Evaluation of Toxicity of Heavy Metals and Surfactants Using Vibrio Fischeri and Daphnia Magna (발광박테리아(Vibrio fischeri)와 물벼룩(Daphnia magna)을 이용한 중금속 및 계면활성제의 혼합독성 평가)

  • In this study, the mixed toxicity of heavy metals (Cu, Pb, Hg) and surfactants (SLS, ALS) was evaluated by using Vibrio fischeri and Daphnia magna. The sensitivity of ecotoxicity to heavy metals was sensitive to daphnia but the case of surfactants were more sensitive to Vibrio fischeri. Experimental results of Vibrio fischeri show that the toxicity value P(O) was lower than the predicted value P(E), and the antagonistic effect was observed when the heavy metal and the surfactant were mixed. It seems that SLS and ALS, which are anionic surfactants, have anionic form on the hydrophilic head, so that they have an antagonistic effect that they are bonded with heavy metal ion which is a cation type and the actual toxicity is lowered. In Daphnia magna, the results showed that antagonistic, additive and synergistic effects were in order as concentrations increased. As the concentration increases, Daphnia magna, which is highly sensitive to heavy metals, seems to have a synergistic effect with a rapid increase in mortality.

HPLC Method for the Determination of Nicorandil in Human Plasma

  • The present study is to determine of sensitive nicorandil analysis method using HPLC and measure the pharmacokinetics parameters (bioavailability, $C_{max}$, $T_{max}$, Ke, $T_{1/2}$) of nicorandil (5 mg, Tab; Choongwae Pharma Corporation). Plasma (500 ul) was mixed with furosemide (internal standard, 500 ug/ml). Detection wavelength was 256 nm. The mixture of 0.01 M ammonium acetate and acetonitrile 80:20 (v/v) was used mobile phase. The HPLC separation was accomplished on ODC reverse HPLC column. The nicorandil was analyzed by a HPLC system, which consists of CAPCELL PAK C18 column (5 ${\mu}$m, 4.6 × 150 mm) and a chromatography data analysis S/W, using a isocratic mobile phase (mixture of 0.01 M ammonium acetate and acetonitrile 80:20 ) at 1.0 ml/min. Its sensitivity, selectivity, accuracy and precision must be adequate for the bioavailabilty study of nicorandil, and the linearity ($r^2$ ≥ 0.9994) of nicorandil was also proved in the range of 0.05 ug/ml . 3 ug/ml. The pharmacokinetic parameters of nicorandil (5 mg) tablets were measured as the follow. AUC: 0.19 ug/ml·hr, $C_{max}$: 0.14 ug/ml, $t_{max}$: 0.58 hr, Ke: 0.11 hr., $t_{1/2\beta}$: 6.76 hrs. This method is simple and sensitive HPLC method using UV detector for determination of nicorandil in human plasma.

An Adaptive Feedback Canceller for Fully Implantable Hearing Device Using Tympanic Membrane Installed Microphone (고막이식형 마이크로폰을 위한 이식형 인공중이 적응 피드백 제거기 구현)

  • Many implantable hearing aids are being developed as alternatives to conventional hearing aids which has inconveniences for use and social stigma that make hearing-impaired people avoid to wear it. Particularly, the fully-implantable middle ear hearing devices (F-IMEHD) are being actively studied for mixed or sensorineural hearing impaired people. In development of F-IMEHD, the most difficult problem is improving the performance of implantable microphone. Recently, Cho et al. have studied the tympanic membrane installed microphone which has better sensitivity and is easier to operate on patient than the microphone implanted under the skin. But, it may cause howling problem due to the feedback signal via oval window and ossicle chain from the transducer on round window in the middle ear cavity, therefore, a feedback canceller is necessary. In this paper, we designed NLMS (normalized least mean square) adaptive feedback canceller for F-IMEHD with tympanic membrane installed microphone and a transducer implemented at round window, and computer simulation was performed to verify its operation. The designed adaptive feedback canceller has a delay filter, a 64 point FIR fixed filter and a 8-tap adaptive FIR filter. Computer simulation of the feedback path is modeled by using the data obtained through human cadaver experiment.

