Prevention of thermal aggregation of the denatured protein by the group II chaperonin from the aerobic hyperthermophilic crenarchaeon Aeropyrum pernix K1 (ApcpnA) has been investigated. ApcpnA exists as a homo-oligomer in a ring structure, which protects thermal aggregation of the chemically denatured bovine rhodanese at 50 oC. ApcpnA alone is not sufficient for chaperonin activity, but the chaperonin activity is greatly enhanced in the presence of manganese ion and ATP. Compared to the mesophilic chaperonin GroEL/GroES, ApcpnA is more activated at a higher temperature and protects the aggregation-prone unfolded state of the denatured rhodanese from thermal aggregation. Binding of ATP is sufficient for ApcpnA to perform the chaperonin function in vitro, but hydrolysis of ATP is not necessarily required. We propose that utilization of Mn2+ and adenosine nucleotide regardless of ATP hydrolysis may be one of peculiar properties of archaeal chaperonins.
PM 2.5/ samples were measured at two sites, Hadan(suburban site) and Anrak (roadside site) in Busan area. PM 2.5/ sampling was performed for 24-hour intervals by the FH9.5 particulate sampler. Aerosol samples were collected on PTFE filter. A total of 60 particulate samples were collected, dad samples were measured for Particulate mass concentration, metallic elements (Cr, Mn, Ni. Cu. Se, Fe, Pb, and Zn) and waer-soluble elements (C $l^{[-10]}$ , N $O_{3}$$^{[-10]}$ ,S $O_{4}$$^{2-}$, N $H_{4}$$^{+}$, $Ca_{2}$$^{+}$, $Mg_{2}$$^{+}$ and $^{+}$.Mass concentration in Hadan ranged 24.23~57.12 $\mu\textrm{g}$/㎥ and 60.22~72.12 $\mu\textrm{g}$/㎥m Yellow Sand Events. Major cations in Hadan and Anrak site is N $H_{4}$$^{+}$and N $a^{+}$ respectively. SO42$^{[-10]}$ was the abundant specie in the PM 2.5 fraction for Hadan site an dAnrak site. Hadan site showed igher concentration in S $O_{4}$$^{2.1}$ and N $H_{4}$$^{+}$ In Anrak site the concentration of S $O_{4}$/sip 2-/and N $a^{+}$ was higher than other ions Prominent metallic elements were Fe and Pb in two sites. Principal component analysis showed that main source of PM 2.5 aerosol particles was non-metal related source which was resulted in relating elements as Cr, Ni, and Pb at Hadan site, Anrak site also has resulted PM2.5 aerosol paricles source, which was related its element like Zn, and Ni,. The SAS package analysis also showed that long-range transport effect at Hadan area due to Yellow Sand Event by the prevailing weaterlies.ling weaterlies.

This study was conducted to estimate the contents of trace metals in vegetable which were produced in Korea. The levels of trace metals determined using a mercury analyzer, and ICP (inductively coupled plasma spectrometer) and an AAS (atomic absorption spectrophotometer) after wet digestion. The values of trace metals [minimum~maximum (mean), mg/kg] in vegetables were a follows ; Hg : 0.0001~0.019 (0.002), Pb : 0.001~0.28 (0.02), Cd : 0.001~0.078 (0.016), As : 0.001~0.06 (0.02), Cu : 0.06~24.81 (0.77), Mn : 0.17~15.12 (2.32), Zn : 0.13~28.70 (2.51). These results showed that metal contents in vegetables on domestic markets were similar to those reported in other countries. The weekly average intakes of lead, cadmium and mercury from vegetables take 2~7% of PTWI (Provisional Tolerable Weekly Intakes) that the FAO/WHO Joint Food Additive and Contaminants Committee had set to evaluate their safeties.
This study was carried out to evaluate the developed microstructures and mechanical properties of friction welded A6063 alloy. For this work, specimens were prepared at a size of 12 mm ${\O}{\times}80mm$, and friction welding was carried out at a rotation speed of 2,000 RPM, friction pressure of $12kgf/cm^2$ and upset pressure of $25kgf/cm^2$. To perform an analysis of the grain boundary characteristic distributions, such as the grain size, orientation and misorientation angle distributions, the electron back-scattering diffraction method was used. In addition, in order to identify the dispersed intermetallic compounds of the base and welded materials, transmission electron microscopy was used. The experimental results found that the application of friction welding on A6063 led to significant grain refinement of the welded zone relative to that of the base material. Besides this, intermetallic compounds such as AlMnSi and $Al_2Cu$ were found to be dispersed with more refined size relative to that of the base material. This formation retains the mechanical properties of the welds, which results in the fracture aspect at the base material zone. Therefore, based on the developed microstructures and mechanical properties, the application of friction welding on A6063 could be used to obtain a sound weld zone.
