Oligomeric Characterization of GroESLx Chaperonin from Symbiotic X-Bacteric in Amoeba proteus

  • GroESLx proteins of symbiotic X-bacteria were overproduced in Escherichia coli and their structural characteristics were assayed after simple purification. The GroESx and GroELx were heat-stable at 8$0^{\circ}C$ and 5$0^{\circ}C$, respectively. After heat-treatment, GroESx was purified by DEAE Sephadex A-50 chromatography and GroELx was purified by step- and linear sucrose density gradient ultracentrifugation. Molecular masses of GroESx and GroELx were 50-80 kDa and 800 kDa, respectively, as estimated by sucrose density gradient ultracentrifugation. In chemical cross-linking analysis, subunits of GroESx were mostly cross-linked by incubation for 3 h in 0.4% glutaralde-hyde and GroESx was found to be composed of homo-heptamer subunits. Those of GroELx were cross-linked within 10 min in 0.3% glutaraldehyde and GroELx was in two stacks of homo-heptamer subunits. On the other hand, GroESx and GroELx proteins in a solution could not be cross-linked even after incubation for 3 h in 0.5% glutaraldehyde. GroELx was stable at 4-37$^{\circ}C$. In the presence of both GroESx and ATP, GroELx$_{14}$ was stable at 37$^{\circ}C$ but not at 4$^{\circ}C$ or 24$^{\circ}C$. Thus, we confirmed the oligomeric properties of GroESx$_{7}$ and GroELx$_{14}$ and their stability to heat and in the interaction with GroESx.x.

Isolation of Novel Alkalophilic Bacillus alcalophilus subsp. YB380 and the Characteristics of Its Yeast Cell Wall Hydrolase

  • An alkalophilic mi.croorganism (strain YB380), which produces yeast cell wall hydrolase extracellulary, was isolated from Korean soil. The rod-shaped cells were 0.3~0.4 by 2~4${\mu}{\textrm}{m}$ long, motile, aerobic, gram-positive, and spore-forming. The color of the colony was light yellow. The temperature range for growth at pH 9.0 was 25 to $45{\circ}C, with optimum growth at $35{\circ}C. The pH range for growth at $35{\circ}C was 8 to 11 with an optimum pH of 9.0. Therefore, the strain YB380 is an obligate alkalophile. The 16S rRNA of strain YB380 has a 99% sequence similarity with that of Bacillus alcalophilus. On the basis of physiological properties, cell wall fatty acid composition, and phylogenetic analysis, we propose that the isolated strain is Bacillus alcalophilus. The yeast cell wall hydrolase from Bacillus alcalophilus subsp. YB380 has been purified and partially characterized. The molecular weight was estimated to be 27,000 daltons with an optimum temperature and pH of $60{\circ}C and 9.0, respectively. The N-terminal amino acid sequence of the enzyme was analyzed as Gln- Thr- Val- Pro- Trp- Gly- Ile- Asn- Arg- Val.

A Simulation for Kaolin Contaminants Accumulation and Varying Characteristics of Leakage Currents (Kaolin 오손물 누적량 모의실험 및 누설전류변화 특성)

  • This study performs a simulation for an accumulation mechanism of contaminants, which were produced in an industrial belt of inland, on the surface of insulators. From the simulation, silicon insulators presented higher accumulation than that of EPDM(Ethylene Propylene Diene Terpolymer : EPDM) insulators on the same distance in the case of the Virgin polymer insulator, and this result presented the same result in the insulator applied in actual fields. In the case of the accumulation test for the Virgin insulator and insulators used in actual fields, it is evident that the Virgin insulator presented more accumulation than that of the insulator used in actual fields. The results can be caused by the generation of LMW (Low Molecular Weight) on the external material of polymer insulators, and the level of the accumulation can be changed according to the degree of the continuous generation of LMW. In order to simulate a certain pollution of an industrial belt, which is located along the coastline, leakage currents were measured by applying the contaminant compulsively that was produced with salts and Kaolin according to the ratio of its weight on the surface of insulators. The more increase in the content of Kaolin pollution, the level of leakage currents on the surface of polymer insulator more increased. In addition, the approaching time to the maximum value of leakage currents presented a nearly constant level regardless of the content of Kaolin. The level of leakage currents significantly decreased according to the passage of time, and the level of leakage currents on the surface maintained a constant level at a specific time regardless of the content of Kaolin.

