Environmental Impacts of the Waste Rump in the Dongjin Gold-Silver-Copper Mine (동진 금·은·동 광산 주변에 방치된 폐석의 환경적 영향)

  • Although the Dongjin Au-Ag-Cu mine had been abandoned since about forty years ago, the results of this study on the dispersion patterns and contamination level of heavy metals in the hydrologic system flowing via the waste rump show that the environmental impacts from the mine wastes are still significant. The stream water in the vicinity of the waste rump is severely acidified (pH 3.8 to 4.4) and highly enriched in various dissolved heavy metals. The heavy metal contents of the stream water and stream sediments are systematically attenuated with increasing distance from the mine area. However, it is worth to note that continuous attenuation of heavy metal contents in both media were reenriched in downstream area more than 800 m apart from the mine because it can be acted as a secondary source of heavy metal pollution. The heavy metals, especially Cd, Cu and Zn of polluted downstream sediments mainly occur in Fe-Mn oxides and organic materials, which indicates that these elements are the main pollutants from the waste rump of the Dongjin mine. The heavy metal contents of crops, such as sesame, perilla, red Pepper and brown rice, collected from the polluted farm land in the downstream area are lower than those of land plants from stream sides, but significantly higher in Cd, Cr, Cu and Zn than those from the unpolluted farm land. Especially, almost all of the crops in polluted farm land have been severly contaminated by Cd (>0.4 ppm). On the other hand, the heavy metal contents of the crops collected from refreshed farm land by means of a soil addition method shows significantly lowered level comparing with those of polluted area, which indicates that a soil addition method was effective for the refreshment of polluted farm land by toxic metallic pollutants. Wormwoods from this area showed very high contents in a11 the heavy metals even in unpolluted area (Cd > 1 ppm, Cr > 1 ppm, Cu > 11 ppm, Pb> 4 ppm, Zn > 55 ppm), indicating that a special caution must be payed when one takes ingest them.

Antiproliferative Effect of Artemisia argyi Extract against J774A.1 Cells and Subcellular Superoxide Dismutase (SOD) Activity Changes

  • The water and methanol extracts of Artemisia argyi showed significant cytotoxicities against J774A.1 cells but not so much against normal leukocytes. The cytotoxicities were found to be dependent on the extract concentration and the incubation time. The concentration of water and methanol extracts inhibiting 50% of cell proliferation ($IC_{50}$) were estimated to be 44.2 mg/ml and 71.6 mg/ml, respectively. In the presence of Artemisia argyi water extract, total superoxide dismutase (CuZnSOD and MnSOD) activities of media, cytoplasmic and mitochondrial fractions of J774A.1 cells increased in accordance with cytotoxicity. MnSOD was found to be the main component of enhanced total SOD activities, particulary in the mitochondrial fraction. In contrast to SOD, catalase and glutathione peroxidase (GPx) were not found in any instance of the current investigation. In addition, substantial amount of $O_2^-$ appeared to be generated in the mitochondrial fraction under the influence of Artemisia argyi. All data put together, it is postulated that Artemisia argyi extracts seem to stimulate $O_2^-$ generation in mitochondria of J774A.1 cells with concomitant increases of SODs. Since $H_2O_2$, the reaction product of SOD on $O_2^-$, is known to be readily converted to very toxic $OH{\cdot}$ in the absence of catalase and/or GPx cooperation, toxicity derived from ROS such as $O_2^-$, $H_2O_2$, and $OH{\cdot}$ may be the main cause of necrosis and/or apoptosis of J774A.1 cells.

Geochemistry of the Major and Trace Elements in a Soil Profile of the Hyangdeung Area, Gwangju City, Korea (광주광역시 향등지역의 토양단면에서 주성분원소 및 미량성분원소의 지화학적 특성)

  • Elemental mobility during the weathering of granite in the Gwangju Hyangdeung area was investigated using ICP-MS analysis. It appeared that Al, Fe, Ti, K were lost from the profile, whereas Si, Ca, Na Mg, P and Mn were immobile during chemical weathering. In less weathered soil, large enrichment of K and Ti were found relative to Al, whereas other elements such as Si, Ca, Na, Mg and P are deplete. Fe content is constant throughout the weathered profiles. Amounts of Rb, Sr, Y, Cs, Pb, Th and U increased toward the surface. Nb, and Co have accumulated in the deepest parts of the weathered soil profile. These results agree with similar published studies. In addition, the analytical data shows that Ba and Ga increased, while Cu, Zn, Cr and Ni were relatively constant in this area. REE tend to increase in most samples, while LREEs, relative to Al, were enriched in the lower and upper saprolite. HREEs were enriched in the lower and upper saprolite.

