Methylobacterium sp. GL-10의 유가식 배양에 의한 Methanol로 부터 Poly-$\beta$-hydroxybutyrate의 생산 (Production of Poly-$\beta$-hydroxybutyrate from Methanol by Fed-batch Cultivation of methylobacterium sp. GL-10)

  • The production of poly-$\beta$-hydroxybutyrate(PHB) from methanol by batch and fed-batch cultivations of Methylobacterium sp. GL-10 was studied. PHB accumulation was stimulated by the nutrients deficiency including, NH4+, SO42-, and K+. The nitrogen deficiency was the most critical factor for PHB accumulation. In batch cultivation, the maximum cell concentration and PHB content were 1.86g/l and 0.62g/l, respectively, with 1.0%(v/v) of methanol and 0.5g/1 of ammonium sulfate. The mass doubling time of Methylobacterum sp. GL-10 was in the range of 4-5 hrs. The cell growth and PHB accumulation were severely inhibited at the methanol concentration over than 2% (v/v). To overcome methanol Inhibition, constant feeding and intermittent feedillg fed-batch cultivations were adopted, using C/N molar ratio as a control factor. In constant feeding fed-batch process, cell concentration was increased up to 2.67g/1, and PHB yield was enhanced from 0.33 of batch culture to 0.53. The relatively low cell concentration was caused by methanol accumulated in culture broth at late growth phase. To prevent methanol accumulation and to maximize PHB production, DO-state intermittent fed-batch cultivation was attempted. The cell and PHB concentration was reached up to 4.55g/1 and 1.80g/1, respectively. It was possible to maintain methanol concentration low and also to feed nutrient of desired C/N molar ratio.

The efficacy of an elevated concentration of lidocaine HCl in impacted lower third molar surgery

  • Background: There have been few studies on the effect of an elevated concentration of lidocaine hydrochloride in the surgical removal of an impacted lower third molar. This study aimed to examine the efficacy of 4% lidocaine along with 1:100,000 epinephrine compared to 2% lidocaine along with 1:100,000 epinephrine as inferior alveolar nerve block for the removal of an impacted lower third molar. Methods: This single-blind study involved 31 healthy patients (mean age: 23 y; range: 19-33 y) with symmetrically impacted lower third molars as observed on panoramic radiographs. Volunteers required 2 surgical interventions by the same surgeon with a 3-week washout period. The volunteers were assigned either 4% lidocaine with 1:100,000 epinephrine or 2% lidocaine with 1:100,000 epinephrine as local anesthetic during each operation. Results: We recorded the time of administration, need for additional anesthetic administration, total volume of anesthetic used. We found that the patient's preference for either of the 2 types of local anesthetic were significantly different (P < 0.05). However, the extent of pulpal anesthesia, surgical duration, and duration of soft tissue anesthesia were not significantly different. Conclusions: Our study suggested that inferior alveolar nerve block using 4% lidocaine HCl with 1:100,000 epinephrine as a local anesthetic was clinically more effective than that using 2% lidocaine HCl with 1:100,000 epinephrine; the surgical duration was not affected, and no clinically adverse effects were encountered.

  • In this study, chromate and cetylperidinium chloride (CPC) removal from artificial wastewater was monitored by using micellar enhanced ultrafiltration (MEUF) and activated carbon fibre (ACF) adsorption hybrid processes. For the efficient chromate removal, molar concentration of the CPC should be five times that of chromate and it should be at least one critical micelle concentration (CMC). The MEUF was found to be effective in the chromate removal while ACF in the CPC adsorption to produce chromate and CPC free effluents. The chromate and CPC removal was 99.8% from MEUF-ACF process. Effluent chromate concentration was exponentially correlated with molar ratio of CPC to chromate and pH.

Influence of the Structural Characteristics of Amino Acids on Direct Methylation Behaviors by TMAH in Pyrolysis

  • Direct methylation behaviors of 20 amino acids with tetramethylammonium hydroxide (TMAH) were studied under diluted conditions with silica. Amino acid concentration was controlled by dilution with silica ($SiO_2$) and the molar ratios of amino acid/silica were 0.20, 0.50, and 2.0. The molar ratios of amino acid/TMAH (0.51 - 4.64) also varied. It was found that arginine, asparagine, aspartic acid, cysteine, glutamic acid, and glutamine did not generate any directly methylated pyrolysis products, whereas alanine, glycine, isoleucine, leucine, methionine, phenylanaline, valine, and proline generated all the directly methylated pyrolysis products. Tri- and tetra methylated products of lysine consisted of two types. Histidine and threonine hardly generated the partly methylated products. Mono- and dimethylated products of serine, tryptophan, and tyrosine were not observed. Relative intensities of the methylated products varied with the amino acid concentration, TMAH concentration, and pyrolysis temperature. Direct methylation behaviors of amino acids were explained by the structural characteristics of amino acids.

