A study on anisotropic characteristics of axial strengths in $\alpha$-quartz by using molecular dynamics simulation and uniaxial compression test (분자동력 학 시뮬레이션과 일축압축강도시험을 이용한 $\alpha$-quartz의 결정축에 따른 강도이방성 검토)

  • We carried out NPT-ensemble (constant-number of particles, pressure, and temperature) Molecular Dynamics (MD) simulations for measuring strength anisotropy under uniaxial compressive stress rotated to the crystallographic axes in $\alpha$-quartz. Uniaxial compressive strengths of a single quartz crystal were measured in directions of the a- and c-axis. Measured uniaxial strength of a single quartz crystal was higher in the direction parallel to the c-axis than that measured in the direction normal to the c-axis. However the reverse was found in calculated uniaxial strengths by MD simulation. The contradictive result of strengths was observed in both cases but was found to be different in origin. Strength anisotropy of defectless $\alpha$-quartz crystal in MD simulation is basically caused by structural difference of quartz. By contrast, anisotropy of measured strength in the uniaxial compression test is related to oriented micro-defects developed during crystal growth.

Spirometra decipiens (Cestoda: Diphyllobothriidae) Collected in A Heavily Infected Stray Cat from the Republic of Korea

  • Morphological and molecular characteristics of spirometrid tapeworms, Spirometra decipiens, were studied, which were recovered from a heavily infected stray cat road-killed in Eumseong-gun, Chungcheongbuk-do (Province), the Republic of Korea (=Korea). A total of 134 scolices and many broken immature and mature proglottids of Spirometra tapeworms were collected from the small intestine of the cat. Morphological observations were based on 116 specimens. The scolex was 22.8-32.6 mm (27.4 mm in average) in length and small spoon-shape with 2 distinct bothria. The uterus was coiled 3-4 times, the end of the uterus was ball-shaped, and the vaginal aperture shaped as a crescent moon was closer to the cirrus aperture than to the uterine aperture. PCR amplification and direct sequencing of the cox1 target fragment (377 bp in length and corresponding to positions 769-1,146 bp of the cox1 gene) were performed using total genomic DNA extracted from 134 specimens. The cox1 sequences (377 bp) of the specimens showed 99.0% similarity to the reference sequence of S. decipiens and 89.3% similarity to the reference sequence of S. erinaceieuropaei. In the present study, we report a stray cat heavily infected with S. decipiens identified by mitochondrial cox1 sequence analysis and morphological examinations of the adult worms.

Composition of Cotton Textile Dyeing Process Wastewater and its Treatment Characteristics by Ionized Gas (면섬유염색폐수의 공정별 폐수성상과 이온화가스에 의한 처리특성)

  • Three types dyeing wastewater (dark, medium, light color) discharged from cotton textile dyeing with reactive dye was collected at each step of process. Each process dying wastewater was analyzed and treated by ionized gas. The analysis focused on $COD_{Cr}$, SS and color. Bleaching & scouring process wastewater has the highest $COD_{Cr}$ value in the three type dyeing wastewater. SS shows the highest value at dyeing process wastewater in dark and medium color but light color has at finishing process wastewater. The result of process wastewater treatment by ionized gas was that the ionized gas was effective in $COD_{Cr}$ removing of bleaching & scouring process and finishing process wastewater but was not good at dyeing process wastewater. From that result it is estimated that the ionized gas could not work in opening the aromatic ring and react only in aliphatic component of the molecule. Because the surfactants contained in bleaching & scouring process and finishing process wastewater have only one aromatic ring in its molecular structure, in contrast with the reactive dye compounds consist of aromatic rings great part of its molecular structure. The color almost removed in 1.5 hrs reaction time but $COD_{Cr}$ removal effiency was only 30.7% through 3hrs in 1500 mL of total dyeing wastewater treated by 10 L/min ionized gas.

