Development of a Novel Bioreactor System for the Treatment of Gaseous Benzene

  • A novel, continuous bioreactor system combining a bubble column (absorption section) and a two-phase bioreactor (degradation section) has been designed to treat a gas stream containing benzene. The bubble column contained hexadecane as an absorbent for benzene, and was systemically chosen considering physical, biological, environmental, operational and economic factors. This solvent has infinite solubility for benzene and very low volatility. After absorbing benzene in the bubble column, the hexadecane served as the organic phase of the two-phase partitioning bioreactor, transferring benzene into the aqueous phase where it was degraded by Alcaligenes xylosoxidans Y234. The hexadecane was then continuously recirculated back to the absorber section for the removal of additional benzene. All mass transfer and biodegradation characteristics in this system were investigated prior to operation of the integrated unit, and these included: the mass transfer rate of benzene in the absorption column, the mass transfer rate of benzene from the organic phase into the aqueous phase in the two-phase bioreactor, the stripping rate of benzene out of the two-phase bioreactor, etc. All of these parameters were incorporated into model equations, which were used to investigate the effects of operating conditions on the performance of the system. Several experiments were conducted to show the feasibility of this system. This process is believed to be very practical for the treatment of high concentrations of gaseous pollutants.

Expression of Adipocyte-Associated Genes in the Mouse Tail Epididymal Fat at Different Postnatal Ages

  • The fat pad defined as the epididymal fat is located at the head part of the epididymis. However, another fat mass is present near the caudal epididymis, named tail epididymal fat. The present research was focused to determine the expression of adipocyte-associated molecules in the mouse tail epididymal fat at different postnatal ages, including 2, 5, 8, and 12 months of age. The quantitative real-time PCR analysis showed continuous increases of expression levels of delta like non-canonical Notch ligand 1, leptin, and resistin as postnatally aged. The transcript level of peroxisome proliferator-activated receptor gamma was the highest at 5 months of age, remaining at a steady level until 12 months of age. Expression levels of fatty acid binding protein 4, leptin, and adiponectin were significantly increased until 8 months of age, persisting the level at 12 months of age. The transcript level of fatty acid synthase was significantly increased at 8 months of age, without a further change of the level at 12 months of age. These findings exhibited the expression of adipocyte-associated genes which were also detected at the ordinary epididymal fat pad. However, expression patterns of these genes in the tail epididymal fat are different with those in the distal and proximal epididymal fat, suggesting distinct characteristics and/or functions of the tail epididymal fat.

Chemosystematics and Molecular Phylogeny of a New Bioflocculant-Producing Aspergillus Strain Isolated from Korean Soil

  • The ubiquinone and G+C contents of the bioflocculant-producing fungus, a new Aspergillus strain, were detennined using high-perfonnance liquid chromatography. The internal transcribed spacers 1 and 2 (ITS1 and ITS2), and the 5.8S ribosomal DNA (rDNA) of the strain were amplified and sequenced. The strain contained ubiquinone-l0($H_2$)as a major quinone and the G+C content was 49 mol%. A phylogenetic analysis of the ITS regions indicated that the strain belonged to the genus Aspergillus according to its previously classified morphological characteristics. Based on a sequence homology search, the strain was most closely related to Petromyces muricatus (anamorph, A. muricatus; accession number, AJ005674). The sequence of a new Aspergillus strain in ITS1 and ITS2, and 5.8S rDNA showed 97% homology to P. muricatus. Therefore, the strain is believed to be a new bioflocculant-producing Aspergillus strain.

  • H3N2 canine influenza virus emerged in South Korea in 2007 and subsequently spread to China and Thailand, causing epidemic or endemic respiratory diseases in dogs. Through intermammalian species transmission, the virus has also infected cats. However, no direct evidence of significant genetic evolution has been reported since its first emergence. Here, we describe in depth the genetic and molecular characteristics of the ancestral strain (i.e., the first virus isolate from South Korea) of the H3N2 canine influenza virus currently circulating in East Asia.

KINETICS OF AUTOTROPHIC DENITRIFICATION FOR THE BIOFILM FORMED ON SULFUR PARTICLES : Evaluation of Molecular Technique on Monitoring Biomass Growth

  • Characteristics of sulfur-based autotrophic denitrification in a semi-continuous type reactor and the kinetic parameters were studied. Enriched autotrophic denitrifying culture was used for the reactor operation. Biomass growth on sulfur particles and in the liquid medium was monitored using the DAPI staining method. From the result of ion concentration changes and the biomass growth, maximum specific growth rate, ${\mu}_{max}$, and the half velocity constant, $K_M$, were estimated as $0.61\;d^{-1}$ and 3.66 mg/L, respectively. Growth yield coefficient, Y values for electron acceptor and donor were found as 0.49 gVSS/g N and 0.16 gVSS/g S. The biomass showed specific denitrification rate, ranging 0.86-1.13 gN/g VSS-d. A half-order equation was found to best simulate the denitrification process in the packed bed reactor operated in the semi-continuous mode.

