  • We fabricated the distributed feedback (DFB) InP/InGaAs/InP grating structures on InP (100) substrates by metal-organic chemical vapor deposition, and their structural properties were investigated by atomic force microscopy and scanning electron microscopy. Self-assembled InAs/InAlGaAs quantum dots (QDs) were grown on the InP/InGaAs/InP grating structures by molecular beam epitaxy, and their optical properties were compared with InAs/InAlGaAs QDs without grating structure. The duty of the grating structures was about 30%. The PL peak position of InAs/InAlGaAs QDs grown on the grating structure was 1605 nm, which was red-shifted by 18 nm from that of the InAs/InAlGaAs QDs without grating structure. This indicates that the formation of InAs/InAlGaAs QDs was affected by the existence of the DFB grating structures.

  • This study was conducted to compare the immunochemical properties of female-specific serum proteins (vitellogenin, VTG) and egg yolk proteins in female fusilier, Caesio diagramma. VTG of fusilier was identified and characterized by using immunochemical analysis. Two types of VTG (VTG1 and VTG2) reacted clearly with antiserum against egg proteins, were confirmed in the serum of mature female. The results of sephacryl S-300 showed that the molecular weights of VTG1 and VTG2 were 560,000 and 410,000, respectively. Yolk proteins, E2 and E3, were isolated from egg extracts, and molecular weights of them were estimated 410,000 and 170,000, respectively. The treatment of $17\beta$-estradiol ($E_2$) to males has induced the synthesis of VTG of which immunological characteristics seems to be similar to the yolk proteins. The results suggest that VTG can be synthesized in the liver by the action of $E_2$ stimulation, and incorporated into the oocytes through the blood circulation. The level of serum $E_2$ was moderately high throughout the spawning period of June. The level of serum VTG was also sustained at high in May and June. The concentration changes of serum $E_2$ and VTG were corelated to the ovarian development in female fusilier. The results indicated that $E_2$ may have some important roles for the vitellogenesis in female fusilier. Also) the VTG can be a precursor protein of yolk not only because it could be synthesized in the liver then incorporated into the oocytes but also because an egg yolk protein had the similiar molecular weights and antigenecity with VTG.

  • Objective: The aim of the present study was to identify genetic variants based on RNA-seq data, obtained via transcriptome sequencing of muscle tissue of pigs differing in muscle histological structure, and to verify the variants' effect on histological microstructure and production traits in a larger pig population. Methods: RNA-seq data was used to identify the panel of single nucleotide polymorphisms (SNPs) significantly related with percentage and diameter of each fiber type (I, IIA, IIB). Detected polymorphisms were mapped to quantitative trait loci (QTLs) regions. Next, the association study was performed on 944 animals representing five breeds (Landrace, Large White, Pietrain, Duroc, and native Puławska breed) in order to evaluate the relationship of selected SNPs and histological characteristics, meat quality and carcasses traits. Results: Mapping of detected genetic variants to QTL regions showed that chromosome 14 was the most overrepresented with the identification of four QTLs related to percentage of fiber types I and IIA. The association study performed on a 293 longissimus muscle samples confirmed a significant positive effect of transforming acidic coiled-coil-containing protein 2 (TACC2) polymorphisms on fiber diameter, while SNP within forkhead box O1 (FOXO1) locus was associated with decrease of diameter of fiber types IIA and IIB. Moreover, subsequent general linear model analysis showed significant relationship of FOXO1, delta 4-desaturase, sphingolipid 1 (DEGS1), and troponin T2 (TNNT2) genes with loin 'eye' area, FOXO1 with loin weight, as well as FOXO1 and TACC2 with lean meat percentage. Furthermore, the intramuscular fat content was positively associated (p<0.01) with occurrence of polymorphisms within DEGS1, TNNT2 genes and negatively with occurrence of TACC2 polymorphism. Conclusion: This study's results indicate that the SNP calling analysis based on RNA-seq data can be used to search candidate genes and establish the genetic basis of phenotypic traits. The presented results can be used for future studies evaluating the use of selected SNPs as genetic markers related to muscle histological profile and production traits in pig breeding.

