  • Studies were carried out to investigate the optimal culture conditions for the production of penicillinase using a strain of Streptomyces sp. isolated from soil, YS-40. Among the carbon and nitrogen sources, glucose and L-asparagine increased the peniciilinase production. The addition of M $n^{++}$, $Ca^{++}$ and L $i^{+}$ increased the enzyme production, but depressed by F $e^{+++}$, F $e^{++}$, $Mg^{++}$, Z $n^{++}$, A $g^{++}$, $Ba^{++}$ and S $n^{++}$. L-Leucine slightly increased the enzyme production but L-histidine, L-methionine depressed. Among the vitamins riboflavine, i-inositol, hesperidine, niacin-amide, biotin, folic acid, DL-$\alpha$-lipoic acid increased the enzyme formation. The addition of cephradine, cephalexin, ampicillin, cloxacillin more increased the enzyme formation than that of other$\beta$-lactam antibiotics and antibiotics. Optimal pH and temperature on the enzyme formation was pH 7.0 and 28$^{\circ}C$ respectively Amount of the enzyme production reached at maximum with incubation for 3 days on the optimal condition.

  • This study was undertaken to analyze proximate compositions, minerals, vitamins, amino acids and fatty acids of Angelica keiskei Koidz. The contents of raw leaf and stem juice were compared with those of aerial whole plant. The aerial whole plant was relatively rich in crude fiber. The leaf juice was rich in lipid, protein, minerals, vitamins and amino acids whereas the stem juice was rich only in sugar and some kinds of fatty acids.

  • This study was undertaken to ascertain food and nutritional evaluating data on the processing of fermented sea cucumber (Stichopus japonicus) entrails. In the experiment, the crude proteolytic enzyme from the entrails tissue of raw and fermented sea cucumber during the days of ripening was extracted. The optimal activity condition for the crude enzyme and the compositional changes of amino acid of the protein and free amino acid in the raw and fermented sample were also investigated. 1. Less than three kinds of proteolytic enzymes that each enzyme has optimal activity condition at pH 3.1 $50^{\circ}C$(A-enzyme), pH 5.7 $50^{\circ}C$(B-enzyme) and pH 7.7 $45^{\circ}C$(C-enzyme), respectively were believed to be exist in the entrails tissue of sea cucumber. 2. A-enzyme and C-enzyme were strongly inhibited with the increase of the salt concentration, and B-enzyme was activated at the 1% salt concentration and was inhibited above the 5% salt concentration. 3. The result of the effect of several salt ions on the proteolytic activity showed that A-enzyme was slightly inhibited in the presence of all salt ions added, B-enzyme was activated in the presence of the all salt ions except $Cu^{2+}$ and C-enzyme was activated in the presence of $Ca^{2+}$ and $Mn^{2+}$, and inhibited by $Cu^{2+}$, $Co^{2+}$ and $Mg^{2+}$. 4. When the effects of the ripening days on the proteolytic activity of the crude enzymes were analysed, the activity of the A-enzyme was slightly weakened with the lapse of the fermentation days, whereas the B-enzyme was not influenced by the fermentation days. 5. In the analysis of amino acid composition of the protein of the samples, the 8 days fermented sea cucumber entrails showed the diminution of all kinds of amino acid. Apparently diminished amino acids were arginine, alanine, glutamic acid, glycine, serine, valine, threonine and lysine etc., and methionine, histidine and isoleucine were slightly decreased. 6. In the analysis of free amino acid composition of the 8 days fermented sample, glutamic acid, aspartic acid, leucine and lysine were rich, while histidine, methionine, proline and tyrosine were poor. The most of free amino acids were increased during the fermentation procedure and especially in lysine, histidine, threonine, glutamic acid, methionine, valine and leucine.

Fatty Acid and Amino Acid Compostions of Gugiseun (Lycuim chinense Miller) Depending on Variety and Harvest Time (채취시기 및 품종에 따른 구기순의 지방산과 아미노산조성)