This study was designed to investigate the effects of silkworm (Bombyx mori L.) powder (SWP) on oxygen radicals and their scavenger enzymes in liver membranes of rats. Sprague-Dawley(SD) male rats(160$\pm$10g) were fed basic diet (control group), and experimental diets(SWP-200 and SWP-400 groups) added 200 and 400 mg/kg BW/day for 6 weeks. Hydroxyl radical (.OH) levels resulted in a consistent decreases (4.0% and 7.2%, 5.0% and 14.1%, respectively) in liver mitochondria and microsomes of SWP-200 SWP-400 groups compared with control group, and O2 radical level was significantly decreased about 12% in liver cytosol of SWP-400 group compared with control group. Lipid peroxide(LPO) levels were significantly decreased (14.4% and 9.1%, respectively) in liver mitochondria and microsomes of SWP-400 group only compared with control group. Oxidized protein (OP) levels were remarkably decreased about 12.7% and 16.3% in liver microsomes only of SWP-200 and SWP-400 groups, but significant difference between liver mitochondria could not obtained. Mn-SOD activities were remarkably increased (15.8% and 25.2%, respectively) in mitochondria of SWP-200 and SWP-400 groups, but significant difference between Cu, Zn-SOD activities in these group could be not obtained. GSHPx activity was significantly increased in liver cytosol of SWP-400 group compared with control group. These results suggest that silkworm powder may play an effective role in a attenuating a oxidative stress and increasing a scavenger enzyme activity in liver membranes.
The study was conducted for quantitive determination of macronutrients, micronutrients and heavy metals present in Spilanthes acmella Hutch and Dalz, a traditionally used important medicinal plant. The results illustrated the presence of substantial amounts of essential nutrient elements in different parts of the plant. K and Mg were detected in the range between 36.35 ± 1.01 to 67.78 ± 1.33 g/kg and 2.17 ± 0.13 to 7.02 ± 0.84 g/kg of DWS respectively. While the essential micronutrients Fe, Na, Mn, Zn and Cu were detected in the range between 62.62 ± 12.72 to 856.95 ± 76.61 mg/kg, 365.47 ± 23.84 to 633.03 ± 38.12 mg/kg, 51.66 ± 7.77 to 186.33 ± 13.92 mg/kg, 41.87 ± 2.85 to 53.89 ± 3.79 mg/kg and 18.49 ± 2.07 to 48.71 ± 4.89 mg/kg of DWS respectively. Besides, heavy metals (Cd, Ni and Pb) detected in some of the plant samples were beyond the maximum permissible limit (MPL) of FAO/WHO for herbal medicines. The concentrations of the essential nutrient elements except zinc were significantly different in different parts of the plant (p < 0.001). Further variation in the concentrations of the elements in the plant was observed with respect to seasonal changes and geographical conditions. Nevertheless the findings corroborate ethnomedicinal significance of the plant and signify the possibilibity of utilizing its standardized products for preparation of food supplements as well as multidimensional therapeutic herbal medicines.
