도까지 중금속 내성을 가지고 있었지만 cadmium에는 전혀 내성을 나타내지 않았다. 또 kanamycin, chloramphenicol, ampicillin, tetracycline, spectinomycin등 조사한 모든 항생제에 대해 전혀 내성을 갖지 않았다. Aeromonas sp. MN44가 생성하는 망간산화물질의 최적 pH는 pH 7.4로 확인되었으며, 이 균이 생성하는 망간 산화 factor는 proteinase K와 가열처리에 의해 저해되는 단백질이고, ammonium sulfate 침전과 ion exchange chromatography 그리고 gel filtration의 단계를 통해 부분 정제한 망간 산화 factor의 분자량은 약 113 kDa로 확인되었다.

호랑나비 유충 혈림프 Transferrin의 특성과 분포 (Characterization and Distribution of Transferrin from the Last Larval Haemolymph of Papilio xuthus)

  • Transferrin은 저장된 철분자를 운반하고 살아있는 유기체에서 철의 항상성을 유지한다. 호랑나비의 종령 유충 혈림프내 철 운반 단백질인 transferrin을 KBr 밀도구배 초원심분리와 Superose 6 HR을 이용한 fast protein liquid chromatography법으로 분리 정제하였으며, transferrin의 조직에 따른 분포는 면역화학적 방법에 따라 확인하였다. 정제된 transferrin의 아미노산 조성은 아스파르트산(Asp), 발린(Val), 루이신(Leu), 글루타민(GIu)이 많이 존재하였으며, subunit의 분자량은 78, 80 kDa으로 확인되었다. Immuno-diffusion을 통해 각 조직에서 분포를 확인한 결과 혈림프와 지방체에서 transferrin이 전시기에 걸쳐 뚜렷한 동질성을 나타냈다. 또한 rocket immuno-electrophresis법에 의해 transferrin의 양적 분포를 살펴보면 혈림프에서 용화 3일과 용화 5일에 증가하였으며, 지방체에서는 전용기와 용화직후에 양적인 증가를 나타냈다.

  • 용매 존재하에서 2-hydroxyethylmethacrylate와 acrylamide, N, N-dimethylamide 및 methylmethacrylate를 함량에 따라 3종류의 공중합체 수화겔막을 제조한 다음 이들 막에 알코올용질의 투과 및 분배계수를 측정하였으며, 이 측정값들을 이용하여 용질의 투과 메카니즘을 논의하였다. 수화겔막에서 유기용질의 투과계수는 그 분자의 크기에 주로 의존하였다. 그러나 수화겔막 내에 존재하는 물의 함량이 낮아지면 막에 대한 용질의 용해도가 점차 중요해짐을 또한 알 수 있었다. 분배계수의 측정 결과에 의하면 수화겔막에서의 용질의 분배는 주로 막과 용질사이에 존재하는 소수성 상호작용에 의해 조절되었다. 그리고 수화겔막에 유기용질의 확산은 전적으로 빈자리에 의존하며, 물분자와 용질 사이에 극성-편극성 및 수소결합도 부분적으로 영향을 미치나 그 정도가 아주 미약함을 확인하였다.

Vapor Permeation Characteristics of TiO2 Composite Membranes Prepared on Porous Stainless Steel Support by Sol-Gel Method

  • Composite membranes with a titania layer were prepared by soaking-rolling method with the titania sol of nanoparticles formed in the sol-gel process and investigated regarding the vapor permeation of various organic mixtures. The support modification was conducted by pressing $SiO_2$ xerogel of 500 nm in particle size under 10 MPa on the surface of a porous stainless steel (SUS) substrate and designed the multi-layered structure by coating the intermediate layer of ${\gamma}-Al_2O_3$. Microstructure of titania membrane was affected by heat-treatment and synthesis conditions of precursor sol, and titania formed at calcination temperature of 300$^{\circ}C$ with sol of [$H^+$]/[TIP]=0.3 possessed surface area of 210 $m^2$/g, average pore size of 1.25 nm. The titania composite membrane showed high $H_2/N_2$ selectivity and water/ethanol selectivity as 25-30 and 50-100, respectively. As a result of vapor permeation for water-alcohol and alcohol-alcohol mixture, titania composite membrane showed water-permselective and molecular-sieve permeation behavior. However, water/methanol selectivity of the membrane was very low because of chemical affinity of permeants for the membrane by similar physicochemical properties of water and methanol.