  • As a part of studies on the development of a Gugisuen(Lycium chinense M.), which is one of the unutilization of waste resources, we investigated the composition of fatty acid and amino acid according as picking period and varieties of Gugisuen. The chemical components of Gugisuen were as follows : 1. Proximate composition of Gugisuen were Cheongyang native had the highest contents of crude lipid, crude protein, total sugar, and the contents of crude protein, total sugar, and reducing sugar was the highest picked on June 20. 2. The contents of $P_2O_5$, $K_2O$, CaO, MgO, Cu, and Mn were higher in Cheongyang native than that of other varieties. And mineral contents of Gugisuen picked on May 10 were the highest. 3. The major fatty acids were linoleic, linolenic acids, and these unsaturated fatty acids composed about 50% of total fatty acids. Fatty acid compositions between each varieties were not significant, and those components were the highest in Gugisuen picked on May 10. 4. Eighteen amino acids were identified from Gugisuen, glutamic acid, the highest content was $10.05\sim10.94%$ and these contents increased in the order to aspartic acid > glycine > alanine > leucine > lysine. The contents of serine, arginine, methionine, cystine-2 and isoleucine were higher in late harvest time and glutamic acid and tryptophane were higher in early harvest time.

  • Chemical components relevant to the characteristic antimicrobial activities of the Korean chestnut (Castanea crenata S. et Z.) leaves and flowers were analyzed. The composition of free sugar were sucrose, maltose in the chestnut flowers and sucrose, glucose, fructose in the chestnut leaves. The contains of tannin were 0.16% in the chestnut flower and 1.98% in the chestnut leaves. In fatty acids case, the linoleic contents were significantly high in the chestnut leaves and flowers. The organic acids showed high composition to succnic and citric acid in the chestnut leaves. The ammo acid compositions showed high contents to aspatic acid, proline, glutamic acid, glycine and methionine in the chestnut leaves and flowers. The total amino acid showed significantly higher in the chestnut leaves than flowers. The major minerals contained in the chestnut leaves and flowers were K, Ca, Ng, Fe, Mn and Al. Ascorbic acids were detected highly in the chestnut leaves and flowers.

Dried Sugarcane Press Residue as a Potential Feed Ingredient Source of Nutrients for Poultry

  • Sugarcane press residue (SPR), a byproduct from the sugar industry was evaluated for it's nutrient and energetic quality in broilers and layers. The composition of SPR included (% DM): CP-11.76 (methionine-2.21, cystine-1.05, lysine-4.85, threonine-5.48% of CP), EE-7.87 (palmitic acid-30.3, stearic acid-4.1, oleic aicd-17.2, linoleic acid-38.0, linolenic acid-5.4% of EE), CF-10.08, TA-21.08 (Ca-3.87, P-1.10, Mg-0.95%, Fe-3500, Mn-284, Zn-113, Cu-61.5, Co-5.0 ppm and AIA-4.93%) and NFE-48.35% indicating that SPR is a valuable source of both organic and inorganic nutrients for poultry. The metabolic trials revealed the average ME of SPR as 749, 842 and 1,270 kcal/kg, respectively in broilers and 844, 936 and 1,031 kcal/kg in layers, at 10, 20 and 30% inclusion levels, respectively. Further, the fortification of SPR incorporated diets with biotechnological products viz., lipid utilizing agents (lipase and lecithin) or NSP degrading enzymes and their combination did not improve the ME content of such diets.

Role of Calcium on Auxin-Induced Ethylene Production in Etiolated Mungbean(Vigna radiata W.) Hypocotyls (녹두(Vigna radiata W.) 하배축의 오옥신 유발 에틸렌 생성 과정에서 $Ca^{2+}$의 작용)

  • The effect of Ca2+ on auxin-induced ehtylene production in etiolated mungbean (Vigna radiata W.) hypocotyls was studied. Auxin-induced ethylene production by mungbean seedlings which had been germinated in the presence of 5-10mM Ca2+ (High Ca2+ ; HC) is greater than that by seedlings which had been germinated in distilled water (Low Ca2+ ; LC). The effect of Ca2+ on auxin-induced ethylene production was greatly increased after 12hr of incubation period. The stimulation of auxin-induced ethylene production by Ca2+ was specific, since divalent cations, such as Mg2+ and Mn2+ did not enhance auxin-induced ethylene production. Calcium also promoted ethylene evoluation induced by methionine and 1-Aminocyclopropane-1-carboxylic acid(ACC). The effect of Ca2+ on auxin-induced ethylene production was not caused by increase in free IAA or ACC contents of hypocotyl tissue. Dimethyl sulfoxide and Triton X-100, that disrupts the emembranes, inhibited ethylene production to a greater extent in LC segments than in HC segments. Addition of Ca2+ to the incubation medium for LC segments resulted in enchancement of ethylene production probalby because the membrane integrity is supported under these conditions. Comparison of activity of Ethylene Forming Enzyme(EFE) in LC and HC hypocotyl segments indicated that the enzyme activity of HC was about 2 times higher than that of L.C. It is suggested that Ca2+ increases the activity of plasma membrane-bound EFE through its stabilizing effect onn the membrane, which in turn brings about promotion of ethylene production.