Substructuring-based Structural Reanalysis by Multilevel Hybrid Approximation (다단계 혼성근사화에 의한 부구조화 기반 구조 재해석)

  • A new solution procedure for approximate reanalysis, using the staged hybrid methods with substructuring, is proposed in this study. Displacements are calculated with two step mixed procedures. First step is to introduce the conservative approximation, which is a hybrid form of the linear and reciprocal approximation, as local approximation. In the next step, it is combined with the global approximation by reduced basis approach. Stresses are evaluated from the displacements by matrix transformation. The quality of reanalyzed quantities can be greatly improved through these staged hybrid approximations, specially for large changes in the design. Overall procedures are based on substructuring scheme. Several numerical examples illustrate the validity and effectiveness of the proposed methods.

Characterization of beer-spoilage microorganism and its rapid detection by specific PCR primer (맥주오염미생물의 동정과 specific PCR primer의한 신속한 검출 방법)

  • Several contaminated bacteria such as Lactobacillus brevis and Pediococcus damnosus in beer production cause beer spoilage by producing off flavours and turbidity. Detection of these organisms is complicated by the strict anaerobic conditions and lengthy incubation times required for their cultivation, consequently there is a need for more rapid detection methods. Recently, two contaminated strains were isolated from vessel of beer production and identified as Lactobacillus species by API kit identificaton as well as 16S-23S ITS sequencing analyses. Two isolated strains were named as Lactobacillus sp. HLA1 and Lactobacillus HLB2, respectively. A polymerase chain reaction (PCR) method was developed for the rapid and specific detection of Lactobacillus sp.. Two sets of primer pairs (HLA1-F/HLA1-R and HLB2-F/HLB2-R) were designed for the amplification of a 1576 base pair (bp) fragment of the HLA1 16S-23S rRNA gene and 1888 bp fragement of the HLB2 16S-23S rRNA. Amplified PCR products were highly specific to detect corresponding bacteria when other contaminated strains were used as PCR templates. However, detection of both strains were limited when $100{\mu}{\ell}$ of cultured samples were mixed with $100m{\ell}$ of beer sample in arbitrary manner. The sensitivity of the assay still needs to be improved for direct detection of the small amounts of bacteria present in beer.

Sensitivity and accuracy for rheological simulation of cement-based materials

  • The flow of freshly mixed cement-based material shows thixotropy, which implies some difficulties on robust measurement of its rheological properties: The flow curve of thixotropic materials depends on the used protocol. For examples, higher viscosity is obtained when the rate of shear strain is more quickly increased. Even though precise measurement and modelling of the concrete rheology needs to consider the thixotropic effect, engineers in the concrete field prefer considering as a non-thixotropic Herschel-Bulkley fluid, even more simply Bingham fluid. That is due to robustness of the measurement and application in casting process. In the aspect of simplification, this papers attempts to mimic the thixoropic flow by the non-thixotropic Herschel-Bulkley model. Disregarding the thixotropy of cement based materials allows us to adopt the rheological concept in the field. An optimized protocol to measure the Bingham parameters was finally found based on the accuracy and reproducibility test of cement paste samples, which minimizes the error of simulation stemming from the assumption of non-thixotropy.

  • Purpose: This article provides an update and overview of a nursing research program focused on understanding the pathophysiology and management of irritable bowel syndrome (IBS). Methods: This review includes English language papers from the United States, Europe, and Asia (e.g., South Korea) from 1999 to 2013. We addressed IBS as a health problem, emerging etiologies, diagnostic and treatment approaches and the importance of a biopsychosocial model. Results: IBS is a chronic, functional gastrointestinal disorder characterized by recurrent episodes of abdominal pain and alterations in bowel habit (diarrhea, constipation, mixed). It is a condition for which adults, particularly women ages 20-45, seek health care services in both the United States and South Korea. Clinically, nurses play key roles in symptom prevention and management including designing and implementing approaches to enhance the patients' self-management strategies. Multiple mechanisms are believed to participate in the development and maintenance of IBS symptoms including autonomic nervous system dysregulation, intestinal inflammation, intestinal dysbiosis, dietary intolerances, alterations in emotion regulation, heightened visceral pain sensitivity, hypothalamic-pituitary-adrenal dysregulation, and dysmotility. Because IBS tends to occur in families, genetic factors may also contribute to the pathophysiology. Patients with IBS often report a number of co-morbid disorders and/or symptoms including poor sleep. Conclusion: The key to planning effective management strategies is to understand the heterogeneity of this disorder. Interventions for IBS include non-pharmacological strategies such as cognitive behavior therapy, relaxation strategies, and exclusion diets.