Diversity of Arbuscular Mycorrhizal Fungi of Woody Plants in Mt. Munan (문안산의 목본식물과 공생하는 수지상균근균의 다양성)

  • This study was conducted to reveal the diversity of arbuscular mycorrhizal fungi (AMF) in Mt. Munan, based on the morphological and molecular characters. Soil and root samples were collected from the rhizosphere of host plants including Lindera obtusiloba, Stephanandra incisa, Styrax obassis and Symplocos sawafutagi and AMF were trap-cultured with Sorghum bicolor as a host plant in a greenhouse. The spores were extracted from the cultured soils and five species were identified using morphological and molecular characteristics; Acaulospora longula, A. mellea, Ambispora leptoticha, Gigaspora margarita and Paraglomus occultum. The distribution of AMF showed different trends according to host plants. The dominant AMF species were A. longula in L. obtusiloba, A. leptoticha in S. incisa, S. obassis and S. sawafutagi.

Physical Characteristics of Silicone Modified Epoxy as a Undercoating Materials (실리콘 변성 에폭시 언더코팅액 물성 연구)

  • The effect of modification conditions on the physical properties of polydimethyl siloxane modified epoxy (PDMSME) was investigated. The number of ring opened epoxy attached to polydimethylsiloxane (PDMS) by silane coupling agent affected the physical properties of undercoating materials. The flexibility of thin coating was enhanced by PDMSME and the increase of ring opened epoxy attached to PDMS resulted in the increase of hardness by the crosslinking in the present with moisture. The higher molecular weight of PDMS caused the lowering of hardness while the surface contact angle increased due to the high silicone content in PDMS. The viscosity of silicone modified epoxy coating materials decreased with increasing of molecular weight of PDMS due to the lowering of entanglement of PDMSME molecules by acetone solvent and consequently, the smooth undercoated surface was obtained.

  • Biodegradable polymers have gained enormous attentions in the pharmaceutical and biomedical applications, especially in drug delivery. In this work, we report the synthesis and characteristics of high molecular weight polyoxalate with ~75000 Da. Hydrolytic degradation kinetics and degradation products were characterized by nuclear magnetic resonance and gel permeation chromatography. Polyoxalate is a semicrystalline and thermally stable polymer with a glass transition temperature of ${\sim}35^{\circ}C$, which is suitable for drug delivery applications. The hydrophobic nature of polyoxalate allows it to be formulated into nanoparticles and encapsulate drugs using a conventional oil-in-water emulsion/solvent displacement method. Polyoxalate nanoparticles also exhibited excellent cytotoxicity profiles. It can be suggested that polyoxalate has great potential for numerous biomedical and pharmaceutical applications.

Systematic Relationships of Korean Freshwater Snails of Semisulcospira, Koreanomelania, and Koreoleptoxis (Cerithiodiea; Pleuroceridae) revealed byMitochondrial Cytochrome Oxidase I Sequences

  • Many freshwater snail taxa are difficult to identify using morphological traits due to phenotypic plasticity. However, using of molecular DNA marker in combination with morphological traits can provide a reliable means for discriminating among freshwater snail taxa including cryptic species. To discriminate among Korean freshwater snail taxa and resolve their systematic relationships, wesequenced a fragment of mtDNA cytochrome oxidase I (COI) gene from 82 specimens collected from ten different sites distributed along the Korean peninsula. We identified more than seven freshwater snail taxa including cryptic species in Korea. Whereas traditional shell morphology of freshwater snails offers only weak discriminatory power for recognizing 'good' taxa, DNA sequence data provided positive and reliable identification. In addition, a major Semisulcospira clade was clearly separated from the remaining lineages observed including cryptic species. However, a phylogenetic tree inferred from the COI gene data did not fully resolve systematic relationships among pleurocerid taxa in Korea. Establishing more robust shell characteristics for identifying taxa unambiguously and hence improving traditional key shell morphology characters for freshwater snail species is an urgent requirement and will require more rigorous examination of all nominal taxa. While molecular data generated here will be useful for species identification and for describing the systematic relationships among Korean freshwater snails, further analysis will be required.

