Quercetin Ameliorates NO Production via Down-regulation of iNOS Expression, $NF{\kappa}B$ Activation and Oxidative Stress in LPS-Stimulated Macrophages

  • Effect of quercetin on NO production and regulation mode of quercetin on oxidative stress, $NF{\kappa}B$ activation, and iNOS expression, possible mechanisms of NO suppression in LPS-stimulated macrophages were investigated. Treatment of RAW 264.7 cells with quercetin significantly reduced lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production dose-dependently ($IC_{50}$, $9.2\;{\mu}M$). Expression of iNOS and specific DNA binding activities of nuclear factor kB ($NF{\kappa}B$) were significantly suppressed by quercetin pretreatment. Quercetin reduced thiobarbituric acid-reactive substances (TBARS) accumulation, enhancing GSH level and antioxidant activities of enzymes, such as superoxide dismutase (SOD) and catalase. These results demonstrate quercetin may ameliorate inflammatory diseases by suppressing NO production through inhibition of iNOS expression, $NF{\kappa}B$ transactivation, and oxidative stress, which may be mediated partially by antioxidative effect of quercetin. Thus, quercetin appears to be used as a potential therapeutic agent for treating LPS-induced inflammatory processes.

연속회분식 처리시스템에서 인공하수를 이용한 Polyhydroxyalkanoates (PHAs)의 생산 (Production of Polyhydroxyalkanoates (PHAs) from Sequencing Batch Reactor Using Synthetic Wastewater)

  • 인공폐수로 연속회분식 하수처리시스템에서 바이오플라스틱인 PHAs의 생산에 대하여 연구하였다. 실험은 하수처리장에서 식종슬러지를 채취하고, 실험실 내에서 제작된 인공하수를 이용하여 4 L 규모의 2조의 연속회분식반응기로 수행하였다. 인공하수의 영양조건(C/N/P)은 42:10:1로 운전되었으며, 연속회분식반응기는 호기상태에서 Feast/Famine 조건을 부여하는 ADF (aerobic dynamic feeding)의 SBR 1과 혐기/호기조건에서 Feast/Famine 조건을 부여하는 AODF (anaerobic/oxic dynamic feeding)의 SBR 2로 운전하였다. PHAs의 생산은 AODF가 ADF 보다 우수하였으며, ADOF 에서 바이오매스 대비 최대 40.0%(w/w)로 높게 생산되었으며, 구조적 및 열적 물성도 우수한 것으로 나타났다.

Enhancement of Geldanamycin Production by pH Shock in Batch Culture of Streptomyces hygroscopicus subsp. duamyceticus

  • Various sequences of pH change were applied in a batch bioreactor to investigate pH shock effects on geldanamycin production by Streptomyces hygroscopicus subsp. duamyceticus JCM4427. In the control culture where the pH was not controlled, the maximum geldanamycin concentration was 414 mg/l. With the pHS1 mode of pH shock, that is, an abrupt pH change from pH 6.5 to pH 5.0 and then being maintained at around pH 5.0 afterward, 768 mg/l of geldanamycin was produced. With pHS2, in which the pH was changed sequentially from pH 6.7 to pH 5.0 and then back to pH 6.0, 429 mg/l of geldanamycin was produced. With pHS3 having a sequential pH change from pH 6.0 to pH 4.0 and then back to pH 6.5 followed by the third pH shock to pH 5.5, no geldanamycin production was observed. Considering that the productivity with pHS1 was about two-fold of that of the control culture with no pH control, we concluded that a more sophisticated manipulation of pH would further promote geldanamycin production.

Hindsiipropane B alleviates HIV-1 Tat-induced inflammatory responses by suppressing HDAC6-NADPH oxidase-ROS axis in astrocytes

  • Human immunodeficiency virus-1 (HIV-1) transactivator of transcription (Tat) is an important viral factor in neuro-inflammation. Hindsiipropane B, present in Celastrus hindsii, possesses various biological mechanisms including anti-inflammatory activity. In this report, we explored the regulatory activity of hindsiipropane B on HIV-1 Tat-mediated chemokine production and its mode of action in astrocytes. Hindsiipropane B significantly alleviated HIV-1 Tat-mediated production of inflammatory chemokines, CCL2, CXCL8, and CXCL10. Hindsiipropane B inhibited expression of HDAC6, which is important regulator in HIV-1 Tat-mediated chemokine production. Hindsiipropane B diminished HIV-1 Tat-mediated reactive oxygen species (ROS) generation and NADPH oxidase activation/expression. Furthermore, hindsiipropane B inhibited HIV-1 Tat-mediated signaling cascades including MAPK, $NF-{\kappa}B$, and AP-1. These data suggest that hindsiipropane B exerts its inhibitory effects on HIV-1 Tat-mediated chemokine production via down-regulating the HDAC6-NADPH oxidaseMAPK-$NF-{\kappa}B$/AP-1 signaling axis, and could serve as a therapeutic lead compound against HIV-1 Tat-associated neuro-inflammation.

