The objective of this research was to evaluate the toxicity of the industial xenobiotic Aroclor 1248 (A) and natural origin substances~elemental sulfur (S80) and oleic acid (OA) and their binary mixtures to V. fischeri bioluminescence during the prolonged exposure time (up to 60 min). The bioluminescence quenching test was used to determine the toxic effects. Full factorial experiment design and multiple regression analysis and the comparison of binary mixture effect with the sum of effects of individual chemicals were used for the evaluation of combined effects of toxicants. The analysis of general trend of mixture toxicity to bioluminescence showed that mixture toxic effects were reversible up to 60 min. Data analysis revealed different joint effects, which were depended on mixture composition. S80 enhanced toxic effect of A and acted additively with synergistic interaction. Hydrophobic OA in mixture with A acted antagonistically and in mixture with sulfur caused an additive effect with antagonistic component of interaction. It was concluded that low concentrations of natural toxic substances present in environmental samples as mixtures of chemicals can define the toxicodynamic character of industrial xenobiotics.
In this research, we tried to develop the application method to water management and treatment using toxicity test method. When we measure the toxicity of environmental samples, we have to decide whether we take some countermeasures to reduce the toxicity or not. The first issue is how to set these action levels in each bioassays. A new idea was attempted to authorize indirect approach of each bioassays through the response characteristics against mixture of chemicals in water quality standard. The significant response in the cell-growth-inhibition bioassay was detected for standards-mixture(STDs). For acute toxicity assay, STDs-based implicit correlation between risks to humans and bioassay data showed a rational approach to set action levels in practical management. A simple model was proposed to describe and predict the changes in the total toxicity based on the concentrations of toxic-controlling chemicals during the ozonation of landfill leachates. On the basis of this simple model, toxicity reduction was predicted for pre-aggregation treatment before ozonation and ozone concentration during the ozonation. The method proposed in this study would be useful in optimizing water treatment processes and their running conditions in terms of the toxicity reduction efficacy.
Background: Organic ultraviolet (UV) filters are widely used in sunscreen products and have been identified as an emerging contaminant. Organic UV filters co-exist with multiple components, but their mixture toxicity remains largely unknown. Objectives: We investigated the toxicity of single and binary mixtures of commonly used UV-filters using the human adrenocarcinoma (H295R) cell line. Methods: After exposure to non-cytotoxic concentrations of avobenzone (AVO), homosalate (HS), octisalate (OS), octinoxate (OMC), and octocrylene (OC), the levels of testosterone (T) and 17β-estradiol (E2) were measured. The median effective concentration (EC50) values for the E2 of the individual substances were used to determine the mixture effect of four binary combinations: OMC+AVB, OMC+HS, OMC+OS, and OMC+OC. The synergistic, additive, and antagonistic effects of the mixture were determined by calculating toxic units (TU). To examine the mechanism of mixture toxicity, eight genes involved in steroidogenesis were analyzed using the real-time polymerase chain reaction. Results: The significant increase in E2 in H295R cells exposed to AVO, HS, OS, OMC, and OC suggest an estrogenic effect of the tested UV-filters. A significant decrease in T was observed in cells exposed to HS and OS. EC50 values for E2 increase were 105 nM for AVO, 110 nM for HS, 120 nM for OS, 55 nM for OMC, and 80 nM for OC. Both binary mixtures consisting of OMC+HS and OMC+OS have synergistic effects. Conclusions: Our results showed that five types of UV-filter substances increase E2 in H295R cells. We examined the mixture toxicity in terms of increased estrogenicity and confirmed that E2 significantly increased when OMC was mixed with a salicylate-based UV-filters. These findings highlight the importance of determining the impact of UV filter mixtures.
Background: Organophosphorus flame retardants (OPFRs) are a group of chemical substances used in building materials and plastic products to suppress or mitigate the combustion of materials. Although OPFRs are generally used in mixed form, information on their mixture toxicity is quite scarce. Objectives: This study aims to elucidate the toxicity and determine the types of interaction (e.g., synergistic, additive, and antagonistic effect) of OPFRs mixtures. Methods: Nine organophosphorus flame retardants, including TEHP (tris(2-ethylhexyl) phosphate) and TDCPP (tris(1,3-dichloro-2-propyl) phosphate), were selected based on indoor dust measurement data in South Korea. Nine OPFRs were exposed to the luminescent bacteria Aliivibrio fischeri for 30 minutes and the human hepatocyte cell line HepG2 for 48 hours. Chemicals with significant toxicity were only used for mixture toxicity tests in HepG2. In addition, the observed ECx values were compared with the predicted toxicity values in the CA (concentration addition) prediction model, and the MDR (model deviation ratio) was calculated to determine the type of interaction. Results: Only four chemicals showed significant toxicity in the luminescent bacteria assays. However, EC50 values were derived for seven out of nine OPFRs in the HepG2 assays. In the HepG2 assays, the highest to lowest EC50 were in the order of the molecular weight of the target chemicals. In the further mixture tests, most binary mixtures show additive interactions except for the two combinations that have TPhP (triphenyl phosphate), i.e., TPhP and TDCPP, and TPhP and TBOEP (tris(2-butoxyethyl) phosphate). Conclusions: Our data shows OPFR mixtures usually have additivity; however, more research is needed to find out the reason for the synergistic effect of TPhP. Also, the mixture experimental dataset can be used as a training and validation set for developing the mixture toxicity prediction model as a further step.