Induction of p21 and apoptosis by C11 in human hepatocarcinoma cells

  • C11, a chloride-containing VK3 analog, acts as a mediator of programmed cell death in SK-Hep-1 cell lines, but its molecular mechanisms linked to cell death are not understood. In this study, we investigated the expression of p21 gene and its relationship to apoptosis induced by C11. In SK -hep-1 cells, the addition of C11 resulted in time-dependent growth suppression and DNA fragmentation characteristics of apoptosis. p21 protein was induced during this process, while the protein level of p53 was not changed at the same condition. This apoptotic cell death with p21 induction was also observed in the Hep3B cells lacking functional p53 after treatment of C11. These results suggest that C11-induced apoptosis is associated with up-regulation of p21 protein in p53-independent pathway. Next, in order to confirm whether the p53-independent p21 induction is required for C11-induced apoptosis, we introduced the p21 gene into Hep3B. Overexpression of p21 did not affect the expression of the bcl-2 gene, but DNA fragmentation and PARa cleavage were significantly increased. These data indicate that p21 is involved in C11-induced apoptosis. Although Bcl-2 has been implicated to interfere with an essential signaling molecule involved in the apoptosis pathway, its molecular mechanism and target molecule are poorly understood. To determine the effects of bcl-2 overexpression on apoptosis and to investigate whether BcI-2 interfers with the p53-independent p21 pathway, we transfected the bcl-2 expression vector into SK - Hep-1 cels. Overexpression of Bcl-2 prevented C11-induced apoptosis. Taken together, C11-induced apoptosis is regulated by p52-independent p21 pathway and bcl-2 may inhibit functional activity of p21, therebe may inhibit the C11-induced apoptosis.ptosis.

Identification of two coliphages from Han-river and its adsorption-elution effect on soil materials (한강에서 분리한 이종 coliphage의 동정과 점토질에 대한 흡착 및 용출효과)

  • Coliphages isolated from Han-River from September 1980 to August 1981 were classified by morphological and physiological characteristics. Effects of soil metrial on the fate of coliphage in nature were investigated. 1. The correlation coefficient between coliphage and E.coli which was host of coliphages in nature was 0.7173 (p=0.004). 2. Coliphage I isolated from Han-River of which DNA molecular weight was $27{\times}10^6$ daltons was identified as $T_1$ phage and coliphage II of which DNA molecular weight $72{\times}10^6$ daltons was classified as $T_5$ phage. 3. Soil material SW was composed of 63.65% silt and 21.92% clay. Clay was consisted of illite, kaolinite and chlorite evenly. Soil material J was composed of 68.92% silt and 11.67% clay. Clay consisted of smectite only. 4. Coliphage was absorbed to soil material J more than soil material SW, and $T_1$ coliphage was absorbed to soil material more than $T_5$ coliphage was. 5. The phage adsorption efficiency to soil material was enhanced at lower pH : the phage adsorption efficiency at pH 4 was 27 time higher than at pH 7. 6. Divalent $(Ca^{2+})\;and\;trivalention\;(Al^{3+})$ enhanced the phage adsorption efficiency to soil material from 4 to 39 and from 17 to 91 times higher than monovalent $ion(Na^+)$, respectively. 7. The concentration of organic compound was inversely related to the phage adsorption efficiency to soil. 8. Adsorption of phage onto soil material, and elution efficiency of elutants was in the order of D.D.W>tap water>river water>seawater. 9. The higher the concentration of organic compound was, the more were adsorbed phages to soil eluted. 10. Coliphages survived longer in sterile soil suspension than in nonsterile soil material suspension.

Endo-1,4-β-xylanase B from Aspergillus cf. niger BCC14405 Isolated in Thailand: Purification, Characterization and Gene Isolation

  • During the screening of xylanolytic enzymes from locally isolated fungi, one strain BCC14405, exhibited high enzyme activity with thermostability. This fugal strain was identified as Aspergillus cf. niger based on its morphological characteristics and internal transcribed spacer (ITS) sequences. An enzyme with xylanolytic activity from BCC14405 was later purified and characterized. It was found to have a molecular mass of ca. 21 kDa, an optimal pH of 5.0, and an optimal temperature of $55^{\circ}C$. When tested using xylan from birchwood, it showed $K_m$ and $V_{max}$ values of 8.9 mg/ml and 11,100 U/mg, respectively. The enzyme was inhibited by $CuSO_4$, EDTA, and by $FeSO_4$. The homology of the 20-residue N-terminal protein sequence showed that the enzyme was an endo-1,4-$\beta$-xylanase. The full-length gene encoding endo-1,4-$\beta$-xylanase from BCC14405 was obtained by PCR amplification of its cDNA. The gene contained an open reading frame of 678 bp, encoding a 225 amino acid protein, which was identical to the endo-1,4-$\^{a}$-xylanase B previously identified in A. niger.