The effects of dentin bonding agent formulas on their polymerization quality, and together with tooth tissues on their microleakage and shear bond strength: an explorative 3-step experiment

  • PURPOSE. Bonding agents (BA) are the crucial weak link of composite restorations. Since the commercial materials' compositions are not disclosed, studies to formulize the optimum ratios of different components are of value. The aim of this study was to find a proper formula of BAs. MATERIALS AND METHODS. This explorative experimental in vitro study was composed of 4 different sets of extensive experiments. A commercial BA and 7 experimental formulas were compared in terms of degree of conversion (5 experimental formulas), shear bond strength, mode of failure, and microleakage (3 experimental formulas). Statistical analyses were performed (${\alpha}$=.05). The DC of selected formula was tested one year later. RESULTS. The two-way ANOVA indicated a significant difference between the shear bond strength (SBS) of two tissues (dentin vs. enamel, P=.0001) in a way that dentinal bonds were weaker. However, there was no difference between the four materials (P=.283). The adhesive mode of failure was predominant in all groups. No differences between the microleakage of the four materials at occlusal (P=.788) or gingival (P=.508) sites were detected (Kruskal-Wallis). The Mann-Whitney U test showed a significant difference between the microleakage of all materials (3 experimental formulas and a commercial material) together at the occlusal site versus the gingival site (P=.041). CONCLUSION. A formula with 62% bisphenol A-glycidyl methacrylate (Bis-GMA), 37% hydroxy ethyl methacrylate (HEMA), 0.3% camphorquinone (CQ), and 0.7% dimethyl-para-toluidine (DMPT) seems a proper formula for mass production. The microleakage and SBS might be respectively higher and lower on dentin compared to enamel.

Detection of Mendelian and Parent-of-origin Quantitative Trait Loci for Meat Quality in a Cross between Korean Native Pig and Landrace

  • This study was conducted to detect quantitative trait loci (QTL) affecting meat quality in an $F_2$ reference population of Korean native pig and Landrace crossbreds. The three-generation mapping population was generated with 411 progeny from 38 $F_2$ full-sib families, and 133 genetic markers were used to produce a sex-average map of the 17 autosomes. The data set was analyzed using least squares Mendelian and parent-of-origin interval-mapping models. Lack-of-fit tests between models were used to characterize the QTL for mode of gene expressions. A total of 10 (32) QTL were detected at the 5% genome (chromosome)-wise level for the analyzed traits. Of the 42 QTL detected, 13 QTL were classified as Mendelian, 10 as paternal, 14 as maternal, and 5 as partial expressed QTL, respectively. Among the QTL detected at 5% genome-wise level, four QTL had Mendelian mode of inheritance on SSCs 5, 10, 12, and 13 for cooking loss, drip loss, crude lipid and crude protein, respectively; two QTL maternal inheritance for pH at 24-h and shear force on SSC11; three QTL paternal inheritance for CIE b and Hunter b on SSC9 and for cooking loss on SSC15; and one QTL partial expression for crude ash on SSC13, respectively. Most of the Mendelian QTL (9 of 13) had a dominant mode of gene action, suggesting potential utilization of heterosis for genetic improvement of meat quality within the cross population via marker-assisted selection.

The three-generation mapping population was generated with 411 progeny from 38 $F_2$ full-sib families, and 133 genetic markers were used to produce a sex-average map of the 18 autosomes. The data set was analyzed using least squares Mendelian and parent-of-origin interval-mapping models. Lack-of-fit tests between the models were used to characterize QTL for mode of expressions. A total of 8 (39) QTL were detected at the 5% genome (chromosome)-wise level for the 17 analyzed traits. Of the 47 QTL detected, 21 QTL were classified as Mendelian expressed, 13 QTL as paternally expressed, 6 QTL as maternally expressed, and 7 QTL as partially expressed. Of the detected QTL at 5% genome-wise level, two QTL had Mendelian mode of inheritance on SSC6 and SSC9 for backfat thickness and bone weight, respectively, two QTL were maternally expressed for leather weight and front leg weight on SSC6 and SSC12, respectively, one QTL was paternally expressed for birth weight on SSC4, and three QTL were partially expressed for hot carcass weight and rear leg weight on SSC6, and bone weight on SSC13. Many of the Mendelian QTL had a dominant (complete or overdominant) mode of gene action, and only a few of the QTL were primarily additive, which reflects that heterosis for growth is appreciable in a cross between Korean native pig and Landrace. Our results indicate that alternate breed alleles of growth and body composition QTL are segregating between the two breeds, which could be utilized for genetic improvement of growth via marker-assisted selection.