Effect of Water Addition on Activity of Gold Catalysts Supported on Metal Oxide at Low Temperature CO Oxidation (일산화탄소 저온 산화에서 금속산화물에 담지된 금촉매의 활성에 미치는 수분첨가의 영향)

  • Gold catalysts supported on metal-oxides were prepared by co-precipitation using the various metal nitrates and chloroauric acid as precursors, and effect of water addition on the catalytic activity in CO oxidation was investigated. Among the various supported gold catalysts, Au/$Co_{3}O_{4}$ and Au/ZnO catalysts showed the excellent activity for CO oxidation. Water in the reactant gas had a negative effect on the oxidation activity over Au/$Co_{3}O_{4}$ catalysts and a positive effect on that over Au/ZnO, which means the activity depends strongly on the nature of support. It was also confirmed that no significant change in the particle size of gold was observed after reaction both in dry and wet conditions. This fact suggested that the deactivated catalyst due to a carbonate species could be regenerated by water addition in the reactant gas.

Relationship of mineral elements in sheep grazing in the highland agro-ecosystem

  • Objective: Minerals are one of the important nutrients for supporting the growth of sheep grazing in the highland, northwest of China. The experiment was conducted to investigate the relationship of both macro and micro minerals in sheep grazing in the highlands of six districts located in the Qilian Mountain of China. Methods: Samples of herbage (n = 240) and soil (n = 240) were collected at random in a "W" shape across the area designated for harvesting from 24 farms, where the sheep commonly graze in October (winter) for mineral analyses. In addition, serum samples were taken via jugular vein from 20 sheep per farm from 24 farms (n = 480 samples in total) for serum minerals analyses. Mean values of macro and micro minerals were statistically compared among districts and the correlations among soil-plant-animal were statistically analyzed and correlations were regressed, as well. Results: The results revealed that there were variations for both macro and micro minerals among districts. Statistical analysis of the correlation coefficients between herbage and sheep were significantly different for most of the minerals but not for P, Cu, and Se. Many correlation regression coefficients were found significantly different among minerals of herbage, soil, and sheep serum especially those of K, Na, Fe, Mn, and Zn (between herbage and sheep serum), and Fe and Mn (between herbage and soil), Na, Fe, Mn, and Zn (between soil and sheep serum), respectively. The regression coefficient equations derived under this experiment for prediction of Ca (R2 = 0.618), K (R2 = 0.803), Mg (R2 = 0.767), Na (R2 = 0.670), Fe (R2 = 0.865),Zn (R2 = 0.950), Mn (R2 = 0.936), and Se (R2 = 0.630), resulted in significant R2 values. Conclusion: It is inferred that the winter herbage minerals in all the districts were below the recommended levels for macro minerals which indicated there would be some mineral deficiencies in sheep grazing the herbage in these regions. Supplemental minerals may therefore play an important role in balancing the minerals available from the herbage in winter and would lead to increased productivity in sheep on the highland areas of China. These findings could be potentially applied to the other regions for improving the livestock productivity.

Characterization of the Starch Degradation Activity of recombinant glucoamylase from Extremophile Deinococcus geothermalis (극한성 미생물Deinococcus geothermalis 유래 재조합 글루코아밀레이즈의 전분 분해 활성 특징)

  • This work focused on characterization of the starch degradation activity from extremophile strain Deinococcus geothermalis. Glucoamylase gene from D. geothermalis was cloned and overexpressed by pET-21a vector using E. coli BL21 (DE3). In order to characterize starch degrading activity of recombinant glucoamylase, enzyme was purified using HisPur Ni-NTA column. The recombinant glucoamylase from D. geothermalis exhibited the optimum temperature as $45^{\circ}C$ for starch degradation activity. And highly acido-stable starch degrading activity was shown at pH 2. For further optimization of starch degrading activity with metal ion, various metal ions ($AgCl_2$, $HgCl_2$, $MnSO_4{\cdot}4H_2O$, $CoCl_2{\cdot}6H_2O$, $MgSO_4$, $ZnSO_4{\cdot}7H_2O$, $K_2SO_4$, $FeCl_2{\cdot}4H_2O$, NaCl, or $CuSO_4$) were added for enzyme reaction. As results, it was found that $FeCl_2{\cdot}4H_2O$ or $MnSO_4{\cdot}4H_2O$ addition resulted in 17% and 9% improved starch degrading activity, respectively. The recombinant glucoamylase from D. geothermalis might be used for simultaneous saccharification and fermentation (SSF) process at high acidic conditions.

The Effect of Additional Elements and Grain Size Distribution on the Magnetic Properties of the Nd-Fe-B Sintered Magnets (Nd-Fe-B계 소결자석의 자기적 특성에 미치는 첨가원소와 결정립 분포의 영향)

  • The influence of casting cooling rate during the preparation of ingots on the grain size distribution and the magnetizing ability of $\textrm{Nd}_{16}\textrm{Fe}_{72}\textrm{V}_{4}\textrm{B}_{8}$ sintered magnets were investigated. Sintered magnets prepared from copper mold which allow a higher cooling rate, show a smaller grain size distribution and god magnetizing ability. The effects of adding bride forming elements, such as Cr, Mn, Nb, and W on the magnetic properties of Nd-Fe-B sintered magnets were also investigated. Both Cr and W addition increase coercivity and the resultant $\textrm{Nd}_{16}\textrm{Fe}_{72}\textrm{Cr}_{4}\textrm{B}_{9}$ alloy exhibits a similar magnetizing ability in comparison with $\textrm{Nd}_{16}\textrm{Fe}_{72}\textrm{V}_{4}\textrm{B}_{8}$ alloy.