Characteristics of IEF Patterns and SDS-PAGE Results of Korean EPO Biosimilars

  • Erythropoietin (EPO) is mainly produced in kidney and stimulates erythropoiesis. The use of recombinant EPOs for doping is prohibited because of its performance enhancing effect. This study investigated whether biosimilar EPOs could be differentiated from endogenous one by iso-electro-focusing plus double blotting and SDS-PAGE for antidoping analysis. The established method was validated with positive control urine. The band patterns were reproducible and meet the criteria, which was made by world anti doping agency (WADA). Isoelectric focusing was conducted in pH range 2 to 6. Recormon (La Roche), Aropotin (Kunwha), Epokine (CJ Pharm Co.), Eporon (Dong-A), Espogen (LG Life Sciences), and Dynepo (Shire Pharmaceuticals) were detected in basic region. All biosimilars showed discriminative isoelectric profiles from endogenous EPO profiles, but they showed different band patterns with the reference one except Epokine (CJ Pharm Co.). Next, SDS-PAGE of biosimilar EPOs resulted in different molecular weight patterns which were distributed higher than endogenous EPO. Commercial immune assay kit as an immune affinity purification tool and immobilized antibody coated magnetic bead were tested for the purification and concentration of EPO from urinary matrix. The antibody-coated magnetic bead gave better purification yield. The IEF plus double blotting and SDS-PAGE with immunoaffinity purification method established can be used to discriminate biosimilar EPOs from endogenous EPO.

Influence of light Regime on Nitrate Reductase Activity and Organic and norganic Solute Composition of Four Sedges (Carex spp.)

  • A survey was conducted on the inorganic and organic solute patterns of plants in connection with nitrate metabolism according to different light regimes (1.9, 16.0, 91.5 $Wm^{-2}$). Besides measuring in vivo NRA, we also quantitatively analyzed ater-soluble inorganic ions, organic acids, low molecular weight carbohydrates, amino aciss and total N (% DW). Among 4 Carex species, C. pilosa is known as shade-adapted species and the others as half (C. gracilis) to full (C. rostrata & C. distans) light-adapted species. Compared to species adapted to high light intensity, shade-adapted C. pilosa showed reduced productivity under the highest light intensity. In general, nitrate and amino acid levels decreased at higher light intensity, while sugar and organic acid concentrations increased. In C. pilosa osmolality tended to rise with increasing light intensity, while in the other species it tended to fall. Under low light intensity, the drop in soluble carbohydrate contents is osmotically compensated for by an enhanced nitrate concentration. It is concluded that competition between nitrate and $CO_2$reduction for reductants and ATP from photosynthesis may have important ecological consequences for the adaptation of plants to low or high light conditions. Additionally, the patterns of ionic changes due to increased light intensities were essentially the same in all selected species, indicating similar characteristics of heir mineral ion and organic acid metabolism as well as in field-grown Carex species.

Forskolin Effect on the Lineage Specification of Trunk Neural Crest Cells in vitro