Effects of Monensin Administation on Mammary Function in Late Lactating Crossbred Holstein Cattle

  • An experiment was carried out to study the effect of monensin administration on mammary functions in crossbred Holstein cattle. Fourteen non-pregnant late lactating crossbred Holstein cattle, approximately 270 days postpartum, were selected for the experiment. They were divided into two groups of 7 animals each. Seven animals in the treated group were given sodium monensin orally in a slow-release capsule. Animals in both control and treated groups were fed the similar diet to maintain milk production and body score at 2.5. Rice straw was fed as a source of dietary fiber throughout the experimental period. After monensin administration, a significant increase in the molar percent of ruminal propionate (p<0.05) and a significant decrease in the molar percent of ruminal acetate (p<0.05) were apparent in comparison to the pretreated period. The ratio of acetate to propionate concentration decreased significantly after monensin administration (p<0.05), while it was maintained at the similar level throughout the period of experiment in the control group. Monensin did not affect the molar percent of ruminal butyrate and valerate. The concentration of milk allantoin between the control group and monensin treated group was not different. An excretion rate of allantoin in milk decreased in animals treated with monensin (p<0.05). Mammary blood flow did not show significant difference between control and monensin treated groups. The plasma glucose concentration, arteriovenous concentration difference and mammary gland uptake of glucose remained constant in both groups. Milk yield of the later stage of lactation in the control group declined during lactation advance while a tendency to increase in the milk yield was apparent after 21 days monensin administration. Milk compositions for concentration of lactose, fat and protein in both control group and monensin treated group did not change throughout the experimental periods. From these results, it can be concluded that the action of monensin could affect the ruminal fermentation pattern. Monensin could not increase milk yield in the late lactating period.

전기펜톤공정을 이용한 석유화학공장 폐활성슬러지의 감량화 가능성 평가 (A study on reduction of excess sludge in activated sludge system from a petrochemical plant using electro fenton process)

  • The reduction of excess activated sludge from petrochemical plant was investigated by the electro fenton (E-Fenton) process using electrogenerated hydroxyl radicals which lead to mineralization of activated sludge to $CO_2$, water and inorganic ions. Factors affecting the disintegration efficiency of excess activated sludge in E-Fenton process were examined in terms of five criteria: pH, $H_2O_2/Fe^{2+}$ molar ratio, current density, initial MLSS (mixed liquid suspended solids) concentration, $H_2O_2$ feeding mode. TSS total suspended solid and $TCOD_{cr}$ reduction rate increased with the increasing $H_2O_2/Fe^{2+}$ molar ratio and current density until 42 and $6.7 mA/cm^2$, respectively but further increase of $H_2O_2/Fe^{2+}$ molar ratio and current density would reduce the reduction rate. On the other hand, as expected, increasing pH and initial MLSS concentration of activated sludge decreas TSS and $TCOD_{cr}$ reduction rate. The E-Fenton process was gradually increased during first 30 minutes and then linearly proceed till 120 minutes. The optimal E-Fenton condition showed TSS reduction rate of 62~63% and $TCOD_{cr}$ (total chemical oxygen demand) reduction rate of 55~56%. Molar ratio $H_2O_2/Fe^{2+} = 42$ was determined as optimal E-Fenton condition with initial $Fe^{2+}$ dose of 5.4 mM and current density of $6.7{\sim}13.3 mA/cm^2$, initial MLSS of 7,600 mg/L and pH 2 were chosen as the most efficient E-Fenton condition.

Single buccal infiltration of high concentration lignocaine versus articaine in maxillary third molar surgery

  • Background: This research evaluated the numbness produced by lignocaine at an equal or higher concentration than that of 4% articaine through a single point of injection for maxillary third molar surgery. This randomized double-blind study was conducted to compare the anesthetic efficiency of 4% lignocaine with that of 4% articaine in impacted maxillary third molar surgery using a single buccal infiltration alone. Methods: The study participants were 30 healthy patients requiring the bilateral surgical removal of symmetrically-positioned maxillary third molars. Using a split-mouth design, each patient randomly received buccal infiltration of 1.7 ml of 4% lignocaine and 1.7 ml of 4% articaine during two separate appointments. After 15 minutes of anesthetic injection, surgery was performed by the same surgeon using a consistent technique on both sides. Pinprick test pain scores of the buccal and palatal gingiva of the maxillary third molar after 10 minutes and 15 minutes latencies, pain scores during the surgery, the need for supplemental anesthesia, and patients' satisfaction with anesthetic efficiency were recorded. Surgery performed without supplemental anesthesia was categorized as successful. Results: The success rates of 4% lignocaine and 4% articaine (83.34% vs. 86.67%, P = 1.00) were not significantly different. Only 5 cases (4 cases in the articaine group and 1 case in the lignocaine group) reported mild pain and pressure sensation (NRS ≤ 1) on probing at the palatal side after 15 minutes of latency (P = 0.25). The pain scores of maxillary third molar surgery in the two groups were not significantly different (P > 0.05). Moreover, the statistical analysis confirmed the comparable patient satisfaction of two study groups (P = 0.284). Conclusion: This study provides evidence that single buccal infiltrations of 4% lignocaine and 4% articaine have comparable anesthetic efficacy and success rates for impacted maxillary third molar surgery. Both 4% lignocaine and 4% articaine can produce effective palatal anesthesia and pain control using buccal infiltration alone after 15 minutes of latency.