Sequential Extraction of Heavy Metals in Soils and A Case Study (토양중의 중금속 연속추출방법과 사례연구)

  • Many researchers have investigated most representative sequential extraction method using various reagents for determining the chemical forms of metals in soils and sediments. In this paper, a newly modified method for sequential extraction scheme based on Tessier's method by Environmental Geochemistry Research, Centre for Environmental Technology, Imperial College, was introduced and examined. In comparison with Tessier's method, originally designed for sediment analysis by Atomic Absorption Spectrophotometry (AAS), the sequential extraction scheme has been developed for the multi-element analysis by Inductively Coupled Plasma Atomic Emission Spectrometry (ICP-AES). The partitioning of particulate trace elements was classified into five fractions: (i) exchangeable, (ii) bound to carbonates or specially adsorbed, (iii) bound to Fe and Mn oxides, (iv) bound to organic matter and sulphides and (v) residuals. The experimental results of the pilot study for in-house reference material (HRM2) and certified international standard reference material (SRM2711) using the modified method showed not only reasonable precision and accuracy but also acceptable overall recovery rates. In addition, mine dump soils sampled in the Dalsung Cu-W mine, Korea were prepared and sequentially extracted using the method. Most of Cu was bound to organic matter/sulphides and residual fractions. The dominant fraction of soil Pb and Zn in the study area was found in the residuals. The fraction of Cd showed a wide variation between samples and could be found bound to the carbonates or specially adsorbed, oxides, organic fraction and residuals. The recovery rates of Cd, however, were poor due to relatively low Cd concentrations in soils. The heavy metals in these mine dumps appear to be in the more inert forms and should not be readily bioavailable. The soils, however, had very low pH values (average 4.1) and had sandy textures; consequently, rapid infiltration of rainfall may increase leaching of Zn and Cd which were found to be around 5 to 10% of the exchangeable fraction. As a result of the investigation of this study, it has been strongly recommended that these mine waste materials should still be considered a significant contaminant source and will need environmental remediation to prevent pollutants from being released into the environment.

Stability of Naphthoquinone Pigments Isolated from the Roots of Lithospermum erythrorhizon by Various Temperatures and Metal Ions (자근(紫根)으로부터 분리한 Naphthoquinone류 색소의 온도 및 금속에 대한 안정성)

  • The pigments of acetylshikonin and isobutylshikonin isolated from the roots of Lithospermum erythrorhizon were determined over a period of storage for their stabilities influenced by temperatures and metal ions. In dark condition, both pigment solution were unstable at 488 and 560 nm; alcoholic solutions of acetylshikonin pigment were stable at $25^{\circ}C$ and $50^{\circ}C$ for 1 hour, but isobutylshikonin showed severe discoloration within 1 hour at $95^{\circ}C$ . Buffered solutions of acetylshikonin tended to be discolored with the addition of $Fe^{++}$, while those of isobutylshikonin with $Fe^{++}$ and $Cu^{++}$. These studies indicated that acetylshikonin and isobutylshikonin can be used in foods as the natural colorant under selected conditions in case of considering storage temperature and metal ions.

Physicochemical Characteristics of Traditional Doenjang with added Lentinus edodes (표고버섯을 첨가한 전통된장의 이화학적 특성)

  • Traditional doenjang with Lentinus edodes added at a concentration of 0, 3, 5 and 10% (w/w) was prepared and its physicochemical characteristics were analyzed during fermentation for 6 months. The moisture content, salt level and pH for doenjang were decreased with increasing addition of Lentinus edodes powder, but crude protein content was increased. Among the free sugars, glucose, galactose, arabinose, sucrose and maltose were identified and quantified, with the first two being the major free sugar. Minerals were detected in the order of Na> K> P> Ca> Mg> Fe while the content of Cu, Mn and Zn exhibited little change. The contents of each amino acid of traditional doenjang with added Lentinus edodes, glutamic, leucine and aspartic acids, had a much higher level. In fatty acid compositions, palmitic, stearic, oleic and linoleic acids were analyzed, with oleic acid being the most abundant fatty acid at $56.4{\pm}0.8\sim57.5{\pm}0.5%$ in all the tested samples. GMP and IMP contents of nucleotides and their related compounds were extremely high. It was thought that these substances contributed to the sweet taste of traditional doenjang with added Lentinus edodes. These results showed the applicability and high quality of traditional doenjang with added Lentinus edodes.