  • Aminoglycoside-3'-phosphotransferase(APH(3'))를 생산하는 균주인 E.coli ATCC 21990을 산업적으로 이용하기 위해서 자외선 조사 및 NTG를 처리하고 고농도의 kanamycin B에 내성을 갖는 변이주인 E.coli M1과 M2를 선별하였다. E.coli M1은 단위 균체당 효소 생산성은 높으나 생육속도가 낮아 실용적이질 못했고 주 질소원인 yeast extract를 사용했을때 E.coli M1보다 E.coli M2가 생육속도가 훨씬 빨랐으며 약 2배의 APH(3')을 얻을 수 있었고 산소 가스를 사용하였을 경우는 약 2.5배의 APH(3')을 얻었다.

중국 현대 클라리넷 작품 중 민족음악 요소 연구 『목마의 노래』와 『파미이지음』을 중심으로 (A Study of Ethnomusicological Elements in Modern Chinese Clarinet focus on 『the Shepherd Horse』 and 『the Voice of the Pamir』)

  • 중국의 오랜 역사 과정에서 풍부하고 소중한 소수민족의 전통음악은 형성되었다. 각 민족은 발전 과정에서 독특한 자연환경과 특수한 생활과 생산 방식에 따라 자기만의 세계관과 음악 문화를 만들어 냈다. 클라리넷은 서양 악기로서, 중국에 전래되는 과정에서 중국 소수민족 문화의 영향을 어느 정도 받아왔다. 따라서 음악가들은 중국 소수민족 음악 중의 여러 요소를 표현하는 데에 서양 악기를 사용하는 방법을 모색했다. 본 논문은 중국 소수민족 음악의 요소를 사용하여 창작된 현대 클라리넷 작품 작곡가 왕염의 『목마의 노래』와 작곡가 호벽정의 『파미이지음』을 중심으로 작품의 형성 유래, 작품에 나타난 예술적인 표현 특징 등 두 측면에서 클라리넷에서 표현되는 민족적인 요소를 분석하고자 한다. 구체적으로는 민족음악의 모드 음계, 박자, 리듬과 선율의 표현기법, 그리고 소수민족 전통 악기를 모방한 소리의 특징 등 다섯 가지 내용으로 구성된다. 또한 지역, 생산, 생활, 민족 정서 등과 접목하여 작품에 사용된 타지크족과 몽골족 음악에 나타난 민족문화의 특징을 분석한다. 본 연구는 중국 현대 클라리넷 작품의 음악 스타일을 습득하고자 하는 창작자와 연주자에게 이론적인 근거를 제공하고 서양 악기로 중국 전통 음악을 연주할 더 좋은 방법을 모색하고자 한다.

리듬 산출 검사 어플리케이션 Beat Processing Device 개발 (Development of Beat Processing Device for Rhythm Production Assessment)

  • 본 연구의 목적은 리듬 산출 시 수행 정확도를 정밀하게 수집, 저장하기 위한 모바일 어플리케이션 'Beat Processing Device(이하 BPD)' 개발에 있다. 이를 위하여 총 3단계의 개발 과정을 거쳤다. 1단계에서는 문헌 조사를 통해 기존 노인 인지 기능 훈련을 위해 사용된 리듬꼴들을 수집, 비교, 분석함으로써, 리듬 산출 과제 구성에 필요한 4개의 리듬꼴 문항을 확정하였다. 2단계에서는 확정한 4개의 리듬꼴 문항이 탑재된 iOS 기반 모바일 어플리케이션을 개발하였으며, 예비 테스트를 통해 디바이스 종류, 사용할 악기 음색, 구현할 실험 종류 등을 최적화하였다. 마지막 단계인 3단계에서는 노인 60명을 대상으로 본 기기와 미디(MIDI)로 수집한 리듬 산출 데이터를 비교하여, BPD의 실효성과 타당성, 정밀함을 검증하였다. 본 연구에서 개발된 BPD는 인지기능 강화를 위한 리듬 수행력 증진을 평가할 수 있는 유용한 도구로 다양한 현장에서 활용될 수 있을 것으로 기대된다.