Pine(Pinus densiflora Sieb et Zucc.) is one of rhe popular plant drugs which has been used as a medicine in Asia. To investigate the effect of pine needle extract (PNE) on oxygen radicals and their scavenger enzymes in brain membranes of Sprague- Dawley (SD), make SD rats were fed basic diets(control group), and experimental diets (PNE group) with 0.5 and 1.0% of PNE 6 weeks. Mitochondrial hydroxyl radical levels in brain of 0.5%-PNE and 1.0%-PNE groups were significantly inhibited to 30% and 25%, respectively, and microsomal hydrogen peroxide levels in brain of 0.5%-PNE and 1.0%-PNE groups were significantly inhibited to 15% compared with control group. Cytosolic superoxide rdical levels in 1.0%-PNE group were significantly inhibited to 20% compared with control group. Lipid peroxide(LPO) levels in brain mitochondria of 0.5%-PNE and 1.0%-PNE groups were significantly lower(25% and 35%) than that in control group. Mn-superoxide disumtase (SOD) activities in brain of 0.5%-PNE and 1.0%-PNE groups were significantly higher(18% and 12%) than those in control groups, but Cu,Zn-SOD activities in brain of 0.5%-PNE were significantly activated to 15% compared with control group. Glutathione peroxidase(GSHPx) activities in brain of 1.5%-PNE and 1.0% PNE groups were significantly higher(14% and 12%) than those in control group. These results suggest that more beneficial effects such as inhibition of oxygen radicals and lipid peroxide(LPO). and oncreases of scavenger enzymes in brain membranes of SD rats may be effectively modulated by administration of pine needle extract (PNE)

This study examined the quenching of ofloxacin (OFL) and flumequine (FLU) fluorescence by $Cuj^{2+}$, $Ni^{2+}$, $Co^{2+}$ and $Mn^{2+}$ in an aqueous solution. The change in the fluorescence intensity and lifetime was measured at various temperatures as a function of the quencher concentration. According to the Stern-Volmer plots, the fluorescence emission was quenched by both collisions (dynamic quenching) and complex formation (static quenching) with the same quencher but the effect of static quenching was larger than that of dynamic quenching. Large static and dynamic quenching constants for both OFL and FLU support significant ion-dipole and orbital-orbital interactions between fluorophore and quencher. For both molecules, the static and dynamic quenching constants by $Cu^{2+}$ were the largest among all the metal quenchers examined in this study. In addition, both the static and dynamic quenching mechanisms by $Cu^{2+}$ were somewhat different from the quenching caused by other metals. Between $Ni^{2+}$ and FLU, a different form of chemical interaction was observed compared with the interaction by other metals. The change in the absorption spectra as a result of the addition of a quencher provided information on static quenching. With all these metals, the static quenching constant of FLU was larger than those of OFL. The fluorescence of OFL was quite insensitive to both the dynamic and static quenching compared with FLU. This property of OFL can be explained by the twisted intramolecular charge transfer in the excited state.
A continuous flow system has been developed to determine trace Cu(II), Mn(II), Ni(II) and Zn(II) in a large volume of water samples by a solvent sublation technique. The mixed solution of 1,10-phenanthroline(phen) and thiocyanate ion was used as ligands for the formation of their ternary complexes. The continuous system was constructed in this laboratory with a peristaltic pump, a mini shaker, three mixing bottles and a flotation cell by connecting each part with a polyethylene tube. The flotation conditions such as the flow rate of sample solution and the injection rates of ligand, buffer and surfactant solutions have been investigated to obtain the best sublation efficiencies. Each solution flowed into the flotation cell through each polyethylene tube by the peristaltic pumps. The ternary complexes were floated and extracted into MIBK in a flotation cell of 2 L by bubbling a nitrogen gas. The absorbances of extracted analytes in MIBK were directly measured by graphite furnace-AAS. The concentrations of 1,10-phenanthroline and thiocyanate ion were $2.6\;{\times}\;10^{-3}$ M and $2.3\;{\times}\;10^{-2}$ M in the mixed solution, respectively. The pH of sample solution was adjusted to 5.0 with a buffer solution and 1%(m/v) sodium lauryl sulphate solution was added as a surfactant to support the effective flotation of the complexes. The $N_2$ gas was bubbled at 30 mL/min for 90 minutes for 20 L of sample. Reproducible results of less than 10% RSD and recoveries of 80-120% could be obtained in real samples.
Removal of subgingival calculus is essential for the success in periodontal treatment. Subgingival instrumentation is used for the removal of all bacterial plaque and calculus. In this study, two types of anterior curet were used on ant. teeth to conduct subgingival scaling and root planing. The remaining amount of calculus was evaluated according to type of instrument, depth of pocket, and tooth surface. 24 teeth extracted from patients being treated at Dan Kook University dept. Perio. were used. 4 surfaces per tooth a total of 96 areas were evaluated. 12 teeth treated with Gracey No. 1-2 was used as the control group and 12 teeth treated with Mini-five curet No. 1-2 was the experimental group. The 4 surfaces of the teeth {buccal, mesial, lingual or palatal, distal) were observed under a stereomicroscope and the images were captured 3 times per surfaced with a CCD. The image were observed on the monitor using a $10{\times]10$ grid produced with the Microsoft power point. The amount of calculus remaining was evaluated 3 times per surface. The results were as follows. 1. There was no significant difference in remaining calculus according to the pre-treatment pocket depth, and tooth position{Mx. or Mn). 2. The Mini-five curet showed better results than the Gracey curet but there was no statistically significant difference. 3. In both Gracey curet group and Mini-five curet group the lingual(or palatal) surface showed significant difference compared to the other surfaces(p<0.05). From the results above, it is thought that when treating ant. teeth consideration of the tooth surface is more important than the choice of instrument.