A Study of the Chemical Components of the Halophyte Suaeda asparagoides $M_{IQ}$ (염생식물 나문재(Suaeda asparagoides $M_{IQ}$)의 이화학적 성분 조성)

  • To obtain basic information on the utilization of Suaeda (S.) asparagoides $M_{IQ}$ as a raw material in food, it's chemical components of were investigated. S. asparagoides $M_{IQ}$ from the Hwaseong-si in Korea was used after drying and grinding with powder. The total dietary fiber, insoluble dietary fiber and soluble dietary fiber contents of S. asparagoides $M_{IQ}$ were found to be 39.41%, 37.63% and 1.78%, respectively. Macromineral components of S. asparagoides $M_{IQ}$ were Na (7.6%), K (2.4%) and Mg (0.6%). High contents of microminerals such as Cu, Mn and Zn were also found. Lysine and methionine which are essential amino acids were detected with the value of 215 mg/100 g and 23.2 mg/100 g, respectively. S. asparagoides $M_{IQ}$ that was harvested in May was higher in crude protein than that harvested in September but the crude fat content of that which was harvested in September was twice as high as that harvested in May.

Cooking Characteristics of Coated Rice with Water Homogenate of Citrus Fruits Peel (감귤과피 물 균질액으로 가공한 유색미의 취반 특성)

  • Cooking characteristics of coated rice with water homogenate of citrus fruit peels (1% for rice) were investigated. The color of the coated rice both before and after cooking was dark yellow. The total content of carotenoids, hesperidin and naringin were 10.74, 2173.12 and 1468.40 mg% for citrus fruit peels, 0.46, 108.65 and 73.35 % for its water homogenate, 0.12, 21.73 and 14.62 mg% for coated rin, and 0.05, 8.67 and 5.87 mg% fur cooked coated rice, respectively. Citrus fruits peel contained 94.22 mg% of asparagine, 24.88 mg% of methionine, 19.64 mg% of alanine, and 15.37 mg% of ${\gamma}$-aminoisobutyric acid as the majority free amino acids, accounting for 70% of the total free amino acids present. Total free amino acid content of the cooked coated rice increased by 15% compared to those of cooked uncoated rice. The majority of minerals in the citrus fruit peels were K and Ca, accounting for 56% of total minerals present. The mineral content of cooked coated rice was generally higher than that of the cooked uncoated rice. The cooked coated rice showed comparable hardness, gumminess and brittleness, but higher cohesiveness and springiness than the cooked uncoated rice. There were no differences in sweet and bitter taste between the cooked uncoated and coated rice. However, the cooked coated rice showed higher sensory scores fur color acceptability, savory taste and overall acceptability than the cooked uncoated rice.

A study on the Active Site of Cytidine Deaminase from Bacillus subtilis ED 213 by Chemical Modification (화학적수식에 의한 Bacillus subtilis ED 213 Cytidine Deaminase의 활성부위에 관한 연구)

  • Essential amino acids involving in the active site ofthe cytidn~e deruninase from Bncillus subtilis ED 213 were determined by chemical modification studies. Tllc purified cytidine deruninase tiom Booillus subtilis ED 213 required the reduced form of Fe(lI)ion. since the enzyme was inhibited 43% by 1 mnM o-phenanthroline. Whereas the enzyme activity was activated up to 28% by 1 1 ethylenediaminetetraacetic acid. The cytidine deaninase activily was completely inhibited by 1 mM N-bromosuccinimide, chloramine-T, and p-chloromercuribenzoic acid (p-CMB), respectively. The enzyme activity was inhibited 36% by 1 mM pyridoxal-S-phosphale, and 31% by 1 mM l-ethy~-3-(3-dirneIhj~laminoprop}~~)c~bodiiamide and glycine inethyl ester. The enzyme activity was strongly inhibited 68% by 1 \mu$M \rho$-CMB and this inhibition of the enzyme activity with 1 \mu$M \rho$-CMB was completely reactivated by 5 mM cysleine as a reducing agent. We speculaled that tyrosine, methionine, cysteuie and/or serine residues are located ui or near ihe active site of the cytidine deruniuase from Bncilus subrilis ED 213 and indirectly related to lysine and/or glycine.