Separation of $H_2$/$N_2$ Gas Mixture by PTMSP/PDMS-PEI Composite Membrane (PTMSP/PDMS-PEI 복합막에 의한 수소/질소 혼합기체 분리)

  • PTMSP/PDMS-PEI composite membrane was prepared by solution casting method. To investigate the characteristics of this membrane, the analytical methods such as FT-IR, $^1$H-NMR, DSC, TGA, GPC, and SEM have been utilized. The number-average((equation omitted)) and weight-average((equation omitted)) molecular weight of PTMSP/PDMS copolymer were 501,516 and 675,560 respectively. The separation of the gas mixture($H_2$/$N_2$) through the composite membrane was studied as a function of pressure. The separation factor($\alpha$, $\beta$, (equation omitted)) of the composite membrane used in this work increased as the pressure of permeation cell increased. The real separation factor($\alpha$), head separation factor($\beta$), and tail separation factor ((equation omitted)) of PTMSP/PDMS-PEI composite membrane were 21.50, 49.14 and 1.84 respectively at $\Delta$P 345.55 kPa and $25^{\circ}C$.

BRCA1 Protein Was Not Expressed in a Normal Human Breast Epithelial Cell Type With Stem Cell and Luminal Characteristics

  • BRCA1 is a tumor suppresser gene in familial cases of breast cancer. It has been controversial whether the subcellular localization of BRCA1 is located in nuclei or cytoplasm in normal human breast cells. We found that a p220 protein was expressed in Type II Normal human breast epithelial cells (NHBEC) but not in Type I NHBEC in Western blot analysis using the 17F8 (3A2) antibody. Immunostaining using the same antibody revealed positive staining in nuclei, cytoplasm and perinuclei of Type II cells and negative staining in Type I NHBEC. The p220 protein, however, was expressed in SV40 immortalized Type I NHBEC and tumorigenic cells derived from them after x-ray and neu oncogene treatment. The subcelluar localization was mostly cytoplasmic and punctate in the nuclei. The breast carcinoma cell lines, MCF-7 and T47D, also expressed the p220 protein. Using RT-PCR, we observed the expression of BRCA1 mRNA in both Type I and Type II NHBEC. This result indicated that there might be mechanisms involved in post-translational or translational regulation of BRCA1 gene. It is speculated that the absence of BRCA1 protein expression in Type I NHBEC might playa role in their susceptibility to neoplastic transformation.

A New species of Elsholtzia (Lamiaceae): E. byeonsanensis M. Kim (향유속(꿀풀과)의 신종: 변산향유(Elsholtzia byeonsanensis M. Kim))

  • A new species, Elsholtzia byeonsanensis M. Kim is named and described from Byeonsan, Jeollabukdo Province, Korea. Molecular data confirmed that this new taxon was distinguished from other congeneric species. Elsholtzia byeonsanensis shares several characteristics (secund spikes, ovate leaves, long corolla length, etc.) with its related species E. splendens, but it is distinct from E. splendens which has a large plant, green stem color, chartaceous leaf textures, non-shiny leaf surfaces, pubescent petioles, pubescent leaf blade surfaces, pubescent bract surface, and open woodland habitats by having a small plant, purple stem color, coriaceous leaf textures, shiny leaf surfaces, glabrous petioles, glabrous leaf blade surfaces, glabrous bract surface, and marine habitats.

A New species of Viola (Violaceae): V. ulleungdoensis M. Kim & J. Lee (제비꽃속(제비꽃과)의 신종: 울릉제비꽃(Viola ulleungdoensis M. Kim & J. Lee))

  • A new species, Viola ulleungdoensis M. Kim & J. Lee is named and described from Is. Ulleung, Gyeongsangbukdo Province, Korea. Molecular data confirmed that this new taxon was distinguished from other congeneric species. The Viola ulleungdoensis shares several characteristics (acaulescent leaves, beardless lateral petals, glabrous petioles and peduncles, etc.) with its related species V. selkirkii, but is distinct from V. selkirkii which has present adventitious buds, same leaves after flowering, small leaves, and high altitude habitats by having absent adventitious buds, larger leaves after flowering, large leaves, and low altitude habitats.