  • Recent evidence has suggested that trunk neural crest cell generally assumed to have equivalent differentiation potentials, demonstrate differentiation bias along the anterior/posterior axis. In amphibian and fish, neural crest cells give rise to three chromatophore types, melanophores, xantho-phores, and iridophores. Each pigment cell type has distinct characteristics but there is speculation about the cellular plasticity that exists among them. Neural crest cells migrate along specific routes, ventromedially and dorsolaterally. Neural crest cells that travel dorsolaterally are the first cells to begin migration in the axolotl and are the major contributors to the visible pigment pattern. Many factors and mechanisms that are responsible for guiding migratory neural crest cells along potential pathways or determining their fate remain unknown. A single lineage of the crest, which becomes restricted to one of the three pigment cell types, gives us the opportunity to examine the existence of neural crest stem cell populations and cellular plasticity. Study presented here showed results from recent in vitro studies designed to identify parameters influencing differentiation events of individual neural crest-derived pigment cell lineages. Melanophore production from neural crest explants originating from different levels along the anterior/posterior axis of wild type-axolotl embryos were compared and demonstrate that the differentiation of melanophores is enhanced in subpopulation of neural crest treated with forskolin. Forskolin (an adenylate cyclase activator) increases intracellular CAMP concentration and eventually activates the protein kinase-A signaling pathway. Melanophore number, melanin content, and tyrosinase activity in explants taken from the anterior-most region of the crest increased significantly in response to forskolin treatment. This study suggests implications of region specific influences and developmental regulation in the development of pigment pattern.

Study and Application of the New Stick Make Up Product Using Clay Minerals as Binder & Buffer.

  • The new stick make-up product was studied by using a gel, which is a viscous complex formed with clay minerals, vitamins A and E and fluorinated liquid polymer with a 1500 molecular weight. The gel cannot be obtained with any random combination of clay minerals and the ingredients described above. It takes the sequential manufacturing method as follows to get this kind of gel. Firstly, clay minerals and liquid polymers have to be pre-mixed in order to saturate the liquid polymers with the clay minerals. Then the on-processed gel has to be finely crystallized. The clay minerals, which are the core elements for this gel, were used as a function of Binder & Buffer and liquid polymer was mixed together for the deterioration of the surface tension of each component and to form a functional film in the gel. This liquid polymer was combined with clay minerals because it is not miscible with most oils and solvents. Waxes have a function of keeping a solid status in the stick. We reduced the usage of waxes by putting clay minerals as buffer in the proportion of 0.5:1 with oil phase. Ceramide takes care of the skin when used regularly and maintains the skin's moisture. Vitamins A and E contribute to preventing skin aging by the activation of skin cells. We could get the stable viscous gel, which has about 80% oil phase using clay minerals and liquid polymer. The crystalline structures of gel were surface-chemically-analyzed using SEM and Image Analyzer and were thermodynamically analyzed using DSC. Surface tension test and softness were done by Rheometer. In the end, these characteristics were verified by consumer panel tests in Seoul, Daegeon and Pusan in Korea and Hokkaido, Osaka and Miyazaki in Japan with correlation to the climate.

Differences and Variations among Anguilla japonica, Muraenesox cinereus and Conger myriaster from the Yellow Sea

  • Genomic DNAs were extracted from the muscle of twenty-one specimens of three eel species collected in Anguilla japonica (AJ), Muraenesox cinereus (MC) and Conger myriaster (CM) from the Yellow Sea, respectively. In the present study, 7 oligonucleotides primers generated 191 specific loci in the AJ species, 226 in the (MC) species and 181 in the CM species, respectively. The primer BION-02 generated the most loci (a total of 83), with an average of 11.86 in the AJ species. The specific loci generated by oligonucleotides primers exhibited inter-individual-specific characteristics, thus revealing DNA polymorphisms. With regard to average bandsharing value (BS) results, individuals from Conger myriaster species (0.808) exhibited higher bandsharing values than did individuals from Muraenesox cinereus species (0.729) (P<0.05). The longest genetic distance (0.430) displaying significant molecular difference was also between individual no. 01 within Anguilla japonica eel species and individual no. 04 within Anguilla japonica species. In this study, the dendrogram resulted from reliable seven oligonucleotides primers, indicating three genetic clusters composed of group I (ANGUILLA 01~ANGUILLA 07), group II (MURAENESOX 08~MURAENESOX 14) and group III (CONGER 15~CONGER 21). The existence of species differentiation and DNA polymorphisms among three eel species were detected by PCR analysis. As mentioned above, a dendrogram revealed close relationships between individual identities within three eel species. High levels of a significant genetic distance among three eel species showed this PCR approach is one of the most suitable tools for individuals and/or species biological DNA studies.