  • Al containing mesoporous molecular sieve (Al-MMS) was synthesized by hydrolysis of $H_2SiF_6$ and $Al(NO_3)_3{\cdot}9H_2O$. The material obtained was characterized by XRD, $N_2$-physisorption. The specific surface area was $981m^2/g$, and the average pore size was uniformity $39{\AA}$. It was confirmed that the acidity of Al-MMS was milder than that of zeolite Y based on the results of $NH_3$-TPD. Active materials, Pt and Pd, were loaded on Al-MMS in order to examine the feasibility of using Al-MMS as a catalyst support in the hydrogenation of aromatic compounds included in the residue oil of a naphtha cracker. The hydrogenation activity of PtPd/Al-MMS has been studied by following the kinetics of the hydrogenation of naphthalene, and by comparing the kinetic parameters obtained with Pt and Pd catalysts supported on the other mesoporous material support and commercial conventional support materials. PtPd/Al-MMS catalyst shows the highest activity of hydrogenation and sulfur resistance. The high activity of PtPd/Al-MMS was confirmed again in the hydrogenation of PGO (pyrolized gas oil), which is residue oil obtained from a naphtha cracker. Therefore, PtPd/Al-MMS can be applied to the hydrogenation of aromatic compounds included in the residue oil of a commercial naphtha cracker commericially.

  • Recovery conditions and characteristics of a high molecular soluble protein from surimi processing wastewater in marine manufacture using calcium powder of cuttle bone treated with acetic acid (ATC) were examined. Judging from results of total-N, pH, COD, turbidity and yields, optimal treatment concentration of ATC for recovery of high molecular soluble proteins from wastewater was $1.0\%.$ The protein recovered from seafood waste (PRW) was macromolecule weight. The COD value in the wastewater treated with ATC was very high. The PRW had a $78.4\%$ in moisture, $1.0\%$ in crude lipid and $5.7\%$ in crude ash. The proximate composition, except the crude ash, of the PRW was similar to that of commercial surimi. The PRW showed white index and similar in the content and in the composition of total amino acid to those of commercial surimi. From the results of sensory evaluation on white index and texture, the heat-induced surimi gel prepared with $5\%$ subsititution of the PRW for bulking agent of commercial surimi was not significantly different compared to that prepared with the original commercial surimi.

  • Phylogeny of the species mainly from the genus Amynthas in family Megascolecidae was inferred at the molecular level using ITS regions in rDNA. With 26 species of earthworms from 10 genera in 2 families, a stretch comprising the 3'-end of the 18S rRNA, ITS1, 5.8S rRNA, ITS2, and 5' end of 28S rRNA was amplified by applying the primers ITS-1, ITS-2. Phylogenetic analyses of nucleotide sequences with a help of MP, NJ, and QP yielded 5 groups similarly. Genus Amynthas was separated largely into two groups, Korean and Philippine origins. Species grouped into the 1st were Amynthas jirensis, A. agrestis, A. gucheonensis, A. sopaikensis, A. bubonis, A. multimaculatus, A. koreanus, A. dageletensis, A. heteropodus, A. odaesanensis, Pontoscolex sp., Pheretima sp. 1, and Dendropheretima banahawensis. Amynthas halconensis, A. isarogensis, A. mindrooensis, Pithemera sp. 2, Pithmera sp. 1, and Pleionogaster sp. clustered into one clade forming the 2nd group. Polypheretima sp. 1 and polypheretima. sp. 2 stayed closely together representing a separate monophyletic status, forming the 3rd group, apart from species in other genera. Archipheretima sp. falls into the 4th group. Distinct morphological characteristics from Archipheretima also coinsides with its branching away from others in the previously reported molecular analyses. Similar to Perionyx excavatus that has been selected as an outgroup, Aporrectodea tuberculata also showed a long branch in the phylogram, but it differed from other 24 species included in the analyses. Unlike others, for example, its habitat is very closely related to that of man.

  • In order to produce xylitol with high yield, experiments were carried out to develope xylitol dehydrogenase(XDH) defective mutant from P. stipitis and to investigate the xylitol fermentation characteristics of mutant strain. After treatment of P. stipitis with EMS, mutant PXM-4 was selected based on te XDH activity and xylitol production capability. Among the tested cosubstrates, galactose was selected as an adequate cosub-strate on xylitol production of mutant PXM-4. But with the increase in the concentration of galactose in the medium, xylitol production was decreased because the transport of xylose into cell was inhibited by galactose. The optimal concentration of galactose for the production of xylitol using 20 g/ι xylose was 20 g/ι Under this condition, maximum concentration of xylitol and yield were 14.4 g/ι and 97%, respectively. In order to prevent the inhibitory effect of xylose transport by galactose, galactose was fed with low concentration and the concentration of xylitol produced was increased up to 25 g/ι.