Arsenite Oxidation by Bacillus sp. Strain SeaH-As22w Isolated from Coastal Seawater in Yeosu Bay

  • This study was conducted to evaluated seawater bacteria and their seasonal characteristics in the arsenic contaminated coastal seawater of Yeosu Bay, the Republic of Korea. Arsenite-oxidizing bacteria play an important role in the seawater of the arsenic contaminated bay, with a variety of arsenic resistance system (ars) genotypes being present during summer. Specifically, Bacillus sp. strain SeaH-As22w (FJ607342), isolated from the bay, were found to contain the arsB, arrA and aoxR type operons, which are involved in arsenic resistance. The isolated bacteria showed relatively high tolerance to sodium arsenite (III; $NaAsO_2$) at concentrations as high as 50 mM. Additionally, batch seawater experiments showed that Bacillus sp. strain SeaH-As22w completely oxidized 1 mM of As (III) to As (V) within 10 days. Ecologically, the arsenic-oxidizing potential plays an important role in arsenic toxicity and mobility in As-contaminated coastal seawater of Yeosu Bay during all seasons because it facilitates the activity of Bacillus sp. groups.

  • As DNA sequences have been known through the Genome project the techniques for dealing with molecule-level gene information are being made researches briskly. It is also urgent to develop new computer algorithms for making databases and analyzing it efficiently considering the vastness of the information for known sequences. In this respect, this paper studies the association rule search algorithms for finding out the characteristics shown by means of the association between promoter sequences and genes, which is one of the important research areas in molecular biology. This paper treat biological data, while previous search algorithms used transaction data. So, we design a transformed association rule algorithm that covers data types and biological properties. These research results will contribute to reducing the time and the cost for biological experiments by minimizing their candidates.

  • The Thermally Stimulated Current(TSC) spectroscopy has been applied to study the influence of the structual cahange and interface on the electrical properties of epoxy composites. Three DGEBA-MeTHPA matrix model samples mixed different ratios and silica($SiO_2$) filled sample and silaln treating-filled sample has been studied. Above room temperature, the relaxation mode $\alpha$ peak associated with Tg has been located at $110^{\circ}C$. Below glass transition temperature(Tg), three relaxation modes are observed in all samples: a $\beta$ mode situated at $10^{\circ}C$, a $\gamma$ mode located at $-40^{\circ}C$ and a $\delta$ mode appeared in $-120^{\circ}C$. The analysis of its fine structure indicates that constitution of elementary processes is characterized by the activation energy and relaxation time. Also the dielectric relaxation properties have been investigated to compare the the change of the molecular structure and motion to the relaxation properties and conduction mechanism in TSC spectra.

  • The second-order nonlinear optical effect, especially the second harmonic generation, in Langmuir-Blodgett(LB) films have get much attention over the past few years. For second harmonic generation(SHG) of the ultrathin organic films, the multilayer structure of the film should have the noncentrosymmetric arrangements of molecules. The Langmuir-Blodgett technique can result in the production of thin films of precisely controlled dimensions and structure. In this paper, n-octadecyl 4-(4'-nitrophenylazo)-1-naphthyl ether, ONNE(azohenzene derivative), was synthesized and the optical properties of ONNE was studied. Nonccntrosymmctric Z-type LB films of ONNE were prepared and SHG intensity of the film were measured. The structural characteristics of floating monolayer(L film) and LB film of ONNE were discussed with $\pi$-A isotherm. UV-visible absorption spectroscopy and spectroscopic ellipsometry. The polarized UV-visible absorption spectroscopy and SHO intensity suggest the molecular orientation in LB film.

Investigation the frequency change in number of layers and fabrication morphology of Arachidic Acid LB Films (Arachidic Acid LB박막의 누적층수에 따른 주파수 변화와 모폴로지 특성)

  • In this study, in order to confirm the application possibility to the molecular electronic device, morphlogical property of the Arachidic acid was investigated. We have investigated morphology by BAM image and AFM. ${\pi}$-A curves investigated surface pressure of this LB film from liquid to solid state ranged between 40 to 45 mN/m. BAM images investigated the different states of Arachidic Acid LB film. When the surface pressure reaches at 40 mN/m, the monolayer was deposited onto the hydrophilic glass substrates by Y-type deposition. We also investigated the frequency characteristics of LB modified glass by QCM. We investigated morphology of arachidic acid by AFM. As a result, we obtained the frequency characteristic and morphology of LB films from controlling the deposited layers.