3종류의 구치부 Uprighting Spring의 초기응력분포에 관한 광탄성학적 연구 (A PHOTOELASTIC STUDY ON THE INITIAL STRESS DISTRIBUTION BY 3 TYPES OF MOLAR UPRIGHITNG SPRING)

  • The aim of this investigation was to identify which suing is more suitable for uprighting of the 30 degree inclined lower second molar in different situations. Between four different molar uprighting springs which were A, B, C type and T-loop spring, the author tested T-loop suing as a control material and the other springs were experimental group. Each spring was fabricated from .017' $\times$ .025' TMA wire and preactivated with 40 degree tip-back bend. Stabilizing unit included from the lower right central incisor to the lower right second premolar which were made by acrylic resin. The photoelastic overview of the lower right quadrant showed that stress concentrations were observed in its photoelastic model. The obtained results were as follows. 1. Higher concentration of compression can be seen clearly at the distal curvature than the mesial of the lower second molar when A type uprighting spring was applied, which can be used as a space regainer. 2. Higher level compression was presented at the mesial root apex area than the distal of the lower second molar in B type uprighting spring, which can be used as a space closer. 3. Areas of higher compression and tension were observed about the mesial and distal root of the lower second moalr than A and B type in C type uprighting spring, which can be used as a partial space regainer.

뇌의 양성자 핵자기공명 분광학을 위한 정량화 방법 개발 (The Development of Quantification Technique for Brain In vivo Proton NMR Spectroscopy)

  • 핵자기공명분광학(NMR Spectroscopy)을 이용해서 우리는 생물체의 조직안에 있는 각종 대사물질들을 측정할 수 있게 되었으며, 이것은 또 지금까지 알려진 여러 방법 중에서 생물체내의 신진대사 물질이나 생화학적인 변화를 비침습적(noninvasive)으로 알아낼 수 있는 유일한 방법에 속한다. 양성자 핵자기공명분광학은 최근들어 사람이나 흑은 동물뇌의 생화학적인 특성을 연구하는데 많이 사용되어오고 있다. 핵자기공명영상은 주로 물분자에 있는 양성자의 밀도와 그들의 이완현상(relaxation phenomena)을 측정하여 영상화하지만 핵자기공명분광학은 생체조직내의 신진대사에 관한 생화학적인 정보를 제공한다. 많은 경우에 있어서 핵자기공명분광학은 CT나 MRI 영상에서는 발견할 수 없는 정보를 제공함으로써 특정질병의 진단에 사용될 수 있으며, 따라서 환자들의 질병상태를 효과적으로 진단하는데 쓰여질 수 있다. 따라서 본 연구에서는 사람의 뇌에서 수소 핵자기공명분광학을 이용하여 대사물질들의 농도를 정량화하는 방법을 시도하였다. 이를 위해 펀텀을 이용하여 대사물질들 각각의 스팩트럼을 구했고, 성인 남녀 16명을 대상으로 insula gray matter 부위의 대사물질들의 농도를 측정하였다. 그리고 이 값들은 다른 연구자들에 의해 밝혀진 값들과 매우 근사한 값을 제공하였다.

Synthesis and Characterization of Yttrium-doped Core-Shell SiO2 Nanoparticles by Reverse Micelle and Sol-gel Processing

  • In this study, yttrium-doped $SiO_2$ nanoparticles are synthesized using a reverse micelle technique combined with metal alkoxide hydrolysis and condensation. Spherical Y-doped $SiO_2$ nanoparticles with a uniform size distribution are prepared using selfassembly molecules in conjunction with the hydrolysis and condensation of organometallic precursors. The water/surfactant molar ratio influenced the Y-doped $SiO_2$ particles distribution of the core-shell composite particles and the distribution of Y doped $SiO_2$ particles was broadened as the water to surfactant ratio increased. The particle size of Y increase linearly as the $Y(NO_3)_3$ solution concentration increased. The average size of the cluster was found to depend on the micelle size, the nature of the solvent, and the concentration of the reagent. The effects of synthesis parameters, such as the molar ratio of water to surfactant and the molar ratio of water to TEOS, are discussed.