  • Objective: A disintegrin and metallopeptidase with thrombospondin motifs type 8 (ADAMTS8) is crucial for diverse physiological processes, such as inflammation, tissue morphogenesis, and tumorigenesis. The chicken ADAMTS8 (chADAMTS8) gene was differentially expressed in the kidney following exposure to different calcium concentrations, suggesting a pathological role of this protein in metabolic diseases. We aimed to examine the molecular characteristics of chADAMTS8 and analyze the gene-expression differences in response to toll-like receptor 3 (TLR3) stimulation. Methods: The ADAMTS8 mRNA and amino acid sequences of various species (chicken, duck, cow, mouse, rat, human, chimpanzee, pig, and horse) were retrieved from the Ensembl database and subjected to bioinformatics analyses. Reverse-transcription polymerase chain reaction (RT-PCR) and quantitative PCR (qPCR) experiments were performed with various chicken tissues and the chicken fibroblast DF-1 cell line, which was stimulated with polyinosinic-polycytidylic acid (poly[I:C]; a TLR3 ligand). Results: The chADAMTS8 gene was predicted to contain three thrombospondin type 1 (TSP1) domains, whose amino acid sequences shared homology among the different species, whereas sequences outside the TSP1 domains (especially the amino-terminal region) were very different. Phylogenetic analysis revealed that chADAMTS8 is evolutionarily clustered in the same clade with that of the duck. chADAMTS8 mRNA was broadly expressed in chicken tissues, and the expression was significantly up-regulated in the DF-1 cells in response to poly(I:C) stimulation (p<0.05). These results showed that chADAMTS8 may be a target gene for TLR3 signaling. Conclusion: In this report, the genetic information of chADAMTS8 gene, its expression in chicken tissues, and chicken DF-1 cells under the stimulation of TLR3 were shown. The result suggests that chADAMTS8 expression may be induced by viral infection and correlated with TLR3-mediated signaling pathway. Further study of the function of chADAMTS8 during TLR3-dependent inflammation (which represents RNA viral infection) is needed and it will also be important to examine the molecular mechanisms during different regulation, depending on innate immune receptor activation.

  • The adsorption dynamics of activated carbon (AC) and carbon molecular sieve (CMS) beds were studied to recover ethylene from FCC fuel gas. In this study, the FCC fuel gas used consisted of six-component mixture ($CH_4/C_2H_4/C_2H_6/C_3H_6/N_2/H_2$,32:15:14:2:12:25 vol.%). And the breakthrough experiments of adsorption and desorption were carried out. The breakthrough sequence in the AC bed was $H_2$ < $N_2$ < $CH_4$ < $C_2H_4$ < $C_2H_6$ while the sequence in the CMS bed was $H_2$ < $CH_4$ < $N_2$ < $C_2H_6$ < $C_2H_4$. The separation performance of the CMS bed during the adsorption step was lower than that of the AC bed. However, due to the characteristics of kinetic separation, the CMS bed could remove $CH_4/N_2$ as well asthe molecules that are larger than $C_2H_6$, which was not easy to be done by the AC bed. Since it was hard to regenerate the adsorption bed by simple depressurization, vacuum regeneration should be adopted. As a result, the pressure vacuum swing adsorption (PVSA) process, consisting of CMS pretreatment process and AC main process, was suggested to recover ethylene efficiently.

  • The molecular epidemiological characteristics of methicillin-resistant Staphylococcus aureus (MRSA) isolates have demonstrated their genetic diversity and evolution. A total of 137 strains of MRSA clinical isolates was collected from Korean healthcare facility in 2007. The MRSA clinical isolates were analyzed by molecular typings (SCCmec element and agr locus typing), virule nce factor gene detections {(Panton-Valentine leukocidin (PVL), enterotoxin, exfoliative toxin and toxic shock syndrome toxin-1), and amplified fragment length polymorphism (AFLP)}. The MRSA clinical isolates were classified as SCCmec type II-agr type 1 (2 strains), type II-agr type 2 (79 strains), type III-agr type 1 (24 strains), type III-agr type 2 (2 strains), type IV-agr type 1 (27 strains), type IV-agr type 2 (2 strains), and non-typable (1 strain, agr type 3). Based on SCCmec types, SCCmec type II (95.1%) and III (88.5%) indicated higher multidrug resistance rate than SCCmec type IV (10.3%) (P<0.001). The most common enterotoxin genes were seg (83.8%), sei (83.1%), and sec (80.2%). The tst gene was present in 86 out of 137 (62.8%) MRSA isolates. All MRSA isolates were negative for PVL and exfoliative toxin genes. The combinations of toxin genes were observed in particular SCCmec types; 97.6% of SCCmec type II strains carried sec, seg, sei and tst genes, 73.0% of SCCmec type III strains carried sea gene, and 89.7% of SCCmec type IV strains carried sec, seg and sei genes. Each of the SCCmec types of MRSA isolates had distinct AFLP profile. In conclusion, SCCmec type II, agr type 1 and 2 have demonstrated to be the most common types in Korea, and the results indicated that the virulence factors are closely associated with their molecular types (SCCmec and agr types).