Biomass and Molecular Characteristics of Multi-tillering Miscanthus Mutants

  • Compared to wide ranges of genetic variation of natural populations, very limited Miscanthus cultivar has been released. This study was the first report on the development of Miscanthus cultivar by means of radiation breeding. Seeds of M. sinensis were initially exposed to gamma rays of 250 Gy for 24 h, generated from a $^{60}Co$ gamma-irradiator. The irradiated seeds were sown and then the highly tiller-producing mutants were selected for this study. Biomass-related parameters including tiller number, plant height, stem diameter, and leaf number were measured. Ploidy level and internal transcribed spacer (ITS) were investigated to characterize the mutants compared to wild type (WT) Miscanthus. Plant height and tiller number were negatively related, where multi-tillering mutants were relatively short after 4 month growth. However stem diameter and leaf number were greater in mutants. All the materials used in this study were diploid, implying that the mutants with greater tiller numbers and stem diameter were not likely related to polyploidization. Based on the sequence of ITS regions, the mutants demonstrated base changes from the gamma irradiation where G+C content (%) was decreased in the ITS1, but increased in ITS2 when compared to WT sequence. ITS2 region was more variable than in ITS1 in the mutants, which collectively allows identification of the mutants from WT. Those mutants having enhanced tillers and allelic variations might be used as breeding materials for enhanced biomass-producing Miscanthus cultivars.

  • The term of Mushrooms means to spread like the April showers. After 18th century, the mycelium and spores of mushrooms were observed by microscope and then was denominated as fungi. About one hundred thousand species of mushrooms in appearance were worldly reported, and in Korea about four thousand species of mushrooms are estimated. In Korea, total of one thousand nine hundred one species of mushrooms have been recorded. Mushrooms belonging to the group of organisms called fungi, which must obtain their food from living plants or animals or from their remains after death. A large number of mushrooms grow in association with the roots of trees and other woody plants, called mycorrhizal fungi, both mushrooms and plants require this relationship for growth and development. And also many Mushrooms are saprobic, living on decayed various fallen leaves, twigs, trees and vegetable remains and etc. some of these million of spores settles on the proper habitat, these spores germinates and grows into a mass of threads, then a mycelium. This is the vegetable part of the mushrooms, what we call mushrooms are the carpophores, all the characteristics of the morphological features are appropriately used to identify species of mushrooms. Recently, identification and classification of mushrooms are newly confirmed by molecular analysis. In 2013, One thousand nine hundred one species of mushrooms in "List of Mushrooms in Korea" which published by the Korean Society of Mycology were recorded. Total of 238species, 107genera, 40families, 13orders, 6Classes belong to phylum Ascomycota. Total of 1,663species, 403genera 81families, 18orders, 7classes belong to phylum Basidiomycota.

Characteristics of Heat Curable Polyorganosiloxane Coating Materials (페닐기 함유 열경화성 폴리유기실록산 코팅제 특성)

  • Polyorganosiloxane having controlled cross-linking density and phenyl group content were prepared by dimethyldimethoxysilane (DMDMS), methyltrimethoxysilane (MTMS) and phenyltrimethoxysilane (PTMS). The effect of cross-linking density and phenyl group content on the physical properties of siloxane resin and its coated film have been invetigated. Si-NMR results confirmed that synthesized siloxane resins have equivalent D $T^{Me}$ $T^{Ph}$ structures according to applied mole ratios of DMDMS, MTMS and PTMS. Polyorganosiloxane having higher cross-linking density with high phenyl content showed the high molecular weight and increasing phenyl content resulted in higher refractive index as well as better thermal stability. Cross-linking density is more important factor than phenyl content to obtain higher pencil hardness of coated film on the glass. Our results concluded that even polyorganosiloxanes having similar siloxane structures show different physical properties as function of cross-linking density and phenyl content in polyorganosiloxane.