Methylisothiazolinone (MIT) has been used in combination with methylchloroisothiazolinone (CMIT) for cosmetic products such as shampoo, body lotion, and skin care products. The mixture of CMIT/MIT has been found to cause allergic contact dermatitis and is thus no longer permitted for use as a preservative in leave-on cosmetics. However, MIT itself was approved as a stand-alone preservative at a maximum concentration of 100 ppm as the toxicity was derived from CMIT rather than MIT. However, in many countries, allergic skin irritation caused by MIT remains a social concern. In this study, skin irritation was assessed for the presence of MIT, propylene glycol, and their mixture using a 3D human skin model $EpiDerm^{TM}$. Although non-diluted MIT causes serious skin toxicity, skin irritation was not observed at a concentration of 100 ppm, the maximum permissible level for cosmetics and personal care products according to European regulations. Propylene glycol, the most widely used vehicle for MIT, did not cause skin irritation in the 3D skin model. The results are expected to provide information for regulatory policies and guidelines on the use of biocides in consumer products.
The object of this study is to investigate the toxicity of fenvalerate [(RS)-$\alpha$-cyano-3 -phonoxybenzyl-(RS)-2-(4-ch1orophenyl)-3-methylbutyrate] and the effect of carbaryl on the toxicity of fenvalerate. Rats were treated with fenvalerate (50 mg/kg, 100 mg/kg), carbaryl (50 mg/kg, 100 mg/kg) or mixtures of the two compounds (fenvalerate+carbaryl: 50 mg/kg+50 mg/kg, 50 mg/kg+100 mg/kg) by oral administration for 1~3 weeks. Control groups were treated with corn oil. The experimental results were summarized as follows. 1. LD$_{50}$ values of fenvalerate and carbaryl in male rats were 385 mg/kg and 625 mg/kg respectively. When 50 mg/kg and 100 mg/kg of carbaryl were administratrd, LD$_{50}$values of fenvalerate were 265 mg/kg and 225 mg/kg respectively. 2. Biochemical parameters such as ALT, LDH and glucose in serum were much more increased in the groups treated with mixture than the groups treated with either one of fenvalerate or carbaryl. 3. The groups treated with carbaryl and mixture for 3 weeks, the contents of cytochrome P-450 in the liver were significantly increased. In renal microsomal fractions, however, no significant changes of drug metabolizing enzyme activities were observed. 4. The activities of aniline hydroxylase in hepatic microsomal fractions were increased in the groups treated with fenvalerate and mixture and activity was much more increased in the groups treated with mixture. 5. The activities of ATPase in the groups treated with fenvalerate were decreased than that of groups treated with mixture. TBA values and the activity of glucose-6 -phosphatase in the liver were not significantly changed. 6. In mixture treated groups, the activities of cholinesterase in serum and in the liver were more decreased than those of carbaryl treated groups. The activities of carboxylesterase in serum in the liver were slightly increased in mixture treated groups, but in fenvalerate treated groups, the activities of carboxylesterase were much more increased than those of control groups. 7. As a result of this study, when carbaryl was as the synergist of fenvalerate, carbaryl inhibited the activities of esterases, so the toxicity of fenvalerate was increased.sed.
Bioluminescent bacteria fusing the stress promoter and lux gene have been developed as a toxicity biosensor. The light emitting bioluminescent bacteria have been used to measure the toxicity of many different chemicals. In this study, specially, DPD2540 (fabA::luxCDABE) was used to detect and classify phenolic toxicity to the cells membrane fatty acids, and then the relationship between phenolic toxicity and the distribution of various phenols in the cell was determined, with a model and equations provided. In addition, to show the possibility of detecting and classifying the toxicity of a chemical mixture, which may be present in wastewater, various bioluminescent bacteria having different stress promoters were used and their distinct response to the sample mixture was measured. To extend the applicable area of these bioluminescent bacteria to field, the portable biosensor using freeze-drying methods was developed and confirmed successfully.
Effect of Synethrin on the Toxicity of Dichlorvos in Rats. Hematological, biological and enzymatic effects were investigated in rats treated with DDVP and synethrin. Mutagenicity was also examined. In serological analysis, LDH and ALP were more significantly increased in rats treated with the mixture of DDVP (10mg/kg) and synethrin (250mg/kg) than with either DDVP or synethrin. DDVP alone slightly increased cytochrome P-450 in the liver while synethrin or the mixture decreased it. Lipid peroxidation was significantly increased in the liver when rats were treated with both DDVP and the mixture. Cholinesterase activity was significantly decreased in the liver and serum when treated with DDVP as well as with the mixture. In mutagenicity test, DDVP was shown to be weakly positive to TA 97a, TA 100 and TA 102, but negative to TA 1537. Synethrin showed negative in all strains tested. These results suggest that the mixture of DDVP and synethrin increased the toxicities but not the mutagenicity.
The test of germination has been used as a good indicator to assess the toxicity of chemicals to plant. This study was carried out to investigate the effect of PAHs, anthracene and phenanthrene, on germination ratio, germination index and germination time of barley, tall fescue and alfafa. Anthracene and phenathrene induced to decrease the seedling length and increase the germination time of the experimental plants. But they did not affect the maximum germination ratio. While it was not correlated between seedling length and final germination ratio, it was shown a close relationship between seedling length and mean germination time of the plants. These results suggested that the mean of germination time and seedling growth could be useful to exmine the phyto -toxicological effect of PAHs. The mixture of anthracene and phenanthrene was examined to investigate the combined effect on seedling length; the mixture induced the reduction of seedling. This was meant that the mixture toxicity of PAHs might be not shown in the sum of each chemical toxicity.