Regulation of NAD+- Specific Isocitrate Dehydrogenase from Pythium ultimum

  • The $NAD^+$-specific activity of a dual coenzyme-specific isocitrate dehydrogenase (IDH; EC 1.1.1.41) from the primitive fungus Pythium ultimum was investigated to elucidate the regulatory factors that may influence the intracellular distribution of carbon and the availability of intermediates, e.g. citrate, for fatty acid synthesis. Inhibition of $NAD^+$-IDH activity by diphospho- and triphosphonucleotides (ATP, ADP, and GTP) reflected the sensitivity of this enzyme to cellular energy charge even though monophosphonucleotides (AMP and GMP) had little effect on activity. NADPH, but not NADH, substantially inhibited $NAD^+$-IDH activity, showing noncompetitive inhibition with isocitrate. Oxalacetate and ${\alpha}$-ketoglutarate showed competitive inhibition with isocitrate, while citrate and cis-aconitate showed mixed-noncompetitive inhibition with isocitrate. Inhibition by these substances ranged from 29 to 46% at 10 mM. The inhibitory effect of oxalacetate was increased synergistically by glyoxylate, which alone caused 31% uncompetitive inhibition at 10 mM, and a mixture of the two substances at 1 mM each showed 98% inhibition of $NAD^+$-IDH activity. The regulation of $NAD^+$-IDH in Pythium ultimum seems to be a complex process involving mitochondrial metabolites. The addition of glyoxylate (3 mM) and oxalacetate (3 mM) to the culture medium resulted in the production of 49% more lipid by P. ultimum.

Highly Sensitive Multichannel Interdigitated Capacitor Based Bitterness Sensor

  • In this study, we propose a multichannel interdigitated capacitor (IDC) sensor for detecting the bitterness of coffee. The operating principle of the device is based on the variation in capacitance of a sensing membrane in contact with a bitter solution. Four solvatochromic dyes, namely, Nile red, Reichardt's dye, auramine-O, and rhodamine-B, were mixed with polyvinylchloride (PVC) and N,N-dimethylacetamide (DMAC), to create four different types of bitter-sensitive solutions. These solutions were then individually inserted into four interdigitated electrodes (IDEs) using a spin coater, to prepare four distinct IDC sensors. The sensors are capable of detecting bitterness-inducing chemical compounds in any solution, at concentrations of approximately $1{\mu}M$ to 1 M. The sensitivity of the IDC bitterness sensor containing the Reichardt's dye sensing-membrane was approximately 1.58 nF/decade. The multichannel sensor has a response time of approximately 6 s, and an approximate recovery time of 5 s. The proposed sensor offers a stable sensing response and linear sensing performance over a wide measurement range, with a correlation coefficient ($R^2$) of approximately 0.972.

Detection of Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus and Listeria monocytogenes in Kimchi by Multiplex Polymerase Chain Reaction (mPCR)

  • We developed an mPCR assay for the simultaneous detection, in one tube, of Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus and Listeria monocytogenes using species-specific primers. The mPCR employed the E. coli O157:H7 specific primer Stx2A, Salmonella spp. specific primer Its, S. aureus specific primer Cap8A-B and L. monocytogenes specific primer Hly. Amplification with these primers produced products of 553, 312, 405 and 210 bp, respectively. All PCR products were easily detected by agarose gel electrophoresis, and the sequences of the specific amplicons assessed. Potential pathogenic bacteria, in laboratory-prepared and four commercially available kimchi products, were using this mPCR assay, and the amplicons cloned and sequenced. The results correlated exactly with sequences derived for amplicons obtained during preliminry tests with known organisms. The sensitivity of the assay was determined for the purified pathogen DNAs from four strains. The mPCR detected pathogen DNA at concentrations ranging from approximately 0.45 to $0.05\;pM/{\mu}l$. Thus, this mPCR assay may allow for the rapid, reliable and cost-effective identification of four potentially pathogens present in the mixed bacterial communities of commercially available kimchi.
